Physiochemical Properties of Carrageenan Hydrolysates by Organic Acids (카라기난의 유기산 가수분해물의 기능 특성)

  • This study was conducted to investigate various physiochemical activities of carrageenan hydrolysates obtained with organic acid treatments. The hydrolysates treated with citrate and malate at $100^{\circ}C,\;110^{\circ}C\;and\;120^{\circ}C$ had antimicrobial activities against Bacillus cereus and Bacillus subtilis. Especially, the hydrolysates with malate at $120^{\circ}C$ for 180 min treatment had the strongest antimicrobial activity to Bacillus subtilis. Regardless of the hydrolysis conditions, inhibition ratios of tyrosinase activity by citrate and malate under $100{\sim}120^{\circ}C$ were over 97%. Especially, the inhibition ratios for tyrosinase activity of the hydrolysates obtained with citrate at $100^{\circ}C$ for 180 min and at $120^{\circ}C$ for 90 min were 99.4% and 98.2%, respectively. Also, the inhibition ratios for tyrosinase activity of the hydrolysates obtained with malate under the same conditions were about 99.5% and 99.3%, respectively. The APTT as anticoagulant activity of carrageenan hydrolysates with 0.3% malate and citrate at $80^{\circ}C$ for 180 min were $2,451{\pm}18(sec)\;and\;1,617{\pm}15(sec)$, respectively.

  • To screen chaperone proteins from Schizosaccharomyce pombe (S. pombe), we prepared recombinant citrate synthase of the fission yeast as a substrate of anti-aggregation assay. Purified recombinant citrate synthase showed citrate synthase activity and was suitable for the substrate of chaperone assay. Several heat stable proteins including aspartyl aminopeptidase (AAP) for candidates of chaperone were screened from the supernatant fraction of heat-treated crude extract of S. pombe. The purified AAP migrated as a single band of 47 kDa on SDS-polyacrylamide gel electrophoresis. The native size of AAP was estimated as 200 kDa by a HPLC gel permeation chromatography. This enzyme can remove the aspartyl residue at N-terminus of angiotensin I. In addition, AAP showed the heat stability and protected the aggregation of citrate synthase caused by thermal denaturation. This study showed that S. pombe AAP is a moonlight protein that has aspartyl aminopeptidase and chaperone activities.

EFFECT OF FEEDING CLOMIPHENE CITRATE ON CARCASS COMPOSITION OF BROILER

  • Four seasonal trials were conducted to study the effect of feeding clomiphene citrate on carcass composition of broilers. It was fed at 5, 10, 15 mg levels per kg feed for a period of two weeks, 3-4 weeks ($S_1$), 4-5 weeks ($S_2$) and 5-6 weeks ($S_3$) of age. Total carcasses were analysed for moisture, ether extract and protein. Clomiphene citrate significantly decreased the moisture with concomitant increase in ether extract content of the carcasses at all the ages and seasons. Carcass protein was significantly decreased at all the ages and seasons. The responses in the different level of clomiphene citrate is dose dependent. The carcass moisture decreased with age while protein and ether extract contents increased in all seasons. The females had more ether extract and less moisture contents than male. The carcass ether extract was higher in summer and rainy seasons followed by spring and winter seasons, and the differences were significant. A concomitant significant decrease of carcass moisture was observed. The effect of seasons on carcass protein varied at different ages but there appeared to be a trend towards decreasing carcass protein in summer and rainy seasons than winter and spring seasons.

  • Sodium lactateand sodium citrate, traditional food preservatives, were added to extend the shelf-life of the loow-salted Myungran-jeot, and various chemical and microbiological analyses were carried out with Muungran-jeot, and various chemical and microbiological analyses were carried out with Myungran-jeot femented at 1$0^{\circ}C$. pH was decreased in the beginning stage of fermentation and then increased, whereas the content of lactic acid was increased during fermentation. But, lactic acid production of the low-salted Myungran-jeot with preservative were lower than control. The NH$_2$-N content of the low-salted Myungran-jot with sodium citrate was increased in the beginning of fermentation and then decreased. Sodium citrate inhibited the productions of VBN and TMA during the fermentation of Myungran-jeot, whereas sodium lactate inhibited the productions of VBN and TBA. Sodium lactate inhibited the growths of proteolytic bacteris and fungi. The estimated shelf-lives of the Myungran-jeot with control, sodium lactate, and sodium citrate were about 11, 13, and 13 days, respectively.

  • The effect of phosphate salt (NafHP04) and sodium citrate on the emulsion stability of soy protein isolate (SPI) in the presence of calcium was investigated in terms of salt concentration and addition order. Both phosphate and citrate salts decreased the solubility of SPI despite their pH enhancing effects. Addition of calcium chloride (CaCl2) significantly decreased ES, which showed nearly negligible at more than 3 mM CaCl2 concentration. When Na2HP04 were added in the presence of 5 mM Cac12, 55 greatly increased up to 20mM concentration, above which however ES decreased. It was found that the addition order of Na2HPO4 and CaCl2 affected ES. The addition of phosphate and subsequent CaCl2 exhibited the higher 55 than the reverse order. In both cases, the overall ES profile was found to be nearly similar to the solubility profile of SPI, indicating the positive relationship between solubility and emulsion stability of SPI in the presence of calcium. Similar trend to the phosphate effect on ES was also observed for sodium citrate in the presence of calcium.

  • The purpose of this study was to investigate the effect of different conditions (osmolity, solution, incubation times, comparison of fresh and frozen/thawed semen and storage times) on the swelling of canine spermatozoa. Employing the hypoosmotic swelling test (HOST), the membrane integrity of spermatozoa in different solutions (sucrose, fructose, latose, Na-citrate, Na-citrate plus sucrose, Na-citrate plus fructose and Na-citrate plus lactose were 61.4%, 66.2%, 62.5%, 68.1%, 62.0%, 68.5% and 60.2%, respectively.   • Coralline algae, the algal whitening phenomenon-causing seaweeds, are characterized by calcareous deposits in the cell wall. The viability of the coralline algae Lithophyllum yessoense and Corallina pilulifera was quantitated using a triphenyltetrazolium chloride assay and eight calcification inhibitors. Among these inhibitors, ferric citrate showed the strongest inhibition of coralline algae viability. The concentrations of ferric citrate conferring 50% inhibition were 1.7 and 3.8 mM for L. yessoense and C. pilulifera, respectively. Thus, at a specific concentration and in a localized area, ferric citrate may be used to prevent the blooming of coralline algae.

  • Three varieties of methyl citrate and 1 -methyl malate were isolated from the fruits of Opuntia ficus-indica var. saboten Makino through in vitro bioassay-guided isolation for the inhibition on monoamine oxidase(MAO). The $IC_50$ values for MAO-B of 1-monomethyl citrate, 1,3-dimethy citrate, trimethyl citrate and 1-methyl malate were 0.19, 0.23, 0.61 and 0.25 mM, respectively. However, on MAO-A, their inhibitions showed only marginal activity.
