  • ETS (environmental tobacco smoke) is composed of exhaled mainstream smoke (MS) from the smoker, sidestream smoke (SS) emitted from the smoldering tobacco between puffs and contaminants that diffuse through the cigarette paper and mouth between puffs. These emissions contain both vapor phase and particulate contaminants. ETS is a complex mix of over 4,000 compounds. This mix contains many known or suspected human carcinogens and other toxic agents. More of these toxic compounds are found in SS than in MS. Workplace exposure to ETS can result in significant smoke intake, and passive smoke exposure may be related to impair respiratory function and an increase risk of lung cancer in nonsmokers. For nonsmokers sharing a work environment with cigarette smokers, the workplace must be considered hazardous independently of any specific industrial toxic exposure. The risk is particularly important when a high percentage of the workers smoke or where smokers and nonsmokers work in poorly ventilated areas. Nicotine is converted in the body to cotinine; cotinine therefore can be used as an indirect measure of a person's recent exposure to tobacco smoke. Levels of nicotine in hair and levels of cotinine in body fluids (saliva and urine) have been shown to increase with increasing environmental nicotine levels and with self-reported ETS exposure. The measurement of nicotine or cotinine in hair may be more appropriate for longer-term exposure to tobacco. The purpose of this study is to comparing airborne nicotine levels and hair cotinine level in restaurant workers. Concentration of airborne nicotine and hair nicotine (and cotinine) is closely related to exposed frequency of sidestream smoke in the workplace. Nicotine in hair is a better predictor of airborne nicotine than hair cotinine. Hair nicotine can be a useful tool to assess ETS exposure interventions. It may have limiting levels of ETS exposure by placing regulatory restrictions on smoking in workplaces and in public spaces.

담배 주류연의 생물학적 활성에 대한 흡연조건의 영향 (Effect of smoking conditions on the biological activity of cigarette mainstream smoke)

  • The objective of this study was to determine the effect of smoking conditions on the in vitro toxicological activity of mainstream smoke. The 2R4F reference cigarette was machine-smoked by International Organization for Standardization(ISO) and Canadian Intense(CI) conditions. Smoke was analysed for chemical composition and in vitro toxicity. The cytotoxic potencies of both the total particulate matter(TPM), which were collected in Cambridge filter pad, and gas/vapor phase(GVP), which was bubbled through in phosphate-buffer saline in a gas-washing bottle, were assessed neutral red up take assay with chinese hamster ovary(CHO) cells. The assessment for genotoxicity of TPMs generated under ISO and CI conditions was determined using Salmonella mutagenicity assay and in vitro micronucleus assay. When calculated on an equal TPM basis, in vitro toxicity of TPM obtained under CI condition was decreased compared to TPM generated under ISO condition. The results of chemical composition analyses revealed that the lower toxicological activity under CI condition than that of ISO condition could be explained by the decreased in the contents of phenols, N-nitrosoamines and aromatic amines of TPM on an equal TPM basis.

필터 종류에 따른 Semi-volatile 화합물 이행 특성 (A study on the Delivery of Semivolatile Components in Cigarette Main Stream Smoke with the Filters)

  • Semivolatile components in mainstream smoke of cigarette attached the different type filter were analysed. Based on the main peak of GC profile, benzene, toluene, acetic acid, limonene, acetamide, pyridine, nicotine, phenol, pyrrole, and furfuryl alcohol were identified. The amount delivered of semivolatile components by each filter was evaluated with the ratio of areas of sample vs area% of ISTD. By comparing dual and triple filter to mono filter, the delivery amounts were 52% benzene, 62% toluene and 74% benzene, 84% toluene, respectively. The delivery amount of limonene which was known of tobacco taste component, were 52%, and 93% by dual and triple filter, respectively. Also, delivery amounts of acetic acid which was one of acidic compounds in smoke, were 86% and 83% by dual and triple filter, respectively. When adding the same amount of active carbon, the amounts delivered of vapour phase such as benzene and toluene by dual filter were lower than that of triple filter. But the delivery amounts of acetic acid and phenol by dual filter were higher than that of triple filter. This results assume that the pH of active carbon in filter affect to the adsorption or absorption of triacetin during filter making process.

오리엔트엽, 판상엽, 팽화엽 첨가에 따른 담배 연기성분 변화 (Changes of Tobacco Smoke Components by Adding Oriental, Reconstituted, and Expanded tobacco leaves)

  • This study was conducted to determine the smoke component changes by adding oriental, reconstituted, and expanded tobacco leaves. 7 different cigarette brands which were mixed with flue-cured, burley, oriental, reconstituted and expanded tobacco leaves were used for this study. 64 kinds of smoke components which were 6 of general components, 34 of semi-volatile and volatile components, 9 of acid components, and 15 of phenolic components were analyzed. All smoke components of mainstream smoke were changed by the different branding. As Tar, nicotine, ammonia, pH, all of acid compounds(except lacatic and glycolic acid) were decreased; HCN, levoglucosame, 4-vinyl phenol, 4-vinyl catechol, quinic acid-r-lactone, acetaldehyde, 2,3-butadiene, stylene were increased by adding oriental tobacco leaves. When the reconstituted tobaccos were added to 20%, the concentration of nicotine, all of acid compounds(except lactic. glycolic, palmitic acid) and all of phenol compounds were reduced; the concentration of ammonia, HNC, CO, aeconitrile, benzene, 2-butanone, moth-acrolene, butyronitrile, stylene, o-xylene were increased. As decreasing ammonia, pH, nicotine, all of acid compounds, all of phenol compounds, Isoprene, acetonitrile, 2-methyl-2-butene, 1,3-pentadiene, 2-nlethyl furane, ethylene cyclopentanone, ethyl bezene; increasing CO concentration were followed by adding expanded tobacco leaves.

항산화 필터 제조 및 주류연중 free radicals 제거 효과 (The Manufacture of Antioxidant Filter and Removal Efficiency of the Free Radicals in Mainstream Smoke)

  • This study describes the manufacturing method of antioxidant filter and evaluate the reduction of free radical and biological activity from mainstream smoke. When we used spray-brush for the application of antioxidant solution to the filter, cv(coefficient of variation) levels of antioxidant added to filter ranged within 3 %, which means stable and constant feeding process. 0.3~0.5mg per cigarette of antioxidant were sprayed to acetate and active carbon part in the filter plug, respectively. It was considered that the use of antioxidant added filter resulted in the reduction of the gaseous compounds of free radicals from 17 % to 19 % in the mainstream smoke. Variation for removal efficiency of free radical in antioxidant filter added to acetate part was more pronounced than that of filter added to active carbon part. Smoking deliveries of tar, nicotine for the antioxidant added filters were similar when comparing to the non-antioxidant added filter, but the delivering amounts of isoprene were significantly reduced. The estimation of consumed GSH(glutathion) showed that the gaseous toxicity of the antioxidant added filter was 14 % ~ 23 % lower than that of non-antioxidant added filter. No significant differences were observed for the total taste quality in sensory evaluation.

Study of Pyrolysis Pattern and Transfer Rate of Organochlorine Pesticide in Tobacco

  • GRLs(Guidance Residue Levels) of agricultural chemicals for tobacco are recommended by the CORESTA Agro-Chemical Advisory Committee guide. In the GRLs list, organochlorine group is one of pesticides commonly used on tobacco cultivation. In this model study, the quantitative correlation in the transfer rate of pesticide residue into tobacco smoke by spiking of organochlorine pesticides to cigarette and pyrolysates were investigated. The spiking concentration referred to the range of GRLs list and the organochlorine pesticides in mainstream smoke were analyzed by GC-MS. For the understanding of the composition variation versus temperature, the behavior of pesticides was investigated by pyrolysis-gas chromatography-mass spectrometry(Py-GC-MS). In this study, the transfer rate of pesticide residue into tobacco smoke at four different spiking concentration and the composition of pyrolysates were analyzed differently. At $10\;{\mu}g/cig$ spiking concentrations, the organochlorine pesticides were transferred into tobacco smoke in $0.02\;{\sim}\;10.19\;%$ each of component and the most of pesticides were pyrolyzed during smoking. It was found that the decomposition compounds from organochlorine pesticides were mainly composed of oxygenated and nitrogenous compounds. This study could estimate that the transfer rate of pesticides into tobacco smoke is very small amount.

CORESTA 모니터링 담배의 비교 분석 (Comparative Smoke Analysis of CORESTA Monitoring Cigarettes)

  • This experiment was conducted as a part of Asia collaborative study on purpose of verifying the difference between CM6 and CM7 including 3R4F for reference. It carried out using various analytical categories for example, main stream, sidestream and Av. smoke. Additional analysis such as physical properties, blending ratio, combustibility and general leaf component analysis also implemented in order to investigate the difference. We complied with ISO standard and CORESTA recommended method during analytical operating procedures. In this study, we described that comparative analytical result for CM6 and CM7 known as reference or monitoring cigarettes including 3R4F for reference. All sample cigarettes were conditioned at $22^{\circ}C$, 60% relative humidity for 48 hours. Av. Smoke, MS and SS smoke analysis were performed over five times with two smoking condition, ISO and Health Canada with the exception of Av. smoke analysis. We complied with ISO standard method during analytical operating procedures. And, we conducted additional analysis, such as physical properties, blending ratio, combustibility and leaf component analysis also in order to investigate the difference. In conclusion, we found out some differences between CORESTA monitoring cigarette No. 6 and No 7. The smoke components such as total particulate matters, NFDPM, nicotine and carbon monoxide contents of CM7 were a little lower than CM6. And, these phenomena were the same as not only main stream smoke but also side stream smoke and Av. smoke. This tendency was consistent with ISO and Health Canada smoking condition. Besides, leaf constituents' color of CM7 was darker than CM6. In case of combustibility, it showed short combustion time approximately 30 seconds.

담배 주류연 중 7개 그룹의 유해성분(HPHC) 분석법 확립 및 유효성 평가 (Establishment of analytical methods for HPHC list of mainstream cigarette smoke)

  • 본 연구를 통하여 담배 주류연에 포함되어 있는 HPHCs (Harmful and potentially harmful constituents) 중 7 개 그룹의 분석법을 확립하였다. 이 7 개 그룹에는 방향족 아민, 휘발성 유기화합물, 중금속, 담배특이니트로사민, 벤조[a]피렌, 암모니아, 카르보닐 그룹이 있다. 시험법 검증을 위하여 표준담배인 1R5F와 3R4F를 사용하여 분석법의 특이성, 직선성, 정성한계, 정량한계, 정확도, 정밀도 및 회수율을 확인하였다. 모든 상관계수(r2)는 0.995 이상 이였고 정성한계는 방향족 아민, 휘발성 유기화합물, 중금속, 담배특이 니트로사민, 벤조[a]피렌, 암모니아, 카르보닐 순으로 0.01-0.04 ng/cig, 0.01-0.16 μg/cig, 0.01-1.27 ng/cig, 0.06-0.28 ng/cig, 0.04 ng/cig, 0.08 μg/cig, 0.78-1.77 μg/cig으로 확인되었다. 또한 일간과 일내 상대표준편차는 모두 15% 이하였고, 회수율의 상대표준편차는 15% 이하, 회수율 범위는 79.2-117.5%로 나타났다. 이러한 결과들을 바탕으로 7 개 그룹에 대하여 확립된 시험법은 각각의 해당성분을 분석하기에 적합하다고 판단된다.

흡연시 발암성 물질의 체내 잔존율에 대한 연구 - 주요 휘발성 유기화합물을 중심으로 (Studies on the body - retaining rate of smoking-related carcinogens using some important volatile organic compounds (VOC))

  • 본 연구에서는 담배 흡연과 관련된 유해성분들의 발생특성을 비교하기 위하여, BTEX를 위시한 주요 방향족 VOC 성분과 포름알데하이드와 아세트알데하이드를 포함하는 카보닐 화합물의 분포특성을 다각도로 비교하고자 하였다. 이를 위해, 직접흡연에 해당하는 exhaled mainstream smoke (EMS)와 간접흡연에 해당하는 side-stream smoke (SS) 시료 등을 분석하고, 이들의 농도분포를 비교하였다. 필터를 제거한 상태에서 흡입되는 담배 연기의 경우, 유해한 성분들이 수 백 ppm 수준의 고농도로 존재한다는 것을 확인하였다. 반면, 흡연자가 비흡연시에도 날숨을 통해 이들 성분을 수 십에서 수 백 ppb의 농도로 배출하는 것을 확인할 수 있었다. 이러한 분석결과와 날숨과 들숨의 관계로부터 주요 흡연성분들의 체내 잔류율 등을 유추해 보았다. 이러한 추정에 의하면, 방향족 및 카르보닐 성분들의 대부분이 99% 이상 체내에 잔류할 수 있다는 것을 확인하였다.
