기내에서 삽목 배양중인 한라구절초[( Chrysanthemum zawadskii ssp. coreanum(Nakai) Y.N. Lee)]의 잎으로부터 효과적인 식물체 재생을 위한 NAA와 BA농도 및 배지고형물의 효과를 조사하였다. 0.2% Gelite를 고형물로 하였을 때 한라구절초의 잎 재분화에는 BA농도보다는 NAA 농도가 더 영향을 미쳤으며, NAA 농도가 1 mg/L 이상이면 모든 조직 에서 재분화를 보였다. 신초수는 NAA 단용 처리에서 많은 경향이었으며 NAA 1 mg/L 처리시가 가장 양호하였다. 배지고형물로 agar과 gelite를 가지고 재분화 효과를 조사한 결과, gelite 첨가시 잎절편 당 신초수가 많았으며 agar 첨가시에는 잎절편당 신초수가 적은 반면에 신초의 길이가 길게 나타났다. 한라구절초의 잎절편으로부터 재분화에는 MS에 3% sucrose, 0.8% agar와 NAA 1 mg/L를 첨가하여 25 $^{\circ}C$, 2,000 lux 정도에서 배양하는 것이 효과적이었다.
Chrysanthemums showing leaf mottling were collected from three southern locations in Korea in 1998. Two kinds of viruses were isolated from the leaves and were identified as tomato aspermy virus ch-TAV) and chrysanthemum virus B (ch-CVB), according to their host range, morphology, intracellular location, agar gel double diffusion test, and double-stranded RNA (dsRNA) analysis. The purified ch-TAV was spherical particles of approximately 29 nm in diameter and ch-CVB was filamentous particles of 685 nm long. Inclusion bodies were not observed in ch-TAV and/or ch-CVB infected chrysanthemum. ch-TAV showed positive serological reaction with TAV antiserum (ATCC-127) but not with CMV-pepper antiserum. In dsRNA analysis, four kinds of viral dsRNA were observed on ch-TAV and one viral dsRNA was shown on ch-CVB. Rate of co-infection with TAV and CVB in commercial chrysanthemums was 20.9%. On the other hand, infection with CVB alone was 97.2%. However, chrysanthemums naturally infected with TAV alone were not found.
This study was carried out to investigate the relationship between endogenous polyamines (PAs) and floral bud differentiation in chrysanthemum (Chrysanthemum morifolium). In this study, PA content (free, bound, and conjugated) in apical buds, leaves, and roots changed appreciably during floral bud differentiation. PAs accumulated during series of processes such as floral induction, differentiation of floret primordia, and crown formation in apical buds; changes in PAs in apical buds may have a relationship with those in leaves and roots. The levels of free PAs and conjugated PAs [putrescine (Put) and spermine (Spm)] in apical buds rapidly increased during the initiation stage of floral bud differentiation, while free and conjugated spermidine (Spd) reached their highest levels at the stage of floret primordium differentiation. In the free, conjugated, and bound PA fractions, the changes in Spm content were negligible compared to those of Put and Spd throughout the experiment. These findings indicate that PAs participate in regulating the process of flower bud differentiation in chrysanthemum.
To elucidate the role of silicon in biotic stress such as pests and diseases, standard chrysanthemum was grown in pots filled with soil without application of pesticide and fungicide. Si treatment was largely composed of three groups: $K_2SiO_3$ (50, 100, and $200mg{\cdot}L^{-1}$), three brands of silicate fertilizer (SiF1, SiF2, and SiF3) and tap water as a control. Si sources were constantly drenched into pots for 14 weeks. Application high concentration $K_2SiO_3$ ($200mg{\cdot}L^{-1}$) and three commercial Si fertilizers for 14 weeks improved growth parameters such as plant height and the number of leaves. In the assessment of disease after 4 weeks of Si treatment, percentage of infected leaves was not significantly different from that of control. After 14 weeks of Si treatment, however, the infected leaves were significantly reduced with a 20-50% decrease in high concentration ($200mg{\cdot}L^{-1}$) of potassium silicate and all commercial silicate fertilizers. Colonies of aphid insect (Macrosiphoniellas anborni) were also reduced in Si-treated chrysanthemum, showing 40-57% lower than those of control plants. Accumulation of silicon (approximately $5.4-7.1mg{\cdot}g^{-1}$ dry weight) in shoots of the plants was higher in Si-supplemented chrysanthemum compared to control plants ($3.3mg{\cdot}g^{-1}$ dry weight). These results indicate that using potassium silicate or silicate fertilizer may be a useful for management of disease and aphid insect in soil-cultivated chrysanthemum.
국화재배 포장에서 시들음증이 발생한 토양 내 식물기생선충의 종류와 발생 밀도 및 국화의 품질과 수량 피해 정도를 구명하였다. 구미 원예수출공사의 대규모 유리온실에서 발생한 식물기생선충의 종류는 사과뿌리썩이선충(Pratylenchus vulnus), 침선충(Paratylenchus sp.), 환선충(Criconemoides sp.)이 검출되었으며, 이중 사과뿌리썩이선충의 평균밀도는 토양 100 g당 667마리, 침선충은 716마리로 나타났고, 토양깊이별 분포는 $0{\sim}30$ cm사이에 87%가 분포하였다. 사과뿌리썩이선충의 국화 생육 기간의 밀도 변동은 정식전 토양 100 g당 854마리에서 정식 10주후에 토양 100 g과 뿌리 1 g에서 14,985마리로 17배 증가하였다. 국화 생육은 정식 10주후 무감염구에서 비해 초장은 8%, 생체중은 24.8% 감소하여 뿌리썩이선충이 국화 품질 및 수량에 영향을 주었다.
In this study, the effects of planting density on the growth of chrysanthemum in a greenhouse were evaluated on two popular varieties (i.e., Sinma and Moonlight). Planting density treatments were as follows: 1) $12cm{\times}12cm$, 2) $6cm{\times}12cm$, 3) $6cm{\times}12cm$ with one-cell vacant, and 4) $6cm{\times}12cm$ with two-cell vacant. Size of each treatments indicate one chrysanthemum was planted in that sized cell that was rectangular shaped field and these treatments were located in a line. Moreover, "one and two-cell vacant" means that it makes middle point of the field empty, offers beside chrysanthemum larger spaces to grow. For the Sinma variety, the results of growth and flowering characteristics at the harvesting stage showed that leaf number, leaf length, flower length, and leaf area were highest when the crop was planted at the $12cm{\times}12cm$ density, and the next preferable density was $6cm{\times}12cm$ with one-cell vacant. For the Moonlight variety, the results showed that stalk height and diameter, leaf number and length, flower length, leaf area, and flower number were highest at the $12cm{\times}12cm$ planting density. For Sinma, ratios of marketable production were 87.5% and 83.3% for the $12cm{\times}12cm$ and $6cm{\times}12cm$ with two-cell vacant, respectively. For Moonlight, ratios were 88.0% and 84.3% for the $12cm{\times}12cm$ and $6cm{\times}12cm$ with two-cell vacant.
Increase of national family income improved demand of high-quality and year-round horticultural products including chrysanthemum. To meet these demand, farmers have introduced protected facilities, such as greenhouses, of which environmental conditions could be monitored and controlled. Environment management up to three weeks after transplanting is critical for chrysanthemum quality. Artificial lighting and light-blocking screen are especially important for long-day (day period > 13 hours) and short-day (night period > 13 hours) treatments. In this study, a web-camera was installed, and the image was obtained and transmitted to mobile phones to monitor the status of 3-wavelength(RGB) lighting environments. RGB pixel values were used to determine malfunctioning of the lighting lamps, and leaking out and incoming illumination status during short-day and long-day treatment periods. Normal lighting lamps provided RGB pixel values of 240~255. During long-day treatment period, G pixel values were useful to detect abnormal lighting conditions (e.g., leaking). During short-day treatment period, R pixel values were useful to determine incoming light (e.g., sun-light). Results of this study would provide useful information for remote monitoring of light conditions for protected chrysanthemum production under artificial lights.

Chrysanthemum is one of the most important floral crop in Korea produced about 7 billion dollars (1 billion for pot and 6 billion for cutting) in 2013. However, it is difficult to breed and to do genetic study because 1) it is highly self-incompatible, 2) it is outcrossing crop having heterozygotes, and 3) commercial cultvars are hexaploid (2n = 6x = 54). Although low-density genetic map and QTL study were reported, it is not enough to apply for the marker assisted selection and other genetic studies. Therefore, we are trying to make high-density genetic mapping using GBS with about 100 $F_1s$ of C. boreale that is oHohhfd diploid (2n = 2x = 18, about 2.8Gb) instead of commercial culitvars. Since Chrysanthemum is outcrossing, two-way pseudo-testcross model would be used to construct genetic map. Also, genotype-by-sequencing (GBS) would be utilized to generate sufficient number of markers and to maximize genomic representation in a cost effective manner. Those completed sequences would be analyzed with TASSEL-GBS pipeline. In order to reduce sequence error, only first 64 sequences, which have almost zero percent error, would be incorporated in the pipeline for the analysis. In addition, to reduce errors that is common in heterozygotes crops caused by low coverage, two rare cutters (NsiI and MseI) were used to increase sequence depth. Maskov algorithm would also used to deal with missing data. Further, sparsely placed markers on the physical map would be used as anchors to overcome problems caused by low coverage. For this purpose, were generated from transcriptome of Chrysanthemum using MISA program. Among those, 10 simple sequence repeat (SSR) markers, which are evenly distributed along each chromosome and polymorphic between two parents, would be selected.
This study examines the dyeability, light fastness, washing fastness, and the antibacterial activity of chitosan and nano silver composite non-woven fabrics dyed with an extracted solution from Chrysanthemum Indicum Linn. The results show that an increase in the chitosan and nano silver percentage resulted in an increase in the $a^*$ values and $b^*$ values; however, the $L^*$ values decreased in the undyed condition. ${\Delta}E$ values of chitosan and nano silver composite non-woven fabrics were higher than cotton 100% non-woven fabrics in the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, and mordant treatments influenced the chrominance change. In the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, an increase in the percentage of chitosan and nano silver resulted in an increase of the K/S values. The dyeability of chitosan and nano silver composite non-woven fabrics increased by mordant treatments. The light fastness and washing fastness of the mordanted non-woven fabrics were better than the non-mordanted. For the antibacterial activity, the bacterial reduction rate of chitosan and nano silver composite non-woven fabrics was 99.9% to Staphylococcus aureus and Klebsiella pneumoniae.
Gray mold caused by Botrytis cinerea is one of the most important postharvest fungal pathogens of cut flowers. Here, gamma irradiation, an alternative for phytosanitary purposes, and sodium dichloroisocyanurate (NaDCC) were used to control B. cinerea in a cut chrysanthemum (Chrysanthemum morifolium Ramat.) cultivar, 'Baekma', one of the cultivars susceptible to B. cinerea. Spore germination and mycelium growth of B. cinerea were inhibited by gamma irradiation in an inversely dose-dependent manner. A dose of 4 kGy completely inhibited the mycelium growth of B. cinerea. A significant change in flower quality (physical properties) on chrysanthemum was shown from gamma irradiation at over 0.2 kGy (p<0.05). Therefore, in this study, the integration of gamma ray (below 0.2 kGy) and NaDCC, an eco-friendly form of chlorine, was investigated to control the disease with low dose of gamma irradiation dose. Interestingly, the gamma irradiated flowers showed more disease severity than the non-irradiated flowers. The combined treatment of gamma irradiation and NaDCC does not affect the severity of the fungal disease, whereas only 70 ppm of NaDCC treatment showed a significantly reduced severity. These results suggest that only chlorination treatment can be applied to control B. cinerea in cut chrysanthemum flowers.
