만성스트레스 검출을 위한 멀티 센서시스템 연구 (A Study on Multi-Sensor System for Detection of Chronic Mild Stress)

  • The development of modern civilization result from the abundance of material. Yet modern people live with chronic mild stress. Excessive chronic mild stress leads to various diseases. From the risk of the disease in order to protect our bodies need to manage chronic mild stress. The purpose of this study is to inspection the effectiveness of detecting in chronic mild stress using the Multi-sensor system. The Multi-sensor system is designed that can be measure three kinds of vital signals of chronic mild stress for the detection. First Photoplethysmogram(PPG), second Electro Dermal Activity(EDA), third Skin Temperature(SKT). The ages and occupations exposed to chronic mild stress, people often use out of this system was applied to dairy products(Pen). In addition, vital signals that occur when the variety of noise was used to remove the accelerometer. Chronic mild stress by the analysis of measured vital signals from Multi-sensor system to the measurement information to a PC to a wireless transmission(Bluetooth). In this study, using Multi-sensor system writing conditions and a variety of situations in the movement to measure vital signals and measurement results verified the accuracy and reliability. Through this measure chronic mild stress in everyday life and managing to maintain will help more healthy lifestyle.

Protective effects of blueberry drink on cognitive impairment induced by chronic mild stress in adult rats

  • BACKGROUND/OBJECTIVES: Stress-induced cognitive impairment is related to the suppression of hippocampal neurogenesis that results from an increase of oxidative stress. Therefore, the aim of this study was to investigate the effects of administration of a blueberry drink, having a high antioxidant power, on the cognitive performance of adult rats exposed to chronic mild stress. MATERIALS/METHODS: Twelve-week-old male Sprague-Dawley rats (n = 48) were randomly divided into four groups: control (CO), stress (ST), control + 5% blueberry drink (CO + B), and stress + 5% blueberry drink (ST + B). After eight weeks, the cognitive performance was assessed using a multiple T-maze water test. Levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and ascorbic acid were measured in the brain, and catecholamine concentrations were measured in plasma. RESULTS: The brain weights of the rats from the ST and ST + B groups were significantly lower than those of the rats from the CO and CO + B groups. The cognitive performance of the ST group was impaired when compared to that of the CO group. This impairment was significantly improved by the blueberry drink supplementation (P < 0.05). The brain SOD and CAT concentrations were not influenced by the stress or by the blueberry drink. However, the brain levels of GPx and ascorbic acid were significantly lower in the ST group than those in the CO group and were increased by the blueberry drink supplementation. The plasma catecholamine concentrations were affected by chronic mild stress and by the blueberry drink. The plasma norepinephrine and dopamine concentrations were decreased by the chronic stress and improved by the blueberry drink supplementation. The plasma epinephrine level was only influenced by the stress. CONCLUSION: These findings suggest that the blueberry drink may protect against the cognitive impairment induced by chronic mild stress.

  • Objective: This study was designed to assess antidepressant effects of Quibitang on an Animal Model of Depression induced by Chronic Mild Stress. Method: The consumption of 1% sucrose solution and active avoidance learning test were used to evaluate antidepressant effect of Quibitang. The consumption of 1% sucrose solution was measured every week for 8 weeks, and active avoidance learning test was executed after 4 weeks treatment of saline or Quibitang. Result: 1. The consumption of 1% sucrose solution was significantly reversed in test group (Quibitang-treated group) at 5th, 7th, 8th weeks, but there was no significant change in control group. 2. Chronic Mild Stress was found to suppress the increase of body weight at 5th, 6th, 7th, 8th weeks. Treatment of Quibitang did not enhanced the body weigt, but it enhanced the consumption of sucrose solution. 3. In order to measure the learning ability of rat which drived to be depressed, we executed active avoidance test. The result revealed that depressed rat showed impaired acquisition than control group, and the treatment of Quibitang restored the learning activity. Conclusion: These results suggest that Quibitang may have antidepressant effects on depression induced by chronic mild stress.

감맥대조탕 복합추출물의 만성스트레스 유발 생쥐의 항우울 작용에 관한 실험연구 (Experimental Study on the Anti-Depressant Effects of Gammakdaejo-tang Complex Extracts in Rats Induced with Chronic Mild Stress)

  • Objectives: To experimentally assess the anti-depressant effects of Gammakdaejo-tang Complex Extracts (GMDJ-Tang) in rats. Methods: Twenty Wistar Hannover (160~170 g) stressed rats were treated with different concentrations of GMDJ-Tang extracts i.e., 0, 62.5 mg/kg, 125 mg/kg, and 250 mg/kg. Chronic mild stress was induced by food deprivation, empty bottles, forced swimming, flickered light, tilt cages, shaking cages, high density breeding, water deprivation, and by soaking the litter cover according to fixed schedule. Blood and brain tissue samples were collected for biochemical analysis. Tests included serotonin and ELISA assays. Results: GMDJ-tang increased the weight of treated rats as well as levels of serotonin, BDNF and TrkB; however, the differences were not significant. In contrast, the extracts significantly decreased blood glucose in stressed rats. GMDJ-tang extracts did not significantly impact Serum AST, ALT, leukocytes, erythrocytes and thrombocytes when comparing treatment groups to control rats. Likewise, hemoglobin, hematocrit and PLT increased in treatment groups following treatment with GMDJ-tang, but this change was without significance. Conclusions: These results suggest that GMDJ-tang can alleviate chronic mild stress in rats, possibly through anti-depressant activity.

우울증(憂鬱症)모델 흰쥐에 대한 조위승청탕(調胃升淸湯)과 fluoxetine의 항우울효과(抗憂鬱效果) 비교 (The effects of Jowiseungchungtang versus fluoxetine in the chronic mild stress model of depression in rats)

  • This study was performed to evaluate the effects of Jowiseungchungtang and fluoxetine in the chronic mild stress(CMS) model of depression in rats. Chronic exposure to mild unpredictable stress was found to depress the consumption of sucrose solution in rats for 5 weeks. These CMS-treated rats were stratified into Jowiseungchungtang group, fluoxetine group and vehicle group. And control rats were also stratified into other CMS-treated group. The change of the consumption of sucrose solution was measured, and open field test were performed to investigate the anti-depression effect of Jowiseungchungtang and fluoxetine. The results were as follows : 1. The consumption of sucrose solution was significantly reversed in Jowiseungchungtang-treated group at 7 week, but there was no significant change in other groups. 2. CMS schedule decreased body weight. Jowiseungchungtang group and fluoxetine group showed significant decrease of body weight after 6 weeks. 3. In open field test, Jowiseungchungtang group and fluoxetine group showed no significant change of exploratory activity.

황칠(黃漆)이 만성 스트레스 유발 백서의 스트레스 및 수면 호르몬에 미치는 영향 (Effect of Resina Dendropanacis Morbiferus on Stress and Sleep Hormone in Chronic Mild Stress-Induced Rats)

  • Objectives: To evaluate effects of Resina Dendropanacis Morbiferus (RDM) on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats. Methods: Twenty-five healthy rats were randomly divided into 5 groups; Normal, CMS (Control), Resina Dendropanacis Mobiferus 50 mg/kg (RDM 50), 100 mg/kg (RDM 100), and 200 mg/kg (RDM 200). All rats except the normal group were exposed to unpredicted stress conditions such as water deprivation, empty bottles, forced thread mill, etc. according to timetable of CMS for 3 weeks. After a week starting experiments, rats in RDM 50, RDM 100, and RDM 200 groups were fed orally once a day for 2 weeks. Afterward, blood samples were taken from rats to analyze complete blood count, AST, ALT, and glucose. Noradrenalin, GABA and Melatonin were measured by ELISA kit. BDNF, CREB and TrkB were measured by RT-PCT. Results: 1. In Noradrenalin content, RDM 100 and RDM 200 groups revealed significant decrease compared to the control group. 2. In GABA content, RDM 50 and RDM 100 groups revealed significant decrease compared to the control group. 3. In Melatonin content, RDM 100 and 200 groups revealed significant decrease compared to the control group. 4. In Activity of BDNF, RDM 100 and 200 groups revealed significant increase compared to the control group. 5. In Activity of CREB and TrkB, RDM 100 group revealed significant increase compared to the control group. 6. In Erythrocyte and Thrombocyte changes, red blood cells and hematocrit significantly increased in RDM 50 and RDM 200 groups than the control group. Hemoglobin and platelet significantly increased in all experimental groups and the control group. 7. In Weight gain content, all RDM groups revealed insignificant increase compared to the control group. 8. In Glucose content, RDM 50, RDM 100, and RDM 200 groups revealed significant decrease compared to the control group. Conclusions: Results suggest that RDM have effects on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats.

Strain Differences in the Chronic Mild Stress Animal Model of Depression and Anxiety in Mice

  • Chronic mild stress (CMS) has been reported to induce an anhedonic-like state in mice that resembles some of the symptoms of human depression. In the present study, we used a chronic mild stress animal model of depression and anxiety to examine the responses of two strains of mice that have different behavioral responsiveness. An outbred ICR and an inbred C57BL/6 strain of mice were selected because they are widely used strains in behavioral tests. The results showed that the inbred C57BL/6 and outbred ICR mice were similarly responsive to CMS treatment in sucrose intake test (SIT) and open field test (OFT). However, the two strains showed quite different responses in forced swimming test (FST) and novelty-suppressed feeding (NSF) test after 3 weeks of CMS treatment. Only C57BL/6 mice displayed the depression- and anxiety-like behavioral effects in response to CMS treatment in FST and NSF test. Our results suggest that there are differences in responsiveness to CMS according to the different types of strain of mice and behavioral tests. Therefore, these results provide useful information for the selection of appropriate behavioral methods to test depression- and anxiety-like behaviors using CMS in ICR and C57BL/6 mice.

LC-MS/MS-based Quantification of Ten Neurotransmitters in Rat Limbic System and Serum: Application to Chronic Unpredictable Mild Stress-Induced Depression Rats

  • As one of the most common mood disorders, numerous studies have shown depression is the main risk factor for non-suicidal self-harm. The pathogenesis of depression is complex, and a comprehensive and rapid measurement of monoamine neurotransmitters and their metabolites will be very helpful in understanding the pathogenesis of depression. Therefore, a rapid and sensitive underivatized liquid chromatography-tandem mass spectrometry method was developed and validated for the simultaneous monitoring of the levels of ten neurotransmitters and their metabolites in rat serum and limbic system and successfully applied to quantify the changes of neurotransmitter levels in chronic unpredictable mild stress-induced rats. The analytes studied were mainly involved in tyrosine metabolism, tryptophan metabolism, and glutamate cycling pathways, which are important in the pathogenesis of depression. It had been verified the method was sensitive and effective, with satisfactory linearity, and met the requirements of biological sample determination. Levels of neurotransmitters in rat serum, hippocampus, amygdala, prefrontal cortex, striatum, and hypothalamus were determined via the method. The results showed serotonin, dopamine, norepinephrine, and their metabolites were decreased, glutamine was increased, and glutamate was disturbed in chronic unpredictable mild stress-induced depression rats. This method provides a new approach to studying the pathogenesis of depression and other neurological disorders.

Protective Effects of Silibinin and Its Possible Mechanism of Action in Mice Exposed to Chronic Unpredictable Mild Stress

  • Silibinin, a natural flavonoid antioxidant isolated from extracts of the milk thistle herb, has recently been identified as having anti-hepatotoxic and anticancer properties. In this paper, we investigated the effects of silibinin on behavior and neuroplasticity in mice subjected to chronic unpredictable mild stress (CUMS). After 5 consecutive weeks of CUMS, the mice were treated with silibinin (100 mg/kg, 200 mg/kg and 400 mg/kg by oral gavage) for 3 consecutive weeks. The results showed that silibinin administration significantly alleviated the CUMS-induced depressive-like behavior, including the total number of squares crossed and the frequency of rearing in the open field test, the immobility time in the tail suspension test and the forced swimming test. Furthermore, silibinin treatment increased the levels of brain-derived neurotrophic factor (BDNF), serotonin (5-HT) and norepinephrine (NE) in the prefrontal cortex and hippocampus. Our study provides new insight into the protective effects of silibinin on the depressive status of CUMS mice, specifically by improving neuroplasticity and neurotransmission.

분심기음의 우울증 모델 흰쥐에 대한 실험적 연구 (The Effects of Punsimgi-eum(fenxinqi-yin) in the Chronic Mild Stress Model of Depression in Rats)

  • Objectives : The aim of this study was to evaluate the effects of Punsimgi-eum(fenxingi-yin) in rat model of depression induced by chronic mild stress (CMS). Methods : The change of the consumption of sucrose solution was measured, and the open field test and passive avoidance conditioning test were performed to investigate the anti-depressive effect of Punsimgi-eum(fenxingi-yin). Results : The results were as follows; I. The consumption of sucrose solution was significantly reversed in the test group at 5, 7 and 8 weeks, but there was no significant change in the control group. 2. There was no significant change of body weight after 8 weeks in each group 3. In the open field test, there was significant increase of exploratory activity in the test group. 4. In the passive avoidance test, the test group showed differences in the maintenance of retention from the control group at the 2-week training-testing interval. Conclusions : The present results suggest that Punsimgi-eum(fenxingi-yin) has an anti-depressive effect.
