Seedless grape (Vitis spp.) cultivars with large berries can be developed from hypo- and hypertetraploid seedlings. The low occurrence of hypo- and hypertetraploid seedlings, however, has impeded the breeding of new hypo- and hypertetraploid grape varieties. In order to establish hypoand hypertetraploid seedlings, we examined the chromosome numbers in seedlings of self-, open-, and cross-pollinated hypotetraploid 'Takao' and hypertetraploid 'RB9127K' grape. Three of the five seedlings (60%) from 'Takao' were aneuploid, including one with 74 chromosomes (2n=4x-2) and two with 75 chromosomes (2n=4x-1). In 'RB9127K', 26 of the 193 seedlings (13.5%) were aneuploid, including three seedlings with 75 chromosomes (2n=4x-1), 18 with 77 chromosomes (2n=4x+1), and five with 78 chromosomes (2n=4x+2). The high frequency of aneuploids from 'Takao' and 'RB9127K' grape indicates that meiosis in hypo- and hypertetraploid female parents is prone to segregation error. These results suggest that various hypo- or hypertetraploid seedlings can be successfully produced using hypo- or hypertetraploid grapes as female parents, which can contribute to the development of new seedless grape varieties with large berries.
Chromosome analysis using mitosis, meiosis and bicolor FISH were carried out in Bupleurum latissimum Nakai, which is one of the endemic plants in Ulleung island of korea. The somatic methaphase chromosomes number of this plant was 2n = 2x = 16 and the chromosome complements consisted of six pairs of metacentrics and two pairs of submetacentrics. The size of chromosomes ranged 2.40${\sim}$4.20 ${\mu}$m and NOR (nucleolus organizer region) chromosome did not observed using conventional staining. In meiosis chromosomes, metaphase-I and anaphase-I were observed. Metaphase-I anaphase-I showed 8 bivalents and chromosomes migration to make two daughter cells. Using bicolor FISH, one pair of 5S and 45S rDNA signals were detected on the centromeric region of chromosome 3 and the end of short of chromosome 2,respectively. We also observed the NOR using 45S rDNA probe.
The uptake of isolated Lyophyllum ulmarium chromosomes by Ganoderma applanatum protoplasts was induced with polyethylen $glycol+CaCl_2+glycine$. Uptake products of chromosomes by protoplasts showed micro-and macrotransgenome type. The former was slowly growing and unstable, the latter was outgrowing and stable mycelial colonies which made thick hyphae of width and segregation of mycelial colony on GCM containing benomyl. A comparison of macrotransgenome type was using isozyme analysis of esterase. The enzyme pattern of two transformants was distinct in position and quantity compared with parents.
The frequency of reicprocal translocation damage in males of D. melanogaster irradiated with X-rays was observed in this study. The frequencies were checked at four periods with two days intervals and duration spermatogenesis after irradiation. (1) Modification in the percentage of the reciprocal translocation damage were not obtained at interval after irradiated with 500r and 1500r of X-rays respectively. (2) In two experimental groups irradiated with 50-0r and 1500r of X-rays, the frequencies showing in the spermatogenesis were 0.50%(500r), 3.85%(1500r) in mature sperm, and 1.59%, 8.10% in the spermatocyte. (3) The frequency of reciprocal translocation between the Y and 3 rd chromosomes was the highest, but in accordance with dosage increase that of 2nd and 3rd chromosomes relatively increased from 9.34 % to 30.49% while decreased from 68.75% to 46.80% in the group of the Y and 3 rd chromosomes. (4) It was supposed that these modifications of the frequency were due to heavy damage of the 2nd chromosomes than other chromosomes in accordance with dosage increase. (5) Spontaneous reciprocal translocations involving the Y, 2nd and 3rd chormosomes was 0.23%.
In counting chromosome number and karyotype study, it is necessary to let chromosomes on metaphase by pretreatment before fixation. For this purpose, colchicine, or 8-oxyquinoline are generally used. The author found out that chromosomes could be scattered by illuminating cyanoferrate complex solution in which root-tips were sunk. As materials, 8 sorts of plant such as Allium fisturosum, allium tuberosum Rottler, Triticum vulgare were used. Their root-tips were sunk on the bottom of beaker in potassium ferricyanide solution $3{\times}10-4M$ and illuminated through the solution by sterilizing lamp for 1~2 hours in dark room, keeping 10 cm distance from light source to the surface of solution and 2cm depth of solution. Then again, they were illuminated to the light which was somewhat weaker intensity than the former (distance, 16cm; depth, 3cm) for 1.5~2 hours after immersed in 1/100N-HCl and washed in water for each 5minutes. By such methods chromosomes could be scattered. About the mechanism of scattering, it is supposed that CN and Fe(CN)x ions $(x {\leq}5)$ which were gradually produced in the process of photodissociation acted together on the scattering of chromosomes.
Polygenic variation of sternopleural bristle number was investigated at the whole chromosome level in a natural population of Drosophila melanogasfer. Fifty pairs of second and third chromosomes were analyzed at $25^\circ{C}$. Since environmental factors such as temperature influence polygenic expression of quantitative traits, whole chromosomal effects of 28 pairs from the larger original sample were measured under cycling temperature, a $10-30\circ{C}$ cycle in 24 hours, to reveal any polygenic alleles whose effects might be masked under the constant temperature. While third chromosomes typically showed a larger contribution to polygenic variation in both environments, second chromosomes showed greater sensitivity to environmental changes. Cluster analyses of second and third chromosomes produced a limited number of clusters. Such a small number of cluster's implies that there may be a small number of genes, or quantitative trait loci (QTLs), having large effects on phenotypic variation. The genetic structure assessed under constant temperature, however, did not show any correlation with the structure under cycling temperature. The discrepancy could be caused by independent response of each polygenic allele to temperature changes. Thus, polygenic structure in natural populations should be thought of as a temporally changing profile of interactions between gene and ever-changing environment.
The chromosome numbers of two species (Culter brevicauda and Erythroculter erythropterus) of Korean Cultrinae are investigated. In two species, the mitotic chromosomes from 24 groups with two chromosomes each indicated that it is a diploid. Their karyotypes and total length of chromosomes were slightly different from each other. The karyotype of C. brevicauda and E. erthropterus is 2n=48 (7M+10SM+7ST), FN=96 and 2n=48 (6M+10SM+8ST), FN=96. Observed chromosomes of C. brevicauda and E. erythropterus ranged from 5.4 to $2.2{\mu}m$ and 6.1 to $2.3{\mu}m$ in length, respectively.

As a series of systematic classification of paramphistomes, the worms in the rumen and reticulum were collected on 214 Korean cattle slaughtered at Jeonju abattoir from January, 1986 to April, 1987 and were classified by means of morphology. Afterwards, the karyotype of Paramphistomum cervi(Zeder, 1790) was detected by means of modified air.drying method from germ cells of the worms. The results were summarized as follows: 1. In the chromosome number of 254 p. cervi, the haploid cell was n:9 and the diploid 2n=18. The meiotic divisions were observed frequently; 1,924 haploid and 32 diploid cells were reliable. Nine pairs of mitotic chromosomes were homologous in the metaphase stage, and the chromosomes were composed of five medium-sized metacentrics (m) , subtelocentrics(st) or submetacentrics(sm) and four small-siRed subtelocentrics(st) or submetacentrics(sm). Meiotic metaphase was composed of five medium and four small chromosomes in size. 2. As a series of C-banding method, C-band was showed in centromeric region from all of the haploid germ cells. Whereas chromosome No. 3 and 5 included heterochromatin on the tip region, chromosome No. 4 on the distal region and No. 6 proximal region. And chromosomes No. 2 and 8 showed a remarkable C-band distinguished from other chromosomes.
This paper describes the design and implementation of personal computer assisted karyotyping system of Giemsa stained chromosomes. The system consists of an Image Acquisition Module being capable of $256 {\times} 256$ pixels and its relevent software modules optimized for karyotyping. The results of karyotyping using this system with an image of chromosomes taken from the Rana Amurensis are acceptable. As a result of this study we can save our load oweing to the conventional hand- karyotyping and the high-cost computer.
