Study on chromosomes survey of Korea native dogs (국내 토종개의 염색체 조사에 관한 연구)

  • The karyotype of the domestic dog is widely accepted as one of the difficult mammalian karyotypes to work. In contrast to many other animals, knowledge about the canine karyotype is quite sparse. The dog has a total of 78 chromosomes; all 76 autosomes are acrocentric in morphology and show only a gradual decrease in length. But appear to be quite small and difficult to identify unambiguously. To purchased standardization of chromosome in Korea native dog, there were analyzed by conventional trypsin/Giemsa staining (GTG-banding techniques), and were compared with 4, 6, 8, 11, 13, 17 chromosome. There were no variations in karyotypes which were analyzed by conventional GTG-banding techniques, but differences were observed in G-banding patterns with Sapsaree, Jindo, Gyeongju DongGyeong dogs, Welshi-Corgi. It is not clear that these disagreements in G-banding patterns between strains of dog were caused by chromosome polymorphism or a difference in interpretation. Comparative analysis of the distribution patterns of conserved segments defined by dog paints in the genomes of the Korea native dogs demonstrates that their differences in the karyotypes of these three species could have resulted from acrocentric banding patterns.

Karyotype Analysis of Liobagrus somjinensis, an Endemic Species in Korea (한국 고유종 섬진자가사리 Liobagrus somjinensis의 핵형 분석)

  • The karyotype analysis of Liobagrus somjinensis, an endemic Korean freshwater fish with restricted waters and a new species of torrent catfish was carried out from nine females and eight males of Somjingang River, Sikjeong-dong, Namwon-si, Jeollabuk-do, Korea. The diploid number of chromosomes was 42, and its karyotype was composed of 28 metacentrics and 14 submetacentrics; 84 FN (fundamental number). Polyploidy and sex dimorphism were not observed in the present species. The chromosome number of L. somjinensis was the same as its congeners, but there was a difference in karyotypes.

  • Morphology and karyotype of Korean Convallaria majalis plants were observed for taxonomic studies. Most plants produced two leaves (70%) and some one leaf (30%), but rarely three leaves. Plant length averaged 34.4 $\pm$ 4.6 cm. Percentage of plants bearing flowers was very low with 3.7% (n=1,346) in the field. Raceme usually grew below leaves and rarely grew over leaves. Plant beared five to ten flowers on a single raceme. Chromosomes were x=19 and diploidy with 2n=38. The chromosomes were composed of 13 pairs of median and 6 pairs of submedian chromosomes. The number of chromosome and karyotype of Korean C. majalis plant agree with those of Japanese and European plants.

  • For the karyotype analysis of Liobagrus hyeongsanensis, an endemic Korean torrent catfish, ten females and five males were used from Yangbuk-myeon, Gyeongju-si, Gyeongsangbukdo, Korea. The diploid number of chromosomes was 42, and its karyotype consisted of 30 metacentrics and 12 submetacentrics; 84 FN (fundamental number), with having no polyploidy and sexual dimorphism. The chromosome number was the same as other closely-related species, L. mediadiposalis and L. somjinensis, but their karyotypes showed a clear difference by species. Such result may be related to chromosomal rearrangements by Robertsonian rearrangement with geographical isolation.

  • CHEF-PFGE(Contour-Clarnped Homogeneous Electric field- Pulsed Field Gel Electrophoresis) was used to identify electrophoretic karyotype for eight strains belonging to the Fzisoriuni section Liseolo. Chromosome numbers were nine to thirteen bands, ranging in size Cram 0.75 to 6.45 Mb. The total genome size was eslimated to range from 38.19 Mb to 43.12 Mb and numerous chromosome-length polymorphisms (CLPs) were observed. For the chromosome localizalion of the gene, 1GS sequence(2.6 Kb) of rDNA from F: moniliforme, chs-2 gene(2.8 Kb) and 4 - 3 gene(3.8 Kb) from Neuuospora cmssa were wed as probes.

Cytogenetic Study of Pleuronectes obscurus, Konosirus punctatus and Pseudoblennius percoides

  • Our objective was to clarify the cytogenetic characteristics, including karyotypes, cellular DNA content, and nuclear size of erythrocytes, of black plaice Pleuronectes obscurus, dotted gizzard shad Konosirus punctatus, and perch sculpin Pseudoblennius percoides, collected from the coastal areas of Jo Island, Busan, Korea. Karyotypes of P. obscurus and K. punctatus both had a diploid number of 48 and a fundamental number (FN) of 48, with a chromosome formula of 48T. The karyotype of p. percoides had a diploid number of 46 and FN of 56, with a chromosome formula of 10SM +36T. No sex-associated heteromorphic pairs were detected for any species. The variation in DNA values (P. obscurus=1.15 pg/nucleus, K. punctatus=1.56pg/nucleus, P. percoides=1.11 pg/nucleus) was positively related to variation in chromosome FN.

  • Studies on the chromosome of Mus musculus have been done by Galton in 1965. In the present studies the authors have analyzed the chromosome pattern of Mus molossinus and found the same pattern of karyotype as observed by Galton. In addition, it was found that there exists no difference in karyotype between different localities and between different species.

  • Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using FISH (fluorescence in situ hybridization) technique were carried out in Nicotiana plumbaginifolia. Somatic chromosome number of N. plumbaginifolia was 2n=20 and kyarotype was composed of three pairs of metacentric chromosomes (1,2 and 7) and seven pairs of submetacentric chromosomes (3, 4, 5, 6, 8, 9 and 10). Length of chromosomes was ranged from 2.29 to 4.50 ${\mu}{\textrm}{m}$. Chromosome 1 and 2 were identified as satellite chromosomes. In FISH experiment, one pair of 5S rDNA signals was detected on the centromeric region of chromosome 2 and one pair of 45S rDNA signals was detected on the terminal region of chromosome 1.

  • The karyotype of F. velutipes Korean cultivar, Fv 3-6, was compared with those of Japanese cultivars, Fv 0-1, Fv 1-5, Fv 11-1, by CHEF gel electrophoresis. The Korean cultivar, Fv 3-6, showed the difference from the three Japanese cultivars in number and size of chromosomes; the Fv 3-6 had two and one more chromosomes then Fv 0-1 and Fv 11-4, and Fv 1-5 had, respectively. The karyotyping by CHEF gel electrophoresis is quite suitable to define new Korean cultivars against Japanese cultivars.

Prenatal diagnosis of an unbalanced translocation between chromosome Y and chromosome 15 in a female fetus

  • We report the prenatal diagnosis of an unbalanced translocation between chromosome Y and chromosome 15 in a female fetus. Cytogenetic analysis of parental chromosomes revealed that the mother had a normal 46,XX karyotype, whereas the father exhibited a 46,XY,der(15)t(Y;15) karyotype. We performed cytogenetic analysis of the father's family as a result of the father and confirmed the same karyotype in his mother and brother. Fluorescence in situ hybridization and quantitative fluorescent-polymerase chain reaction analysis identified the breakpoint and demonstrated the absence of the SRY gene in female members. Thus, the proband inherited this translocation from the father and grandmother. This makes the prediction of the fetal phenotype possible through assessing the grandmother. Therefore, we suggest that conventional cytogenetic and molecular cytogenetic methods, in combination with family history, provide informative results for prenatal diagnosis and prenatal genetic counseling.
