Determination of Voglibose in Pharmaceutical Preparation by Gas Chromatography (기체크로마토그래피를 이용한 제제 중의 보글리보스 정량)

  • Voglibose is a new antidiabetic agent currently being devoloped by the research and development division of Takeda chemical industries Ltd. in japan. Basic research for the determination of voglibose in pharmaceutical preparation by gas chromatography. This method utilizes voglibose to give its acyl compound with acetyl anhydride in pyridine solution. The calibration curve for voglibose was linear over the concentration range of 0.08∼0.32 $\mu\textrm{g}$/${mu}ell$ with correlation coefficient of 0.996. The detection limit for voglibose was 0.016 $\mu\textrm{g}$/${mu}ell$ and the result of recovery test was 101.6% with relative standard deviation of 2.0% by standard addition method.

  • 한국에서 사용하고 있는 carbamate계 살충제의 사용율(%)은 1968년경에는 2.1%인데 비해서 현재는 급증되고 있는 실정에 있다. 따라서 이로 인한 자타살 사건, 환경오염, 살포시 급만성 중독 등의 사고가 빈번히 발생하고 있는 동시에 법화학적 분석에도 많은 문제점이 되두되고 있으므로 1차적으로 시판되고 있는 carbamate계 살충제를 thin layer chromatography에 의해서 성분의 분리 확인 시험을 실시한 결과 1) Silica Gel G-60 및 Silica Gel F-254에서 적합하였고 2) 전개용매는 chloroform: benzeneP: carbontetra chloride (60:30:25), cyclohexane: acetone (80:20), cyclohexane:acetone(70:30), ether:hexane(80:20)에서 9종의 살충제가 잘 분리되었으며 3) UV light (2537 ${\AA}$) 및 2.6-dibromoquinone-4-chlorimide는 전개는 Rf치를 육안으로 구분하는데 가장 양호하였고 4) Carbamate계 살충제 중 Bux-2와 Sevin은 Silica Gel G-60에서 UV light (2537 ${\AA}$)에서 특이한 청보라 및 청색형광을 관찰할 수 있었다. 5) 이상의 결과를 보아 법화학적 시료중에 함유된 카바메이트계 농약의 분석은 가능할 것이며 또한 이 방법의 적용이 현재의 실정으로서는 적합한 것으로 사료되는 바이다.

Genetical and Biochemical Studies on White Egg Strains of Silkworm, Bombyx mori (누에 백란 계통의 유전적 특성구명을 위한 유전.생화적 연구)

  • To elucidate genetical and biochemical characters of the white egg strains preserved in National Sericultural and Entomology Research Institute(NSERI), RDA of Korea, the genetic segregation ratios in egg colours were investigated by crossing test, and egg pigment precursors were also analyzed by paper chromatography and UV-spectrophotometer scanning. The result obtained by crossing test between the white egg strains and normal one illustrated that the most of white egg strains showed typical segregation ratio of white egg-2(w-2), while maternal inheritance which can be seen in white egg-1(w-1) was not found in any white egg strains. Paper chromatographic analysis showed that egg extracts of all the white egg strains contained 3OH-kynurenine, while kynurenine which is known to existed in white egg-1(w-1) could not be detected at all. From the results of these experiments, it was found that all the white egg strains preserved in NSERI were classified as the white egg-2(w-2) strain.

Screening Test on the Ingredient of the Genus Thalictrum Roots in Korea (한국산 Thalictrum속 식물 지하부의 성분검색)

  • This study is about the five species of the genus Thalictrum growing abundantly in South Korea. Dried roots of plants; T. aquilegifolium var. japonica, T. simplex var. affine, T. actaefolium, T. tuberiferum were compared by morphological observation and screening test on the ingredient extracted by methanol, ether etc.. The experiments by thin-layer chromatography was at tempted to find out the similarity and the difference among them. In conclusion, the author found that: 1) A special component similar to coumarin was found under UV-ray from T. actaefolium. 2) By thin-layer chromatography, they show the corresponding spot supposed to be that of berberine and magnoflorine according to the $T_{ABLE}\;IV$. Therefore it is worthwhile considering that Thalictrum species would be a substitution for Epimedium species used as a tonic. 3) Thalictrum species has resemblance of ingredient and chemotaxonomy gives more help to classify them than the morphology.

  • 독사 또는 곤충의 독 30여종을 대상으로 SNU-1 위암세포에 대하여 MTT test를 실시한 결과 세포 독성 활성이 제일 높은 킹코브라(Ophiophagus hannah)의 venom을 가지고 세포 독성 물질을 정제하였다. Gel Filtration Chromatography와 Anion Exchange Chromatography로 정제한 4번째 peak만이 MTT/SRB test결과 IC$_{50}$ value 0.947$\mu\textrm{g}$/ml이었다. 음이온 교환 크로마토그라피로 정제한 단백질을 PRO-RPC로 더 분리하여 순수한 단일성분을 얻었으며 맹장암, 대장암, melanoma, fibrosarcoma 세포에 대해 독성을 확인하였고, 광학 및 전자 현미경에 의해 암세포의 분화와 성장이 억제됨을 재확인하였다. 또한 thymidine uptake asaay에서 암세포의 증식이 억제되었고, 또한 EDTA, $Zn^{++}$, $Ba^{++}$ 첨가로 세포 독성 활성이 증가되었다. The Comparative Study on Compositional Pattern Analysis of Decoction of Extracted Artemisia argyi by Different Extraction Time (전탕 시간에 따른 애엽의 성분패턴 비교연구)

  • Objectives: This study was conducted to find out the optimal extraction time for Artemisia argyi. Methods: The compositional pattern was compared with HPLC (High Performance Liquid Chromatography) and GC (Gas-Chromatography) by decocting Artemisia argyi 10, 60, 120 minutes respectively. Results: With longer extraction time, the contents of reference compounds were extracted 1.1 times more when 3,4-dicaffeoylquinic acid was extracted for 60 minutes than when extracted for 10 minutes in HPLC test, but the contents were reduced when extracted for 120 minutes compared to 60 minutes extraction time. 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, 4,5-di-O-caffeoylquinic acid, jaceosidin, and eupatilin showed the largest yield rate when extracted for 10 minutes, and it decreased as time passed. The contents of chlorogenic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid, jaceosidin, scoparone, and eupatilin were detected only in 10 minutes extraction but not in 60 or 120 minutes extraction according to GC test. Conclusions: The results show that extraction time could affect the physicochemical characteristic or composition of Artemisia argy extracted. Thus, short extraction time could be useful for decoction of Artemisia argyi.

  • The suppressing effects of vinegar and ginger for fish odor were investigated by sensory test and gas chromatography. According to the results of sensory test. the fishy odor gradually decreased as the vinegar concentration increased. But the addition of more than 16m1 of vinegar per 100 g of mackerel tasted very sour and tough. The addition of 6 ml of vinegar 100go of mackerel tasted acceptable, TMA content of 100 g of mackerel was found to be $0.4{\sim}0.7{\;}mg$. It was identified by gas chromatography that the TMA content reduced as the vinegar concent ration increased. The amount of TMA in the mixture of TMA and phenolic fraction of ginger was reduced to 67.5 % after standing 4 hours.

  • A dense, pellicular UpFront adsorbent ($p=1.5 g/cm^3$, UpFront Chromatography, Cophenhagen, Denmark) was characterized in terms of hydrodynamic properties and protein adsorption performance in expanded bed chromatography. Cibacron Blue 3GA was immobilised into the adsorbent and protein adsorption of bovine serum albumin (BSA) was selected to test the setup. The Bodenstein number and axial dispersion coefficient estimated for this dense pellicular adsorbent was 54 and $1.63{\times}10^{-5}m^2/s$, respectively, indicating a stable expanded bed. It could be shown that the BSA protein was captured by the adsorbent in the presence of 30% (w/v) of whole-yeast cells with an estimated dynamic binding capacity $(C/C_o = 0.01)$ of approximately 6.5 mg/mL adsorbent.

Anti-inflammatory Activity of Opuntia ficus-indica (손바닥선인장의 항염증 활성)

  • Cactus (Opuntia ficus-indica var saboten Makino) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. We previously found that the ethanol extract of cactus stem showed anti-inflanunatory action. This investigation was designed to isolate the active fraction of anti-inflanimatory action from cactus stem by solvent extraction and colunm chromatography. Carrageenan-induced paw edema in rats and acetic acid-induced writhing test in mice were used as animal models to search anti-inflammatory and analgesic activities. respectively. The ethanol extract of cactus stem was consecutively extracted with hexane, ethyl acetate, and n-butanol. The hexane fraction was the most effective in carrageenan-induced paw edema, and then was separated in colunm chromatography of silica gel by the elution with hexane/ethyl acetate mixture. The most effective fraction 1 was separated in a second colunm chromatography by eluting with hexane/diethyl ether mixture. The most effective fraction 1-5 was obtained, and separated in a third column chromatography by eluting with hexane/chloroform mixture. It produced the most effective fraction 1-5-1. Moreover, fraction 1-5-1 showed an inhibitory effect on acetic acid-induced writhing in the doses of 30mg/kg and 60mg/kg,p.o.,indicating that it also contained analgesic activity.

Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli (알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy)

  • The aim of this study was to separate or purify some bioactive compounds from flowers of alfalfa(Medicago sativa L.) and to test of the isolated compounds on alfalfa for their autotoxicity and on Echinochloa crus-galli for their allelopathy for seed germination and seedling weight. Using thin layer chromatography(TLC) of $CHCl_3$ extracts, the most inhibitory band to alfalfa seed germination was determined. Germination inhibition of this extract suggested a complex chemical interaction. Separation and purification of compounds with CHCl$_3$ extract of fresh alfalfa flowers were conducted by a silica gel TLC, and microcrystalline cellulose TLC(MCTLC), followed by droplet countercurrent chromatography(DCCC) bioassay. Preliminary identification was done by high perfomance liquid chromatography(HPLC) on the most inhibitory fractions in DCCC. Ferulic acid, caffeic acid, vanillic acid, rutin, narringin were identified in fraction 5 and ferulic acid, caffeic acid, vanillic acid, rutin, coumarin in fraction 6. The phytotoxicity of their individual compound was tested on alfalfa and Echinochloa crus-galli seed germination and seedling weight. Coumarin and ferulic acid showed the most inhibitory effect on alfalfa seed germination and Echinochloa crus-galli seedling fresh and dry weight. These compounds may be, at least in part, involved in autotoxicity and allelopathy.
