Determination of halogen elements in plastics by using combustion ion chromatography (연소IC를 이용한 플라스틱 중 할로겐 물질 정량)

  • For plastics samples, a method using combustion ion chromatography was selected as a method for rapid low-cost analysis to test whether hazardous substances are contained or not. Using combustion ion chromatography, a verification test for F, Cl and Br compounds generated a linear calibration curve with a correlation coefficient of $r^2$ = 0.999~1.000 in the calibration range from 0.5 to 4.0 mg/kg. The detection limits were found to be 0.005~0.024 mg/kg and quantitative limits were found to be 0.014~0.073 mg/kg. The recoveries of combustion ion chromatography using certified reference material (CRM) were found to be 95.5~104.9%. Based on these results, a proficiency test was conducted together with several laboratories in and out of the country, to make comparative analysis of the results from each laboratory. As a result, the data supported the use of combustion ion chromatography as an effective analysis method to deal with regulations for halogen-free electronic products and for other hazardous substances in the electronic products.

  • These experiments were carried out to develop new techniques for In Vitro separatin of X-and Y-bearing spermatozoa. The bull semen was applied to the various Gel-Columns filled with swellen Sephadex G-50 Fine and then elutriated wtih Locke solution (elutriation rate; 1ml/3-4min., 1ml/1-2min.). Elutriated solution was fractionated into 1ml by automatic Fraction Collector and spermatozoa included in each fraction were subjected to the estimation of viability and recovery rate, and to B-body test. The results obtained in these experiments were summarized as follows: 1. When the column size and the elutriation rate were adjusted to 15$\times$1.6cm and 1ml/3-4min., respectively, the highest sperm concentration was obtained from the 8th to the 12th fraction. 2. As a trend, the viability of spermatozoa was improved by chromatography, and the degree of improvement ranged 5 to 10 percentage. 3. The average recovery rate of spermatozoa applied to column was 73.2 percentage and ranged 52.6 to 81.3 percentage. 4. The lowest rate of B-body bearing spermatozoa following chromatography was obtained when the column size and the elutriation rate were adjusted to 15$\times$0.8cm and 1ml/1-2min., respectively.

  • Metabonomic analysis using Liquid Chromatography-Mass Spectrometry (LC-MS) was employed to test the feasibility to predict chemical-induced toxicity. Time-dependent metabolic variations were evaluated in rats treated with the model hepatotoxin, ${\alpha}$- naphthylisothiocyanate (ANIT). Urine samples of ANIT treated group and control group were collected up to 7 days postdose. Urine samples were analyzed by gradient HPLC combined with electrospray mass spectrometry. The chromatographic results were data-reduced and analyzed using principal component analysis to show the time dependent biochemical variations induced by ANIT toxicity. (omitted)

  • For the purpose of obtaining microorganisms producing high amount of erythritol, the screening test was carried out. Productivity of erythritol was analyzed by paper chromatography and HPLC' methods. Among more than two hundred isolates, one strain(KJX1) was selected as an erythritol prtducer from thc soil of corn shock. The isolated strain was identified as Pmicilliurn sp. KJ81 from the morphological and physiological characteristics. Penicillium sp. KJ81 showed white to green colony color, two- to three-stage branching conidiophcvc and flask-shaped phialides.

  • The Anthocyanins in the Korean native Brassica juncea, which were favorably used directly for food or as condiments, were investigated. Two kinds of anthocyanin were separated from Korean native Brassica juncea by T.L chromatography and then Robinson test, spectrophotometry, T.L. chromatography, partial hydrolysis were tested for the structural elucidation of these two anthocyanins. It was known that two pigments were peonidin-3-glucoside and peonidin-3-galactoside.

Optical Purity Determination of (S)-Ibuprofen in Tablets by Achiral Gas Chromatography

  • An optical purity test was indirectly performed on (S)-ibuprofen as its diastereomeric (R)-(+)-1-phenylethylamide derivative using achiral gas chromatography (GC). The method for the determination of trace (R)-ibuprofen (optical impurity), within the range 1.0 to 50 ng, from a racemic ibuprofen standard was linear (r=0.9997) with acceptable precision (% $RSD{\leq}5.3$) and accuracy (% RE=0.7~-3.9). Similar results were obtained with the method validation for the quantification of (S)-ibuprofen within the range 0.1 to 2.0 $\mu\textrm{g}$ using a (S)-ibuprofen stan-dard. When applied to seven different commercial (S)-ibuprofen products, their optical purities (98.7~99.1%) were determined with good precision (% $RSD{\leq}4.0$).

  • An attempt was carried out to purify the cell components of Bordetella bronchiseptica by the mild procedures. From the supernatant of sonic treated colls, the K-agglutinogen, hemagglutinogen and skin necrotizing factor were separated by a successive use of DEAE-cellulose column chromatography and ammonium sulfate precipitation. The specific activity of purified K-agglutinogen was demonstrated by a gel-diffusion test and was also found to be free from other biologically active substances of B. bronchiseptica: namely hemagglutinin, skin-necrotizing factor and O-antigen.

  • Phenolic compounds such as gallic acid, ferulic acid, p-hydroxybenzoic acid, vanillic acid, salicylic acid, tannic acid, and hydroquinone were identified from the aqueous extracts and volatile substances of tomato plant by paper chromatography, high performance liquid chromatography and gas chromatography. The seed germination and seedling growth of the experimental species, lettuce and egg plant, were severely inhibited in 5$\times$10-3M of phenolic reagents identical to those identified from tomato plant. Germination and growth rate of test species in 5$\times$10-4M and 5$\times$10-5M were higher than that of 5$\times$10-3M. Therefore, 5$\times$10-3M of phenolic compounds would be assumed to be threshold concentration for allelopathic effects.

Purity Test of $Hippuran^{131}$I by Chromatography (크로마토그래프법에 의한 $Hippuran^{131}$-I의 순도시험)

  • The purified .omicron.-iodobenzoic acid-$^{131}$ I, sodium iodide-$^{131}$I and mixtures of those compounds were applied to radio paper or radio thin layer chromatography to select the developing solvents with useful separation efficiency. The separation efficiencies were checked by radiochromatogram sacnnings. It has been found that the two dimensional radio paper chromatogrphy using the solvent systems of 8% phenol and butanol-glacial acid-water (4:1:1 v/v) and the radio thin layer chromatography using alumina gel plate and butanol- glacial acetic acid- water (4:1:1 v/v) are both efficient. In this method, the Rf value of .omicron.-iodobenzoic acid-$^{131}$ I and the unbound $^{131}$ I is 0.48, 0.85 and 0.15, respectively.
