녹두(Phaseolus aureus L.) 첨가 식이가 고지혈증 흰쥐의 지질성분 개선 및 단백질 농도에 미치는 영향 (Mung Bean (Phaseolus aureus L.) Improves Lipid Composition Protein Concentration in Hyperlipidemic Rats)

  • This study aimed to investigate the improvement effect of 5% (w/w) mung bean (Phaseolus aureus L.) on lipid composition and protein concentration in hyperlipidemic rats. Seven-week-old male (Sprague-Dawley) rats were divided into four groups (n=6, each), and fed experimental diets containing mung bean meal [basal diet+5% (w/w) mung bean (BM), basal-hyperlipidemic diet+5% (w/w) mung bean (BHM)], basal-hyperlipidemic diet (BH), and control (basal diet, BD). Lipid composition in the mung bean diet groups (BM and BHM) evidenced significant reductions in serum total cholesterol, low-density lipoprotein cholesterol, atherosclerotic index, cardiac risk factor, triglyceride, phospholipid, free cholesterol, cholesteryl ester, blood glucose, nonesterified fatty acid, and an elevation in high-density lipoprotein cholesterol. The serum albumin/globulin ratio was increased in mung bean diet-supplemented rats compared to that in hyperlipidemic rats (p < 0.05). Total calcium, phosphorus and potassium concentrations in sera were higher in the BM, BHM and BD groups than in the BH group. Concentrations of sodium and chlorine in sera were lower in the mung bean diet-supplemented groups than in the hyperlipidemic group. The in vivo experiments showed that ingestion of mung bean was effective in improving lipid composition, protein concentration, and blood glucose.

Preparation of a Hydrophobized Chitosan Oligosaccharide for Application as an Efficient Gene Carrier

  • To prepare chitosan-based polymeric amphiphiles that can form nanosized core-shell structures (nanopar­ticles) in aqueous milieu, chitosan oligosaccharides (COSs) were modified chemically with hydrophobic cholesterol groups. The physicochemical properties of the hydrophobized COSs (COSCs) were investigated by using dynamic light scattering and fluorescence spectroscopy. The feasibility of applying the COSCs to biomedical applications was investigated by introducing them into a gene delivery system. The COSCs formed nanosized self-aggregates in aqueous environments. Furthermore, the physicochemical properties of the COSC nanoparticles were closely related to the molecular weights of the COSs and the number of hydrophobic groups per COS chain. The critical aggregation concentration values decreased upon increasing the hydrophobicity of the COSCs. The COSCs effi­ciently condensed plasmid DNA into nanosized ion-complexes, in contrast to the effect of the unmodified COSs. An investigation of gene condensation, performed using a gel retardation assay, revealed that $COS6(M_n=6,040 Da)$ containing $5\%$ of cholesteryl chloroformate (COS6C5) formed a stable DNA complex at a COS6C5/DNA weight ratio of 2. In contrast, COS6, the unmodified COS, failed to form a stable COS/DNA complex even at an elevated weight ratio of 8. Furthermore, the COS6C5/DNA complex enhanced the in vitro transfection efficiency on Human embryonic kidney 293 cells by over 100 and 3 times those of COS6 and poly(L-lysine), respectively. Therefore, hydrophobized chitosan oligosaccharide can be considered as an efficient gene carrier for gene delivery systems.

콜레스테릴과 아크릴로일 그룹을 지닌 하이드록시프로필 셀룰로오스들의 합성 및 특성 (Synthesis and Characteristics of Hydxoxypropyl Celluloses Containing Cholesteryl and Acryloyl Groups)

  • 하이드록시프로필 셀룰로오스를 6-콜레스테릴옥시카보닐펜타노일 클로라이드와 반응시켜 에스터화도가 2.25-2.91 범위의 값을 갖는 6-콜레스테릴옥시카보닐펜톡시프로필 셀룰로오스들을 합성하였다. 또한 6-콜레스테릴옥시카보닐펜톡시프로필 셀룰로오스의 아크릴산 에스터들과 액정질서를 지닌 이들의 광가교 필름들을 합성하였다. 미가교와 가교시료들의 열방성 액정의 특성과 아세톤 중에서의 가교시료들의 팽윤거동을 검토하였다. 하이드록시프로필 셀룰로오스는 쌍방성 콜레스테릭 상을 형성하는 반면 모든 콜레스테릴을 지닌 미가교 시료들은 단방성 콜레스테릭 상을 형성하였다. 6-콜레스테릴옥시카보닐펜타노일 클로라이드는 단방성 스멕틱 상도 형성하였다. 하이드록시프로필 셀룰로오스는 광학 피치 (λ$_{m}$ )가 온도상승에 의해 증가하는 우측방향의 나선을 형성하는 반면 모든 미가교 유도체들은 λ$_{m}$ 이 온도상승에 의해 감소하는 좌측방향의 나선들을 형성하였다. 6-콜레스테릴옥시카보닐펜톡시프로필 셀룰로오스와 이의 아크릴산 에스터의 액정상의 열적 안정성과 동일한 온도에서의 λ$_{m}$ 의 크기는 에스터화도가 증가함에 따라 감소하였다. 모든 가교시료들은 6-콜레스테릴옥시카보닐펜톡시프로필 셀룰로오스의 아크릴산 에스터들과 달리 반사색깔을 나타내지 않았으며 액정질서를 지닌 가교젤의 특징적인 이방성 팽윤을 나타냈다.

Penicillium griseofulvum F1959, High-Production Strain of Pyripyropene A, Specific Inhibitor of Acyl-CoA: Cholesterol Acyltransferase 2

  • Acyl-coenzyme A: cholesterol acyltransferase (ACAT) catalyzes cholesterol esterification and plays an important role in the intestinal absorption of cholesterol, hepatic production of lipoproteins, and accumulation of cholesteryl ester within cells. During the course of screening to find ACAT inhibitors from microbial sources, the present authors isolated pyripyropene A from Penicillium griseofulvum F1959. Pyripyropene A, an ACAT2-specific inhibitor, has already been produced from Aspergillus fumigatus. Yet, Aspergillus fumigatus is a pathogen and only produces a limited amount of pyripyropene A, making the isolation of pyripyropene A troublesome. In contrast, Penicillium griseofulvum F1959 was found to produce approximately 28 times more pyripyropene A than Aspergillus fumigatus, plus this report also describes the ideal conditions for the production of pyripyropene A by Penicillium griseofulvum F1959 and its subsequent purification.

산국의 잎과 줄기에서 ACAT 저해활성을 가지는 Guaianolides의 분리 (Isolation of Guaianolides with ACAT Inhibitory Activity from the Leaves and Stems of Chrysanthemum boreale Makino)

  • Acyltransferase (AT) catalyzes the transfer of an acyl moiety from acyl-coenzyme A (acyl-CoA) to an acceptor. ATs play important roles in the maintenance of homeostasis in the human body and have been linked to various diseases; therefore, several ATs have been proposed as potential targets for the treatment or prevention of such diseases. The AT family includes acyl-CoA:cholesterol AT (ACAT), diacylglycerol AT, and monoacylglycerol AT for the metabolism of lipids. Furthermore, recent molecular biological studies revealed the existence of their isozymes with distinct functions in the body. ACAT plays a critical role in the formation of cholesteryl esters from cholesterol and fatty acids, and is a potential target for treating hypercholesterolemia. During an experiment designed to discover biologically active compounds from herbal medicines, we isolated two known guaianolide sesquiterpene lactones from Chrysanthemum boreale Makino (Compositae). The lactones were characterized from their spectroscopic data (NMR, IR, MASS). These compounds were subjected to ACAT inhibition assay. Here, we report the isolation and structural elucidation of the compounds 8-o-acetyl-2-methoxy-10-hydroxy-3,11(13)-guaiadiene-12,6-olide and 8-acetyl-3,10-hydroxy-4(15),11(13)-guaiadiene-12,6-olide. In the ACAT inhibition assay, compound 1 showed strong inhibitory activity, with an $IC_{50}$ value $45{\mu}g/mL$, whereas compound 2 did not exhibit significant inhibitory activity with an over $100{\mu}g/mL$.

식용버섯과 식물성 유지의 혼합급여가 흰쥐 간장의 지질성분 및 지방산 조성에 미치는 영향 (Effects of the Feeding Mixture of Mushrooms and Vegetables Oils on the Lipid Component and Fatty Acid Composition of Liver in Rats)

  • This study was designed to observe the effects of the mixed diets of edible mushrooms and vegetables oils on the lipid component and fatty acid composition in liver of the diet induced hydpercholesterolemic rats. Ten groups of male S.D. rats were fed a basal diet supplemented with 5% of one of three mushrooms(G.I, L.e, A.j) and 10% of one of three vegetable oils (olive ,safflower perilla) for three weeks. In liver, total cholesterol concentration was significantly low in group 3 (olive oil 10 % + L. edodes 5%) and 6 (safflower oil 10 % $_2$L. edodes 5%) , triglyceride concentration was low in groups 8 (perilla oil 10 % + g. lucidum 5%) and 9 (perilla oil 10% + L. edodes 5%) and phospholipid concentration was significantly low in groups 3, 5, (safflower oil 10 % + G.lucidum 5%), 6, 7 (safflower oil 10 % + A .judae 5%) 8, 9 and 10 (perilla oil 10% + a. judae 5%). in the fatty acid composition of total lipid inliver, monounsaturated fatty acid (MUFA) concentration s were high in groups 2 (olive oil 105 + g. lucidum 5%), 3, and 4 (olive oil 10% + A. judae 5%) and all the perilla oil groups, polyunsaturated fatty acid (PUFA) and linoleic aicd concentrations were signifciantly high in all the safflower oil groups. In the fatty acid composition of liver phospholipid , PUFA concentrations were ghih but MUFA concentrations were low. In the triglyceride component, MUFA were some more than saturated fatty acid (SFA) . In the cholesteryl ester component, MUFa concentrations were significantly high. In the fatty acid composition of liver lipid components, linholeic acid was high in the PUFA and so it was major fatty acid. Eicosapentaenoic acid (EPA) of phospholipid component in liver was significantly high.

동충하초(Cordyceps militaris)가 당뇨의 혈당 및 동맥경화 개선에 미치는 영향 (Effects of Cordyceps Militaris on the Improvement Actions of Blood Glucose and Atherosclerosis in Diabetes)

  • The purpose of this study was to assess the effects of Cordyceps militaris extract supplementation on the improvement of blood glucose, lipid compositions and hepatic functional enzyme activities in the serum of streptozotocin(STZ, 50 mg/kg BW, IP injection)-induced diabetic rats fed the experimental diets for 5 weeks. The concentrations of blood glucose and lipid compositions(total cholesterol, ${\beta}$-lipoprotein, free cholesterol, cholesteryl ester, triglyceride and phospholipid) and atherosclerotic index in serum were significantly lower in the DM+CM group than in the DM group, whereas the ratio of HDL-cholesterol concentration to total cholesterol in the DM+CM group were higher than in the DM group. The activities of hepatic functional enzymes(aminotransferase, LDH and ALP) in serum were lower in the Cordyceps militaris extract administration group(group DM+CM) than in the hyperglycemic DM group. The results shown above suggested that Cordyceps militaris extract supplementation effectively improves blood glucose and atherosclerosis in the serum of diabetic rats.

Synthesis of Reconstituted High Density Lipoprotein (rHDL) Containing apoA-I and apoC-III: the Functional Role of apoC-III in rHDL

  • Apolipoprotein (apo) C-III is a marker protein of triacylglycerol (TG)-rich lipoproteins and high-density lipoproteins (HDL), and has been proposed as a risk factor of coronary heart disease. To compare the physiologic role of reconstituted HDL (rHDL) with or without apoC-III, we synthesized rHDL with molar ratios of apoA-I:apoC-III of 1:0, 1:0.5, 1:1, and 1:2. Increasing the apoC-III content in rHDL produced smaller rHDL particles with a lower number of apoA-I molecules. Furthermore, increasing the molar ratio of apoC-III in rHDL enhanced the surfactant-like properties and the ability to lyse dimyristoyl phosphatidylcholine. Furthermore, rHDL containing apoC-III was found to be more resistant to particle rearrangement in the presence of low-density lipoprotein (LDL) than rHDL that contained apoA-I alone. In addition, the lecithin:cholesterol acyltransferase (LCAT) activation ability was reduced as the apoC-III content of the rHDL increased; however, the CE transfer ability was not decreased by the increase of apoC-III. Finally, rHDL containing apoC-III aggravated the production of MDA in cell culture media, which led to increased cellular uptake of LDL. Thus, the addition of apoC-III to rHDL induced changes in the structural and functional properties of the rHDL, especially in particle size and rearrangement and LCAT activation. These alterations may lead to beneficial functions of HDL, which is involved in anti-atherogenic properties in the circulation.

단일지방산 첨가에 의한 간세포의 지질조성과 지방산조성에 미치는 영향 (Effects of Stearic, Oleic and Elaidic Acid on Cellular Lipids and Their Fatty Acid Composition in Hep-$G_2$ Cells)

  • The effects of stearic(18 : 0, SA), oleic(18 : 1 cis, OA) and elaidic acid(18 : 1 trans, EA) on the cell growth, contents of cellular lipids, and the fatty acid composition of cellular and medium lipids in Hep-G$_2$cells were evaluated. The cells were incubated in serum-free medium containing 25, 50, 100 and 200$\mu$M of a fatty acid combined with albumin for 2 days. The fatty acid concentration up to 100$\mu$M showed the normal growth, but the cell growth decreased in the presence of 200$\mu$M fatty acid. The treatment of cells with 100$\mu$M of a fatty acid for two days significantly(p<0.05) increased the cellular triglyceride(TG) content in all fatty acid groups compared to control, but TG contents was not significantly different among all treatment group, but total cholesterol(TC) was the highest level in EA group. The level of free cholesterol(FC) and cholesteryl ester(CE) was similar to those of TC in all fatty acid treated group. The cellular phospholipid(PL) contents were similar between the control and all fatty acid groups. The treatment of cells with SA has no notable effects on the fatty acid composition of TG, CE and PL. The OA treatment caused significant increases in CE(51.2%) and PL(29.8%), but not in TG. The EA treatment resulted in 10.1, 10.7 and 7.8% of $C_{18:1\;trans}$ content in cellular TG, CE, and PL. The TG, CE and PL of medium were relatively similar between SA and OA groups. In EA treated group, TG, CE and PL of medium contained 17.0%, 0.7% and 5.6% of $C_{18:1\;trans}$, respectively.

Effects of Palm Kernel Cake on Performance and Blood Lipids in Rats

  • Palm kernel cake (PKC), a by-product of oil palm seeds after extraction of their oil. The aim of this study was to investigate the effects of different levels of PKC on growth performance and blood lipids in rats. A total of 64 Sprague-Dawley (8 weeks of age) male rats were assigned individually to four treatments with different levels of PKC in the diet: 0, 15, 20 and 25%. No differences (p<0.05) were found in daily feed intake (6-8 g/day), body weight, growth rate and epididymal fat weight for all the dietary groups. Plasma protein and very low density lipoprotein (VLDL) triacylglycerol (TG) were higher (p<0.05) for 20% PKC fed rats than the control rats. Conversely, the plasma cholesterol and TG and VLDL-phospholipid (PL) concentrations of the control rats were higher (p<0.05) than those of PKC fed rats. The VLDL-protein, total cholesterol, free cholesterol (FC) and cholesteryl ester (CE) were not significantly different (p>0.05) among the treatment groups. Rats fed PKC had greater (p<0.05) ratios of total surface to core lipid components [(FC+PL)/(CE+TG)] than control rats. The results reflect dissimilarities of VLDL particle size between PKC treatment and control rats, where the plasma of the PKC treated rats contained more lipid rich VLDL. In conclusion, there was no adverse effect on growth performance when inclusion of PKC up to 25%. However, fibre content may affect the plasma lipid concentrations.
