Isolation and Identification of Antimicrobial Compound from Sancho (Zanthoxylum Schinifolium) (산초로부터 항균성 화합물의 분리 및 동정)

  • Antimicrobial activity of Sancho (Zanthoxylum schinifolium) was investigated. Methanol extract of dried Sancho was fractionated to hexane, chloroform, ethylacetate, butanol, and aqueous fractions. Chloroform fraction among these fractions showed the highest inhibitory effect on the microorganisms such as Escherichia coli, Bacillus subtilis, Staphylococcus aureus and Lactobacillus plantarum at 1000 $\mu\textrm{g}$/$m\ell$. Chloroform fraction was further fractionated into 4 fractions by silica gel column and thin layer chromatography (TLC). The fraction 3 on TLC exhibited the highest antimicrobial activity. In the 2nd fractionation, subfration 2 was identified as hexadecanoic acid by MS, $^1$H-NMR and IR.

  • The cytotoxic activity of medicinal plants was screened using A549 human lung cancer cell line. Plant materials were extracted with 80% methanol and fractionated to chloroform and water layers. Each methanol, chloroform, and water extract of thirty-two medicinal plants was tested for cytotoxic activity in A549 cell culture system and the cell viability was measured by SRB assay.

  • The analgesic and the antipyretic activity of the methanol, chloroform, hexane and acetonitrile extracts of the aerial parts of Stachys schimperi Vatke were investigated in mice. The nociceptive response was tested using acetic acid-induced writhing and tail flick method; while hypothermia affect was examined via yeast-induced fever test. The chloroform extract at 500 mg and hexane and acetonitrile extracts at 250 mg produced significant analgesic and antipyretic activity.

Isolation of Antioxidative Components from the Bark of Rhus verniciflua STOKES Screened from Some Chinese Medicinal Plants (한약재로부터 선발된 옻나무 수피 추출물로부터 항산화 활성물질의 분리)

  • To develop new natural antioxidants, antioxidative activity of ethanol (75%) extracts from 50 edible or medicinal plants were examined on lard and palm oil by Rancimat method ($120^{\circ}C$, 20 L/hr). The extracts from Rhus verviciflua STOKES showed comparatively strong antioxidative activity on test. Of the solvents used for extraction, chloroform extract exhibited the strongest antioxidant activity. AI (antioxidant index: induction period of oil containing extract/induction period of control oil) of chloroform extract was higher than that of commercial antioxidant, such as BHT, BHA and ${\delta}-tocopherol$. Free phenolic acid fraction (200 ppm) of the chloroform extract from 75% EtOH extract of Rhus verniciflua STOKES (RCF) showed stronger activity than that of BHT, BHA, and ${\delta}-tocopherol$ at the same concentration. RCF-11 and RCF-13 fractions separated by silicagel column chromatography from the RCF showed stronger activity than other fractions by the Rancimat method.

  • Water extract, and methanol extract, its chloroform and hexane fractions, and estragole from Agastache rugosa O. Kuntze were tested to find the inhibition effect on the growth of several microorganisms. The organisms used were Escherichia coli ATCC 1129, Staphylococcus aureus IAM 1011, Vibrio parahaemolyticus WP, Bacillus subtilis ATCC6633, Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240. Water and methanol extracts at the concentration of 0.5%, and chloroform and hexane fraction at the concentration of 0.05% inhibited the growth of microorganisms from 1/5 to 2/3 of the control group. Estragole identified from the hexane faction as a major component, its authentic compound completely inhibited the growth of Vibrio parahaemolyticus completely at the concentration of 0.03%, and the other bacteria were at 0.05%.

  • For the biological survey, effects of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima(Simaroubaceae) on epoxide hydrolyzing enzymes were checked. The methanolic extract and its chloroform fraction were shown to activate the liver metabolizing enzyme system including epoxide hydrolase system which was monitored by activities of transaminase, lactate dehydrogenase, alkaline phosphatase and epoxide hydrolase system in bromobenzene treated rats. But they showed no effect on glutathione S-transferase activity.

  • In the course of screening for new natural compounds having antioxidant activity from microbials, mushrooms as well as plants. We have isolated novel sesquiterpenoid and officinalic acid from methanolic extracts of Stereum ostrea. The dried fruit body( 450 g) of Stereum ostrea was extracted with methanol, then methanolic extract was partitioned between water and hexane, chloroform, and ethyl acetate, subsequently. The chloroform extract was subjected on silica gel, Sephadex LH-20 and RP-18 column chromatography, successively. And two compounds were isolated and identified as new triterpenoids and officinalic acid using $^1$H - NMR, $\^$13/C - NMR as well as Mass spectra.

  • Effects of the ultrasonic treatment on plant represents antifeeding activity and chitin synthesis inhibition activity against diamondback moth (Plutella xylostella L.) by a leaf-dipping method at a concentration of 5,000ppm. Methanol extracts of leaves of the ultrasonic treatment on plant was sequentially partitioned with n-hexane, chloroform, ethlyacetate, butanol and water fractions, and bioassayed. Antifeeding activities were observed in n-hexane, chloroform and ethlyacetate fractions. N-hexane fraction has 94.6% of strongest antifeeding activity against P. xylostella. From the n-hexane sequentially partitioned showed H6 and H8 fractions of antifeeding activities against P. xylostella. And chitin synthesis inhibition activity against P. xylostella was obtained the methaol, n-hexane and water fractions.

Studies on the Cytotoxicity and Antimicrobial Effects of the Extract of Ajuga multiflora Bunge (조개나물 추출물의 세포독성과 항균효과)

  • This study was carried out to evaluate cytotoxic effects of Ajuga multiflora Bunge extracts on murine leukemia tumor $(P388D_1)$ cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The comparison of $IC_{50}$ values of Ajuga multiflora Bunge extracts in L1210 and $P388D_1$ cell lines showed that their susceptibility to these extracts decreased in the following order: Adriamycin>methanol extract>chloroform extract>ethyl acetate extract>hexane extract>water extract by the MTT assay. In order to develop an antimicrobial agent, dried Ajuga multiflora Bunge was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activities of amocla and ketoconazole as references were compared to those of extracts of $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi $(MIC>200\;{\mu}g/ml)$. These results suggest that the methanol soluble extract of Ajuga multiflora Bunge may be a valuable choice for the studies on the treatment of murine leukemia tumor cell lines.

Effect of Mugwort Extract on the in vitro Mutagenicity, Desmutagenicity. (쑥 추춤물의 항돌연변이 활성효과)

  • Mugwort has been known as a traditional substitutive foodstuff and as showing a physiologically beneficial function to a human being. Therefore, effect of mugwort extract in terms of mutagenicity and desmutagenicity was investigated to berify its function. Ethanol extract from mugwort did not exhibit any mutagenicity. On the contrary, inhibitory effects of the ethanol extract were observed on mutagenicity induced by aflatoxin $B_{1}(AFB_1)$, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-P-1), 3-amino-1-methyl-5H-pyrido[4,3-b]indole(Trp-P-2) and 2-nitroflourene(2NF) using Salmonella typhimurium reversion assay. On direct-acting mutagen(2NF, 3${\mu}$g/plate), ethanol extract showed a slight inhibitory effect of 19.7~22.9%, however on indirect-acting mutagen such as AFB1(2${\mu}$g/plate), Trp-P-1(1${\mu}$g/plate) and Trp-P-2(1${\mu}$g/plate), we observed higher inhibitory effect of 47.9~61.2%, 64.1~70.7%, 67.4~78.7%, respectively. Step-wise fractionation of the ethanol extract was done by using hexane, chloroform, ethyl acetate and water to obtain effective fraction. Among them, hexane, chloroform, and ethyl acetate fractions showed high inhibition of 63.0~80.0%, 77.5~82.1%, and 68.5~83.1%, respectively on the mutagenicity of $AFB_1$ in Sal. typhimurium TA98. Consequently, these results indicated that mugwort extract contains some compound(s) which may show desmutagenicity.
