Investigation of Antioxidative Activity from Apple (Malus pumila Miller) Peel by Various Extraction Solvents

  • Consumption of fruits and vegetables has been conducted to be effective in the prevention of chronic diseases. In this study, 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used as solvents in the extraction of apple peels. The total phenol content, total flavonoid content and antioxidant activity of various extracts were investigated using in vitro assays. The extract obtained by 70% methanol showed the highest total phenol content ($20.87{\pm}0.17mgCAE/g$), ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity and ferric reducing antioxidant power. However, 70% ethanol extract possessed the strongest antioxidant activity assayed by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ${\beta}$-carotene bleaching method. And CM extract was found to show the highest total flavonoid content with the value of $9.26{\pm}0.06mgQE/g$. These results indicated that apple peels can be used in dietary applications with a potential to reduce oxidative stress.

Studies on Licorice in Drug Preparations(I) Determination of Glycyrrhizin and Glycyrrhetinic acid by HPLC. (생약중의 감초에 관한 연구(I) HPLC에 의한 Glycyrrhizin 및 Glycyrrhetinic acid의 정량)

  • Glycyrrhizin (GA) content in licorice was determined by a couple of methods using HPLC, respectively. In Method(I), GA content itself was determined from the licorice aqueous extract, while in Method (II) glycyrrhetinic acid (GHeA ; the aglicone of GA) content corresponding to the quantity of GA was measured from the chloroform extract of the hydrolyzed product of licorice aqueous extract. A reverse phase column Hibar Lichrosorb RP-18 (E. Merck) was used as the stationary phase. As the mobile phase MeOH: $H_{2}O$(0.05M-$NaH_{2}PO_{4}$)=58 : 42 solution in Method (I), and MeOH: $H_{2}O $: AcOH=78; 19: 3 solution in Method (II) were suitable, respectively. The value obtained by Method (II) appeared slightly higher than that by Method (I). The effect of some other herbal drugs on the assay of GA quantity in mixed sample was also observed in both above two methods. By Method (I) Cassiae Cortex, Rehmaniae Rhizoma, Paeoniae Radix, and Angelicae Radix gave the subtractive effect on the amount of GA compared with the value from licorice alone. In the case of Method (II) Cassiae Cortex and Rehmaniae Rhizoma appeared to have subtractive effect but Paeoniae Radix and Angelicae Radix scarcely showed any influence. Pachymae Fungus did not affect the GA content at all. It seems that glycyrrhizin in licorice interacts with certain components of other herbal drugs.

Total Flavonoid Content and Antioxidant Activities of Turmeric (Curcuma longa L.) Extracts in Jindo Korea (진도산 울금(Curcuma longa L.) 추출물의 총 플라보노이드 함량 및 항산화 활성)

  • The present study were conducted to determine physiological activities and antioxidant effects [2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, reducing power, Ferric Reducing Antioxidant Power (FRAP) and Fe2+ (ferrous ion) chelating capacity] of 70% methanol, chloroform:methanol, 2:1 volume ratio (CM) and ethyl acetate extract of turmeric (Curcuma longa L.). Bioactive compound of tannin $0.125{\pm}0.007mg$ Catechin Equivalent (CE)/g dry weight. Turmeric extracts yield were 70% methanol 16.54%, CM 5.64% and ethyl acetate 4.14%, respectively. Antioxidant activity of the samples exhibited a dose-dependent increase. Results showed that extraction solvent had significant effects on total flavonoid content and antioxidant effects of ethyl acetate. But ferrous ion-chelating capacity of 70% methanol extract was higher than CM and ethyl acetate extract. From the results of this study, turmeric can be utilized as a valuable and potential nutraceutical for the functional food industry.

Antimicrobial Effect of Extract of Glycyrrhiza uralensis on Methicillin-Resistant Staphylococcus aureus (감초 추출물이 항생제 내성균주의 항균활성에 미치는 영향)

  • Antimicrobial drug-resistance is natural response to antimicrobial stress based on selection, which weakens chemotherapy effect. Introduction of large numbers of chemotherapeutic agents to clinical practice has generated strains of microorganisms that survive and multiply in vivo with high-drug concentrations. Methicillin-resistant Staphylococcus aureus (MRSA), bacteria found in normal daily life, can be easily ingested through milk vegetables, and meats, etc. MRSA emerged in many port of the world, increasing complex clinical problems. Therefore, new agents are needed to treat MRSA. Glycyrrhiza uralensis was extracted using 80% MeOH to investigate its antimicrobial activity against MRSA stains KCCM 11812, 40510, and 40512 through bacterial measurement, disc diffusion, and O.D. methods, MIC values, MRSA gene expression investigation, and scanning electron microscope observation. Results revealed MecA, Mecl, MecRI, and FemA were the most highly manifested MRSA genes. Methanolic extract of G. uralensis significantly inhibited MRSA and thus could be used in development of antibacteria.

Evaluation of Antioxidant Capacity and Antimutagen Activity of Bulbil Extracts of the Dioscorea japonica Decaisne and Dioscorea batatas Decaisne (단마와 장마 영여자의 항산화능 및 항돌연변이 활성 검정)

  • The bulbil of the $Dioscorea$ $species$ is produced, the amount of which is 2,000 tons annually, but it has been discarded without specific use. In this study the antioxidant and antimutagenicity of bulbil of the $Dioscorea$, which compared to bulbil of Danma($Dioscorea$ $japonica$ Decaisne) and Jangma($Dioscorea$ $batatas$ Decaisne), a major domestic cultivation species. The study was done by extracting bulbil of the Dioscorea methanol and the methanol extracts was re-extracted with chloroform, ethyl acetate, $n$-butanol and water. In methanol extract of Danma and Jangma, polyphenolic compounds contained 2.2 and 3.9 mg/g extract, respectively. The ethyl acetate fraction of Danma and Jangma had higher polyphenolic contents of 33.9 and 39.1 mg/g, whereas water fractions were much lower at 2.4 and 5.8 mg/g. Determination of antioxidant activity showed that the ethylacetate fraction strong DPPH radical scavenging activity and ABTS radical scavenging activity. The inhibitory effects of methanol extracts and chloroform, ethylacetate, $n$-butanol, water fraction from bulbil of Danma and Jangma on the mutagenicity in 1-NP, $AFB_1$, Trp-P-1 were investigated using $S.$ $typhimurium$ TA98. Danma and Jangma cultivars decreased the reverse mutation induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$ $typhimurium$ TA98. The fraction of chloroform and ethylacetate showed strong inhibitory effects, in a dose dependant manner against the mutagenicities induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$ $typhimurium$ TA98.

Bioactivities of feral peach (Prunus persica Batsch var. davidiana Max.) flower extracts and Prunus mume flower extracts (개복숭아꽃과 매화꽃 추출물의 생물활성)

  • In this study, we investigated the antibacterial, antioxidant, and whitening effects of Prunus persica Batsch var. davidiana Max. (Feral Peach) and Prunus mume (white and pink) flowers. The extracts of the three kinds of flowers showed antibacterial activity against Staphylococcus. aureus. The chloroform fractions of the white Prunus mume, Feral Peach, and pink Prunus mume flowers exhibited antibacterial activities of 84, 49, and 30%, respectively, against Staphylococcus. aureus at a concentration of 0.5 mg/mL. The flower extracts of the three species also exhibited antibacterial effects against Pseudomonas. aeruginosa. The chloroform fractions of the Feral Peach and pink Prunus meme flowers exhibited antibacterial activities of 36 and 30%, respectively, at a concentration of 0.5 mg/mL. These extracts did not exhibit any significant antibacterial activity against Staphylococcus. epidermidis and Escherichia. coli. The extracts of the three kinds of flowers did not significantly affect the survival of HaCaT cells. The distilled water fraction of the pink Prunus mume flower extract exhibited antioxidant effects at concentrations of both 20 and 40 ㎍/mL. The ethyl acetate fraction of the pink Prunus mume flower extract exhibited an antioxidant activity superior to glutathione at a concentration of 40 ㎍/mL. The flower extracts did not significantly affect the survival rate of B16F10 cells. The chloroform fraction of the Feral Peach flower exhibited a whitening effect of 18% at a concentration of 40 ㎍/mL. Based on these results, we conclude that the three kinds of flower extracts are raw materials exhibiting antibacterial, antioxidant, and whitening effects.

  • The cytotoxic and antitumor activity of Herba cratalariae sessiliflorae on cultured NIH 3T3 fibroblast and human oral epitheloid carcinoma cells were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured mouse fibroblast and human oral epitheloid carcinoma cells (KB). These results were obtained as follows; Ethyl acetate, chloroform and hexane extracts showed a significant cytotoxicity in NIH 3T3 fibroblast, but the other extracts did not show. All extracts exhibited a significant antitumor activity in human oral epitheloid carcinoma cells, but ethanol extract did not show a antitumor activity. Hexane extract showed low cytotoxic effect, but exhibited the most antitumor activity. The MTT absorbance in NIH 3T3 fibroblast was significantly decreased by treatment with chloroform, ethyl acetate and hexane extracts respectively. Human oral epitheloid carcinoma cells was significantly decreasd by treatment with all extracts with the exception of ethanol extract. The difference in MTT absorbance in two cell Types was most remarkable when treated with water and hexane extracts. Cholroform and hexane extracts showed the strongest effect in growth inhibition of human oral epitheloid carcinoma cells. These results indicated that water extract possessed no cytotoxicity and a strong antitumor activity.

Anti-oxidant Property and Tyrosinase Inhibition Activity of Various Extracts from plants in Compositae plants (국화류 추출물의 항산화 및 멜라닌 생성 억제 활성)

  • The research was conducted to identify the antimicrobial effect, anti-oxidative effect and tyrosinase inhibitory effect of MeOH 80% extract and n-hexane, chloroform, ethyl acetate, n-butanol and water fractions from the extract of six kinds of compositae plants, which are naturally grown across the nation. In the antimicrobial effect, the extract and chloroform fraction of Arctium lappa and hexane/ethyl acetate fractions of Taraxacum platycarpum exhibited significant inhibition. In case of antioxidant effect, the extract of Artemisia capillaries showed the highest effect and ethyl acetate/butanol fractions of all plants showed about 90%, which fractions were more polar than the fractions that showed antimicrobial effect. In case of tyrosinase activity, only the MeOH 80% of Arctium lappa among the extracts showed a potent inhibition, and butanol fraction of Chrysanthemum indicum, as well as ethyl acetate/water fractions of Artemisia capillaries showed 48, 38, and 37% respectively, which were higher than control group (arbutin). These active fractions in tyrosinase inhibition also were higher polarity than those that showed antimicrobial effect. In MeOH 80% extracts, only Arctium lappa was found to have antimicrobial, anti-oxidant and tyrosinase inhibitory activity, however there was no fraction to show effects commonly in the three assay system.

Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts (누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성)

  • This study investigated the antioxidant activities, whitening effects, and antimicrobial activities of fractions from Pleurospermum kamtschaticumin extract. Total polyphenolic contents of fractions from Pleurospermum kamtschaticumin extract were 116.44~382.73 mg/g GAE (gallic acid equivalent), with the highest value in the ethyl acetate fraction. Fractions of Pleurospermum kamtschaticumin showed the highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ($IC_{50}$=0.04 mg/mL) and ABTS radical scavenging activity (98.86% at 0.5 mg/mL), which was similar to ascorbic acid. Further, reducing power and FRAP activity were significantly high in the ethyl acetate and n-butanol fractions. The ethyl acetate and n-butanol fractions showed significantly high SOD-like activities (1 mg/mL, 86.93%, and 78.23%, respectively) compared to other fractions. Tyrosinase inhibition activities of the n-butanol fraction and 80% ethanol extract were 43.52% and 41.64%, respectively. Antimicrobial activity against Escherichia coli was only observed in the chloroform fraction (16.7 mm in 0.5 mg/disc). These results suggest that fractions from Pleurospermum kamtschaticumin extract show comparatively high antioxidant activity and thus could be used as a food additive or cosmeceutical ingredient.

Antioxidative Effect of Some Edible Plant Solvent Extracts with Various Synergists (패모, 어성초, 쇠비름 및 들깨박 에탄올 추출물의 순차용매 분획별 항산화 효과)

  • The antioxidative effect of the 75% ethanol extracts of preliminary selected Fritillaria ussuriensis Max(Fs), Houttuynia cordata Thunb(Hc), Portulaca oleracea L.(Po) and Perilla frutescene Var javanica Hara cake(Pf) were tested on palm oil and lard by rancimat test. Each solvent fraction of chloroform, ethyl acetate(EtOAc), butanol and water, was also evaluated its antioxidative effect with some synergists, like as ascorbic acid, citric acid and ${\delta}-tocopherol$. Po extract showed higher antioxidative effect on lard and the fraction of all test plants were the most effective on palm oil and lard with ascorbic acid and ${\delta}-tocopherol$. When 200 ppm of EtOAc fraction of Fs, He and Po extract each with 200 ppm of ascorbic acid were added to palm oil, the antioxidative index(AI, induction time of oil containing each extract/induction time of test oil) were 1.60, 1.53 and 1.47 respectively and 200 ppm of EtOAc fraction of Po and Pf extract each with 200 ppm of tocopherol to lard, the AI were 3.19 and 3.63 respectively.
