Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

Detection of Endolichenic Fungi Producing Antifungal Compound (항진균성 물질을 생산하는 지의류 내생 곰팡이의 탐색)

  • To isolate a novel antifungal compound, we obtained 100 kinds of endolichenic fungi from Korean Lichen & Allied Bioresources Center and examined their antifungal capability. Three fungi Usnea rigidula (2326), Parmotrema pseudotinctorum (2202) and Myelochroa sp. (2292) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these three fungi with chloroform and then with ethyl acetate. Chloroform fraction exhibited the highest antifungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. The chloroform faction was on further analysis with $C_{18}$ column chromatography to see whether the inhibitors are already known or not. Two peak fractions were collected from 4-day culture extract for Usnea rigidula and from 6-day culture extract for Parmotrema pseudotinctorum on the HPLC. A peak fraction from chloroform extracts of 4-day culture filtrate of Parmotrema pseudotinctorum showed higher antifungal activities against C. albicans and C. glabrata than another peak fraction. It appears that the antifungal materials are relatively nonpolar as usnic acid often found in lichenic fungi.

  • A methanol extract of the roots of bupleurum longiradiatum (Umbelliferar) showed antiinflammatory activity in rats. Hexane, chloroform, butanol, and aqueous fractions from the methanol extract were tested with the result that only the hexane fraction exhibited positive activity. Fractionation of the hexane extract resulted in isolation of arborinone as an active principle.

  • The antioxidative action of petroleum ether, chloroform, chloroform-methamol (2 : 1, v/v), methanol and methanol-water extracts of Panax ginseng roots in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUFA) was investigated in vitro. All of the extracts of Panax ginseng roots possessed antioxidative activity and inhibited the formation of hydroperoxides in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUEA). The induction period in the autoxidation of MEUFA was extended by the addition of each extract. The antioxidative activity was more prominent with chloroform and chloroform-methanol (2 : 1, v/v) extracts than with other extracts. The antioxidative activity of each extract estimated by Olcott's oven test and the measurement of peroxide value (POV) showed a similar tendency. From the results obtained, it was conclude that extracts of Panax ginseng roots had remarkable antioxidative activity in the autoxidation of MEUFA in vitro.

Protective Effects of Extracts of Diospyrus kaki Folium against Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (감잎 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 효과)

  • This study was done to investigate the protective effects of Diospyrus kaki Folium on carbon tetrachloride intoxicated rats. Male Sprague-Dawley rats (200~230g) was used. Experimental groups were divided into normal group and $CCl_{4}$-treated group and then $CCl_{4}$-treated groups were divided into 7 groups: only $CCl_{4}$-treated group, methanol extract pretreated group, butanol fraction pretreated group and $H_{2}O$ fraction pretreated group, $CCl_{4}$ treated groups were injected with $CCl_{4}$ 0.6 mg/kg (ip). The activities of ananine aminotransferasein hexane, chloroform and $H_{2}O$ fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group and also activity of aspartate aminotransferase and hepatic malondialdehyde content in $H_{2}O$ fraction pretreated group were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The activity of alkaline phosphatase in chloroform and $H_{2}O$ fraction pretreated groups were decreased compared to the only $CCl_{4}$-treated group but the difference was not statistically significant. The activity of ${\gamma}$-glutamyltranspeptidase in hexane, chloroform and ethylacetate fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The contents of cholesterol and triglyceride were not influenced in any fraction pretreated groups, but HDL-cholesterol content in hexane and chloroform fraction pretreated groups were significantly increased (p<0.05) compared to the only $CCl_{4}$-treated group. Therefore, these results demonstrate a possible hepatoprotective role of $H_{2}O$ fraction of Diospyrus kaki Folium methanol extract in carbon tetrachloride intoxicated rats.

Antibacterial and Antioxidative Activities of Various Extracts from Basidiomycetes (담자균 추출물의 항균작용 및 항산화작용)

  • The water and methanol extract were obtained from fruit body of 8 kind of edible mushrooms. The antibacterial activity of extracts on the growth of pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O 157:H7 and Salmonella typhimurium) was determined. Methanol fraction of Gyrophora esculenta showed excellent antibacterial activity alai t 5 strains of pathogenic bacteria. The 80% methanol extract of Gyrophora esculenta and Phelinus were fractionated with diethylether, chloroform, ethyl acetate, butanol and water. The diethylether, ethyl acetate and butanol fractiorl of Gyrophora esculenta had excellent antibacterial activity and ethyl acetate and butanol fraction of Phellinus linteus had weak antibacterial activity against 5 strains of pathogenic bacteria. Electron donating ability of each fraction of Gyrophora esculenta was increased in order of ethylacetate, chloroform, butanol, diethylether and water. Nitrite scavenging ability was observed in ethyl acetate fraction of Gyrophora esculenta and other fractions showed no activities.

Potential of Benthic Diatoms Achnanthes longipes, Amphora coffeaeformisand Navicula sp. (Bacillariophyceae) as Antioxidant Sources

  • Recently, interest in plant-derived food additives has developed natural antioxidants, in order to alternate syn-thetic antioxidants with several disadvantages. In the present study, different organic fractions from solvent parti-tions of 80% methanol extract from Jeju benthic diatoms, Achnanthes longipes, Navicula sp. and Amphora coffeaeformis was assessed for their potential antioxidant effects. Among the solvent fractions tested, n-hexane (80.4%) and 80% methanol extract (76.6%) from A. longipes, chloroform (63.2%) from Navicula sp. and n-hexane (67.4%) from A. cof-feaeformis were effective in DPPH free radical scavenging. Fractions of chloroform (53.4%) and n-hexane (53.1%) from A. longipes exhibited higher activities on $H_2O_2$ scavengin. Fraction of n-Hexane from A. longipes exhibited the highest hydroxyl radical scavenging activity and NO. scavenging activity (56.5% and 75.6%, respectively). Aqueous residue from A. coffeaeformis (75.6%) showed the highest metal chelating effect. chloroform and ethyl acetate frac-tion of all the diatoms exhibited significant antioxidant activities in lipid peroxidation inhibitory activity. In particu-lar, both chloroform and the ethyl ecetate fraction from A. longipes and A. coffeaeformis exhibited lipid peroxidation inhibitory activity significantly higher than that of $\alpha$- tocopherol. These data suggest that the Jeju benthic diatoms tested are rich in hydrophilic and hydrophobic antioxidative compounds with different antioxidative properties that can be applied in food industry.

The Inhibitory Effects of Chloroform Fraction Extracted from the Dandelion (Taraxacum platycarpum D.) against Lactic Acid Bacteria and Yeast Related to Kimchi Fermentation (포공영 클로로포름 분획의 김치 유산균 및 효모에 대한 저해 효과)

  • This study was conducted to investigate the effects of solvent extracts from medicinal plants on the fermentation of kimchi. Five microorganisms related to kimchi fermentation were selected and the antimicrobial activities of solvent fractions from medicinal plants were investigated. The chloroform fraction from the methanol extract of dandelion (Taraxacum platycarpum D.) exhibited inhibitory activity against five strains such as Lactobacilli plantarum, Lactobacilli brevis, Leuconostoc mesenteroides, Enterococcus faecalis and Saccharomyces cerevisiae. The chloroform fraction from the methanol extract of Dandelion inhibited the growth of E. faecalis and Leu. mesenteroides at the concentration of 80 mg/mL. Scanning electron micrographs of Leu. mesenteroides and E. faecalis treated with chloroform fraction 80 mg/mL exhibited morphological changes, including irregularly contracted cell surface.

Antioxidant Activity of Solvent Extracts from Sargassum thunbergii (지충이 용매 추출물의 항산화 활성)

  • In this study, we investigated the antioxidant activity of solvent extracts from Sargassum thunbergii by measuring electron-donating ability (EDA), nitrite- scavenging ability, superoxide dismutase (SOD)-like activity, reducing power and antioxidant activity in $\beta-carotene-linoleate$ model system. Total phenolic compound contents of chloroform and methanol extracts from Sargassum thunbergii were $22.5{\pm}0.8$ mg/100 mL and $20.1{\pm}0.6$ mg/100 mL, respectively. The EDA were proportionally increased with concentration of the extracts except hexane extract. The EDAs were $70.4\%\;and\;63.2\%$ in methanol and water at concentration of $1000{\mu}g/mL$, respectively. The nitrite-scavenging ability of methanol extract (pH 2.5, $1000{\mu}g/mL$) was $89.4{\pm}0.9\%$ that was higher than those of other extracts. SOD-like ability of chloroform extract was stronger than other extracts. Reducing power of chloroform extracts at $1000{\mu}g/mL$ were 0.75 as O.D. value of 700 nm and antioxidant activities measured with $\beta-carotene-linoleate$ model system were in order of methanol $(78.4\%)>BHT\;(72.1\%)>chloroform\;(62.2\%)>water\;(51.4\%)>hexane\;(43.2\%)$. These results indicated that methanol extract from Sargassum thunbergii showed the highest activities on the EDA, nitrate- scavenging ability and antioxidant activities while the chloroform extract had the highest effect on the SOD-like activity and reducing power.

Study on the Antimicrobial Activity of Thalictrum uchiyamai Nakai (Thalictrum uchiyamai Nakai의 항균작용에 관한 연구)

  • The methanol extract of Thalictrum uchiyamai Nakai was fractionatedi nto four parts; (1) ether soluble nonphenolic fraction (2) phenolic tertiary bases fraction, (3) chloroform soluble tertiary bases fraction and (4) aqueous quaternary bases fraction. The fractions of this plant showed a total of twenty-four spots on the silica gel TLC. Ether soluble nonphenolic fraction and ether soluble phenolic fraction showed nine alkaloid spots respectively, chloroform soluble fraction showed three alkaloid spots and quaternary bases fraction showed three alkaloid spots. The antimicrobial activity of the four extracts was shown as follows; nonphenolic tertiary bases inhibited the growth of Micrococcus dutica at the concentration of $50\mu$g/ml, in phenolic tertiary bases $500\mu$g/ml concentration was inhibitory to Micrococcus dutica and to Staphyloccus aureus. In aqueous quaternary bases, $500\mu$g/ml concentration was inhibitory to Klebsiella pneumonae. Chloroform soluble tertiary fraction showed slight activity. All of bases showed reproducible activity against tested microorganisms.
