납의 흡입독성 연구를 위한 에어로졸 발생장치의 고안 및 실시간 모니터링을 이용한 성능평가 (Design of Aerosol Generator for Inhalation Toxicology Study of Lead and Evaluation with Real Time Monitoring)

  • This paper was the design of aerosol generator for inhalation toxicology study of lead and evaluation with real time monitoring, and applied several engineering methodology to classical aerosol generator to cope with it's disadvantages. According to the testing conditions, source temperature 50℃ and inlet-duct band heater temperature 150℃, aerosol generation results for sodium chloride and lead acetate were as followings: CPM(Count Per Minute) for Sodium chloride that used for the testing material in aerosol generation and inhalation system was decreased in the 2nd and the 3rd hour's serial trials, but CVs(coefficient of variation) were maintained within 10%. CPMs for 5 and 2.5 gram of lead acetate that used for aerosol generation and inhalation exposure of lead showed similar results because of the sedimentation of lead acetate on piezoelectric crystal with time. For that reason, heating and mixing of nebulizing solution will be needed to generate lead aerosol with stable profile and maximum generation efficiency. Fluctuations of 10 and 5 gram lead acetate were low but 2.5gram was high. However, CVs for 10, 5, and 2.5gram lead acetate were within 10%. Considering the theoretical efficiencies for sodium chloride and lead acetate, 5gram sodium chloride and 2.5gram lead acetate were appropriate choice. Aerosol generation characteristics for two materials with 1 hour interval were different with respect to the fluctuation of CPM and the decrease to 10gram in it's material. For that reason, sodium chloride can not be used to estimate the aerosol generation and it's related parts for lead acetate. According to the testing conditions, source temperature 20, 50, 70℃, and inlet-duct band heater temperature 20, 50, 100, 150, 200℃, aerosol generation results for sodium chloride and lead acetate were as followings: Excluding inlet-duct band temperature 200℃, maximum CPM for sodium chloride was manifested in source temperature 70℃ with each inlet-duct band temperature conditions. We suggest that this condition was the optimum in the design of aerosol generator, inhalation system, and the testing. Maximum CPMs for 10, 5, and 2.5gram sodium chloride were from source temperature 70℃ and inlet-duct band temperature 20℃. Excluding inlet-duct band temperature 50, 200℃, maximum CPMs for lead acetate were indicated in source temperature 50℃ with each inlet-duct band temperature conditions. We suggest that this condition was the optimum in the design of aerosol generator, inhalation system, and the testing for lead inhalation study. Source and inlet-duct band temperatures for 10, 5, 2.5gram lead acetate were 50 and 100℃, 50 and 100℃, 50 and 150℃, respectively. In conclusion, considering above 2 paragraphs of results for aerosol generation, 5gram efficiencies for sodium chloride, lead acetate were higher than 2.5gram's. If inlet-duct band temperature was same, aerosol generation was increased with increase of source temperature. To get maximum aerosol generation will be the conditions that set the appropriate inlet-duel band temperature for each materials and increase the source temperature.

PVC재질 프로파일의 생산, 발생 및 재활용, 처리에 기반한 물질흐름도 검토 (Investigation on Material Flow Diagram for PVC(poly vinyl Chloride) Profile Based Production, Generation, Recycling and Treatment)

  • 본 연구에서는 현실적으로 실현가능한 플라스틱 제품의 재활용률을 산정하기 위하여 PVC프로파일에 대한 물질흐름도를 작성하는 것을 목적으로 연구가 수행되었다. 이를 위해 합성수지 및 제품별 생산 현황 조사, 폐기물 발생 및 재활용 현황 조사, 재활용 실태 현황 조사, 제품별 물질흐름 검토를 거쳐 내구성 플라스틱의 발생부터 폐기까지의 조사를 실시하여 물질흐름도를 작성하였다. 제품별 재활용이 가능한 비율을 산정한 결과, PVC바닥재와 PVC프로파일의 경우 국내에서 사용된 제품의 내수량의 합이 525,448톤이고, 폐기물 배출량은 105,853톤으로 나타났다. 재생제품과 재활용원료의 생산량 합계는 76,004톤으로 제품의 내수량에 대비하면 2009년 의무량 8.5%에 비교하여 연구를 통한 재활용량은 추정량 14.46%로 산출되었다. 향후 각 품목의 연도별 사용량은 해당제품의 생산량이 현재와 크게 차이가 없을 것으로 가정하고 연도별 생산량에 대입한 후 5~20년 뒤의 물질수지도를 작성한 결과, 재활용률은 지속적으로 상승하여 바닥재 및 프로파일은 2013년에 20% 수준의 재활용률에 도달될 것으로 예측되었다.

Stimulation of Platelet-Activating Factor (PAF) Synthesis in Human Intestinal Epithelial Cell Line by Aerolysin from Aeromonas encheleia

  • Aeromonas encheleia, a potential human intestinal pathogen, was shown to infect a human intestinal epithelial cell line (Caco-2) in a noninvasive manner. The transcriptional profile of the Caco-2 cells after infection with the bacteria revealed an upregulated expression of genes involved in chloride secretion, including that of phospholipase A2 (PLA2) and platelet-activating factor (PAF) acetylhydrolase (PAFAH2). This was also confirmed by a real-time RT-PCR analysis. As expected from PLA2 induction, PAF was produced when the Caco-2 cells were infected with the bacteria, and PAF was also produced when the cells were treated with a bacterial culture supernatant including bacterial extracellular proteins, yet lacking lipopolysaccharides. Bacterial aerolysin was shown to induce the production of PAF.

CLINICO-HAEMATOLOGICAL AND BIOCHEMICAL ALTERATIONS IN ETHYLENE GLYCOL INDUCED ACUTE NEPHROTOXICITY IN COW CALVES

  • Ethylene glycol was given orally in 6 crossbred male cow calves @ 12 ml/kg b wt for 2 days continuously to develop acute nephrotoxicity and monitor blood chemicals profile in affected calves. Progressive depression, hypersalivation, ataxia, incoordination, staggering gait, grinding of teeth, recumbency, coma, convulsions and death were prominent symptoms in affected calves. Respiration and pulse rates were increased whereas body temperature and rumen movements were low. Haematological investigations revealed increase in total erythrocyte count, platelets count and packed cell volume till death and total leukocyte count up to day 3 which decreased on day 4 and 5. These calves revealed azotaemia, reduction in calcium, chloride and potassium and rise in sodium and AST, ALT and alkaline phosphatase enzymes activity.

  • The production of reactive oxygen species on addition of hexavalent chromium (potassium dichromate, $K_2Cr_2O_7$ ) to lung cells in culture was studied using flow cytometer analysis. A Coulter Epics Profile flow cytometer was used to detect the formation of reactive oxygen species after $K_2Cr_2O_7$ was added to A549 cells grown to confluence. The cells were loaded with the dye, 2',7'-dichlorofluorescein diacetate, after which cellular esterases removed the acetate groups and the dye was trapped intracellularly. Reactive oxygen species oxidized the dye, with resultant fluorescence. Increased doses of Cr(VI) caused increasing fluorescence (10-fold higher than background at 200 gM). Addition of Cr(III) compounds, as the picolinate or chloride, caused no increased fluorescence. Electron paramagnetic resonance (EPR) spectroscopic studies indicated that three (as yet unidentified) spectral "signals" of the free radical type were formed on addition of 20, 50, 100 and 200 gM Cr(VI) to the A549 cells in suspension. Two other EPR 'signals" with the characteristics of Cr(V) entities were seen at field values lower than the standard free radical value. radical value.

Immunotoxicity Following Pre- and Post-natal Aluminum Exposure in Rats

  • The present study was designed to explore the immunotoxic effects of orally administered aluminum (AI) on pregnant rats (n = 60) and their growing fetuses and consequently on the animal wealth. The animals were randomly allocated into three equal groups of 20 rats each. The first group has no treatment and kept as a control (G1). The second and third groups of pregnant rats were treated orally with aluminum chloride at 345 mg/Kg b.wt. The second group (G2) received the tested compound from the $6^{th}$ day of gestation to the end of weaning, whereas the third group (G3) received the tested compound from the $15^{th}$h day of gestation to the end of weaning. Control and treated animals (dams and offspring) were immunized ip with (0.5 ml) 20% sheep red blood cell (SRBC) suspension seven days before the end of experiments. At the end of exposure, ten dams and ten offspring from each group were used for assessment of cell-mediated immunity and a similar number of animals were sacrificed for evaluating the humoral immune response and serum protein profile. Aluminum chloride exposure of dams ($G_2&G_3$) caused significant suppression of both cell mediated and humoral immune responses in the obtained offsprings compared to the control group ($G_1$) without any significant effect on the immune responses of these dams. Moreover, the serum total globulins, albumin/ globulin (A/G) ratio and gamma globulin fraction were significantly decreased in the treated dam's offsprings compared to the corresponding controls while the serum total protein and all serum protein fractions showed non significant difference between the control and treated dams and between the two treated dam groups themselves. There were no histopathological changes observed in thymus, spleen and liver of the control and treated dams. Thymus of treated dam's offsprings (G2) showed lymphoid depletion in both cortex and medulla. Their spleens showed lymphoid depletion in the white pulps and congestion with hemosiderosis in the red pulps. Liver of treated dam's offsprings showed dilation and congestion of its central vein with degenerative changes in the hepatocytes. These histopathological changes were more severe in G2 than in G3 offsprings. It can be concluded that gestational and/ or lactation exposure of pregnant dams to AI chloride caused suppression of both cellular and humoral immune responses of their offsprings.

  • 유포자 유산균인 Lactobacillus sporogenes 가 특이하게 균체외 $\beta$-galactosidase를 다량 생산하는 것을 발견하여 효소생산을 위한 영양요구성과 배양조건을 조사하였다. 영양요구성으로는 탄소원으로서 lactose 1%, 유기질소원으로서 peptone 1.5%, 무기질소원으로서 ammonium sulfate 0.2%, phosphorus 원으로 ammonium phosphate, dibasic을 0.8%. mineral은 potassium chloride 0.05%, ferric chloride 0.001% 첨가했을 때 최대의 효소생산을 나타내었다. 배지의 최적 initial pH는 7.0, 최적배양온도는 37$^{\circ}C$, aeration효과는 500$m\ell$용 진탕 flask에 배지량을 50~200$m\ell$ 주입, 140 strokes/min (진폭 7cm)으로 진탕 배양하였을 때 최고의 효소생산을 나타냈다. 상기의 최적조건에서 균체증식은 24시간 배양에 최고에 달하는 반면 $\beta$-galactosidase 생산은 균체내 효소생산의 경우 균체증식 속도와 비례적으로 증가하여 배양 24시간 전후에서 30U/$m\ell$로 최고치를 나타내고 균체외 효소는 배양 40시간 전후 즉, 균증식의 후기 정지기에서 38U/$m\ell$로 가장 높은 효소생산량을 보였다. 또한 fermentor 실험 역시 flask배양과 거의 비슷한 배양 양상을 보였으며 균체외 효소역가는 45U/$m\ell$로 진탕 flask 배양 결과보다 다소 양호하였다.

Ground-based model study for spaceflight experiments under microgravity environments on thermo-solutal convection during physical vapor transport of mercurous chloride

  • For $P_B=50Torr,\;P_T=5401Torr,\;T_S=450^{\circ}C,\;{\Delta}T=20K$, Ar=5, Pr=3.34, Le=0.01, Pe=4.16, Cv=1.05, adiabatic and linear thermal profiles at walls, the intensity of solutal convection (solutal Grashof number $Grs=7.86{\times}10^6$) is greater than that of thermal convection (thermal Grashof number $Grt=4.83{\times}10^5$) by one order of magnitude, which is based on the solutally buoyancy-driven convection due to the disparity in the molecular weights of the component A ($Hg_2Cl_2$) and B (He). With increasing the partial pressure of component B from 20 up to 800 Torr, the rate is decreased exponentially. It is also interesting that as the partial pressure of component B is increased by a factor of 2, the rate is approximately reduced by a half. For systems under consideration, the rate increases linearly and directly with the dimensionless Peclet number which reflects the intensity of condensation and sublimation at the crystal and source region. The convective transport decreases with lower g level and is changed to the diffusive mode at $0.1g_0$. In other words, for regions in which the g level is $0.1g_0$ or less, the diffusion-driven convection results in a parabolic velocity profile and a recirculating cell is not likely to occur. Therefore a gravitational acceleration level of less than $0.1g_0$ can be adequate to ensure purely diffusive transport.

Effects of Diverse Water Pipe Materials on Bacterial Communities and Water Quality in the Annular Reactor

  • To investigate the effects of pipe materials on biofilm accumulation and water quality, an annular reactor with the sample coupons of four pipe materials (steel, copper, stainless steel, and polyvinyl chloride) was operated under hydraulic conditions similar to a real plumbing system for 15 months. The bacterial concentrations were substantially increased in the steel and copper reactors with progression of corrosion, whereas those in stainless steel (STS) and polyvinyl chloride (PVC) reactors were affected mainly by water temperature. The heterotrophic plate count (HPC) of biofilms was about 100 times higher on steel pipe than other pipes throughout the experiment, with the STS pipe showing the lowest bacterial number at the end of the operation. Analysis of the 16S rDNA sequences of 176 cultivated isolates revealed that 66.5% was Proteobacteria and the others included unclassified bacteria, Actinobacteria, and Bacilli. Regardless of the pipe materials, Sphingomonas was the predominant species in all biofilms. PCR-DGGE analysis showed that steel pipe exhibited the highest bacterial diversity among the metallic pipes, and the DGGE profile of biofilm on PVC showed three additional bands not detected from the profiles of the metallic materials. Environmental scanning electron microscopy showed that corrosion level and biofilm accumulation were the least in the STS coupon. These results suggest that the STS pipe is the best material for plumbing systems in terms of the microbiological aspects of water quality.

MOCVD Deposition of AlN Thin Film for Packaging Materials

  • New single-source precursor, [AlCI3:NH2tBu] was synthesized for AlN thin f film processing with AICI3 (Aluminum Chloride) and tBuNH2 (tert-butylamine). AlN thin films for packaging aspplication were deposited on sapphire substrate by a atmosph하ie-pressure MOCVD. In most of other study methyl-based AI precursors w were used for source, But herein Aluminum Chloride was used for as AI source i in order to prevent the carbon contamination in the films and stabilize the p precursor. New precursor showed the very high gas vapor pressure so it allowed to m make the film under atmospheric-pressure and get the high purified film. High q quality AlN thin film was obtained at 700 to $900^{\circ}C$. The new precursor was p purified by a sublimation technique and help to fabricate high purity film. It s showed high vapor pressure, which is able to a critieal factor for the high purity a and atmospheric CVD of AlN. High Quality AIN thin film was obtained at $700-900^{\circ}C$.
