Effect of Chitosan Oligosaccharide on Melanin Production in B16 Melanoma Cells (B16 Melanoma 세포에서 Chitosan Oligosaccharide가 Melanin 생성에 미치는 영향)

  • To investigate the effect of chitosan oligosaccharide on melanin synthesis, we measured tyrosinase activity and melanin production in B16 melanoma cells. Chitosan oligosacchaide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-dependently increased melanin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. These results suggest that chitosan oligosaccharide-induced melanin production may be independent on tyrosinase activity in B16 melanoma cells. From the above results. chitosan oligosaccharide dose-dependently appears to increase melanin production in B16 melanoma cells, suggesting that chitosan oligosaccharide may be used as a tanning agent.

Chitosan Oligosaccharide Inhibits $^{203}HgCl_2-Induced$ Genotoxicity in Mice: Micronuclei Occurrence and Chromosomal Aberration

  • The purpose of this study was to investigate the safety of chitosan oligosaccharide and the effects of chitosan oligosaccharide on mercury induced genotoxicity in mice using the micronuclei and chromosome aberration. The micronuclei test was performed by microscopic examination $(\times1,000,\;stained\;using\;a\;May-Grunwald\;solution)$ after administering 0.01, 0.1, and $1\%(10\;mg/mL)$ chitosan oligosaccharide for 7, 60, and 180 days ad libitum in mice. Total micronuclei of 1,000 polychromatic erythrocytes were recorded for each group. There was no difference between the untreated and experimental groups. The intake periods and concentrations of chitosan oligosaccharide did not affect the occurrence of micronuclei in bone marrow cells (P>0.05). The chromosomal aberration test was performed by microscopic examination $({\times}1,000,\;stained\;using\;a\;4\%\;Giemsa\;solution)$ after administering the same concentration of chitosan oligosaccharide to mice, in $F_1,\;F_2,\;F_3$ generations and parents. The frequency of chromosomal aberrations was defined as [Ydr=(D+R)/total number of counted lymphocytes]. Similar to the micronuclei test, there was no difference between the untreated and treated groups. These results showed that the intake periods and concentrations of chitosan oligosaccharide did not affect chromosomal aberrations in bone marrow cells (P>0.05). To investigate the effect of chitosan oligosaccharide on mercury-induced chromosome aberration, mice in each condition were supplied with $^{203}HgCl_2$ and chitosan oligosaccharide ad libitum. Chitosan oligosaccharide significantly inhibited $^{203}HgCl_2-induced$ chromosome aberration in mice. Based on the results of this study, it may be concluded that the chitosan oligosaccharide is a nontoxic material that could be used as a suppressor of heavy metal-induced genotoxicity.

A Study on the Quality Characteristics of Soybean Dasik by Addition of Chitosan-Oligosaccharide (키토산 올리고당 첨가량에 따른 콩다식의 품질 특성)

  • Along with development of modem medicine, the average life span has been increased but diseases of lifestyle such as cancer, obesity atheroscerosis, cardiac disorder become social issues. This study was designed to examine the availability of the functional food by adding chitosan-oligosaccharide to Korean traditional food, Soybean Dasik. For the study, 0, 30, 50 and $70\%$ of chitosan-oligosaccharide power that contains $23\%$ of chitosan-oligosaccharide were added to soybean Dasik in proportion to the total powder weight These were compared with the Soybean Dasik itself in relation to the physicochemical characteristics, sensory evaluation, and mechanical characteristics. The results of the research are as follows. Physicochemical characteristics showed that chitosan-oligosaccharide soybean Dasik had less ash, K, and Mn. As for the result of sensory evaluation, Dasik added $0\%$ chitosan-oligosaccharide was evaluated very good in its color. Sweetness and softness was good on $0\%\;30\%\;and\;50\%$ ones, while moistness and overall quality on $30\%\;and\;50\%$ ones. As the test results of mechanical characteristics, the chitosan-oligosaccharide soybean Dasik was good for hardness, cohesiveness, gumminess, and brittleness, especially on $0\%\;30\%\;and\;50\%$ ones. Springiness was good in all groups. In color, L value of chitosan-oligosaccharide soybean Dasik decreased but a and b values increased as the substitution percentage increased Therefore, the right amount of addition fit for the production properties was $30-50\%$ chitosan-oligosaccharide of total powder weight.

  • Tyrosinase (monophenol. 3.4-${\beta}$-dihydroxyphenylalanin oxygen oxidoreductase. EC 1.14.18.1 J. which plays a pivotal role in melanogenesis. It is single chain glycoprotein catalyzing the hydroxylation of tyrosine to ${\beta$\mid$$-3.4-dihydroxyphenylalanin (DOPA) and the oxidation of DOPA to DOPA quinone. To investigate whitening effect of chitosan oligosaccharide. we obtained chitosan oligosaccharide [(glucosamine)2-6] by NaNO2 oxidation and measured the effect of chitosan oligosaccharide on tyrosinase activity. Chitosan oligosaccharide dose-dependently inhibited tyrosinase (2 unit) activity and inhibited by 18.8% at dose of 100${\mu}$g/ml. Vitamin C. arbutin and kojic acid that are well known to be inhibitor of melanin production dose-dependently inhibited tyrosinase (2unit) activity. These results suggest that chitosan oligosaccharide may be used as inhibitor of melanin production in melanocyte. which will be further studied.

  • The mutagenic activity of chitosan oligosaccharide and its antimutagenic effect against 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) were investigated using the Salmonella/Ames test. No mutagenic activity was found in the Salmonella typhimurium strains TA 98 and TA 100, either with or without S9 activation. In contrast, chitosan oligosaccharide showed an inhibitory effect on the mutagenic activity of the cooked food mutagen, MeIQx, in the presence of S9. The influence of chitosan oligosaccharide on the genotoxicity of MeIQx was examined using a host-mediated assay in mice. The oligosaccharide was administered for 14 consecutive days (intragastric application at doses of 0.1 or 0.5 g/kg body wt) to mice. S. typhimurium TA 98 was given intravenously before an oral dose of MeIQx (4.5 mg/kg body wt.). The number of $his^+$ revertants were determined from the Ever of mice. The intragastric application of oligosaccharide led to a 47% reduction in the number of mutants induced by MeIQx (p<0.05). These results suggested that chitosan oligosaccharide had antimutagenic properties against MeIQx in vitro and in vivo.

Antioxidative Effects of Water-Soluble Chitinous Compounds on Oxidation of Low Density Lipoprotein in Macrophages (대식세포에서 지단백 산화에 대한 수용성 Chitinous Compounds의 항산화 효과에 대한 연구)

  • It has been proposed that oxidative modification of LDL (oxLDL) plays a significant role in the pathogenicity of atherogenesis. We tested the hypothesis that chitin and chitosan may function as antioxidants with respect to 0.1 mg cholesterol/ml LDL incubated with 5 $\mu$ M Cu$^2$$^{+}$alone or in the P338Dl mouse macrophage system using L-ascorbic acid as a standard classical antioxidant. The degree of oxLDL formation was ascertained by the relative electrophoretic mobility (rEM) in the combination of thiobarbituric acid reactive substances (TBARS) levels, and the cytotoxicity of oxLDL was detected by macrophage viability. The oxLDL uptake and foam cell formation of macrophages were measured by Oil Red O staining. Incubation with Cu$^2$$^{+}$and macrophages increased rEM of LDL and stimulated TBARS formation. Culture of macrophages with LDL in the presence 5 $\mu$ M Cu$^2$$^{+}$induced macrophage death. In cell-free system 200 $\mu$g/ml water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation. Water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation near-completely relative to L-ascorbic acid, whereas water-soluble chitin and chitin-oligosaccharide had no measurable antioxidant effect. In macrophage system water-soluble chitosan and chitosan-oligosaccharide blocked oxidation of LDL with a significant increase in cell viability, and decreased TBARS in medium. As for the inhibitory effect on macrophage foam cell formation, chitosan and its oligosaccharide, but not watersoluble chitin, revealed the effectiveness. The endothelial expression of lectin-like oxLDL receptor-1 (LOX-1) was tested by Western blot analysis, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide blocked LOX-1 expression. These results indicate that water-soluble chitosan and its oligosaccharide showed the inhibitory effect on Cu$^2$$^{+}$-induced LDL oxidation of macrophages, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide had blocking effect on oxLDL receptor expression in the human umbilical vein endothelial system. Thus, water-soluble chitosan and its oligosaccharides possess anti-atherogenic potentials possibly through the inhibition of macrophage LDL oxidation or endothelial oxLDL receptor expression depending on chemical types.l types.

  • To investigate the effect of chitosn oligosaccharide on skin care, we measured tyrosinase activity and melanin production in B16 melanoma cells, and elastase and hyaluronidase activity. Chitosan oligosaccharide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-depednetly increased mealnin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. (omitted)

Effects of Chitosan Oligosaccharide Supplementation on Blood Glucose, Lipid Components and Enzyme Activities in Hyperglycemic Rats (키토산 올리고당이 당뇨성 흰쥐의 혈당과 혈중 지질성분 및 효소활성에 미치는 영향)

  • The principal objective of this study was to assess the effects of chitosan oligosaccharide supplementation on the improvement of blood glucose, lipid components and enzyme activities in the serum of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglycerides(TG) and phospholipids(PL) in serum were remarkably higher in the hyperglycemic group(group BSW) and STZ(I.P.)+chitosan oligosaccharide supplementation group(group ECW) than those in the control group(group BW, basal diet+water). However the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, tree cholesterol, cholesteryl ester ratio, TG and PL in serum were lower in the ECW group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ECW group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ECW group than in the hyperglycemic BSW group. The results shown above suggested that chitosan oligosaccharide supplementation effectively improves blood glucose, lipid composition and enzyme activities in the sera of STZ-induced hyperglycemic rats.

  • Chitosan oligosaccharide with an average degree of polymerization 2-3 was prepared by degradation reaction using sodium nitrite, and the resulting aldehyde group was reduced to hydroxy group. N-palmitoyl, O-palmitoyl chitosan oligosaccha was obtained from the reaction between palmitoyl chloride and oligosaccharide under DMAP catalyst. Alkaline hydrolysis was carried out to remove the O-palmitoyl group. After dispersion and ultrasonication in aqueous solution, N-palmitoyl chitosan oligosaccharide gives vesicle structure. Its structure and size was analyzed by TEM.

  • Chitosan oligosaccharide, chitosan polysaccharide and monolaurin were used to impart the antimicrobial properties onto paper for food products . Staphylococcus aureus (ATCC 25923) was used as test microorganism and shake flask test was carried out in order to examine the antimicrobial activity of each sample. Chitosan oligosaccaride showed the greatest antimicrobial effect among them and gave the promising result even at the concentration of 0.005%. Its antimicrobial power was enhanced when it is used with nomolaurin together. Chitosan oligosaccharide improved the tensile strength of antimicrobial paper.
