Chitosan film has potential applications in agriculture, food, and pharmacy. However, films made only from chitosan lack gas barrier and have poor mechanical properties. For enhanced gas barrier and mechanical properties, chitosan/clay nanocomposites have been prepared with montmorillonite (MMT) which is a layered structure of clays and chitosan. The cationic biopolymer, chitosan is intercalated into $Na^+-montmorillonite$ through cationic exchange and hydrogen bonding process. Diluted acetic acid is used as solvent f3r dissolving and dispersing chitosan. Chitosan was intercalated or exfoliated in MMT and it was confirmed by X-ray diffraction method. D-spacing of the characteristic peak from MMT plate in chitosan/clay nanocomposites was moved and diminished. The thermal stability and the mechanical properties of the nanocomposites are measured by TGA and Universal Testing Machine. Gas permeability through the chitosan/clay nanocomposites films decreased due to increased tortuosity made by intercalation of clay in chitosan.
Porous chitosan and chitin membranes were prepared by using silica particles as porogen. Membrane preparation was achieved via the following three steps: (1) chitosan film formation by casting an chitosan solution containing silica particles, (2) preparation of porous chitosan membrane by dissolving the silica particles by immersing the film into an alkaline solution and (3) preparation of porous chitin membrane by acetylation of chitosan membrane with acetic anhydride. The optimum preparation conditions which could provide a chitosan and chitin membranes with good mechanical strength and adequate pure water flux were determined. To allow protein affinity, a reactive dye (Cibacron Blue 3GA) was immobilized on porous chitosan membrane. Binding capacities of affinity chitosan and chitin membranes for protein and enzyme were determined by the batch adsorption experiments of BSA protein and lysozyme enzyme. The maximum binding capacity of affinity chitosan membrane for BSA protein is about 22 mg/mL, and that of affinity chitin membrane for lysozyme enzyme is about 26 mg/mL. Those binding capacities are about $several{\sim}several$ tens times larger than those of chitosan and chitin-based hydrogel beads. Those results suggest that the porous chitosan and chitin membranes are suitable in affinity filtration chromatography for large scale separation of proteins.
Applications of chitosan are related to molecular weight and degree of deacetylation(DOD) of chitosan completely. The molecular weight and DOD were greatly affected by the concentration of solution time and temperature. The degree of demineralization was not significantly different at $50^{\circ}C\;and\;70^{\circ}C$ after 30 minutes. Deproteinization decreased as process time increased. The nitrogen content was reached to 6.92% after 90 minute at $80^{\circ}C$, which is similar to theoretical nitrogen content of chitin. The DOD was 82.84% after 2 hours reaction and increased as the reaction time increased in the process. Viscosity and molecular weight are increased as recycling number of concentrated NaOH solution increased. Chemical, biological and physical properties of chitosan depend on the DOD and molecular size of the molecule. Tensile strength of the films from acetic acid solutions was between $28.9{\sim}33.6$ MPa and was generally higher than that of the films from lactic acid. Elongation of the films from lactic acid was between $97.0{\sim}109.7%$ and was generally higher than that of the films from the acetic acid. Water vapor permeability of the films prepared from lcetic acid solutions was between $1.9{\sim}2.3ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and was generally higher than that of the films from the acetic acid.

The aim of this study was to evaluate the antibacterial effect on Enterococcus Faecalis and physical properties of chitosan added calcium hydroxide canal filling material. Low, medium, high molecular weights of chitosan powder were mixed with calcium hydroxide canal filling material. Also, for each molecular weight group, 1.0, 2.0, 5.0 wt% of chitosan powder were added. An overnight culture of E. faecalis was adjusted to 1 × 106 CFU/ml. For test of antibacterial effect, three different molecular weights of 2.0 wt% chitosan and three different concentrations of high molecular weight chitosan were mixed with calcium hydroxide canal filling material. The absorbance of plates was analyzed using spectrophotometer at 570 nm with a reference wavelength of 600 nm. Physical properties such as flow, film thickness and radiopacity were examined according to ISO 6876 : 2012. All molecular weight type of chitosan containing material showed inhibitory effect against E. faecalis growth compared to non-chitosan added calcium hydroxide canal filling material group (p < 0.05). High molecular weight chitosan containing material showed the most antibacterial effect. Also, the antibacterial effect decreased as the incorporated amount of chitosan decreased (p < 0.05). Every molecular weight group of material containing chitosan had a tendency for reduced flow and radiopacity, increased film thickness according to amount of chitosan. Low molecular weight of 1.0 wt% chitosan addition did not show any significant difference of physical properties compared to conventional calcium hydroxide canal filling material. In conclusion, for reinforcement of antibacterial effect against E. faecalis and for favorable physical properties, 2.0 wt% of chitosan adding is recommended. Considering its antibacterial effect of chitosan, further studies are required for clinical application of chitosan in endodontics and pediatric dentistry.

A preparative method has been proposed for obtaining chitosan products which have a desired degree of deacetylation (DD, %) without much decrease in molecular weight. Deacetylation was prepared by two methods. The method I which was proposed by Domard etc. was that DMSO/thiophenol was used repeatedly only during the deacetylation, the method II was that DMSO/thiophenol was used after the alkali treatment. Then, effective deacetylation was performed by the method ll. To determine the degree of deacetylation, potentiometric titration method was adopted. According to the increase of DD, the viscosity average moleclar weght $(\overline{M\upsilon}$) and the DP of the chitosan products were decreased. The intensity of FT-lR peak appeared at $1655cm^{-1}, which was characteristic peak of amide I (Nacetyl secondary amide group), was gradually decreased with deacetylation. X-ray analysis showed that the lattice-spacing in the chitosan film increased with the degree of deacetylation, but the difference of the transparency of the chitosan products was not diverged significantly.
Chitosan(CS) and cellulose acetate(CA) composite films were prepared using formic acid as a cosolvent by casting, solvent evaporating and neutralization method. This study examines if the blending method, which uses formic acid as a cosolvent is efficient in improving the mechanical properties of CS film, especially wet strength and elongation. Formic acid is an effective cosolvent for the blend of CS and CA. Under wet condition, tensile strength and elongation of the composite films were obviously higher than those of the films made from pure CS. FTIR, DSC, and X-ray diffraction showed that the composite films exhibit a high level of compatibility and that strong interaction between the CS and CA was caused by intermolecular hydrogen bonding. The affinity series of composite film to transition metal ions are Cu(II) > Cd(II) > Cr(III). The adsorption of Cu((II) ion was shown to be highly pH sensitive.
The objective of present study was to evaluate the effects of the application of chitosan and chitosan/whey protein on the chemical, microbial and organoleptic properties of Göbek Kashar cheese during ripening time (on 3rd, 30th, 60th and 90th d). Difference in microbiological and chemical changes between samples was found to be significant (p<0.05) during ripening period. Cheese samples with edible coating had statistically lower mould counts compared to the uncoated samples. Furthermore the highest and lowest mould counts were determined in control (4.20 Log CFU/g) and other samples (<1 Log CFU/g) at 60th and 90th d of storage. All samples exhibited higher levels of water soluble nitrogen and ripening index at the end of storage process. At the end of 90 day storage period, no signicant dierences in salt and fat values were observed among the cheeses studied. The edible coatings had a beneficial effect on the sensory quality of cheese samples. In the result of sensory analysis, while cheese C and the chitosan coated cheese samples were more preferred by the panellists, the chitosan/whey protein film-coated cheese samples received the lowest scores. This study shows coating suggests could be used to improve the quality of cheese during ripening time.
Chitosan films were prepared using four types of chitosans with different molecular weight and the effect of molecular weight of chitosan on selected film properties such as color, water vapor permeability (WVP), water solubility (WS), tensile strength (TS), and elongation at break (E) was investigated. Generally, the total color difference (${\Delta}E$) and WS of the films decreased, while TS and E of the films increased as molecular weight of chitosan increased. WVP of the films did not show any significant relationship with molecular weight of chitosan.

We have developed a rigorous heat treatment method to improve the biocompatibility of chitosan as a tissue-engineered scaffold. The chitosan scaffold was prepared by the controlled freezing and lyophilizing method using dilute acetic acid and then it was heat-treated at 110$^{\circ}C$ in vacuo for 1-3 days. To explore changes in the physicochemical properties of the heat-treated scaffold, we analyzed the degree of deacetylation by colloid titration with poly(vinyl potassium sulfate) and the structural changes were analyzed by scanning electron microscopy, Fourier transform infrared (FT-IR) spectroscopy, wide-angle X-ray diffractometry (WAXD), and lysozyme susceptibility. The degree of deacetylation of chitosan scaffolds decreased significantly from 85 to 30% as the heat treatment time increased. FT-IR spectroscopic and WAXD data indicated the formation of amide bonds between the amino groups of chitosan and acetic acids carbonyl group, and of interchain hydrogen bonding between the carbonyl groups in the C-6 residues of chitosan and the N-acetyl groups. Our rigorous heat treatment method causes the scaffold to become more susceptible to lysozyme treatment. We performed further examinations of the changes in the biocompatibility of the chitosan scaffold after rigorous heat treatment by measuring the initial cell binding capacity and cell growth rate. Human dermal fibroblasts (HDFs) adhere and spread more effectively to the heat-treated chitosan than to the untreated sample. When the cell growth of the HDFs on the film or the scaffold was analyzed by an MTT assay, we found that rigorous heat treatment stimulated cell growth by 1.5∼1.95-fold relative to that of the untreated chitosan. We conclude that the rigorous dry heat treatment process increases the biocompatibility of the chitosan scaffold by decreasing the degree of deacetylation and by increasing cell attachment and growth.
Poly(vinyl alcohol)(PVA)/chitosan blend films with non-toxicity, biodegradability, and biocom-patibility were prepared by solution casting. Variation of the physicochemical properties of the blend films was investigated through to several analysis methods. Examination of antibacterial properties revealed that bacterio-static ratios of all blend samples containing chitosan more than 10 wt% were greater than 99.9%. Moisture regain was increased with increasing chitosan content but the degree of swelling was decreased. Up to chitosan content 15 wt%t, the melting and crystallization temperature of blend films was increased with chitosan content. The blends containing chitosan content 10 and 15 wt% gave melting temperature 229 and 228$^{\circ}C$, respectively. However, the melting temperature was decreased if chitosan content exceeded 20 wt%. The mechanical properties of the blend films were increased with increasing chitosan content in both dry and wet states. The blend film including 15 wt% chitosan exhibited unusually high tensile strength.
