Evaluation of the quality characteristics of nitrogen gas-stunned chicken meat and small intestine

  • This study aimed to confirm the applicability of the new nitrogen (N2) gas stunning method in the broiler slaughtering process by comparing the meat and small intestine quality following different stunning methods (electrical, carbon dioxide (CO2), N2, and halal). Four treatments were compared: (i) electrical stunning (Elec), (ii) 80% CO2 gas stunning (CO2-gas), (iii) 98% N2 gas stunning (N2-gas), and (iv) the non-stunning method (halal). N2 gas stunning (98%) and the halal method were conducted at the pilot plant abattoir of the national institute of animal science, Korea, and electrical and 80% CO2 stunning were performed on the nearest commercial slaughter house. Meat pH24h, color (lightness, redness and yellowness), proximate composition, water holding capacity (WHC), cooking loss, and Warner-Bratzler shear force (WBSF) were measured, and in the small intestine, pH24h, color, thickness, and WBSF were measured. The Elec treatment showed high lightness, yellowness, and low redness in both meat and the small intestine, indicated by a pale color; the CO2-gas treatment showed high redness, low lightness, and low yellowness, and the coloration of meat from the N2-gas treatment was intermediate between Elec and CO2-gas. For other quality traits, the N2-gas showed good results and was between Elec and CO2-gas. Additionally, severe stress (low pH in both meats), low WHC in meat, and cracked small intestine with numerous apertures were observed in the CO2-gas, and pale colored hemorrhagic breast meat was found in the Elec. Therefore, in view of animal welfare and quality traits of meat and the small intestine, 98% N2 gas can be considered in broiler stunning.

Comparison of Carcass Characteristics, Meat Quality, and Blood Parameters of Slow and Fast Grown Female Broiler Chickens Raised in Organic or Conventional Production System

  • The objective of the study was to compare the carcass characteristics, meat quality, and blood parameters of slow and fast grown female broiler chickens fed in organic or conventional production system. The two genotypes tested were medium slow-growing chickens (SG, Hubbard Red JA) and commercial fast-growing chickens (FG, Ross 308). Both genotypes (each represented by 400 chickens) were divided into two sub-groups fed either organic (O) or conventional (C) systems. Chickens of each genotype and system were raised in a semi environmentally controlled poultry house until 21 d of age and were assigned to 5 pens of 40 chickens each. Then, O system chickens were transferred into an open-side poultry house with an outdoor run. At 81 d of age, 10 female chickens from each genotype and from each production system (n = 40) were randomly chosen to provide material for analysis, and were weighed and brought to the slaughterhouse to assess carcass characteristics and meat quality. The blood parameters were determined by using 5 female chickens from each genotype and from each production system (n = 20). FG had the higher live weight, along with carcass, breast, and thigh-drumstick weights compared to SG (p<0.05). FG had the higher breast yield, whereas SG had the higher thigh-drumstick yield (p<0.05). The O system resulted in a higher amount of abdominal fat (p<0.05). In addition, the O system values were higher for dry matter, crude ash, crude protein, and $pH^{15}$ values in breast meat, and for crude ash, crude protein, and $pH^{15}$ values in drumstick meat (p<0.05). In addition, total saturated fatty acids, total mono-unsaturated fatty acids, and total omega 3 were significantly higher in the O system than in the C system. Thus, the O system showed a positive advantage compared to the C system regarding female chicken meat quality, primarily within the ash, protein, and total omega 3 fatty acid profiles. In conclusion, the present study indicated that the main factor affecting the carcass characteristics of female chickens was genotype, whereas the organic system contributed to enhanced meat quality. These findings provide a better understanding of the relative roles of genotype and production systems in female broiler characteristics, and might aid producers in designing their facilities to optimize yield and quality while maintaining acceptable animal welfare standards.

  • The aim of this experiment was to compare the characterization of fatty acid digestion of Beijing Fatty (BF) and Arbor Acres (AA) chickens. One-day-old male AA and BF chickens were raised in the same house, and fed with the same diet. We first evaluated utilization of dietary fatty acids in chickens by the total collection procedure, and chickens were then killed to compare the abundance of intestinal mRNA expression of liver-fatty acid binding protein (L-FABP) and intestinal-fatty acid binding protein (I-FABP) by Real-time PCR, and also the pH of intestinal mucosa at 3 and 6 weeks of age. Another group of chickens were sampled at 6 weeks of age to compare the total bile acid concentration in serum, and lipase activity in contents of the small intestine. Results showed that compared to AA chickens, BF chickens had higher lipase activity in the content of the small intestine (p<0.05), greater total bile acid content in portal vein blood (p<0.05) at 6 weeks of age, lower intestinal mucosal pH at both 3 weeks (p<0.05) and 6 weeks (p<0.05) of age, and higher abundance of liver-fatty acid binding protein (L-FABP) mRNA expression in intestine tissues at 6 weeks of age (p<0.05), and higher digestibility of fatty acids at both 3 and 6 weeks (p<0.05) of age. There was no difference in I-FABP mRNA expression between AA and BF chickens at either age. Thus, BF chickens had greater fatty acids utilization than AA chickens that was associated with L-FABP, lipase activity, bile acid content and intestinal mucosal pH.

Antibacterial Activities of Mushroom Liquid Culture Extracts Against Livestock Disease-Causing Bacteria and Antibiotic Resistant Bacteria (가축질병 세균 및 항생제 내성 세균에 대한 버섯 균사체 배양 추출물의 항균 활성)

  • The ethyl acetate extracts from the liquid cultures of Coriolus versicolor, Phellinus linteus, and Hericium erinaceus showed significant antibacterial activities against Escherichia coli K88, E. coli K99, E. coli 987P, and Salmonella typhimurium 14058 causing bacterial diarrhea in Korean house pigs and chicken. Of these extracts, Coriolus versicolor extract showed the highest antibacterial activity. In addition, these extracts also showed significant growth inhibition against Staphylococcus aureus CARM3230 and E. coli CARM1381 which are known as kanamycin and ampicillin-resistant strains. These results showed that the mushroom extracts could be developed as a livestock feed additives that can replace commercial antibiotics, and also could be good resources for the development of a new antibacterial agent.

Application of ATP Bioluminescence Method for Measurement of Microbial Contamination in Raw Meat, Meat and Dairy Processing Line (식육 및 육가공 . 유가공 생산라인의 환경미생물오염도 측정을 위한 ATP 방법의 이용)

  • This study was conducted to investigate the application of ATP bioluminescence to measure the degree of microbial contamination from raw meat, meat processing and milk processing lines. Samples collected from slaughter house, meat and milk processing plants were tested for estimation of bacterial number by using ATP bioluminescence and conventional method. The former result was transffered to R-mATP value(log RLU/ml), and the latter transffered to CFU(log/ml). Correlation coefficient(r) between aerobic counts(CFU, log/ml) and R-mATP(log RLU/ml) value was 0.93(n=408). R-mATP of aerobic counts from beef, pork, chicken was 0.93(n=220), and that was 0.93(n=187) between meat processing and dairy processing plants. In addition, Correlation coefficient(r) between aerobic counts and R-mATP was 0.87(n=252) under 1$\times$10${^5}$/ml of bacterial count and 0.74(n=152) over 10${^5}$ respectively.

Outbreak of Fowl Cholera in native chickens (토종닭에서 가금콜레라(Fowl Cholera) 발생)

  • Fowl cholera(FC), which has been classified as reportable animal disease in Korea, occurred in the backyard farm located in Chungju, Chungbuk province. The farm was not ordinary domestic poultry farm but backyard farm with main cultivation of fruits. The farmer reported unusual mortality of chickens which were grazed in his own fruit farm without any poultry house. There was a lots of opportunities to contact with wild animals including wild birds freely. Clinical signs observed in the chickens were torticolis, mucous excretion from mouth and greenish diarrhea. On the necropsy we found multifocal necrosis on the surface of liver, hemorrhages in the epicardium and ovary and mucopurulent exudates in the joint. The causative agents was isolated from the liver of chickens collected from the farms and identified as Pasteurella multocida using biochemical tests of bacteria. Also, the isolated bacteria were inoculated into specific pathogen free chickens to evaluate the pathogenicity. High mortality and similar pathological lesions compared to those of chickens died in the farm were observed. We report the first case of fowl cholera of the chickens in Korea and further studies are needed to evaluate the serotype, genotype and pathogenicity of isolated Pasteurella multocida.

  • Methicillin-resistant coagulase-negative staphylococci (MRCNS) were isolated from the respiratory sites of chickens in 4 farms and slaughter house located in Chungnam provinces. Isolation of coagulase-negative staphylococci (CNS) was positive for 61 (26.6%) of the 229 chickens tested, and isolation of MRCNS was positive for 17 (27.9%) of the isolated CNS. A total of 17 MRCNS isolates were selected and subjected to identification. Of the 17 MRCNS isolates selected, 6 were identified as Staphylococcus cohnii, 2 as S. saprophyticus, 3 as S. simulans, 3 as S. lentus, 2 as S. carnosus, and 1 as S. xylosus. The MRCNS isolates were resistant to many beta-lactam antibiotics, and some isolates were also resistant to macrolide and aminoglycoside antibiotics. The mecA gene was detected in some isolates of each MRCNS strains. The mecA-positive isolates were classified into five staphylococcal cassette chromosome mec (SCCmec). SCCmec types I to IV were detected in isolates from chickens.

Effects of Production Performance, Immunity and Egg Quality by Raising on Exercise Yard in Laying Hens (산란계 운동장 사육이 생산성, 면역성 및 계란의 품질에 미치는 영향)

  • The present study examined effects of production performance, immune activity and egg quality by raising on exercise yard in laying Hens, the results of which could be used as baseline data to enhance animal welfare and the safety of livestock products. A total of 90 Hy-line-Brown laying hens of 13 weeks old were used in the experiment for 38 weeks. The cage group (Cage group) was raised in a cage, where an area of $0.084m^2$ was assigned to two hens, while hens in the exercise yard $1.1m^2$ group ($1.1m^2$ group) was assigned to a combination of a chicken house ($0.11m^2$) and a exercise yard ($1.1m^2$) per a hen. Hens in the exercise yard $2.2m^2$ group ($2.2m^2$ group) was assigned to a combination of a chicken house ($0.11m^2$) and a exercise yard ($2.2m^2$) per a hen. Treatment was replicated 3 times with ten birds per replication. Ten birds were arranged according to randomized block design. While initial egg production rate was significantly higher in the Cage group, $1.1m^2$ group exhibited a slightly higher rate in the mid- and late-stage of the experiment, although the difference was not statistically significant. Exercise yard treatment groups exhibited a higher feed intake rate than the Cage group up until the hens were 39 weeks old (P<0.05), but the difference dissipated from that age on. The age at first egg in the exercise yard treatment groups was 16 days later than that for the Cage group (P<0.05), although differences in the quality of the eggs were not observed. The results of immune activity test showed that IgA in the exercise yard treatment groups was significantly higher than that in the Cage group (P<0.05). IgG, IgM, and corticosterone were also higher in the exercise yard treatment groups. The soil in exercise yard increased of organic matter and decreased of cation exchange capacity (CEC) in raised hens. In sum, raising hens in a exercise yard raise style decreased the rate of initial egg production, which was followed by a slight increase during the latter part of the experiment. The exercise yard raise hens' immune activity was heightened.

Prevalence of Salmonella Species Isolated from Old Hen Delivery Trucks in Korea and Application of Disinfectant for the Reduction of Salmonella Contamination (국내 노계 운반 전용 차량에서의 살모넬라 오염율 및 차량소독에 따른 오염도 감소 효과)

  • Salmonella is closely related with human health of modern society which has concern increased in livestock goods consumption as well as give economic damage throughout the chicken industry such as farm, hatchery, slaughter house and processing plant. From 2007 to 2011, this study investigated Salmonella prevalence from 200 old hen delivery trucks which deliver old egg-laying hens and broiler breeders in Korea. The prevalence of Salmonella species was 38.0% in old hen delivery truck. Serogroup C1 was the most frequently detected serogroup of Salmonella, followed by the serogroups D1, C2 and B. A total of 25 serotypes were identified and Salmonella infantis was the most frequently isolated serotype. In addition, we applied disinfectant to old hen delivery truck for the reduction of Salmonella contamination. The disinfectant consists of formaldehyde, glutaladehyde and quaternary ammonium compound was applied to the trucks. Salmonella isolation rate was significantly decreased after disinfection from 38.0% to 7.5%. Disinfectant could not effectively reduce Salmonella contamination at a dilution of 1:200 which is recommended by manufacturer, but Salmonella isolation rate was significantly decreased at a dilution of 1:50. Since old hen delivery truck could be a potential vector to carry Salmonella into farms and abattoirs, chicken delivery truck should be disinfected thoroughly and sufficiently to control contamination of farms and abattoirs.
