Production of Specific Egg Yolk Antibodies in Chicken against Recombinant Fowl Adenovirus Fiber 2 Protein (재조합 가금 아데노바이러스 Fiber 2 단백질을 이용한 특이 난황 항체 생산)

  • Fowl adenovirus (FAV) is an important cause of several diseases, which result in considerable economic losses to the poultry farm. An outer capsid protein of FAV, fiber 2 is essential for virus growth, assembly or spread. This study was performed to produce about 22 kDa of recombinant fiber 2 protein and to immunize in laying hens to acquire the specific IgY antibody against the recombinant fiber 2. Laying hens were immunized with the recombinant fiber 2 intramuscularly in the breast muscle by injection 4 times at intervals of three weeks. At 12 weeks, serum- and egg yolk-antibody titers of hens against fiber 2 were increased up to 430,000 and 414,000, respectively. The recombinant fiber 2 could be recognized be the anti-His monoclonal antibody. Anti-fiber 2-IgY antibody could recognize the fiber 2 specifically in western blot analysis. These results suggested that the recombinant fiber 2 antigen could be used as an immunogen to elicit IgY antibody against fiber 2 and the anti-fiber 2-IgY could neutralize fowl adenovirus fiber 2 effectively.

Pasteurella anatipestifer infection in ducklings in Korea (오리의 Pasteurella anatipestifer 감염증 발생)

  • This study was conducted to investigate the cause of a new duck disease occured in southern part of Korea. A meat type duck farm located in Kangjin, Chonnam Province had experienced outbreaks of septicemic disease at around 20 days of age in nearly every batch of ducklings from early spring to early summer in 1989. Main symptoms of the birds were eye and nasal discharge, depression, inappetence, diarrhea and nervous signs such as tremor and ataxia. Some birds died suddenly without any signs. Mortality reached from 20% to 80% in severe cases. The autopsy findings of the affected ducklings revealed consistantly severe airsacculitis, fibropericarditis, perihepatitis and occasionaly enteritis and distended ureter with urate deposit. A rod shaped gram-negative bacterium was isolated purely from brain and liver of the diseased ducks by culturing the specimens on blood agar for 48 hours in candle jar. The isolate neither produced hemolysis nor grew on MacConKey Agar. It formed colony relatively slowly being recognizable at least 36 hours after culturing, reaching colony size of about 1mm in diameter at 48 hours culture. The colony looked iridescent under oblique light and had muddy odor. The isolate did not ferment carbohydrates tested but produced gelatinase, hippuricase and oxidase which were considered as characteristics of P anatipestifer. The isolate induced similar signs and lesions when infected experimentally into ducks of 3 to 38 days age via intraperitoneum or intratrachea. However it did not produce any clinical signs wen inoculated via intranasal route. It produced only mild signs in chicken just injected with a very large dose. The bacteria did not produce any signas or lesions in mice. It was concluded through biochemical and physiological tests and animal inoculation tests that the new disease was caused by P anatipestifer.

  • Sawdust, peat, and sawdust+peat were used as bulking agent in the compost production process using three different origin of manure; cow, pig, and chicken. The organic content and individual N, P, K content of the final manure compost were higher when peat or peat+sawdust were used to control the moisture. The carbon to nitrogen ratio and moisture content were low when peat or peat+sawdust were employed. In the case of cow and pig manure compost produced with peat or peat+sawdust, beneficial microorganism content was also higher than that of the manure samples produced with sawdust only. These results indicate that peat can be a useful component in the production of high quality manure compost.

  • The aim of this study was to apply the new agricultural technique developed for three years($'96{\sim}'98$) by demonstration at an model farm of environmental agriculture located in Bangsan-ri, Dagang-myun, Namwon-city. Rice growth of environmental farming plot, which supplied with oil cake and compost fermented with chicken manure was reduced compared to that of conventional farming plot, which supplied with chemical fertilizers. Weed control by using of Ampullarius insularus in the rice paddy field was effective compared to that of control, which treated with chemical herbicide twice. The yield of rice was decreased of 35% in both environmental and eclectic farming polt compared to that of conventional farming plot. The water quality of environmental not and eclectic farming plot were good compared to conventional farming plot. Density of water flea was high in environmental and eclectic farming plot compared to that of the conventional farming plot.

Angiotensin I-Converting Enzyme Inhibitor Activity on Egg Albumen Fermentation

  • Lactobacillus plantarum is used for fermentation of fish products, meat and milk. However, the utilization of these bacteria in egg processing has not been done. This study was designed to evaluate the potential of fermented egg albumen as a functional food that is rich in angiotensin I-converting enzyme inhibitors activity (ACE-inhibitor activity) and is antihypertensive. A completely randomized design was used in this study with six durations of fermentation (6, 12, 18, 24, 30, and 36 h) as treatments. Six hundred eggs obtained from the same chicken farm were used in the experiment as sources of egg albumen. Bacteria L. plantarum FNCC 0027 used in the fermentation was isolated from cow's milk. The parameters measured were the total bacteria, dissolved protein, pH, total acid and the activity of ACE-inhibitors. The results showed that there were significant effects of fermentation time on the parameters tested. Total bacteria increased significantly during fermentation for 6, 12, 18, and 24 h and then decreased with the increasing time of fermentation to 30 and 36 h. Soluble protein increased significantly during fermentation to 18 h and then subsequently decreased during of fermentation to 24, 30, and 36 h. The pH value decreased markedly during fermentation. The activities of ACE-inhibitor in fermented egg albumen increased during fermentation to 18 h and then decreased with the increasing of the duration of fermentation to 24, 30, and 36 h. The egg albumen which was fermented for 18 h resulted in a functional food that was rich in ACE-inhibitor activity.

Study on Utilization of Heterosis in Layer Chicken (산란계종의 잡잡강세리용에 관한 연구)

  • This study was carried out to estimate combining abilitie or economic traits in layer chickens. The data used in this study were the record of 10 single crosses produced by half diallel cross of 5 lines of Single Comb White Leghorns, such as A, B, C, K and S lines. Total 720 progenies of the crosses were reared at the Poultry Breeding Farm, College of Agriculture, Seoul National University from Feb. 1979 to August, 1980. Combining abilities were estimated by Grilling's mathematical model for the traits; age the first egg, total egg number, egg weight and body weight. The results attained from the studies were summarized as follows; In estimate of combining ability, an age at first egg of BS cross was largely due to significiantly higher general combining ability (G. C. A.) effect of B and S strains than Cand K strains in G. C. A. effect, and to specific combining ability (S. C. A) effect of B and S-trains. AB and BS crosses showed the highest egg Production. AB cross performance was result from high G.C.A. effect of A ana B strains. BS cross performance was result from high G. C. A. effect of B and high S. C. A. effect of BS cross. Specific combining ability effect in egg. weight was not statiscally significiant, but S strain showed high G. C. A. effect. A and B strains in body weight showed significantly low C. C. A. effect. From the above results, BS cross in an age at first egg, AB and BS crosses in egg Production, S strain in egg weight and AB cross in body weight were superior to other strains or crosses.

  • Bovine mastitis is costly infectious disease of dairy cattle, being responsible for significant economic losses all over the world. Also, mastitis has troubled about resistance to antibiotics. The purpose of this study was to screen a novel antimicrobial strain from various sources (raw milk and feeds (from farm of Paju, Dangjin, and Hwasung), commercial milk, Korean traditional fermented foods, and chicken feces). The isolate was screened using triple agar layer method and deferred method was used for confirmation of antimicrobial effect. Seventy six of isolates were screened using triple agar layer method. In these strain, 42 isolates were shown a broad spectrum of autimicrobial activity against mastitis pathogens. Especially, fourteen isolates were shown over 20 mm inhibition zone against S. aureous ATCC 25923. These results suggest that these novel antimicrobial strains could be used for the alternative of antibiotics.

Genotyping of avian pathogenic Escherichia coli by DNA fragment analysis for the differences in simple sequence repeats

  • Avian pathogenic E. coli (APEC) causes severe economic losses in the poultry farms, due to systemic infections leading to lethal colisepticemia. It causes a variety of diseases from air sac infection to systemic spread leading to septicemia. Secondary infection contains opportunistic infections due to immunosuppression disease. Collibacillosis causes the great problems in the poultry industry in Korea. Thus, it is necessary to identify and classify the characteristics of E. coli isolate of chicken origin to confirm the diversity of symptoms and whether they are transmitted among the farms. Fragment analysis is identify the difference in the number of Variable-Number Tandem-Repeats (VNTRs) for genotyping. VNTRs have repeating structure (Microsatellite, Short tandem repeats; STR, Simple sequence repeats; SSR) in the chromosome. This region can be used as a genetic marker because of its high mutation rate. And various lengths of the amplified DNA fragment cause the difference in the number of repetition of the DNA specific site. The number of repetition sequences indicates the separated size of fragments, so the each fragments can be distinguished by specific samples. The results of the sample show that there is no difference in six microsatellite loci (yjiD, aidB, molR_1, ftsZ, b1668, yibA). There are differences among the farms in relation of the number of repetitions of other six microsatellite loci (ycgW, yaiN, yiaB, mhpR, b0829, caiF). Four (ycgW, yiaB, b0829, caiF) of these six microsatellite loci show statistically significant differences (P<0.05). It means that the analysis using four microsatellite loci including ycgW, yiaB, b0829, and caiF can confirm among the farms. Five E. coli samples in one farm have same SSR repetition at all markers. But, there are significant differences from other farms at Four (ycgW, yiaB, b0829, caiF) microsatellite loci. These results emphasize again that the four microsatellite loci makes a difference in the amplified DNA fragments, enabling it to be used for E. coli genotyping.

Variability of laying hen behaviour depending on the breed

  • Objective: For many generations, most species of farm animals have been subjected to intense and strictly targeted selection for improvement of their performance traits. This has led to substantial changes in animal anatomy and physiology, which resulted in considerable differences between the current animal breeds and their wild ancestors. The aim of the study was to determine whether there is breed-specific variability in behaviour as well as differences in emotional reactivity and preferences of laying hens. Methods: The investigations involved 50 Green-legged Partridge, 50 Polbar, and 50 Leghorn hens. All birds were kept in the same conditions, and the behavioural tests were carried out at 30 weeks of age. We used the tonic immobility test and a modified open-field test including such objects as water, commercial feed, feed enriched with cereal grains, finely cut straw, and insect larvae, a sandpit, a mirror, and a shelter imitating a hen nest. Results: The research results demonstrate that the birds of the analysed breeds differ not only in the excitability and emotional reactivity but, importantly, also in the preferences for environment-enriching elements. Ensuring hens' well-being should therefore be based on environmental modifications that will facilitate acquisition of essential elements of chickens' behaviour. The greatest emotional reactivity was found in the Leghorn breed, which may be a result of correlated selection aimed at an increase in chicken productivity. Conclusion: The differences in the behaviour of the birds from the analysed breeds indicate that laying hens cannot be regarded as one group of animals with the same environmental requirements.

Prevalence of Salmonella Species Isolated from Old Hen Delivery Trucks in Korea and Application of Disinfectant for the Reduction of Salmonella Contamination (국내 노계 운반 전용 차량에서의 살모넬라 오염율 및 차량소독에 따른 오염도 감소 효과)

  • Salmonella is closely related with human health of modern society which has concern increased in livestock goods consumption as well as give economic damage throughout the chicken industry such as farm, hatchery, slaughter house and processing plant. From 2007 to 2011, this study investigated Salmonella prevalence from 200 old hen delivery trucks which deliver old egg-laying hens and broiler breeders in Korea. The prevalence of Salmonella species was 38.0% in old hen delivery truck. Serogroup C1 was the most frequently detected serogroup of Salmonella, followed by the serogroups D1, C2 and B. A total of 25 serotypes were identified and Salmonella infantis was the most frequently isolated serotype. In addition, we applied disinfectant to old hen delivery truck for the reduction of Salmonella contamination. The disinfectant consists of formaldehyde, glutaladehyde and quaternary ammonium compound was applied to the trucks. Salmonella isolation rate was significantly decreased after disinfection from 38.0% to 7.5%. Disinfectant could not effectively reduce Salmonella contamination at a dilution of 1:200 which is recommended by manufacturer, but Salmonella isolation rate was significantly decreased at a dilution of 1:50. Since old hen delivery truck could be a potential vector to carry Salmonella into farms and abattoirs, chicken delivery truck should be disinfected thoroughly and sufficiently to control contamination of farms and abattoirs.
