  • For the quantitative detection of algal toxin, microcystin, a chemiluminescence immunochromatographic assay method was developed. The developed system consists of four parts, chemiluminescence assay strip (nitrocellulose membrane), horse radish peroxidase labeled microcystin monoclonal antibodies, chemiluminescence substrate (luminol and hydrogen peroxide), and luminometer. The performance of the chemiluminescence immunochromatographic assay system was compared with high performance liquid chromatography (HPLC) detection. The detection limit of chemiluminescence immunochromatographic assay system is several orders of magnitude lower than with HPLC. The chemiluminescence immunochromatography and HPLC results correlated very well with the correlation coefficient ($r^2$) of 0.979.

  • We have studied the interaction of 3-nitrotyrosine with peroxynitrite using three different methods; chemiluminescence, spectrophotometry and HPLC. Peroxynitrite-induced luminol or lucigenin chemiluminescence were significantly decreased by 3-nitrotyrosine, in concentrationdependent manners. The intensity of the peroxynitrite spectrum was also markedly reduced in the presence of 3-nitrotyrosine in the spectrophometric assay. However, there was no attenuation of the 3-nitrotyrosine signal in the HPLC assay after mixing with peroxynitrite. The interaction of 3-nitrotyrosine and hypochlorous acid (HOCI) was also studied via the chemilumines-cence assay, where the HOCI-induced responses were markedly inhibited by 3-nitrotyrosine. These results suggest that caution should be taken when studying the levels or interactions of 3-nitrotyrosine.

  • Sphingolipids has been known to induce apoptosis, cell proliferation, differentiation and migration in a variety of cell types. Recently, its phosphate form was suggested that they may act both as an agonist ligand to SlPRs and a second messenger in intracellular action. Phytosphingosine(PHS) is not easily detected due to trace component of cellular lipids in mammalian and human tissues while this is a major sphingolipid in yeast and plants. We therefore developed highly sensitive and reproducible analytical method for PHS and its phosphate by oxalic acid bis(2,4,6-tri-chlorophenyl) ester(TCPO)-hydrogen peroxide(H$_2$O$_2$) chemiluminescence. • Soybean sauce fermented with soybean and wheat, has been a major condiment of Korean diets from centuries ago. Melanoidin, a brown pigment generally found in various food systems, is a final product produced in amino-carbonyl reaction during soybean sauce processing. Antioxidative activities of soybean sauce and melanoidin were investigated in vitro system using linoleic acid emulsion. Soybean sauce and glucose-lysine model melanoidin showed the stronger antioxidative effect than control by ferric thiocyanate and conjugated diene assays. In addition, DPPH radical scavenging effect of soybean sauce was higher than melanoidin, which was ascribed to soluble peptide and low molecular protein existing in soybean sauce. To ascertain antioxidative effect of dietary soybean sauce and melanoidin in vivo, the male Wister rats were fed 10% soybean sauce or 10% glucose-lysine model melanoidin with corn oil or fish oil for 5 weeks. Fatty acid compositions in liver and plasma were influenced by oil source. Therefore, EPA and DHA contents of fish oil group were higher than those of corn oil group. When the inhibitory effect of soybean sauce and melanoidin on lipid peroxidation using TBARS methods was measured, fish oil group (FC) showed higher malondialdehyde (MDA) content than corn oil group (CC). However, supplementation of soybean sauce and melanoidin to fish oil group attenuated MDA formation. In the levels of phosphatidyl choline hydroperoxide (PCOOH) in liver and plasma by CL (chemiluminescence)-HPLC method, PCOOH in FC group was significantly higher than that of CC group both in liver and plasma. Supplementation of soybean sauce to fish oil groups significantly inhibited the formation of PCOOH in plasma and liver, while melanoidin suppressed hepatic PCOOH formation. Based on these results, it can be suggested that soybean sauce possesses stronger antioxidative potential than melanoidin.

  • The antioxidant effect of capsaicin, the pungent principle of red pepper and oleoresin extracted from red pepper was investigated by measuring TBA(Thiobarbituric acid) value and hydroperoxide value using CL-HPLC(Chemiluminescence-high performance liquid chromatography) during storage at $30^{\circ}C$. The antioxidant activity of capsaicin and oleoresin was compared with erythorbate already used. The antioxigenic effect of capsaicin and oleoresin was very effective to the preservation of ground bacon belly meat. Especially, oleoresin have a remarkable effect to prevent the peroxidation of ground bacon belly meat. Capsaicinoids were known as the main additives in Korea, but the antioxidant activity of meat products has not been reported. So, we suggest that capsaicin, especially, oleoresin combined with other natural antioxigenic substances as like tocopherol may be effective to prevent the oxidation of ground bacon belly meat.

  • Total suspended particulates (TSP) in the atmosphere were collected for 2 weeks during winter in Muroran, Hokkaido, Japan, a typical iron-manufacturing city. The concentrations of polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs) in TSP were determined by high-performance liquid chromatography (HPLC) using fluorescence and chemiluminescence detectors, respectively. No relationship was observed between the atmospheric PAH and NPAH concentration, or the atmospheric PAH and TSP concentration. However, there was a tendency that the atmospheric PAH concentration was higher when the wind blew from the coke-oven plant. Furthermore, the concentration ratios of 1-nitropyrene to pyrene, which is a suitable indicator of the contribution made by automobiles and coal combustion systems to urban air particulates, were smaller in Muroran and the values were close to those observed in particulates from coal combustion systems. Therefore, these results show that the PAH and NPAH compositions for Muroran are characteristic of an iron-manufacturing city.

  • Hazardous polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs) are mainly originated from imperfect combustion of fossil fuels such as petroleum and coal. The consumptions of not only petroleum but also coal have been increasing in the East Asian countries. This review describes the result of international collaboration research concerning characteristics and major contributors of atmospheric PAHs and NPAHs in cities in Japan, Korea, China and Russia. We collected airborne particulates in ten cities in the above countries and six PAHs and eleven NPAHs were determined by HPLC methods using fluorescence and chemiluminescence detections. The total PAH concentrations were much higher in Chinese cities (Fushun, Tieling, Shenyang and Beijing) than those in other cities (Vladivostok, Busan, Kanazawa, Kitakyushu, Sapporo and Tokyo). The total NPAH concentrations were also higher in Chinese cities than those in the other cities. The [NPAH]/[corresponding PAH] ratios are much larger in diesel-engine exhaust particulates than those in coal-burning particulates. The [1-nitropyrene]/[pyrene] ratio of airborne particulates was much smaller in the four Chinese cities, suggesting that coal combustion systems such as coal heaters were the main contributors. On the other hand, the ratios were larger in Korean and Japanese cities, suggesting the large contribution of diesel-engine vehicles.

  • Crude catechin extracts were prepared using ethyl acetate from Korean Puerariae Radix (PR) and their biological effects on the alcoholic liver damage were investigated in male Sprague-Dawley rats (6 weeks old). Ethanol (5g ethanol/kg body weight/day) administration was performed for 8 weeks and after ethanol consuming rats were treated with one or two% catechin extracts of diet for 8 weeks. At the end of experimental period, lipid hydroperoxides in liver were analyzed using a chemiluminescence-high performance liquid chromatography (CL-HPLC) method. Compared with control animals, ethanol consumed rats showed lighter body weights, lower ratios of liver/body weight, higher activities of GOT and GPT, and increased lipid hydroperoxide amount in liver. With one or two% catechin extracts treatment, GOT and GPT activities returned to normal ranges. Lipid hydroperoxide contents in liver of on or two% PR treated rats lowered to 20% or 25% respectively, compared with the levels of those ethanol consumed animals without catechin extracts treatment. Therefore, we concluded that on or two% PR crude catechins treatment could be effective for alcoholic liver damage caused by lipid peroxidation.
