Minimally processed Products readily deteriorate in quality, especially color and texture, as a result of endogeneous enzyme enhanced respiration and microorganisms which lead to reduced shelf Iife. As a result, accelerated senescence and enzymatic browning OCCUI', Recent l'esearch on minimally processed fl'uits and vegetables has mainly focused on methods to inhibit browning, due to ban on use of sulfur dioxide, In order to retard or prevent these physiological changes, val'ious al ternatives, reducing agents. acidulants, chelating agents and inol'ganic sal ts have been evaluated for use on fresh cut fl'ui ts. Al though some agents were effective replacement for sulfur dioxide. consum$\textregistered$I'S demandless use of chemical on such products. Shel~ life of minimally processed products has been extended by inhibition of metabolic reactions associated with loss of quality and by inhibition of aerobic spoilage caused by wide variety of microorganisms. Appl ication of ~I.-\ packaging, including vacuum packaging, retards the rate of respiration, prevents growth of aerobic spoilage organisms, inhibits oxidation and color deterioration. Tissue softening is another major problem in minimally processed products because enzymes re 1 a ted to ce 11 wa 11 degrada t i on are not inactivated. Various treatments have been investigated for retardation of the softening of sliced products. Some studies have concentrated on the application of an active packaging system with ~I, l. packaging and calcium infi 1 tration as possible measures to retain firmness of processed products. In my opinion, one important step for production of minimally processed frui ts wi th favorabl e color of cut surface and firm texture is the selection of better cultivar. As the view, changing tendency of fresh color by apple cultivars and relationship between the tendency and PPO activity will be discussed in the seminar. In addition to the topic, research result on quality enhancement of fresh apple slices by heat shock treatment will be introduced.

Antioxidant and anti-inflammatory activities of fruiting bodies of Dyctiophora indusiata (흰망태버섯 자실체의 메탄올 및 열수추출물의 항산화 및 항염증 활성)

  • Dictyophora indusiata is an edible mushroom belongs to Family Phallaceae of Phallales, Basidiomycota. The purpose of this study was to investigate the antioxidant and anti-inflammatory activities of methanol and hot water extracts prepared from fruiting bodies of Dictyophora indusiata. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity for those extracts. To measure the anti-inflammatory activities for the extracts, nitric oxide(NO) production from lipopolysaccharide(LPS) treated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that the extracts have excellent DPPH scavenging and chelating activity on the ferrous ions compared with positive control. The nitric oxide(NO) production in LPS-stimulated RAW 264.7 macrophage cells were decreased as we increased the concentration of the mushroom extracts. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. Therefore, the experimental results suggested that methanol and hot-water extracts of Dictyophora indusiata fruiting bodies might be used for natural sources of antioxidant and anti-inflammatory agents.

A Study on the Adsorption and Recovery of Metal Ions by Amberlite XAD Resins Impregnated with Oxime Compounds (Oxime 화합물을 침윤시킨 Amberlite XAD 수지에 의한 금속이온의 흡착 및 회수에 관한 연구)

  • The adsorption behaviors of some oxime compounds well known as metal chelating agents on the Amberlite XAD resins were compared by measuring their distribution coefficients (log Kd) in various media, respectively. Among the oxime compounds, salicylaldoxime (SAO) and $\alpha-benzoinoxime(${\alpha}$-BzO)$ which showed large log Kd values were chosen. The characteristics of XAD-4 resins impregnated with SAO and ${\alpha}$-BzO have been studied to apply them for the adsorption and recovery of minute quantities of metal ions in aqueous solution. The optimum conditions for adsorption of SAO and ${\alpha}$-BzO on the resin were 30% methanol media having pH range of 1~8(for SAO) and 1~9 (for ${\alpha}$-BzO), respectively. The distribution coefficients of two oxime compounds were decreased as temperature increased. From the adsorption enthalpy data of SAO and ${\alpha}$-BzO, ranging from 4.96 to 6.66 Kcal/mol, it is suggested that their adsorption mechanism on XAD-4 resin is likely due to molecular adsorption equivalent to dipole-dipole interaction. The impregnated resins were considerably stable in the aqueous solutions of pH 5.0~10.0 and in 0.1~5M hydrochloric acid solutions. The former is the medium for adsorption of metal ions, while the latter is for recovery of the adsorbed metal ions. The adsorption mole ratio of Mn(II), Co(II), Ni(II), Zn(II) ions on SAO-XAD-4 and ${\alpha}$-BzO-XAD-4 resins were about 1 : 2 at the optimum conditions, respectively. The adsorbed metal ions were recovered completely by eluting with 3M HCl-50% methanol solution

Partial Purification and Some Properties of Carboxymethyl Cellulases from Alkalophilic Cephalosporium sp. RYM-202 (호알칼리성 Cephalosporium sp. RYM-202가 생산하는 carboxymethyl cellulase의 부분정제 및 특성)

  • An alkalophilic Cephalosporium sp. RYM-202 capable of producing cellulase components was isolated from soil. This organism grew best at an initial pH 9.0 and produced cellulase maximal at an initial pH 9.5-10.0. Three carboxymethyl cellulases(CMCases), P-I-I, P-I-II and P-II-I, were partially purified by DEAE-Sephadex A-50 ion exchange column followed by Sephadex G-150 gel filtration. The optimum pH values for activity were 7.5 for P-I-I, 8.0-9.5 for P-I-II and 7.5-10.0 for P-II-I. All CMCases were stable between pH 4.5 and 12.0. Temperature optima for activity ranged between 40 and $60^{\circ}C$ and more than 50% of the maximum activity was observed at $20^{\circ}C$ for both of P-I-I and P-II-I. The activity of CMCases was significantly stable in the presence of various laundry components, such as, surfactants, chelating agents and alkaline proteinases.

Antioxidant and Antimicrobial Activities of Fruiting Bodies of Phellinus gilvus Collected in Korea (국내에서 수집된 마른진흙버섯 자실체의 항산화 및 항균 효과)

  • This study was initiated to evaluate the antioxidant and antimicrobial activities of methanol extract (ME) and hot water extract (HWE) obtained from the fruiting bodies of medicinal mushroom, Phellinus gilvus. The free radical scavenging activity of ME from P. gilvus on 1,1-diphenyl-2-picrylhydrazyl (DPPH) were 93.65% at 2 mg/mL, which was comparable with the positive control, butylated hydroxytoluene (BHT, 96.97%) at the same concentration. The ferrous ion-chelating ability of ME and HWE was significantly higher than that of BHT at all concentration levels. The antimicrobial assay of ME was performed against six bacteria and one species of fungus. ME exhibited antibacterial activity against 5 out of 6 bacteria: Staphylococcus aureus, Streptococcus mutans, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa; whereas, ME did not show antimicrobial activity against gram-negative bacterium Vibrio vulnificus and fungal species Candida albicans. The minimum inhibitory concentration (MIC) of ME against 5 strains of bacteria, such as S. aureus, S. mutans, B. subtilis, E. coli, and P. aeruginosa, was 100, 100, 50, 100, 200 mg/mL, respectively. The results suggest that good antioxidant and microbial activities of P. gilvus fruiting bodies might be used for natural antioxidant and antimicrobial agents.

Free radical scavenging, anti-inflammatory and melannin synthesis inhibitory activities of Gloeostereum incarnatum (느릅나무버섯 자실체의 메탄올과 열수추출물의 항산화, 항염증 및 멜라닌 생성저해 활성)

  • Gloeostereum incarnatum is an edible and medicinal mushroom belongs to Family Cyphellaceae of Polyporales, Basidiomycota. The purpose of this study was to investigate the free radical scavenging, anti-inflammatory, and melanin synthesis inhibitory activities of fruiting bodies of Gloeostereum incarnatum. In the free radical scavenging activities, the mushroom extracts showed good 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and chelating activity on the ferrous ions compared with the positive control, BHT. The mushroom extract suppressed nitric oxide (NO) production in LPS-induced RAW 264.7 macrophage cells in dose dependant manners. Significant reduction of paw edema of rats were observed at 2~6 h after administration with 50 mg/kg of the methanol and hot-water extracts, which were comparable with treatment of 5 mg/kg of indomethacin, the positive control. The melanin synthesis of Melanoma B16/F10 cells treated with $100{\mu}g/mL$ of the methanol and hot water extracts decreased melanin concentration to 50% and 45% compared with the control, arbutin. Therefore, the experimental results showed that methanol and hot-water extracts of Gloeostereum incarnatum fruiting bodies might be used for good sources of anti-inflammatory, free radical scavenging, and skin whitening agents for human health.

Effects of Antioxidative Activities and Antibrowning of Extracts from Onion, Apple and Mandarin Orange Peel as Natural Antibrowning Agents (천연 갈변 억제제 개발을 위한 양파, 사과 및 감귤 과피 추출물의 항산화 및 갈변 저해 효과)

  • This study was carried out to investigate antioxidative properties of various extracts and antibrowning effects of extracts in apple slices were investigated by ${\Delta}E$ value and PPO relative activity. Apples were cut into 1.5 cm thickness and they were dipped in 1% extract solutions(OW: water extracts of onion, OE: 80% EtOH extracts of onion, AW: water extracts of apple, AE: 80% EtOH extracts of apple, MW: water extracts of mandarin orange peel, ME: 80% EtOH extracts of mandarin orange peel) for 1 min. OW showed higher than the other treatments for total phenolic contents(94.35 mg/g), PPO inhibition(74.00%). And the highest DPPH free radical scavenging activity(40.27%) measured in ME. ${\Delta}E$ value of apple slices dipped in MW was 2.37 whereas ${\Delta}E$ value of apple slices dipped in AW was 12.12. These results suggest that onion and mandarin orange peel extracts should be a potential source for controlling browning during storage of apple slices.

Removal of Radiostrontium ($^{85}Sr$) from the Rat by Water Soluble Chitosan (수용성 카이토산에 의한 체내 방사성스트론튬의 제거)

  • Chitosan is known to be one of the natural chelating agents. It is derived from chitin which is a cellulose-like biopolymer distributed widely in nature, especially in shellfish, insects, fungi, and yeast. There are two forms of chitosans, water soluble and insoluble, The purpose of the present study is to investigate whether water soluble chitosan can be applied to reduce the bioabailability of radios-trontium in foods. We compared the effect of water soluble and insoluble chitosans on the absorption of ingested radiostrontium ($^{85}Sr$). Three percent water soluble and insoluble chitosan solutions were given orally, and immediately after $^{85}SrCl_2$ ($0.2{\mu}Ci$) was administered to rats using a orogastric tube. In one group water soluble chitosan solution was given for additional 4 days. And in control group no chitosan was given. Each group consisted of 6 rats. The whole-body retention of $^{85}Sr$, determined by in vivo counting method, was lower in water soluble chitosan group than that of water insoluble chitosan group and that of control. Urinary excretion of $^{85}Sr$ in chitosan-treated rats was higher than that of control. And 5 day ingested group of water soluble chitosan showed least whole body retention of $^{85}Sr$. In conclusion water soluble chitosan was more effective in reducing bioavailability of ingested radiostrontium in the gastrointestinal tract than insoluble chitosan.

Antioxidant and Anti-lipase Activity in Halocynthia roretzi Extracts (우렁쉥이 추출물의 항산화 및 리파아제 저해활성 효과)

  • Halocynthia roretzi is one of the most important cultured marine species on the southern coast of Korea. Samples were extracted using methanol (ME), ethanol (EE) and water (WE) to evaluate the antioxidant activities and antilipase activity in Halocynthia roretzi extracts. Antioxidant potentials of the samples were determined by poly-phenol content, flavonoid content, free radical scavenging activity, reducing potential, and chelating activity. The ME showed significant scavenging activity (1176 ${\mu}g/mL$ IC50 for DPPH, and 895 ${\mu}g/mL$ IC50 for ABTS assay). The IC50 for lipase inhibition activity was 12,021, 6,004, and 14,979 ${\mu}g/mL$ in the ME, EE, and WE, respectively. In conclusion, Halocynthia roretzi extracts exhibited antioxidant activities and anti-lipase activity. These results suggest that Halocynthia roretzi extracts can be potentially used as a source of antioxidant and antiobesity agents.
