This paper reports the results of a field evaluation which used sampling and analytical methods to determine the levels of airborne hexavalent chromium Cr(VI) in major industrial complexes in Korea over a seven year period (2007-2013). Cr(VI) concentrations were determined using cellulose filter sampling and ion chromatography analysis. In order to validate the analytical performance of these methods, studies were also carried out to investigate data quality control (QC) parameters, such as the method detection limit (MDL), repeatability, and recovery efficiencies. The average concentrations of Cr(VI) for the nine industrial complexes in Korea were in the range of 0.09 to $1.40ng/m^3$, which is similar to of the concentrations in other industrial areas around the world. The impacts of Cr(VI) emissions from industrial areas on Cr(VI) concentrations in neighboring-residential areas were considerably low, and the dispersion of Cr(VI) from industrial areas to residential areas was estimated to be 'not-significant'. Cr(VI) levels were not affected by seasonal variation, which suggests that chromium was emitted continuously from the industrial sources throughout the year. The concentration of Cr(VI) measured accounted for 0.7 to 9.4 percent of the total chromium level, which is a low percentage compared to those in other urban areas around the world. This is the first report in an international journal of a field study conducted in Korea to determine the concentration of Cr(VI) in the ambient air of industrial and residential areas.

The purpose of this study was to investigate for filtration efficiency of several dust masks, comparing with filtration efficiency certified by KOSHA(Korea Occupational Safety & Health Agency), and to require of the right use of protective respirators. Using a welding fume generator and chamber, several dust masks, which were widely used in the workplaces in korea, were tested for their filtering efficiency for stainless steel arc welding fume. The filtration efficiency testing system consisted of a welding fume generator, a chamber and a filtration unit. The filtration unit was made of a mask which was inserted into the sampling cassette and another sampling cassette, which contained mixed cellulose ester filter paper. These two cassettes were connected with tubing. Stainless steel arc welding fume generator was delivered into an chamber. The welding fume in the chamber was passed into the filtration unit with flow rate of 30 liter/min. The welding fume filtration efficiency was evaluated by gravimetric measurement. Metal concentrations in the welding fume before and after filtration were measured with inductive coupling plasma analyzer. Following results were obtained: Filtration efficiency of welding fume for common hygienic mask was 63.82% and the average efficiencies for A, B, C, D, E, F and G masks were 94.62%, 96.58%, 83.20%, 82.76%, 77.25%, 86.55% and 93.22%, respectively. Our results indicate that dust masks used widely in the welding workplaces in korea are not proper for protecting worker's health and then the use of fume mask should be required.
A new endo-$\beta$-1, 4-glucanase was purified from the culture filtrate of thermophilic anaerobic Clostridium thermocellum. The purification procedure included two steps of ion exchange chromatography with DEAD-Sephadex A-50 and gel filtration chromatography with Sephadex G-75. Even though the 56 fold increase in CMCase specific activity was obtained, the actually recovered enzyme activity was relatively lower level of 0.7%. Judging from the two bands in SDS-polyacrylamide gel electrophoresis, the endo-$\beta$-1, 4-glucanase consists of two subunits whose M.W. are 38,000 and 58,000, respectively. The optimum pH and temperature were determined to be 5.0 and $65^{\circ}C$, respectively. The enzyme was stable up to $70^{\circ}C$, but inactivated at $80^{\circ}C$. The kinetic parameters of the separated fraction were also determined. The purified enzyme did not show any significant hydrolytic activity against the highly ordered crystalline cellulose as well as filter paper.
An alkalophilic bacterium, the strain AC-711 as a potent producer of alkaline cellulase, was selected among many isolates from soil environments. Morphological, physiological and chemical characteristics of the strain AC-711 suggested that it belongs to the genus Pseudomonas according to the Bergey's Manual of Systematic Bacteriology, however the G+C mol% (54.43) of its chromosomal DNA is lower than the normal values of the genus. The major cell wall fatty acids were determined as 15:0 and 17:0 anteiso. The production of alkaline CMCase by the strain was maximal when grown on the mediun containing 1% carboxymethyl cellulose, 0.1% $KH_2PO_4$, 0.02% $CoCl_2$, 0.02% Tween 80, 0.5% $Na_2CO_3$, 0.8% yeast extract, pH 10.3 at $30^{\circ}C$ for 3 days, and the most of enzyme was excreted into culture mediun.
Elevated near-UV radiation, containing a large amount of UV-B and a small amount of UV-C, inhibited the development of leaves and tillers, the increase in biomass production, the elongation of plant height, the photosynthetic rate and the chlorophyll content in rice plants in a phytotron. Elevated UV-B radiation filtered through cellulose diacetate film or UV-31 cut filter (transmission down to 290 nm) similarly suppressed each growth component above. Near-UV radiation-caused injuries were alleviated either by elevated CO$_2$ atmosphere or by exposure to high irradiance-visible radiation. On the basis of these findings, we examined cultivar differences in the resistance to UV radiation-caused injuries among 198 rice cultivars belonging to 5 Asian rice ecotypes ( aus, aman, boro, bulu and tjeleh) from the Bengal region and Indonesia and to Japanese lowland and upland rice groups. It was shown that .various cultivars having different sensitivities to the effects of near-UV radiation were involved in the same ecotype and the same group, and that the Japanese lowland rice group and the boro ecotype were more resistant. Among Japanese lowland rice cultivars, Sasanishiki (one of the leading varieties in Japan) exhibited more resistance to near-UV radiation, while Norin 1 showed less resistance, although these two cultivars are closely related. It was thus indicated that the resistance to the inhibitory effects of near-UV radiation of rice cultivars is not simply due to the difference in the geographical situation where rice cultivars are cultivated. From the genetic analysis of resistance to the inhibitory effects of UV radiation on growth of rice using F$_2$ plants generated by reciprocally crossing Sasanishiki and Norin 1 and F$_3$ lines generated by self-fertilizing F$_2$ plants, it was evident that the resistance to the inhibitory effects of elevated near-UV radiation in these rice plants was controlled by recessive polygenes.
High levels of xylanase activity (143.98 IU/ml) produced by the newly isolated Paenibacillus campinasensis G1-1 were detected when it was cultivated in a synthetic medium. A thermostable xylanase, designated XynG1-1, from P. campinasensis G1-1 was purified to homogeneity by Octyl-Sepharose hydrophobic-interaction chromatography, Sephadex G75 gel-filter chromatography, and Q-Sepharose ion-exchange chromatography, consecutively. By multistep purification, the specific activity of XynG1-1 was up to 1,865.5 IU/mg with a 9.1-fold purification. The molecular mass of purified XynG1-1 was about 41.3 kDa as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Sequence analysis revealed that XynG1-1 containing 377 amino acids encoded by 1,134 bp genomic sequences of P. campinasensis G1-1 shared 96% homology with XylX from Paenibacillus campinasensis BL11 and 77%~78% homology with xylanases from Bacillus sp. YA-335 and Bacillus sp. 41M-1, respectively. The activity of XynG1-1 was stimulated by $Ca^{2+}$, $Ba^{2+}$, DTT, and ${\beta}$-mercaptoethanol, but was inhibited by $Ni^{2+}$, $Fe^{2+}$, $Fe^{3+}$, $Zn^{2+}$, SDS, and EDTA. The purified XynG1-1 displayed a greater affinity for birchwood xylan, with an optimal temperature of $60^{\circ}C$ and an optimal pH of 7.5. The fact that XynG1-1 is cellulose-free, thermostable (stability at high temperature of $70^{\circ}C{\sim}80^{\circ}C$), and active over a wide pH range (pH 5.0~9.0) suggests that the enzyme is potentially valuable for various industrial applications, especially for pulp bleaching pretreatment.

This study was conducted for the purpose of obtaining the fundamental data on improvement of working environment and contributing to health improvement of workers who dealed with metal by assessing the metal concentration in air of industries located in Chang-Won Industrial Complex. Authors measured the concentration of metals(Al, Cd, Cr, Cu, Mn, Ni, Pb, Sn and Zn) is the air to 25 working processes of 73 industries by flame atomic absorption spectrometry from February to December 1994. Objective: Fibronectin 3 (FN3) and immunoglobulin like modules (Ig) are usually collocated beside modular cellulase catalytic domains. However, very few researches have investigated the role of these modules. In a previous study, we have sequenced and analyzed bacterial metagenomic DNA in Vietnamese goats' rumen and found that cellulase-producing bacteria and cellulase families were dominant. In this study, the properties of modular cellulases and the role of a FN3 in unique endoglucanase belonging to glycosyl hydorlase (GH) family 5 were determined. Methods: Based on Pfam analysis, the cellulases sequences containing FN3, Ig modules were extracted from 297 complete open reading frames (ORFs). The alkaline, thermostability, tertiary structure of deduced enzymes were predicted by AcalPred, TBI software, Phyre2 and Swiss models. Then, whole and truncated forms of a selected gene were expressed in Escherichia coli and purified by His-tag affinity column for assessment of FN3 ability to enhance enzyme activity, solubility and conformation. Results: From 297 complete ORFs coding for cellulases, 148 sequences containing FN3, Ig were identified. Mostly FN3 appeared in 90.9% beta-glucosidases belonging to glycosyl hydrolase family 3 (GH3) and situated downstream of catalytic domains. The Ig was found upstream of 100% endoglucanase GH9. Rarely FN3 was seen to be situated downstream of X domain and upstream of catalytic domain endoglucanase GH5. Whole enzyme (called XFN3GH5 based on modular structure) and truncate forms FN3, XFN3, FN3GH5, GH5 were cloned in pET22b (+) and pET22SUMO to be expressed in single and fusion forms with a small ubiquitin-related modifier partner (S). The FN3, SFN3 increased GH5 solubility in FN3GH5, SFN3GH5. The SFN3 partly served for GH5 conformation in SFN3GH5, increased modules interaction and enzyme-soluble substrate affinity to enhance SXFN3GH5, SFN3GH5 activities in mixtures. Both SFN3 and SXFN3 did not anchor enzyme on filter paper but exfoliate and separate cellulose chains on filter paper for enzyme hydrolysis. Conclusion: Based on these findings, the presence of FN3 module in certain cellulases was confirmed and it assisted for enzyme conformation and activity in both soluble and insoluble substrate.

Impression cytology refers to application of cellulose acetate filter material to the ocular surface to remove the superficial layers of the conjunctival epithelium. The technique is non-invasive, is easy to perform, causes minimal discomfort to the patient, and can be used to follow changes in the conjunctival ocular surface over time. With this method, the morphology of the conjunctival ocular surface can be studied and the degree of squmaous metaplasia assessed. This study was performed to evaluate the conjunctival surface by impression cytology in dry eye patients. A total of 70 students with no contact lens wearing history were recruited. Subjects were required to fill in a McMonnies dry eye symptom questionnaire. The non-invasive tear thinning time(TIT) test of each subject was measured, followed by Schirmer tear test(STI), tear film break-up time(TBUT) tests and Rose-bengal staining were performed as a baseline. Conjunctival epithelial cells from the inferior bulbar conjunctiva were harvested by the impression cytology technique. The specimens collected were labelled and stained with PAS(Periodic Acid Schift)-haematoxylin. The goblet cells and conjunctival epithelial cells were observed under a light microscope of 400x magnification. The specimens were classified according to the Nelson Grading scale which was based on the degree of squamous metaplasia such as changes of goblet cells density, size/form, N:C(nucleus : cytoplasm) ratio. Dry eye patients were observed morphological changes of the epithelial cells, different nuclear alterations, decrease of the goblet cells density. The degree of cytological changes was related to severity of dry eye conditions. When the epithelial cell morphology was graded according to the system described by Nelson, specimens from the control group revealed 91.43% of the eyes to be grade 0 and 8.57% to be grade 1, whereas of the dry eye patients, 20% were grade 0, 42.86% grade 1, 34.29% grade 2 and 2,86% grade 3. Impression cytology represents a non- or minimally invasive biopsy of the ocular surface epithelium with no side effects or contraindications. It has demonstrated to be a useful diagnostic aid for a wide variety of processes involving the ocular surface. This technique is a safe, simple method and may help increase understanding of various ocular surface alterations in dry eye patients.
Trickling filter has been extensively studied for the domestic wastewater treatment especially for the small scale plants in rural area. The purpose of this research is to survey the physical and microbial characteristics of wood chip media and the removal efficiency of animal wastewater using wood chip trickling filter system. The trickling filtration system comprises a filtration bed packed with wood chip media having a particle dia. of 5~7cm. The method comprises natural air from the bottom of the bed. The system also comprises a control mechanism including a time a constant discharge pump for controlling supply of the wastewater into the bed. The following conclusions were obtained from the results of this research. 1. The specific surface area of wood chip was 0.4123 $m^2$/g, pore volume was 0.0947 $cm^3$/g, density was 0.49 g/$cm^3$. It has forms of parallelogram and oblong which have numerous small pore space. This wood chip has been good condition for microorganism's habitat, having very larger specific surface area by complex the three dimension structure of cellulose at wood's major ingredients. 2. The total counts of in attached aerobic microbes were ranged from $10^6$ to $10^8$ CFU/g, and anaerobes microbial numbers were from $10^4$ to $10^7$. The aerobic microbial numbers appeared to be much more than those of anaerobic microbial numbers. 3. The average efficiency of $BOD_5$ and CODcr were 74.5% and 51.5%, respectively. The removal efficiency of T-N and T-P were 61.4%, 56.2%, respectively. But SS removal levels remain 19.3%.
