Since cellulose is the only organic material that is annually replenishable in very large quantities, we must explore ways to utilize it as a source of energy, food and chemicals. For the utilization of this resource, it is first enzymatic hydrolyzed to glucose, then the glucose can be used as a food, converted single cell protein by microorganism, fermented to clean burning fuel and other chemicals. Cellulolytic enzyme, cellulase, consists of two or three major components, $C_1-cellulase$, $C_x-cellulase$ and ${\beta}-glucosidase$. $C_x-cellulase$ are fairly common but $C_1-cellulase$ are quite rare. Trichoderma viride is the best source of active cellulose, especially $C_1-enzyme$. Saccharification rate of cellulose in greatly influenced by the degree of crystallinity and extent of lignification. But by the pretreatment the substrate with cellulose swelling agent, delignifying reagent and physical treatment, the degree of saccharification is enhanced. Thus, glucose syrups of 2 to 10% concentration are realized from milled newspaper. The enzymatic hydrolysis of such energy rich material, such as cellulose, to glucose is technically feasible and practically achievable on a very large scale.
The structural properties of cellulose are significantly changed with the progress of hydrolysis reaction. The effects of changes on such properties of cellulosic substrate as crystallinity, amicessibility of enzyme to the active site of cellulose surface, and particle size on the kinetics of enzymatic hydrolysis have been studied. Among those physical studies, the apparent surface active site of cellulose particle was found to have the most significant effect on the hydrolysis kinetics. Based on the experimental results, the adsorption affinity of enzyme and hydrolysis rate were mainly influenced by the surface roughness of cellulose particle. The extent of accesssible active site may be expressed as the change of particle diameter. The Langmuir isotherm was proposed in terms of enzyme activity to explain the actual action of enzyme protein.
Simple methods of conductometric titration and infrared spectroscopy were used to quantify the surface carboxyl content of cellulose fibrils isolated by 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO)-mediated oxidation. The effects of different cellulose sources, post or assisted-sonication oxidation treatment, and the amount of sodium hypochlorite addition on the carboxyl content of cellulose were reported. This study showed that post sonication treatment had no influence on the improvement of surface carboxyl charge of cellulose macrofibrils (CMFs). However, the carboxyl content increased for the isolated cellulose nanofibrils (CNFs). Thus the carboxyl content of CNFs is different from those of their corresponding bulk oxidized cellulose and CMFs. Filter paper as a CNF source imparted a higher surface charge than did hardwood bleached kraft pulp (HWBKP) and microcrystalline cellulose (MCC). It was considered that the crystallinity and microstructure of the initial cellulose affected oxidation efficiency. In addition, the carboxyl content of cellulose was successfully controlled by applying sonication treatment during the oxidation reaction and adjusting the amount of sodium hypochlorite.
Substrates used were hardwood-Suwon poplar-(Populus alba${\times}$glandulosa L.) and softwood-pitch pine-(Pinus rigida M.). And these substrates were steam exploded then treated with sodium chlorite at 75$^{\circ}C$ with occasional stirring in order to obtain samples which had different lignin contents and crystallinity. And then this resulting samples incubated with a commercial cellulase derived from Trichoderma ressei. The contents of Klason lignin were decreased as the increasing of the ratio of sodium chlorite in the two species. The effect of hardwood was more effective than that of softwood in the same ratio of sodium chlorite. The minimum contents of Klason lignin were 0.8% and 5.1% respectively. And the crystallinities of cellulose were increased very little as increasing of the ratio of sodium clorite. The hydrolysis extent of the two species were increased as the increasing of delignification. Especially, the hydrolysis extent of hardwood was more higher than that of softwood. The maximum hydrolysis extent were 89.8% and 71.1%, respectively.
Radial and vertical variations of relative crystallinity and crystallite width of cellulose within stems of three softwoods (Pinus densiflora S. et Z., P. koraiensis S. et Z, P. rigida Mill.) grown in Korea were examined by an X-ray diffraction method. The mean of relative crystallinity was 61.7% in P. koraiensis, 60.6% in P. densiflora and 49.4% in P. rigida. The degree of crystallinity in earlywood and latewood increased with the age from pith to about 10~15 years, and then remained almost constant value. The relative crystallinitiy of latewood was slightly higher than that of earlywood. The relative crystallinity in P. densiflora was a little lower at the base of stem, but no significant difference by height was shown in P. koraiensis and P. rigida. The crystallite widths in the stems were 2.8 to 3.0 nm, but were not significantly different in earlywood and latewood by height. In conclusion, the relative crystallinity appeared to be a useful index for separating juvenile wood from adult wood in the softwoods of P. densiflora, P. koraiensis, and P. rigida grown in Korea.
An effective method for production of glucose was developed using enzymatic hydrolysis of Suwon poplar by the cellulase. Enzymatic hydrolysis of wood is the reaction to produce glucose from wood using enzyme which derives from microorganism. Glucose can be transferred easily to ethanol by fermentation. Ethanol is the starting material for producing acetone, butanol, citric acid and lactic acid. The mechanism of the enzymatic hydrolysis of cellulose are reasonably explained in terms of the sequential action of three different types of enzymes, endo-cellulase, ex-cellulase, and $\beta$ -glucosidase. The goal of this work was to investigate the cellulose hydrolysis pretreated polar with various concentration NaOH, the crystallinity of cellulose, lignin contents and the degree of hydrolysis.

The study aimed at the theoretical analysis of vapor sorption properties of, pp.rmaking fibers. Water vapor affinity and sorption thermodynamic properties of fiber constituents were evaluated based on Henry's law and Hildebrand's solubility theory. Theoretical equilibrium moisture content(ThEMC) on fiber surface was estimated using functional group contribution. Crystallinity of cellulose in fiber significantly controlled the water vapor solubility. Comparisons of the measured equilibrium moisture content data and the estimated ThEMC data coincidently suggested the fact that crystallinity of cellulose in fibers was around 60% to 70%. Carbohydrates constituents including amorphous cellulose and hemicellulose in fibers showed higher vapor solubility than lignin molecules. High correlation existed between ThEMC and vapor solubility as well as between ThEMC and solubility parameter. In the thermodynamic analysis on water-vapor sorption process in fibers, the sorption enthalpy increased as RH increased, whereas sorption entropy and free energy decreased with increasing RH.
This study demonstrated that the brown rot basidiomycete Fomitopsis palustris was able to degrade crystalline cellulose (Avicel). This fungus could also produce the three major cellulases (exoglucanases, endoglucanases, and $\beta-glucosidase$) when the cells were grown on $2.0\%$ Avicel. Avicel degraded by F. palustris showed a decrease in relative crystallinity from $83\%\;to\;78.5\%$ after 14 days of incubation. The characterization study indicated that optimum pH was 4.5 and optimum temperature was $70^{\circ}C$ for exoglucanase (cellobiohydrolase) activity. Hydrolysis of Avicel by the crude enzyme from F. palustris yielded 1.6 mg/ml of glucose after 43 h, which corresponded to a cellulose conversion degree of $3.2\%$. Therefore, this study revealed for the first time that the brown rot basidiomycete F. palustris produces cellulases capable of yielding soluble sugars from crystalline cellulose.
The aim of this study is to investigate cane molasses pretreatments for the production of cellulose by Acetobacter sp. V6, which has excellent bacterial cellulose (BC) producing capacity in the shaking culture. Among pretreatments of cane molasses, 1% (w/v) tricalcium phosphate (TP) treatment was more efficient in BC production. The physico-chemical properties of BCs that were produced in static and shaking cultures were also investigated. Although BC had an emulsifying ability, its emulsion stability was low. Water holding capacity (WHC) of BC was high; the WHC of BC produced in static culture was 14 times higher than that of $\alpha$-cellulose. In addition, the viscosity of BC was higher than that of $\alpha$-cellulose. Composition analysis by FT-IR showed no difference in composition between BC and plant cellulose. In the crystallinity analysis by XRD, all BC samples showed crystallinity. All BC samples showed reticulated structures consisting of ultrafine cellulose fibriles. Microfibriles of cellulose from static culture were especially more compact than those of cellulose from shaking culture.
The present study was aimed at evaluating the effect of variable sodium carbonate ($Na_2CO_3$) loading during wet air oxidation (WAO) pretreatment of rice straw in reducing biomass recalcitrance. The research study was intended to increase the cellulose recovery, hemicellulose solubilization, lignin removal in the solid fraction and limiting the generation of inhibitors in the liquid fraction while reducing the chemical input. The operating condition of $169^{\circ}C$, 4 bar, 18 min and 6.5 g/L $Na_2CO_3$ loading resulted in maximum cellulose recovery of 82.07% and hemicellulose solubilization and lignin removal of 85.43% and 65.42%, respectively, with a total phenolic content of 0.36 g/L in the liquid fraction. The crystallinity index increased from 47.69 to 51.25 along with enzymatic digestibility with an increase in $Na_2CO_3$ loading from 0 to 6.5 g/L as a result of removal of barriers for saccharification via effective cleavage of ether and ester bonds cross-linking the carbohydrates and lignin as indicated by FT-IR spectroscopy. A further increase in the $Na_2CO_3$ loading to 9.5 g/L did not significantly increase the sugar release. Thus, it was concluded that 6.5 g/L $Na_2CO_3$ during WAO is sufficient to increase the delignification and deacetylation, leading to significant changes in apparent cellulose crystallinity inter alia improvement in cellulose accessibility and digestibility of rice straw.
