Isolation of Sangivamycin from Streptomyces sp. A6497 and its Herbicidal Activity

  • During the screening for the inhibitors of cellulose biosynthesis as herbicides, we discovered a Streptomyces sp. A6497 with a selective antifungal activity against cellulose containing Phytophthora parasitica, but not against cellulose lacking Candida albicans. The inhibitor was isolated and identified, using a series of chromatographies. Based on structure analyses with UV spectrophotometry, mass and various NMR, the inhibitor was identified as sangivamycin. The compound exhibited strong antifungal activities against P. parasitica (MIC; 3.125 $\mu$g/ml). In particular, it showed strong herbicidal activities against various weeds in the greenhouse experiment. Taken together, these results suggest that sangivamycin is a useful lead compound for the development of new herbicides.

셀룰러 오토마타 기반 해쉬 함수 분석 (Analysis of hash functions based on cellular automata)

  • 해쉬 함수란 임의의 길이의 비트 열을 입력으로 하여 고정된 길이의 비트 열을 출력하는 함수이다. 셀룰러 오토마타는 유한상태머신으로서 인접한 셀과의 결합 논리로 의사난수를 효과적으로 생성할 수 있는 특성을 가지고 있다. 신상욱 등$^{[1]}$ 과 Mihaljevic 등$^{[7]}$ 은 하드웨어 구현에 효율적인 셀룰러 오토마타에 기반한 해쉬 함수를 제안하였다. 본 논문에서는 [1]과 [7]에서 제안된 셀룰러 오토마타 기반 해쉬 함수에 대한 충돌 쌍을 각각 0.46875와 0.5의 확률로 찾을 수 있음을 보인다.

  • CDMA 셀룰라 시스템은 같은 시간에 같은 주파수를 모든 사용자들이 같이 사용하므로 자기 신호 외의 다른 사용자의 신호는 간섭으로 나타나 통신 품질에 영향을 미친다. 이 간섭의 양에 따라 단위 셀당 사용자 수로 정의되는 시스템의 용량이 결정되며 간섭량의 정확한 계산이 이루어져야 시스템 성능 평가를 정확히 할 수 있다. 본 논문은 임의의 겹(tiers) 구조를 갖는 다중 셀 구성의 CDMA 셀룰라 시스템의 타셀 간섭량을 계산하기 위해 Riemann-Zeta 함수를 이용하여 임의의 전파 감쇄 지수에도 적용할 수 있는 근사식을 제안하였고, 제안된 식의 수치 결과와 시뮬레이션 결과를 비교하여 그 효용을 살펴보았다. 제안된 근사식을 이용해 계산한 타셀 간섭량과 시스템 용량은 시뮬레이션을 통해 얻은 결과를 중심으로 상한과 하한을 이루고 있으며 겹 수에 따른 값의 변화가 평균 간섭 및 용량 계산치와 일치하는 결과를 얻었다. 제안된 타셀 간섭 근사식은 복합적인 전파 환경이 공존하는 계층셀(Hierarchical Cellular) 시스템에서의 간섭 및 용량 계산과 알고리즘 검증에 유용하게 사용될 수 있을 것으로 생각된다.

배양된 흰쥐 대뇌 피질 astrocytes의 세포기능에 대한 화학적 무산소증 유도물의 효과 (Effects of Chemical Anoxia Inducers on Cellular Functions of Cultured Rat Cortical Astrocytes)

  • The effects of antimycin A(AA), dodium azide ($NaN_3$) and 2,4-dinitrophenol (DNP), which inhibit mitochondrial ATP production, on cellular functions of cultured astrocytes were studied. High concentrations of AA $(50{\;}\mu\textrm{g}/ml),{\;}NaN_3$ (100mM) and DNP (20mM) significantly decreased 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) reduction, which was known to be related to mitochondrial function and then cel viability. AA ($50{\;}\mu\textrm{g}/ml$) increased lactate dehydrogenase (LDH) release and decreased [$^3H$] glutamate uptake, suggesting severe damage of cellular function by the concentrations of the compounds. Meanwhile, low concentrations of AA $(\leq{;\}10{\;}\mu\textrm{g}/ml),{\;}NaN_3{;\}(\leq{\;}50mM)$ and DNP ($\leq{\;}5mM$) significantly increased MTT reduction, the effect of which was specific to astrocytes. AA (5 and $10{\;}\mu\textrm{g}/ml$) did not affect LDH release and [$^3H$] glutamate uptake, indicating that these compounds increased MTT reduction at the low concentrations without cellular membrane damage. However, the low concentrations of AA produced significant decrease of MTT reduction in a glucose-free medium. Low concentrations of AA (1 and $5{\;}\mu\textrm{g}/ml$) did not change ATP production of astrocytes in the medium containing 10 mM glucose, but completely inhibited in a glucose-free medium, suggesting marked increase of cytosolic ATP production by the blockade of mitochondrial ATP production with low concentrations of AA. These results suggest that astrocytes have ability to enhance neuronal function or survival under conditions of incomplete ischemia or early by enhancement of glycolysis, and that cellular reduction of MTT occurs not only mitochondrially but also extramitchondrially.

Isolation of Candida albicans Chitin Synthase 1 Inhibitor from Streptomyces sp. A6705 and Its Characterization

  • In the course of searching for potent chitin synthase 1 inhibitors from natural resources, Streptomyces sp. A6705 was found to exhibit potent inhibitory activity against the chitin synthase 1 from C. albicans (CaCHS1p). As a result, the inhibitor was isolated and identified using a series of chromatographies. Through chemical analyses with UV spectrophotometry, MS spectrometry, and various NMR techniques, the inhibitor was identified as N,N-bis(2-phenylethyl)urea. The compound exhibited strong inhibitory activity against the chitin synthase 1 from C. albicans with an $IC_{50}$ of 14 ${\mu}g$/ml, representing a similar inhibitory activity to that of the well-known chitin synthase inhibitor, polyoxin D ($IC_{50}$ 15 ${\mu}g$/ml). However, the compound showed no inhibitory activity against the chitin synthase 2 of Saccharomyces cerevisiae up to 280 ${\mu}g$/ml, which is structurally and functionally analogous to CaCHS 1 p. In addition, the compound exhibited weak antifungal activities against Cryptococcus neoformans and Rhizoctonis solani.

  • In the course of screening for selective growth inhibitors against the mycelial form of Candida albicans, we isolated a Streptomyces sp. A6792 from soils. The inhibitor was isolated from the above bacterium and identified through several spectral analyses with UV and mass spectrophotometries, and various NMR. The compound was determined to be a macrocyclic dilactone antibiotic, IKD-8344 (molecular weight: 844, molecular formula: $C_{48}H_{76}O_{12}$). The compound selectively inhibited the growth of mycelial form of C. albicans with an MIC of 6.25 ${\mu}g/ml$. It also exhibited strong inhibitory effect preferentially on the mycelial form of various Candida spp. including C. krusei, C. tropicalis, and C. lusitaniae, with MICs ranging from 1.56 to 25 ${\mu}g$/ml. Furthermore, the compound showed no significant toxicity against SPF ICR mice up to 60 mg/kg. These results suggest that IKD-8344 is a useful lead compound for the development of novel antifungal agents, based on the preferential growth inhibition against Candida spp.

A Quantitative Study of the Quality of Deconvolved Wide-field Microscopy Images as Function of Empirical Three-dimensional Point Spread Functions

  • In this work, for the first time, the quality of restoration in wide-field microscopy images after deconvolution was analyzed as a function of different Point Spread Functions using one deconvolution method, on a specimen of known size and on a biological specimen. The empirical Point Spread Function determination can significantly depend on the numerical aperture, refractive index of the embedding medium, refractive index of the immersion oil and cover slip thickness. The influence of all of these factors is shown in the same article and using the same microscope. We have found that the best deconvolution results are obtained when the empirical PSF utilized is obtained under the same conditions as the specimen. We also demonstrated that it is very important to quantitatively check the process' outcome using several quality indicators: Full-Width at Half-Maximum, Contrast-to-Noise Ratio, Signal-to-Noise Ratio and a Tenengrad-based function. We detected a significant improvement when using an indicator to measure the focus of the whole stack. Therefore, to qualitatively determinate the best deconvolved image between different conditions, one approach that we are pursuing is to use Tenengrad-based function indicators in images obtained using a wide-field microscope.

  • Objective: A case report on the improvement of patients with intestinal metaplasia by Ortho-Cellular Nutrition Therapy (OCNT). Methods: A 55-year-old Korean male suffering from the symptoms of the reduced digestive function, bloating, and hypoacidity. Results: The severity of the symptoms improved after Ortho-Cellular Nutrition Therapy (OCNT), and the patient was finally cured of intestinal metaplasia. Conclusion: Ortho-Cellular Nutrition Therapy (OCNT) is effective in relieving the symptoms of patients with intestinal metaplasia.
