Molecular biological approaches to study the function and regulation of citrate synthase genes in saccharomyces cerevisiae

  • Almost all of the aerobic organisms contain citric acid cycle (or, tricarboxylic acid cycle). This cycle is involved both in energy metabolism and biosynthetic reactions; generation of NADH which derives the synthesis of chemical energy, ATP, and provision of intermediates needed for the biosynthesis. Because of its importance in the cellular metabolism, the regulation of the TCA cycle and its component enzymes has been extensively studied by many biologists (7,28). Citrate synthase is resposible for the initial step of the cycle and has been recognized as the rate limiting step (14,121,26). Understanding of the mechanism of the expression of citrate synthase should be a key step for the elucidation of the regulation of the TCA cycle in the cell metabolism.

Histone tail cleavage as a novel epigenetic regulatory mechanism for gene expression

  • Chromatin is an intelligent building block that can express either external or internal needs through structural changes. To date, three methods to change chromatin structure and regulate gene expression have been well-documented: histone modification, histone exchange, and ATP-dependent chromatin remodeling. Recently, a growing body of literature has suggested that histone tail cleavage is related to various cellular processes including stem cell differentiation, osteoclast differentiation, granulocyte differentiation, mammary gland differentiation, viral infection, aging, and yeast sporulation. Although the underlying mechanisms suggesting how histone cleavage affects gene expression in view of chromatin structure are only beginning to be understood, it is clear that this process is a novel transcriptional epigenetic mechanism involving chromatin dynamics. In this review, we describe the functional properties of the known histone tail cleavage with its proteolytic enzymes, discuss how histone cleavage impacts gene expression, and present future directions for this area of study.

폐주기(肺主氣)에 관한 고찰 (Study on the Pye(Fei)/Lungs Control Ki(Qi))

  • The Pye is one of the organs in oriental medicine, which has functions of controlling Ki. The Lungs is also a organ that has a function of respiration, in western medicne. Ki means the air in the first notion. Therefore, the functions of the Lungs and Pye are no differences. However, Ki doesn't mean the air only. Respiration in western medicne has two meanings. One of them is a external respiration, ang the other is a cellular respiration. In that process, the Lungs supervise oxygen and oxidation of protein, carbohydrate and fat and prduce ATP as energy. Two of Functions in Ki are Choodong(推動;Tuidong) and Onhoo(溫煦;Wenxu). Choodong means making energy ang Onhoo means making heat, that indicate a similarity functions of Pye and Lungs from the view of making energy.

Establishment of New Method for the Assay of Glutamate-cysteine Ligase Activity in Crude Liver Extracts

  • As the antioxidant and free radical scavenger, glutathione (GSH) participates in the preservation of cellular redox status and defense against reactive oxygen species and xenobiotics. Glutamate-cysteine ligase (GCL; also known as ${\gamma}$-glutamylcysteine synthetase, EC 6.3.2.2) is the rate limiting enzyme in GSH synthesis. In the present study, the accurate method for determination of GCL activity in crude liver extracts was developed by measuring both ${\gamma}$-glutamylcysteine and GSH from cysteine in the presence of glutamate, glycine and an ATP-generating system. We added glycine to promote the conversion of ${\gamma}$-glutamylcysteine to GSH, and to minimize the possibility of ${\gamma}$-glutamylcysteine metabolism to cysteine and oxoproline by ${\gamma}$-glutamylcyclotransferase. We established optimal conditions and substrate concentrations for the enzyme assay, and verified that inhibition of GCL by GSH did not interfere with this assay. Therefore, this assay of hepatic GCL under optimal conditions could provide a more accurate measurement of this enzyme activity in the crude liver extracts.

Cleavage of the Star Strand Facilitates Assembly of Some MicroRNAs into Ago2-containing Silencing Complexes in Mammals

  • In animals, microRNAs (miRNAs) and small interfering RNAs (siRNAs) repress expression of protein coding genes by assembling distinct RNA-induced silencing complexes (RISCs). It has previously been shown that passenger-strand cleavage is the predominant mechanism when siRNA duplexes are loaded into Argonaute2 (Ago2)-containing RISC, while an unwinding bypass mechanism is favored for miRNA duplexes with mismatches. Here I present experimental data indicating that some mammalian miRNAs are assembled into Ago2-containing RISC by cleaving their corresponding miRNA star strands. This phenomenon may depend on the secondary structure near the scissile phosphate of the miRNA duplex. In addition, I show that ATP is not required for star-strand cleavage in this process. Taken together, the data here provide insight into the miRNA-loading mechanisms in mammals.

허혈 및 재관류한 흰쥐 심장에 미치는 인삼의 영향 (Effects of Ginseng on Global Myocardial Ischemia and Reperfusion in the Rat Heart)

  • The effect of Ginseng on global myocardial ischemia and reperfusion was examined in isolated perfused rat hearts. The Ginseng ethanol extract (100mg/kg/day) was administered orally for 10 days. The rat hearts were removed and perfused at 75cm $H_{2}O$ by the Langendorff method. After 25 min. of global ischemia, the hearts were reperfused. The myocardial contents of adenosine 5'-triphosphate, creatine phosphate, and calcium were assayed. There no differences in ATP levels in all group of normal and Ginseng-treated hearts. Both in non-ischemic and ischemic heart, Ginseng increased significantly tissue creatine phosphate levels compared with control. Whereas, in ischemic-reperfused heart, there was no significant difference. In the control groups, myocardial calcium contents in the ischemic hearts were decreased compared with the non-ischemic hearts. But, in the Ginseng-treated groups, the calcium contents in the ischemic herts were not changed with the nonischemic hearts. Therefore, Ginseng appears to exert its protective effect against ischemic heart condition, not against ischemic-reperfused heart condition, by regulating energy metabolism and maintaing cellular function.

Flavonoids: An Emerging Lead in the P-glycoprotein Inhibition

  • Multidrug resistance is a major obstacle in cancer chemotherapy. Cancer cells efflux chemotherapeutic drug out of cell by means of transporter and reduce the active concentration of it inside cell. Such transporters are member of the ATP binding cassettes (ABC) protein. It includes P-gp, multiple resistant protein (MRP), and breast cancer resistant protein (BCRP). These proteins are widely distributed in the human cells such as kidney, lung, endothelial cells of blood brain barrier etc. However, there are number of drugs developed for it, but most of them are getting transported by it. So, still there is necessity of a good modulator, which could effectively combat the transport of chemotherapeutic agents. Natural products origin modulators were found to be effective against transporter such as flavonoids, which belongs to third generation modulators. They have advantage over synthetic inhibitor in the sense that they have simple structure and abundant in nature. This review focuses on the P-gp structure its architecture, efflux mechanism, herbal inhibitors and their mechanism of action.

  • Secretion of surfactant phospholipid can be stimulated by a variety of agonists acting via at least three different signal transduction mechanisms. These include the adenylate cyclase system with activation of cAMP-dependent protein kinase; activation of protein kinase C either directly or subsequent to activation of phosphoinositide-specific phospholipase C and generation of diacylglycerols and inositol trisphosphate; and a third mechanism that involves incresed $Ca^{2+}$ levels and a calmodulin-dependent step. ATP stimulates secretion via all three mechanisms. The protein kinase C pathway is also coupled to phopholipase D which, acting on relatively abundant cellular phospholipids, generates diacylglycerols that further activate protein kinase C. Sustained protein kinase C activation can maintain phosphatidylcholine secretion for a prolonged period of time. It is likely that interactions between the different signaling pathways have an important role in the overall physiological regulation of surfactant secretion.

Protection by Ginseng Total Saponin of Rat Hepatocytotoxicity Induced by Toxic Chemicals

  • The present experiment was perf'3rmed to investigate the protective effects of ginseng total saponin (GTS) and possible mechanisms on the hepatocytotoxicity induced by tert-butylhydroperoxide (t-BuOOH), 4-Bromo-calciumu ionophore A23187 (Br-A23187) and KCN. Hepatocytes were isolated by collagenase perfusion of livers from fasted male Sprague Dawley rats and cultured overnight. After various treatments in Krebs-Ringer-HEPES buffer at pH 7.4, cell viability was determined by propidium iodide using fluorocytometry. GTS (5-20 ${\mu}$M) inhibited cell killing induced by t-BuOOH, and KCN, dose-dependently. However, GTS did not inhibit Br-A23187-induced cell killing. These findings support that GTS could protect the hepatocytoxicity induced by some toxic chemicals. The mechanisms of these protective effects by GTS seem to be associated with antioxidant activity and increase of cellular ATP.

AMPK activation and adenine nucleotide content in skeletal muscle following different types of high fat feeding

  • We investigated the role of fatty acid availability on skeletal muscle AMPK activity and adenine nucleotide content. To investigate the chronic effects of elevated fatty acid in vivo Sprague-Dawley rats were fed a chow diet (15% fat) or a diet high in saturated (SAFA, 52% fat) or polyunsaturated (PUFA, 52% fat) fat for eight weeks. High fat diets increased (P < 0.05) plasma FFA levels by 25%. AMPK activity was increased in SAFA and PUFA rats and occurred in the absence of changes in ATP, AMP, phosphocreatine and glycogen content. These results suggest that increasing fatty acid availability increases AMPK activity independent of changes in the cellular energy charge, and implicate the regulation of AMPK by a covalent mechanism. These data also support the contention that increasing fatty acid availability can increase subsequent fatty acid oxidation by an AMPK-mediated process.
