Production of Organic Acids from Food By-Products - Mass Production of Organic Acids by Continuous Flow Ceil Recycling Fermentation - (식품부산물로부터 유기산의 대량생산공정에 관한 연구 - 세포재순환식 연속발효를 이용한 유기산의 대량 생산 -)

  • Fermentation studies were conducted in batch and continuous flow cell-recycle reactors with food by-products as substrates. The genus Propionibacterium acidipropionici ATCC 4965 was utilized in the production of organic acids. Good performance was achieved in the batch fermentation using hydrol as a carbon source and corn steep liquor (CSL) as nitrogen and vitamin sources. Product yields and productivity based on maximum values were 0.80 g total acids/g glucose and 0.26 g total acids/L/h, respectively, when $3\%$, (w/v) of hydrol and $2.5\%$, (w/v) of CSL were utilized. Continuous fermentation with cell-recycling system using the optimum amounts of substrates resulted in dramatic increase in cell concentration (X) and maximum productivity (P). Compared to the batch fermentation, X and P were increased by as much as 21 and 13 times, respectively, at the dilution ratio of $0.2\;hr^{-1}$, indicating that cell recycling fermentation of food by-products provides valuable means for the mass production of organic acids as well as utilizing cell mass as good nutrient resources.

  • In this paper, we development and operated a two-stage continuous stirred tank reactor (CSTR)/trickling biofilter(TBF)system for the long-term continuous treatment of trichloroethylene (TCE) using Burkholderia cepacia G4. In this reactor system. CDTR with cell recycle from TBF was coupled to the TBF for the reactivation of the cells deactivated during TCE degradation. The critical elimination capacity was determined to be 25.3 mg TCE/L day and the reactor has been stably operated for more than 1 months, which clearly represented that CSTR/TBF system can be used for long-term treatment of TCE.

Optimization of Semi-Batch Process for Ethanol Production (에타놀 생산을 위한 Semi-batch 발효 공정의 최적화)

  • As flocculent strains are likely to have considerable potential for internal cell recycle, kinetic studies on glucose medium with flocculent Saccharomyces uvarum were carried out in batch and continuous culture. Using a mathematical model, the kinetic parameters at each temperature and pH were estimated in order to establish optimal conditions. It was found that an overall optimum temperature for growth and ethanol production in the range 33-35$^{\circ}C$ was desirable. With regard to the effect of pH, ethanol production by S. uvarum was found to be relatively insensitive to pH value between 4 and 6, with an optimum pH of around 5. At these optimal conditions a maximum ethanol productivity of 12 g/$\ell$/h was determined using semi-batch process together with 5. uvarum.

  • Organic acids which were produced from biomass wastes streams by cell-recycle fermentation using Propionibacterium acidipropionici ATCC 4965 were extracted by Membrane Contactor using TOA/MIBK system. Maximum productivity was 3.32g organic acid/L/hr at the dilution rate of 0.2/hr in the results of continuous fermentation. The diluted organic acids in the fermenter were selectively separated by Membrane Contactor extraction using 30%(w/w) trioctylamine(TOA) dissolved in methylisobutylketone(MIBK). The flow rate of aqueous phase is 200ml/min and that of extraction phase is 100ml/min. The degree of Acetic acid and Propionic acid extraction from fermentation broth was reached 56.25%, 72.41% respectively.

  • The crystal transformation from cellulose I to cellulose II during alkaline swelling of waste wood, which has been used for cultivating oak mushroom(Cortinellus edodes (Berk.) Ito et Imai), was investigated and compared to that of normal wood by a series of X-ray diffraction analysis. When the sapwood of cultivated wood was treated with 20% NaOH solution for 2 hours, the cellulose I can be easily transformed into Na-cellulose I than normal wood or heartwood of cultivated wood. Certainly the formation of Na-cellulose in wood is proportional to alkali swelling duration, and the formation of cultivated sapwood was faster than that of the other woods. Cellulose I in the sapwood of cultivated wood was easily transformed into cellulose II during mercerization, but the sapwood of normal wood and the heartwood of cultivated wood hardly converted to cellulose II. Namely, most of Na-cellulose I in normal wood can be reconverted to cellulose I in the process of washing and drying. Therefore, it can be concluded from this study that in cell wall lignin and hemicellulose can prevent the alkaline swelling of cellulose in wood and the transformation from cellulose I to cellulose II as well.

Recycling of Fermented Sawdust-based Oyster Mushroom Spent Substrate as a Feed Supplement for Postweaning Calves

  • The objective of this study was to find the way to prolong the storage time of sawdust-based oyster mushroom (Pleurotus osteratus) spent substrate (OMSS) by fermenting with potential probiotic microorganisms to recycle the otherwise waste of mushroom farms. To this purpose, lactic acid bacteria (LAB) were screened to select the best lactic acid-producing strains. Three strains of LAB (Lactobacillus plantarum Lp1', Pediococcus acidilacticii Pa193, L. plantarum Lp2M) were selected and in mixture they lowered the pH of the fermented OMSS to 3.81. fOMSS (fermented sawdust-based oyster mushroom spent substrate) could be stored at room temperature for at least 17 days without any deterioration of feed quality based on the pH, smell, and color. In dry matter disappearance rate in situ, commercial TMR (total mixed ration), OMSS and OMMM (oyster mushroom mycelium mass) showed no significant differences between the samples after 6, 12 and 24 h incubation except for 48 h. Two separate field studies were performed to test the effects of fOMSS supplement on the growth performance of postweaning Holstein calves. Field trials included groups of animals feeding calf starter supplemented with: Control (no supplement), AB (colistin 0.08% and oxyneo 110/110 0.1%), fOMSS (10% fOMSS) and fConc (10% fermented concentrate) and DFM (direct-fed microbials, average $10^9$ cfu for each of three LAB/d/head). Growth performance (average daily gain and feed efficiency) of the fOMSS supplement group was higher than that of AB followed by fConc and DFM even though there was no statistically significant difference. The Control group was lower than any other group. Various hematological values including IgG, IgA, RBC (red blood cell), hemoglobin, and hematocrit were measured every 10 days to check any unusual abnormality for all groups in trial I and II, and they were within a normal and safe range. Our results suggest that sawdust-based OMSS could be recycled after fermentation with three probiotic LAB strains as a feed supplement for post-weaning calves, and fOMSS has the beneficial effects of an alternative to antibiotics for a growth enhancer in dairy calves.

A Pilot Study for Microfiltration of Alcohol Stillage Condensate and Permeate Recycle to Fermentation Broth (알코홀 증류폐액의 Pilot Scale 정밀여과와 여과액의 발효 재활용에 대한 연구)

  • Distillation condensate generated from downstream processing of microbial alcohol fermentation imposes a serious burden to biological wastewater treatment or anaerobic digestion due to its high contents of SS (suspended solids) and TN (total nitrogen), A pilot scale microfiltration of the stillage condensate with a stainless steel SCEPTER membrane of 0.1 ${\mu}$m pore size was carried out to remove SS which was mostly composed of microbial cell residue. A stable permeate flux was achieved when the decanter effluent containing 0.7% of SS was filtered under the conditions of X10 VCR (volume concentration ratio), 2.5 bar of TMP (transmembrane pressure), and 60$^{\circ}C$. When stillage condensate with 2.6% SS was treated directly with microfiltration, VCR below X3 was recommended for a long duration of filtration. The permeate and retentate obtained from microfiltration were recycled to make-up medium of fermentation. Adding permeate or retentate up to 30% of fermentation volume showed no distinguished undesirable influence during the course of alcohol fermentation. Although only slight improvements in the final amount of CO$_2$ evolution and alcohol content were observed, fermentation rate increased so that the required time to reach 450 L/ton of CO$_2$ evolution was shortened to 72% of that with normal media.

  • Recovery method of silicon wafer from defective products generated from manufacturing process of silicon solar cells was studied. The removal effect of the N layer and antireflection coating (ARC) of the waste solar cell were investigated at room temperature ($25^{\circ}C$) by variation of concentration of $H_3PO_4$, $NH_4HF_2$, and concentration and types of chelating agent. Removal efficiency was the best in the conditions; 10 wt% $H_3PO_4$ 2.0 wt% $NH_4HF_2$, 1.5 wt% Hydantoin. Increasing the concentration of $H_3PO_4$, the surface contamination degree was increased and the thickness of the silicon wafe became thicker than the thickness before surface treatment because of re-adsorption on the silicon wafer surface by electrostatic attraction of the fine particles changed to (+). The etching method by mixed solution of $H_3PO_4$-$NH_4HF_2$-chelating agents was expected to be great as an alternative to conventional RCA cleaning methods and as the recycle method of waste solar cells, because all processes are performed at room temperature, the process is simple, and less wastewater, the removal efficiency of the surface of the solar cell was excellent.

2-Stage Optimal Design and Analysis for Disassembly System with Environmental and Economic Parts Selection Using the Recyclability Evaluation Method

  • Promotion of a closed-loop supply chain requires disassembly systems that recycle end-of-life (EOL) assembled products. To operate the recycling disassembly system, parts selection is environmentally and economically carried out with non-destructive or destructive disassembly, and the recycling rate of the whole EOL product is determined. As the number of disassembled parts increases, the recycling rate basically increases. However, the labor cost also increases and brings lower profit, which is the difference between the recovered material prices and the disassembly costs. On the other hand, since the precedence relationships among disassembly tasks of the product also change with the parts selections, it is also required to optimize allocation of the tasks in designing a disassembly line. In addition, because information is required for such a design, the recycling rate, profit of each part and disassembly task times take precedence among the disassembly tasks. However, it is difficult to obtain that information in advance before collecting the actual EOL product. This study proposes and analyzes an optimal disassembly system design using integer programming with the environmental and economic parts selection (Igarashi et al., 2013), which harmonizes the recycling rate and profit using recyclability evaluation method (REM) developed by Hitachi, Ltd. The first stage involves optimization of environmental and economic parts selection with integer programming with ${\varepsilon}$ constraint, and the second stage involves optimization of the line balancing with integer programming in terms of minimizing the number of stations. The first and second stages are generally and mathematically formulized, and the relationships between them are analyzed in the cases of cell phones, computers and cleaners.

Biodegradation of Endosulfan by Klebsiella oxytoca KE-8 Immobilized on Activated Carbon

  • Endosulfan degrading ability of Klebsiella oxytoca KE-8 immobilized by entrapment with activated carbon was examined. Endosulfan degradation by the immobilized bacterial strains on several different activated carbon based support materials was investigated. Based on results, activated carbon ($8\times30$ mesh) was chosen as a support material. The immobilized Klebsiella oxytoca KE-8 with the cell density of 4 mg $g^{-1}$ (dry weight) degraded 22.18 ug $ml^{-1}$ endosulfan within 5 days at pH 7.0, $30^{\circ}C$ in batch shake flask cultures. Also, we an experimented recycle packed bed column mode and continuous packed bed column mode for endosulfan degradation. Under optimum operation condition, the immobilized cells in a laboratory scale pack bed column with support beads were able to degrade endosulfan completely in defined minimal salt medium at a maximum rate of 129.6 ug $ml^{-1}$ per day. Moreover, the endosulfan degradation activity could be demonstrated at $4^{\circ}C$ for one month without significant decrease in activity. Results of this study suggest that immobilized cells of Klebsiella oxytoca KE-8 might be applicable to endosulfan contaminated site.
