이온 빔 조사 처리된 키토산 스펀지의 세포적합도에 관한 연구 (A study on cytocompatibility of ion beam-irradiated chitosan sponges)

  • Chitosan is a biodegradable and non-toxic material with a molecular weight of 800-1,500Kd which can be obtained in various forms with extraordinary chemical structures and biological characteristics of which enables it to be used in many fields as a biomaterial. Ion irradiation is a useful tool to modify chemical structures and physical properties of high molecular weight polymers. The basic hypothesis of this study is that when surface properties of chitosan in a sponge form are modified with ion beam-irradiation and cell adhesion properties of chitosan would improve and thereby increase the regenerative ability of the damaged bone. The purpose of this study was to illuminate the changes in the cytocompatibility of chitosan sponges after ion beam-irradiation as a preliminary research. Argon($Ar^+$) ions were irradiated at doses of $5{\times}10^{13}$, $5{\times}10^{15}$ at 35 keV on surfaces of each sponges. Cell adhesion and activity of alkaline phosphatases were studied using rat fetal osteoblasts. The results of this study show hat ion beam-irradiation at optimal doses($5{\times}10^^{13}\;Ar^+\;ion/cm^2$) is a useful method to improve cytocompatibility without sacrificing cell viability and any changing cell phenotypes. These results show that ion beam-irradiated chitosan sponges can be further applied as carriers in tissue engineering and as bone filling materials.

일정한 주름을 갖는 친수성 PDMS 제작 및 응용 (Fabrication of Hydrophilic Poly(dimethylsiloxane)with Periodic Wrinkling Surface and Its Application)

  • This paper presents a fabrication of hydrophilic Poly(dimethylsiloxane) (PDMS) with periodic wrinkling surface. The proposed periodic wrinkling surface was fabricated using the sequential processes of typical curing process of PDMS, cutting process, platinum deposition process, and wrinkling transfer process. The surface morphology of the fabricated wrinkling surface was observed by using optical and dynamic atomic force microscopy and discussed. The measured period and amplitude of wrinkling was about $2.2{\mu}m$ and $0.31{\mu}m$, respectively. And, the contact angles of water droplets on the wrinkled surface were measured in order to understand effect of the wrinkling surface on surface modification of hydrophobic PDMS. Our new finding was that the proposed wrinkling surface was hydrophilic and the measured contact angle was about $62^{\circ}$. Moreover, it was found out from the simple cell culture test that the fabricated wrinkling surface was more effective for cell spreading and adhesion than the case of native PDMS substrate.

  • A lipopolysaccharide (LPS) defective mutant of Bradyrhizobium japonicum was characterized in terms of its cell surface hydrophobicity (CSH). By monitoring the kinetics of adhesion to hexadecane the LPS mutant was found to be far more hydrophobic than the wild type strain; the removal coefficients were 4.65 $min^{-1}$ for the mutant, as compared with only 2.40 $min^{-1}$ for the wild type. The possible role of cell surface hydrophobicity of B. japonicum in nodulation process is discussed.

Anti-metastatic Effects on B16F10 Melanoma Cells of Extracts and Two Prenylated Xanthones Isolated from Maclura amboinensis Bl. Roots

  • Inhibitory effects of Maclura amboinenesis Bl, one plant used traditionally for the treatment of cancers, on metastatic potential of highly metastatic B16F10 melanoma cells were investigated in vitro. Cell proliferation was assessed using the MTT colorimetric assay. Details of metastatic capabilities including invasion, migration and adhesion of B16F10 melanoma cells were examined by Boyden Chamber invasion and migration, scratch motility and cell attachment assays, respectively. The results demonstrated that n-hexane and chloroform extracts exhibited potent anti-proliferative effects (p<0.01), whereas the methanol and aqueous extracts had less pronounced effects after 24 h exposure. Bioactivity-guided chromatographic fractionation of both active n-hexane and chloroform extracts led to the isolation of two main prenylated xanthones and characterization as macluraxanthone and gerontoxanthone-I, respectively, their structures being identified by comparison with the spectral data. Interestingly, both exhibited potent effective effects. At non-toxic effective doses, n-hexane and chloroform extracts (10 and $30{\mu}g/ml$) as well as macluraxanthone and gerontoxanthone-I (3 and $10{\mu}M$) significantly inhibited B16F10 cell invasion, to a greater extent than $10{\mu}m$ doxorubicin, while reducing migration of cancer cells without cellular cytotoxicity. Moreover, exposure of B16F10 melanoma cells to high concentrations of chloroform ($30{\mu}g/ml$) and geratoxanthone-I ($20{\mu}M$) for 24 h resulted in delayed adhesion and retarded colonization. As insights into mechanisms of action, typical morphological changes of apoptotic cells e.g. membrane blebbing, chromatin condensation, nuclear fragmentation, apoptotic bodies and loss of adhesion as well as cell cycle arrest in the G1 phase with increase of sub-G1 cell proportions, detected by Hoechst 33342 staining and flow cytometry were observed, suggesting DNA damage and subsequent apoptotic cell death. Taken together, our findings indicate for the first time that active n-hexane and chloroform extracts as well as macluraxanthone and gerontoxanthone-I isolated from Maclura amboinensis Bl. roots affect multistep of cancer metastasis processes including proliferation, adhesion, invasion and migration, possibly through induction of apoptosis of highly metastatic B16F10 melanoma cells. Based on these data, M. amboinensis Bl. represents a potential candidate novel chemopreventive and/or chemotherapeutic agent. Additionally, they also support its ethno-medicinal usage for cancer prevention and/or chemotherapy.

Rho-associated Kinase is Involved in Preimplantation Development and Embryonic Compaction in Pigs

  • The first morphogenetic event of preimplantation development, compaction, was required efficient production of porcine embryos in vitro. Compaction of the porcine embryo, which takes place at post 4-cell stage, is dependent upon the adhesion molecule E-cadherin. The E-cadherin through ${\beta}$-catenin contributes to stable cell-cell adhesion. Rho-associated kinase (ROCK) signaling was found to support the integrity of E-cadherin based cell contacts. In this study, we traced the effects of ROCK-1 on early embryonic development and structural integrity of blastocysts in pigs. Then, in order to gain new insights into the process of compaction, we also examined whether ROCK-1 signaling is involved in the regulation of the compaction mediated by E-cadherin of cellular adhesion molecules. As a result, real-time RT-PCR analysis showed that the expression of ROCK-1 mRNA was presented throughout porcine preimplantation stages, but not expressed as consistent levels. Thus, we investigated the blastocyst formation of porcine embryos treated with LPA and Y27632. Blastocysts formation and their qualities in LPA treated group increased significantly compared to those in the Y27632-treated group (p < 0.05). Then, to determine whether ROCK-1 associates embryonic compaction, we explored the effect of activator and/or inhibitor of ROCK-1 on compaction of embryos in pigs. The rate of compacted morula in LPA treated group was increased compared to that in the Y27632-treated group (39.7 vs 12.0%). Furthermore, we investigated the localization and expression pattern of E-cadherin at 4-cell stage porcine embryos in both LPA- and Y27632-treated groups by immunocytochemical analysis and Western blot analysis. The expression of E-cadherin was increased in LPA-treated group compared to that in the Y27632-treated group. The localization of E-cadherin in LPA-treated group was enriched in part of blastomere contacts compared to that Y27632-treated group. ROCK-1 as a crucial mediator of embryo compaction may plays an important role in regulating compaction through E-cadherin of the cell adhesion during the porcine preimplantation embryo. We concluded that ROCK-1 gene may affect the developmental potential of porcine blastocysts through regulating embryonic compaction.

이차원 전기영동을 이용한 Lactobacillus acidophilus Strains의 Shiga Toxin-producing E. coli (STEC) 부착 억제와 관련된 단백질 발현 변화 분석 (Comparison of Specific Proteins of Shiga Toxin-producing E. coli (STEC) Adhesion by Lactobacillus acidophilus Strains Using Two Dimensional Gel Electrophoresis)

  • 최근 들어 병원성 미생물의 저감화를 위하여 기존의 항생제 계열의 항균물질이 아닌 새로운 개념의 신소재 개발이 활발하게 진행 중에 있다. 특히, 이러한 신개념의 병원성 저감화 소재 중 인간의 장내에 존해하는 probiotics 균주의 특성을 이용하여 병원성 미생물을 예방하는 것은 보다 효과적인 방법 중의 하나가 될 수 있을 것으로 판단된다. 본 실험에서는 HT-29 cell을 대상으로 L. acidophilus 균체와 세포 파쇄물을 대상으로 STEC ATCC 43894의 장 상피세포 부착 억제능력을 측정하였다. 10 mg/mL의 세포 파쇄물이 존재하였을 때 $10^9cfu/mL$의 균체가 존재했을 때와 유사한 수준으로 STEC ATCC 43894의 부착 저해 효과가 관찰되었다. 하지만, L. acidophilus A4의 상등액에서는 그 저해 효과가 세포 파쇄물의 $5{\sim}10%$ 정도 수준으로 관찰되어 그 효과는 매우 적은 것으로 판단되었다. 또한, L. acidophilus A4의 세포 파쇄물이 STEC의 부착에 미치는 영향을 관찰하기 위하여 10mg/mL의 세포 파쇄물이 첨가된 배지에서 STEC의 단백질발현 양상을 확인하였다. 각 gel의 image에서 평균적으로 800개의 spot을 관찰할 수 있었으며 이중 2배 이상의 발현차이를 보이는 13개의 spot을 선발하였다. 7개의 spot은 세포파쇄물이 첨가되었을 때 발현이 증가하였으며 3개의 spot은 발현이 감소하였다. 흥미롭게도 3개의 단백질 spot은 세포파쇄물이 존재할 때만 발현되는 것을 확인하였다. 명확하지는 않지만 이러한 L. acidophilus A4의 세포 파쇄물에 존재하는 물질은 (1)STEC의 부착과 관련된 특정 단백질의 발현을 저해하거나 (2)STEC과 장상 피세포에서의 수용체 경합을 통해 부착을 억제하는 것으로 생각된다. 앞으로 이와 관련된 보다 세부적인 작용 메카니즘 연구 및 생화학적연구가 필요할 것으로 판단된다.

방사선 가교된 유착방지용 Carboxymethyl Cellulose/Porcine Cartilage Acellular Matrix 수화젤 필름의 물리적 특성 및 부착 방지 평가 (Radiation-Crosslinked Carboxymethyl Cellulose/Porcine Cartilage Acellular Matrix Hydrogel Films to Prevent Peritoneal Adhesions with physical properties and anti-adhesivity)

  • 본 실험은 방사선을 이용하여 유착방지용 Carboxymethyl cellulose sodium salt (CMC)/Porcine Cartilage Acellular Matrix (PCAM) 수화젤 필름에 대해 연구하였다. 방사선 조사시 CMC/PCAM의 농도 및 혼합비율에 따른 필름의 형태학적 구조, 젤화율, 젤강도, 팽윤도 등을 분석하였다. 방사선에 의해 가교된 CMC/PCAM 필름은 CMC 필름보다 물리적인 특성인 젤화율이 낮게 측정되었다. 또한, 가교된 CMC 필름보다는 CMC/PCAM 필름에서 인간 혈관내피세포의 부착 및 증식율이 감소하였다. 우리는 PCAM에 함유한 세포의 항부착 성분이 도입된 CMC/PCAM 필름과 CMC 필름은 세포의 접착 및 증식율을 낮추는 것을 확인하였다. 결론적으로, 본 연구에서는 방사선 가교된 CMC/PCAM 수화젤 필름은 유착방지 향상을 위한 유착방지제로서 응용이 기대된다.

Disodium Kromoglycate와 Ketotifen의 사람정장 유도 비만세포 활성화 억제작용 (Inhibitory Effect of Disosium Cromoglycate and Ketotifen on Human Seminal Plasma-Induced Mast Cell Activation)

  • Background: Human seminal plasma (HSP)-induced hypersensitivity is one of the serious complications with sexual intercourse. The clinical manifestations of HSP-induced hypersensitivity may be related to the release of vasoactive mediators from mast cell induced by HSP. It has recently been reported that HSP modulates immune systems and induces mast cell degranulation and histamine release from rat peritoneal mast cells (RPMC). Ketotifen and disodium cromoglycate (DSCG), anti-asthmatic and anti-allergic drugs, have a role of mast cell stabilization and inhibit mast cell-induced leukocyte rolling and adhesion. But the inhibitory agents of HSP-induced mast cell activation are unknown. This study was performed to investigate the effects of DSCG and ketotifen on the HSP-induced mast cell activation. Methods: For this, influences of DSCG and ketotifen on the human seminal plasma-induced degranulation, histamine release and morphological changes of RPMC were observed. Results: The mast cell degranulation and histamine release of RPMC by HSP were induced in a dose-dependent fashion. The HSP-induced cytomorphological changes such as swelling, intracellular vacoules, and interrupted cell boundary were significantly inhibited by pretreatment with DSCG or ketotifen. DSCG and Ketotifen inhibited the HSP-induced degranulation and histamine release from RPMC. Conclusion: From the above results, it is suggested that DSCG and ketotifen have a inhibitory effect of the HSP-induced mast cell activation. DSCG and ketotifen may be used for treatment of HSP-induced hypersensitivity.

TAGLN2-mediated actin stabilization at the immunological synapse: implication for cytotoxic T cell control of target cells

  • Actin dynamics is critical for the formation and sustainment of the immunological synapse (IS) during T cell interaction with antigen-presenting cells (APC). Thus, many actin regulating proteins are involved in spatial and temporal actin remodeling at the IS. However, little is known whether or how actin stabilizing protein controls IS and the consequent T cell functions. TAGLN2 − an actin-binding protein predominantly expressed in T cells − displays a novel function to stabilize cortical F-actin, thereby augmenting F-actin contents at the IS, and acquiring leukocyte function-associated antigen-1 activation following T cell activation. TAGLN2 also competes with cofilin to protect F-actin in vitro and in vivo. During cytotoxic T cell interaction with cancer cells, the expression level of TAGLN2 at the IS correlates with the T cell adhesion to target cancer cells and production of lytic granules such as granzyme B and perforin, thus expressing cytotoxic T cell function. These findings identify a novel function for TAGLN2 as an actin stabilizing protein that is essential for stable immunological synapse formation, thereby regulating T cell immunity. Transforming growth factor β1 enhances adhesion of endometrial cells to mesothelium by regulating integrin expression

  • Endometriosis is the abnormal growth of endometrial cells outside the uterus, causing pelvic pain and infertility. Furthermore, adhesion of endometrial tissue fragments to pelvic mesothelium is required for the initial step of endometriosis formation outside uterus. $TGF-{\beta}1$ and adhesion molecules importantly function for adhesion of endometrial tissue fragments to mesothelium outside uterus. However, the function of $TGF-{\beta}1$ on the regulation of adhesion molecule expression for adhesion of endometrial tissue fragments to mesothelium has not been fully elucidated. Interestingly, transforming growth factor ${\beta}1$ ($TGF-{\beta}1$) expression was higher in endometriotic epithelial cells than in normal endometrial cells. The adhesion efficiency of endometriotic epithelial cells to mesothelial cells was also higher than that of normal endometrial cells. Moreover, $TGF-{\beta}1$ directly induced the adhesion of endometrial cells to mesothelial cells through the regulation of integrin of ${\alpha}V$, ${\alpha}6$, ${\beta}1$, and ${\beta}4$ via the activation of the $TGF-{\beta}1/TGF-{\beta}RI/Smad2$ signaling pathway. Conversely, the adhesion of $TGF-{\beta}1-stimulated$ endometrial cells to mesothelial cells was clearly reduced following treatment with neutralizing antibodies against specific $TGF-{\beta}1-mediated$ integrins ${\alpha}V$, ${\beta}1$, and ${\beta}4$ on the endometrial cell membrane. Taken together, these results suggest that $TGF-{\beta}1$ may act to promote the initiation of endometriosis by enhancing integrin-mediated cell-cell adhesion.
