A semi-automatic cell type annotation method for single-cell RNA sequencing dataset

  • Single-cell RNA sequencing (scRNA-seq) has been widely applied to provide insights into the cell-by-cell expression difference in a given bulk sample. Accordingly, numerous analysis methods have been developed. As it involves simultaneous analyses of many cell and genes, efficiency of the methods is crucial. The conventional cell type annotation method is laborious and subjective. Here we propose a semi-automatic method that calculates a normalized score for each cell type based on user-supplied cell type-specific marker gene list. The method was applied to a publicly available scRNA-seq data of mouse cardiac non-myocyte cell pool. Annotating the 35 t-stochastic neighbor embedding clusters into 12 cell types was straightforward, and its accuracy was evaluated by constructing co-expression network for each cell type. Gene Ontology analysis was congruent with the annotated cell type and the corollary regulatory network analysis showed upstream transcription factors that have well supported literature evidences. The source code is available as an R script upon request.

  • The pars distalis of the Korean frogs (Rana dybowskii Guenther) during hibernating and active periods was observed with the electron microscope. Seven cell types were classified according to the size and shape of secretory granules and to the ultrastructural characteristics. There were many differences between hibernating and active frogs in type 5 cells. Therefore the following results were observed. Cell type 1; This type cell contains spherical secretory granules, $375{\sim}687m{\mu}$ in diameter. Cell type 2; This type cell contains various secretory granules, $250{\sim}437m{\mu}$ in diameter Cell type 3; Spherical and rod-shaped granules, $l25{\sim}187m{\mu}$ in diameter were observed. Cell type 4; In this type cell, the electron density is the lowest and the density of granules is the highest of all type cells. This type cell contains various secretory granules and large secretory granules, $2l0{\sim}420m{\mu}$ in diameter, were also observed. Cell type 5; The electron density of this cells is similar to that of type 4 cells. The density of granules is lower than that of type 4 cells. And the shapes of the secretory granules are similar to those of type 4 cells. But many rod shaped granules, $200{\sim}863m{\mu}$ in diameter, were also observed. Cell type 6; This type was similar to type 2. The electron density of cytoplasm is very low. Spherical granules, $232{\sim}316m{\mu}$ in diameter, were observed. Cell type 7; This type of cell has no secretory granules. This cell is not developed very well. The type 5 cells in hibernating frogs are different from cells in active frogs. In type 5 cells, many secretory granules were observed during active period. But the number of secretory granules were greatly declined and there were many vacuoles in cytoplasm during hibernating period.

  • This paper presents analysis of temperature and light intensity characteristics of PV modules with solar cell type. Taking the effect of sunlight irradiance on the cell temperature, the first experiment takes ambient temperature as reference input and uses the solar insolation as a unique varying parameter. Then taking the effect of the cell temperature on sunlight irradiance, the second experiment takes 1000W/$m^2$ as reference input and uses the cell temperature as a unique varying parameter. As a result, varying sunlight irradiance, the Cell-Type with the smallest change in output is HIT and the Cell-Type with the biggest change in output is a-Si. Varying the cell temperature, the Cell-Type with the smallest change in output is a-Si and the Cell-Type with the biggest change in output is Single-Si. And considering both temperature and light intensity characteristics, the Cell-Type with the smallest change in output is HIT.

Metamorphic Changes of the Neurosecretory Cells in the Brains of Wax Moth Gafferfa mellonella during Metamorphosis (꿀벌부채명나방 뇌신경분비세포의 변태에 따른 변화)

  • The paraidehyde-fuchsin(PAE)-positive neurosecretory cells in the brains of wax moth Galleria mellonella have been morphologically examined at the six different metamorphic stages. During the metamorphosis, neurosecretory cells in the brain can be found in the five unclei of pars intercerebralis, lateral region of protocerebrum, optic lobe, deutocrebrum and tritocerebrum. The five nuelel include one to seven neurosecretory cells. On the bases of cell sizes and histochemical specificities of neurosercretion within cells, all the PAF-positive neurosercretory cells included in the six different metamorphic brains can be recognized as four species of neurosecretory cells as follows; (1) large (about 25 $\mu$m), neurosecretion-parcked cell (type I neurosecretory cell), (2) large, granule-dispersed cell (type II neurosecretury cell), (3) small (about 15 $\mu$m), neurosecretion-packed cell (type III neurosecretory cell), and (4) small, granule-dispersed cell (type IV neurose-cretory cell). The three tb seven medial neurosecretory cells are included in the pars intercerebralis of the six different metamorphic brains. With the increase of days from the late larva to the adult the type I cells of medial neurosecretory cells gradually decrease in number, but the respective three type II neurosecretory cells appear in the five different metamorphic brains except in pupa 2 day before the emeregnce of the adult which has only one type II. The one to five lateral neurosecretory cells are observed in the lateral region of protocerebrum from thepupa just after pupation to the adult. The type IV neurosecretory cells are the most in number of lateral neurosecretory cells. The one type Ineurosecretory cells are included near the optic lobe of only the 4-day-old pupa. the one deutocerebral neurosecretory cell, type II, appears only in the adult. The tritocerebrum includes both three neurosecretory cells in the late larva and one neuresecretory cell in the adult. In the late larva the two tritocerebral neurosecretory cells are type Ill neurosecretory cell and the one is type IV. The remaining one tritocerebral neurosecretory cell is type IV.

  • A histochemical and ultrastructural study on the epithelia of some selected digestive tracks such as esophagus, crop, intestine of a land snail N. samarangae was carried out during the period of June 1997 to may 1998. The epithelium of digestive tract are simple columnar epithelium and consisted of five types of columnar cells. Type 1 cell which is majority in number has a brush border with microvilli on the free surface of the cell and contains numerous secretory granules supposed to be neutral mucopolysaccharide. Type 2 cell, elongated conical in shape, is rarely found in the epithelium. This cell also has a brush border with microvilli on its free surface and contains well developed rough surfaced endoplasmic reticulum, Golgi bodies, and secretory granules in various electron densities. This cell seems to produce both of acid and neutral mucopolysaccharides. Type 3 cell, which is morphologically similar to the Type 1 cell, has microvilli and cilia on the free surface and exists in group only in the limited regions of the intestine. Type 4 cell, typical goblet cell containing secretory granules in high electron density. Type 5 cell rarely found in the digestive tract. This cell contain inconspicuous materials.

  • A fuel-cell power system among various alternative power sources has many advantages such as comparatively independable circumstances, high-efficient, and heat-recyclable, thus it is now able to be up to hundreds MWh-scaled through improving feasibility and longevity of it. During the last few decades, numerous research results has been investigated to expand interest in fuel-cell technology. This study presents pressure distribution on the geometrical manifold structures, which are U-type and Z-type, of a planar-type fuel-cell stack by simulated with computational fluid dynamics(CFD). Then, electrical performance of a 200W fuel-cell stack, which is U-type, was diagnosed after pre-conditioning operation. The stack has electrical characteristics ; 22V, 10A, 220W, and current density $200mA/cm^2$.

And it is advantageous from economic due to adjust the indoor temperature. It is appropriate for the atrium type, curtain wall type, window type and etc.

  • The brain is an organ that consists of various cell types. As our knowledge of the structure and function of the brain progresses, cell type-specific research is gaining importance. Together with advances in sequencing technology and bioinformatics, cell type-specific transcriptome studies are providing important insights into brain cell function. In this review, we discuss 3 different cell type-specific transcriptome analyses i.e., Laser Capture Microdissection (LCM), Translating Ribosome Affinity Purification (TRAP)/RiboTag, and single cell RNA-Seq, that are widely used in the field of neuroscience. Clinical and Cytomorphological Studios on Bovine Leukosis (우백혈병의 임상 및 세포형태학적 연구)

  • Clinical and cytomorphological studies were carried out in 32 leukotic cattle from Tokachi and Kushiro districts in Hokkaido during the 12 year period from 1969 to 1980. The leukotic cattle were examined :and divided into four types(15 cases of the adult, 11 cases of the thymic, 4 casas of the calf and 2 cases of the skin types). The results obtained were as follows : 1. As for the frequency of the main clinical signs in each type, In the adult type, the main clinical signs (of decreasing order) are as follows: swelling of the superficial lymph nodes>depression and loss of weight>tachycardia>anorekia, anemia of the visible mucous membrane and tachypnea. Those of the thymic type were swelling of the thymus>swelling of the medial iliac lymph nodes> swelling of the superficial lymph nodes>tachypnea. Those of the calf type were swelling of the auperficial lymph nodes>depression and emaciation>tachypnea>anorexia, tachycardia, anemia of the visible mucous membrane and recumbency. Those of the skin type were generalized urticaria-like lesions in skin and swelling of superficial lymph nodes>and depression and loss of weight in the decreasing order of frequency. In addition, large tumor mass in the pelvic cavity and swelling of the medial iliac lymph nodes were detected through rectal palpation in 33.3% and 100% in the adult type cases, respectively. 2. As for the hematological findings, The frequency of occurrence of decreased erythrocyte counts in the decreasing order were as follows : adult>calf>thymic>and skin types. The increase in the total leukocyte count in the order of decreasing frequency were as follows: calf>thymic>adult>and skin types. The increase in the absolute lymphocyte counts was found to be at a low rate, 62.5% of all the cases examined. By contrast, the increase of 5% or more of abnormal lymphocyte rates was observed at a high rate, 96.9% of the total cases. 3. Abnormal lymphocytes were found in all cases examined for lymph nodes biopsied. 4. From the cytomorphological point of view, leukotic cells were divided into 3 types: reticulum cell, lymphoid cell and monocytic cell types. The adult type leukotic cattle were divided with reticulum cell type (66.7%), the lymphoid cell type(22.6%) and monocytic cell type(6.7%). The thymic type was lymphoip cell type(72.7%) and reticulum cell type(27.3%). In the calf type, all were lymphoid cell type while all of the skin type were reticulum cell type only. 5. The leukotic cattle had higher NP frequency in the blood and lymphoid tissue than non-1 eukotic cattle. Especially the adult type had the highest NP frequency. However, it was not recognized that NP were characteristic of leukotic cattle alone. 6. The above findings lead to the conclusion that the most effective diagnostic methods for bovine leukosis are the confirmation of swelling of the superficial and internal lymph nodes and thymus in addition to appearance of abnormal neoplastic cell in the peripheral blood and lymph nodes biopsied.

Study on the tree-mimic array of solar cell modules (수목형상에 따른 태양전지 모듈의 배열 연구)

  • This study is about the installation of the solar cell modules. The solar cell modules are built by the tree-mimic structure, and the performance is compared with that of the flat-plate type solar cell module installation. The mathematical tree model, which was suggested by Fisher and Honda, is utilized to determine the location of the solar cell modules for the tree-mimic type. The experiment shows that the generated electric power of the flat-plate type is higher than that of the tree-mimic type by 30% for one month of July. This lower performance for the tree-mimic type comes from the shading effects among the solar cell modules. The theoretical calculation for the absorbed solar radiation on the two types of solar cell installation shows that the tree-mimic type is higher than the flat-plate type by 8.5%. The shading area for the tree-mimic model is calculated with time by using the 3D-CAD, which will be utilized for the optimization of the tree-mimic model in the future.
