Effect of Lithium Bis(Oxalato)Borate Additive on Thermal Stability of Si Nanoparticle-based Anode (리튬 이차 전지용 실리콘 나노입자 음극의 고온 열안정성에 대한 Lithium bis(oxalato)borate첨가제의 효과)

  • Silicon (Si) has been investigated as promising negative-electrode (anode) materials because its theoretical specific capacity of 4200 mAh/g for $Li_{4.4}Si$ is far higher than that of carbonaceous anodes in current commercial products. However, in practice, the application of Si to Li-ion batteries is still quite challenging because Si suffers from severe volume expansion and contraction and lead to a continuous solid electrolyte interphase (SEI)-filming process by cracking of Si. This process consumes the limited $Li^+$ source, builds up thick and unstable SEI layer on the Si active materials, and will eventually disable the cell. Since unstable SEI reduces electrochemical performance and thermal stability of the Si anode, the surface chemistry of the anode should be modified by using a functional additive. It is found that lithium bis(oxalato)borate (LiBOB) as an additive effectively protected the Si anode surface, improved capacity retention when stored at $60^{\circ}C$, and alleviated exothermic thermal reactions of fully lithiated Si anode.

  • To enhance electrochemical performances of anode materials, the surface of coal tar pitch (CTP) was modified by incorporating heteroatoms through chemical treatment with phosphoric acid ($H_3PO_4$). The prepared anode materials with modified CTP was analyzed by XRD, FE-SEM and XPS. The electrochemical performances of modified CTP were investigated by constant current charge/discharge test, rate performance, cyclic voltammetry and impedance tests using the electrolyte of $LiPF_6$ dissolved in the mixed organic solvents (ethylene carbonate : dimethyl carbonate = 1 : 1 vol% + vinylene carbonate 3 wt%). The coin cell using modified CTP ($H_3PO_4/CTP$ = 3 : 100 in weight) has better initial capacity and initial efficiency (489 mAh/g, 82%) than those of other composition coin cells. Also, it was found that the capacity retention was 86% after 30 cycles and the rate capability was 87% at 2 C/0.1 C.

  • The $\alpha$1,6-mannosyltransferase encoded by Saccharomyces cerevisiae OCH1 plays a key role for the outer chain initiation of the N-linked oligosaccharides. A search for Hansenula polymorpha genes homologous to S. cerevisiae OCHI (ScOCH1) has revealed seven open reading frames (ORF100, ORF142, ORF168, ORF288, ORF379, ORF576, ORF580). All of the seven ORFs are predicted to be a type II integral membrane protein containing a transmembrane domain near the amino-terminal region and has a DXD motif, which has been found in the active site of many glycosyltransferases. Among this seven-membered OCH1 gene family of H. polymorpha, we have carried out a functional analysis of H. polymorpha ORF168 (HpOCH2) showing the highest identity to ScOCH1. Inactivation of this protein by disruption of corresponding gene resulted in several phenotypes suggestive of cell wall defects, including hypersensitivity to hygromycin B and sodium deoxycholate. The structural analysis of N-glycans synthesized in HpOCH2-disrupted strain (Hpoch2Δ) and the in vitro $\alpha$1,6-mannosyltransferase activity assay strongly indicate that HpOch2p is a key enzyme adding the first $\alpha$1,6-mannose residue on the core glycan Man$_{8}$GlcNAc$_2$. The Hpoch2Δ was further genetically engineered to synthesize a recombinant glycoprotein with the human compatible N-linked oligosaccharide, Man$_{5}$GlcNAc$_2$, by overexpression of the Aspergillus saitoi $\alpha$1,2-mannosidase with the 'HDEL” ER retention signal.gnal.

  • Most of the cholesterol is synthesized by liver in the body while about one of third is taken via dietary. The main functions of cholesterol is to protect membranes in cell surface, avoid the arterial bleeding by hypertension, and prolong the life of erythrocytes, and so on. However, overload of cholesterol leads to arteriosclerosis associated with leading death cause. Lack of physical activity, emotional and environmental stress, and low intake of protein or vitamin E induce the unbalance between HDL- and LDL-cholesterol so become a basis of ischemic disorders in heart, brain and elsewhere in the body. So far, four major classes of medications for hyperlipidemia are HMG-CoA reductase inhibitors (statins), bile acid sequestrants, nicotinic acid, and fibric acids. The statins can lower LDL and levels triglyceride, but may induce myopathy and an elevation of liver enzyme levels. The bile acid sequestrants lower LDL levels and raise HDL levels with no effect on triglyceride levels but side effects of gastrointestinal (GI) distress, constipation, and a decrease in the absorption of other drugs. Nicotinic acid and fibric acids lower LDL and triglyceride levels with showing flushing, hyperglycemia, hyperuricemia, GI distress, and hepatotoxicity dyspepsia, gallstones, myopathy, and unexplained noncardiac death as adverse effects. Above western drugs lower cholesterol by 15 to 30% while all have notable adverse effects. In traditional medicine, hyperlipidemia is regarded as retention of phlegm and fluid disease. Etiology and pathogenesis of hyperlipidemia is basically based on Spleen-Deficiency and Phlegm-Stagnation, accumulation and stasis of -heat, and Qi & blood stagnation induced by Phlegm-damp, water-dampness, and blood stasis. Thereby, strengthening Spleen and dissolving Phlegm, clearing away heat and diuresis, and supplementing Qi and activating blood circulation are commonly used therapeutic methods for hyperlipidemia. The traditional herbal medicine, have been used for patients with CVA, hypertension or hyperlipidemia in Oriental hospital or Oriental clinic. The lock and key theory is used to develop most of western medicine, however many diseases are caused by mixed factors in body-complex system. We expect that Oriental pharmacological theory could be newborn as a novel drug showing high advantage of blood levels of lipidsand QOL of performance without side effects.

  • The biological carbon dioxide fixation using microalgae has been known as an effective carbon dioxide reduction technology. With many environmental factors influencing microalgal productivity, the desirable cultivation factors were investigated using a green alga, Euglena gracilis. It has the high protein and vitamin E to be used as fodder. In batch culture with a photobioreactor, initial pH, temperature, carbon dioxide concentration and light intensity in the optimum cultivation condition were 3.5, $27^{\circ}C$,5-10% and $520{\mu}mol/m^2/s$, respectively. After that, the optimum hydraulic retention time (HRT for the continuous cultivation was 4 days at carbon dioxide concentration of 10%. In this condition, the final dry cell weight was 1.2g/l.

Effects of Replacing Ground Corn with Cassava Chip in Concentrate on Feed Intake, Nutrient Utilization, Rumen Fermentation Characteristics and Microbial Populations in Goats

  • Ten male crossbred (Thai $Native{\times}Anglo$ Nubian) goats with average live weight of $27{\pm}2$ kg were randomly assigned according to a $5{\times}5$ replicated Latin square design to receive five diets, $T_1$ = concentrate with 0% cassava chip (CC), $T_2$ = 25% CC, $T_3$ = 50% CC, $T_4$ = 75% CC and $T_5$ = 100% CC. Fresh elephant grass (FEG) was offered ad libitum as the roughage. A metabolism trial lasted for 21 days during which liveweight changes and feed intakes were measured. Based on this experiment, there were no significant differences (p>0.05) among treatment groups regarding DM intake and digestion coefficients of nutrients (DM, OM, NDF and ADF), except for $T_5$ (100% CC) which was lowest (p<0.05) in digestion coefficient of CP than $T_1$ and $T_3$. Rumen parameters (ruminal pH, $NH_3$-N and volatile fatty acids), blood urea nitrogen, blood glucose and packed cell volume were similar among treatments. Moreover, rumen microorganism populations were not affected (p>0.05) by cassava inclusion. The amount of N absorption and retention were similar among treatments, except for $T_5$ which tended to be slightly lower. Based on this experiment, it could be concluded that the optimal level of cassava inclusion to replace corn in goat diets was in the range of 25-75% of CC when fed with FEG and it was a good approach in exploiting the use of local feed resources for goat production.

Effects of Sago Palm Pith as Replacement for Corn Grain on Intake, Rumen Fermentation Characteristics and Microbial N Supply of Cattle Fed Paspalum plicatulum Hay

  • To investigate the effects of sago palm pith (SPP) substitution of corn in the diets on intake, digestibility, rumen fermentation characteristics, nitrogen balance and microbial N Supply, five ruminally fistulated Southern indigenous cattle (mean initial BW = 226${\pm}$5 kg) were randomly assigned to a 5${\times}$5 Latin Square Design to receive five diets, $T_1$ = concentrate with 0% SPP, $T_2$ = 25% SPP, $T_3$ = 50% SPP, $T_4$ = 75% SPP and $T_5$ = 100% SPP, of dietary dry matter, respectively. Plicatulum hay (PH) was offered ad libitum as the roughage. A metabolism trial lasted for 21 days during which liveweight changes and feed intakes were measured. Based on this experiment, there were no significant differences (p>0.05) among treatments groups regarding total DM intake (OMI, NDFI and ADFI) and digestion coefficients of nutrients (DM, OM, CP, NDF and ADF), while total DM intake (% BW) was significantly (p<0.05) higher as higher levels of SPP were incorporated into diets. Rumen parameters (ruminal temperature, pH, glucose, packed cell volume, volatile fatty acid and rumen microorganism populations) were similar among treatments (p>0.05), whereas $NH_3-N$, blood urea nitrogen and molar proportion of propionate concentrations were significantly (p<0.05) higher as higher levels of SPP were incorporated into diets. The amount of N absorption, N retention and microbial protein synthesis were similar among treatments. These results indicate that SPP can be included in diets for Southern indigenous cattle to supply up to 100% of supplemental corn when fed with PH without negative impact on animal performance and it was a good approach in exploiting the use of local feed resources for beef cattle production.

Effects of Chongmyung-tang, Polygalae Radix and Acori Graminei Rhizoma on $A{\beta}$ Toxicity and Memory Dysfunction in Mice (총명탕, 원지, 석창포가 베타아밀로이드로 유발된 학습과 기억장애에 미치는 영향)

  • Objectives : This study investigated the protective effects of the water extracts of Chongmyung-tang, Polygalae Radix, and Acori Graminei Rhizoma in an in vivo Alzheimer's disease (AD) mouse model. Methods : Memory impairment was induced by an intraventricular injection of $A{\beta}25-35$ peptides and subsequently Chongmyung-tang, Polygalae Radix, or Acori Graminei Rhizoma extract were administered orally for 14days. Results : In the water maze task, Chongmyung-tang, Polygalae Radix, and Acori Graminei Rhizoma extracts improved learning ability during the acquisition period and significantly increased memory scores during the retention period versus $A{\beta}-injected$ controls. Furthermore, the toxicity of $A{\beta}25-35$ on hippocampus was assessed immunohistochemically (Tau, MAP2, TUNEL, Bax) and by in vitro study. Chongmyung-tang, Polygalae Radix, and Acori Graminei Rhizoma demonstrated significant neuroprotective effects against oxidative damage and apoptotic cell death of hippocampal neurons damaged by $A{\beta}25-35$. Conclusions : These results suggested that Chongmyung-tang, Polygalae Radix and Acori Graminei Rhizoma extract improve memory impairment and reduce Alzheimer's dementia via anti-apoptotic effects and by modulating the expressions of Tau and MAP2 protein in the hippocampus.

Effect of Ozone Injection on Dewaterability and Thickening of Sewage Sludge (하수 슬러지 농축 및 탈수성에 미치는 오존의 영향)

  • It is required to propose an alternatives for appropriate sludge treatment owing to persistent construction of sewage treatment plant and increase of sewage sludge quantity. In order to treat sludge more efficiently, the methods which reduce the cost of sludge treatment have been studied such as sludge reduction and conditioning. Especially ozone treatment reduces solid quantity and improves separation of solid-liquid at the same time. Therefore ozone treatment have a positive effect on reduction and stabilization of sludge. So, this study applied ozone to sewage sludge and induced cell destruction of sludge. By comparing with the correlation between thickening and dewatering, and evaluating moisture content and solubilization of cake, this study verificates the effect of process improvement for ozone pre-treatment. In J-STP case, according to ozone dose solid flux increased about 12 times from $1kg/m^2{\cdot}h$ to $12kg/m^2{\cdot}h$. Also this plant were capable to shorten thickening time from 40 minutes to 6~7 minutes. Thus it is expected to reduce volume and retention time of thickener. On pH effect factor, dewatering at pH4 was more than at pH11, $3.05{\times}10^{11}$ and $3.82{\times}10^{11}(m/kg)$. But effect of pH was analogous to ozone, $2.81{\times}10^{11}(m/kg)$. The effect of pH on thickening was similar to law sludge, $0.68(kg/m^2{\cdot}h)$, and the effect of ozone injection on thickening was the biggest, $3.45(kg/m^2{\cdot}h)$. The COD solubilization rate improved from about 5 to 30%. So it is judged that we are able to utilize most solubilized sludge to another sewage treatment plants.

Hypoxia Induced Multidrug Resistance of Laryngeal Cancer Cells via Hypoxia-inducible Factor-1α

  • Objectives: To investigate whether hypoxia has an effect on regulation of multidrug resistance (MDR) to chemotherapeutic drugs in laryngeal carcinoma cells and explore the role of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$). Methods: Laryngeal cancer cells were cultured under normoxic and hypoxic conditions. The sensitivity of the cells to multiple drugs and levels of apoptosis induced by paclitaxel were determined by MTT assay and annexin-V/propidium iodide staining analysis, respectively. HIF-$1{\alpha}$ expression was blocked by RNA interference. The expression of HIF-$1{\alpha}$ gene was detected by real-time quantitative RT-PCR and Western blotting. The value of fluorescence intensity of intracellular adriamycin accumulation and retention in cells was evaluated by flow cytometry. Results: The sensitivity to multiple chemotherapy agents and induction of apoptosis by paclitaxel could be reduced by hypoxia (P<0.05). A the same time, the adriamycin releasing index of cells was increased (P<0.05). However, resistance acquisition subject to hypoxia in vitro was suppressed by down-regulating HIF-$1{\alpha}$ expression. Conclusion: HIF-$1{\alpha}$ could be considered as a key regulator for mediating hypoxia-induced MDR in laryngeal cancer cells via inhibition of drug-induced apoptosis and decrease in intracellular drug accumulation.
