This study was designed to investigate the protective effect of Bojungmyunyuk-dan(BJMY-Dan) on the cisplatin-induced cytotoxicity of primary rat astrocytes. BJMY-Dan is an oriental herbal prescription for its ability to recover protective effects against anti-cancer chemotherapies. After astrocytes were treated cisplatin, MTT assay was performed for cell viability test. To explore the mechanism of cytotoxicity, I used the several measures of apoptosis to determine whether this processes was involved in cisplatin-induced cell damage in astrocytes. Also, astrocytes were treated with BJMY-Dan and then, followed by the addition of cisplatin. Cisplatin decreased the viability of astrocytes in a dose and time-dependent manner. BJMY-Dan increased the viability of astrocytes treated cisplatin. Astrocytes treated cisplatin were revealed as apoptosis characterized by nuclear staining and flow cytometry. BJMY-Dan protected astrocytes from cisplatin-induced nuclear fragmentation and chromatin condensation. Also, caspase-3 and caspase-9 proteases were activated in astrocytes by cisplatin. BJMY-Dan inhibited the activation of caspase proteases in cisplatin-treated astrocytes. Cleavage of [poly(ADP-ribose) polymerase](PARP) was occurred at 12hr after treatment of cisplatin in astrocytes. BJMY-Dan recovered the cleavage of PARP in cisplatin-treated astrocytes. Also, BJMY-Dan inhibited the activation of pro-apoptotic factor, Bak by cisplatin. Lastly, astrocytes stained with JC-1 and Rhodamine 123 were photographed by fluorescence microscope to visualize changes of mitochondrial membrane permeability transition(MPT) during treatment with cisplatin for 24hr. BJMY-Dan recovered the change of MPT by cisplatin in astrocytes. According to above results, BJMY-Dan may protect astrocytes from cytotoxicity induced by chemotherapeutic agents, including cisplatin.
Translocator protein 18 kDa (TSPO) is a mitochondrial outer membrane protein and is abundantly expressed in a variety of organ and tissues. To date, the functional role of TSPO on vascular endothelial cell activation has yet to be fully elucidated. In the present study, the phorbol 12-myristate 13-acetate (PMA, 250 nM), an activator of protein kinase C (PKC), was used to induce vascular endothelial activation. Adenoviral TSPO overexpression (10-100 MOI) inhibited PMA-induced vascular cell adhesion molecule-1 (VCAM-1) and intracellular cell adhesion molecule-1 (ICAM-1) expression in a dose dependent manner. PMA-induced VCAM-1 expressions were inhibited by Mito-TEMPO ($0.1-0.5{\mu}m$), a specific mitochondrial antioxidants, and cyclosporin A ($1-5{\mu}m$), a mitochondrial permeability transition pore inhibitor, implying on an important role of mitochondrial reactive oxygen species (ROS) on the endothelial activation. Moreover, adenoviral TSPO overexpression inhibited mitochondrial ROS production and manganese superoxide dismutase expression. On contrasts, gene silencing of TSPO with siRNA increased PMA-induced VCAM-1 expression and mitochondrial ROS production. Midazolam ($1-50{\mu}m$), TSPO ligands, inhibited PMA-induced VCAM-1 and mitochondrial ROS production in endothelial cells. These results suggest that mitochondrial TSPO can inhibit PMA-induced endothelial inflammation via suppression of VCAM-1 and mitochondrial ROS production in endothelial cells.

Previously, we reported LVH peptiede derived from Aurelia aurita as cosmecetuticals with anti-winkle activity. In this study, we synthesized pal-LVH using palmitic acid to enhance skin permeability of LVH and examined the effects as cosmeceuticals of pal-LVH. To evaluate these effects, we performed cell toxicity, wound healing, and patch test for skin irritation with LVH and pal-LVH and compared these results for their effects. As a result. pal-LVH was not showed in cytotoxicity and allergenic effect like as LVH. Besides, pal-LVH had almost same excellent anti-ageing properties in high concentration and anti-winkle effect in low concentrationwas as LVH. These results suggested synthesis of palimitic acid and LVH did not affect any functions as cosmeceuticals with increasing skin permeability. Therefore, pal-LVH can be adaptable as new cosmecetuticals with anti-winkle and anti-ageing materials and applied in the development of medicine through various convergence study.

In this study, the cellular protective effect of isoquercitrin against $H_2O_2$ and rose bengal-indued HaCaT cell damage was investigated. The ethosome and elastic liposome for enhanced transdermal delivery were prepared. Particle size, loading efficiency and cumulative permeated amounts of them were evaluated. Isoquercitrin didn't show any characteristic cytotoxicity at 50 ${\mu}M$. When HaCaT cells were treated with 5 mM $H_2O_2$ and 25 ${\mu}M$ rose bengal, isoquercitrin protected the cells against the oxidative damage in a concentration dependent manner (6.25 ~ 50 ${\mu}M$). The size of 0.03 % isoquercitrin loaded ethosome was 222.85 nm and the loading efficiency was 82.26 %. The ethosome loaded with 0.03 % isoquercitrin was stable and maintained the constant particle size for 2 weeks after being prepared. The ethosome exhibited more enhanced skin permeability than general liposome and ethanol solution. The optimal ratio of lipid to surfactant of 0.1 % isoquercitrin loaded elastic liposomes was observed to be 89 : 5 through evaluating particle size (341.2 nm), deformability index (59.89), loading efficiency (54.3 %), and skin permeability (54.4 %).
Vanadium redox flow batteries (VRFBs) using the electrolytes containing various vanadium ions in sulfuric acid as supporting solution are one of the energy storage devices in alternatively charging and discharging operation modes. The positive electrolyte contains $V^{5+}/V^{4+}$ and the negative electrolyte $V^{2+}/V^{3+}$ depending on the operation mode. To prevent the mixing of two solutions, proton exchange membranes are mainly used in VRFBs. Nafion 117 could be the most promising candidate due to the strong oxidative property of $V^{5+}$ ion, but causes high crossover of electroactive species to result in a decrease in coulombic efficiency. In this study, the composite membranes using Nafion ionomer and porous polyethylene substrate were prepared to keep good chemical stability and to decrease the cost of membranes, and were compared to the properties and performance of the commercially available electrolyte membrane, Nafion 117. As a result, the water uptake and ionic conductivity of the composite membranes increased as the thickness of the composite membranes increased, but those of Nafion 117 slightly decreased. The permeability of vanadium ions for the composite membranes significantly decreased compared to that for Nafion 117. In a single cell test for the composite membranes, the voltage efficiency decreased and the coulombic efficiency increased, finally resulting in the similar energy efficiency. In conclusion, the less cost of the composite membranes by decreasing 6.4 wt.% of the amount of perfluorinated sulfonic acid polymer due to the introduction of porous substrate and lower vanadium ion permeability to decrease self-discharge were achieved than Nafion 117.
In this study, polymer composite membranes based on sulfonated poly(arylene ether sulfone) (SPAES) were prepared using a solution casting method with different amount of vermiculite (VMT) content. The dispersion of VMT particles in the SPAES matrix was confirmed by means of a scanning electron microscopy observation. The composite membrane containing less than 1 wt% of VMT has a smooth skin on the top and bottom, which means there is a good dispersion of VMT in the matrix. The water uptake of the composite membranes gradually increases as the temperature increases, and the results confirm that all the adsorbed water is bound water because VMT has a strong water affinity on account of its high cation exchange value. A composite membrane with a VMT content of less than 1 wt% increases the proton conductivity and reduces the methanol permeability. Of all the composite membranes, the membrane SPAES/VMT 1.0 has the best fuel cell performance in terms of membrane selectivity. The performance value of SPAES/VMT 1.0 is double that of Nafion 112, which suggests that SPAES/VMT1.0 could be an excellent candidate for direct methanol fuel cells.

The aim of this study was to investigate whether a novel formulation of an herbal extracts has an inhibitory effect on obesity. To determine its anti-obesity effects, we performed anti-obesity-related experiments in vitro and in vivo. Thus, our present study was carried out to evaluate the anti-obesity effect of herbal extracts using a high fat diet (HFD)-induced obese mouse model and 3T3-L1 adipose cells. The effects of each herbal extracts on lipid accumulation in 3T3-L1 cells were examined using Oil Red O staining. Results showed that treatment with each herbal extracts at $10{\sim}100{\mu}g/ml$ had no effect on cell morphology and viability. Without evidence of toxicity, herbal extracts treatment decreased lipid accumulation compared with the untreated adipocytes controls as shown by the lower absorbance of Oil Red O stain. Futhermore, compared with control-differentiated mature adipocytes, each herbal extracts significantly inhibited lipid accumulation in mature 3T3-L1 adipocytes. In the HFD-fed obese mice, body weight, liver weight and white adipose tissue weights were significantly reduced by mixture of herbal extracts administration in mouse skin. Futhermore, we found that mixture of herbal extracts administration suppressed serum triglyceride (TG), and total cholesterol (TCHO) in HFD-induced obese mouse model. The mixture of herbal extracts of permeability was estimated by measuring the transepithelial electrical resistance (TEER) value in pig skin. The optimized formulations of herbal extracts (Test 3 formulation) showed skin permeation. However, test 1 formulation containing essential oil as enhancer showed maximum skin permeation. After confirming the enhanced skin permeability, in vivo studies were performed to assess whether skin irritation potential on the basis of a primary irritation index (PII) in rabbit skin. Reactions were scored for erythema/edema reactions at 24 h, 48 h and 72 h post-application. It was concluded that the test 1 formulation was not irritation (PII = 0). The present study suggests that the test 1 formulation might be of therapeutic interest with respect to the treatment of obesity.

Somatic cell nuclear transfer (SCNT) has generally demonstrated that a differentiated cell can convert into a undifferentiated or pluripotent state. In the SCNT experiment, nuclear reprogramming is induced by exposure of introduced donor nuclei to the recipient cytoplasm of matured oocytes. However, because the efficiency of SCNT still remains low, a combination of SCNT technique with the ex-ovo method may improve the normal development of SCNT embryos. Here we hypothesized that treatment of somatic cells with extracts prepared from the germinal vesicle (GV) stage Siberian sturgeon oocytes prior to their use as nuclear donor for SCNT would improve in vitro development. A reversible permeability protocol with $4{\mu}g/mL$ of digitonin for 2 min at $4^{\circ}C$ in order to deliver Siberian sturgeon oocyte extract (SOE) to porcine fetal fibroblasts (PFFs) was carried out. As results, the intensity of H3K9ac staining in PFFs following treatment of SOE for 7 h at $18^{\circ}C$ was significantly increased but the intensity of H3K9me3 staining in PFFs was significantly decreased as compared with the control (p<0.05). Additionally, the level of histone acetylation in SCNT embryos at the zygote stage was significantly increased when reconstructed using SOE-treated cells (p<0.05), similar to that of IVF embryos at the zygote stage. The number of apoptotic cells was significantly decreased and pluripotency markers (Nanog, Oct4 and Sox2) were highly expressed in the blastocyst stage of SCNT embryos reconstructed using SOE-treated cells as nuclear donor (p<0.05). And there was observed a better development to the blastocyst stage in the SOE-treated group (p<0.05). Our results suggested that pre-treatment of cells with SOE could improve epigenetic reprogramming and the quality of porcine SCNT embryos.

Ceramic foams are prepared as positive images corresponding to a plastic foam structure which exhibits high porosities (85-90%). This structure makes the ceramic foams attractive as a catalyst in a dry reforming process, because it could reduce a high pressure drop problem. This problem causes low mass and heat transfers in the process. Furthermore, the reactants would shortly contact to catalyst surface, thus low conversion could occur. Therefore, this research addressed the preparation of dry reforming catalysts using a sol-gel catalyst preparation via a polymeric sponge method. The specific objectives of this work are to investigate the effects of polymer foam structure (such as porosity, pore sizes, and cell characteristics) on a catalyst performance and to observe the influences of catalyst preparation parameters to yield a replica of the original structure of polymeric foam. To accomplish these objectives industrial waste foams, polyurethane (PU) and polyvinyl alcohol (PVA) foams, were used as a polymeric template. Results indicated that the porosity of the polyurethane and polyvinyl alcohol foams were about 99% and 97%. Their average cell sizes were approximate 200 and 50 micrometres, respectively. The cell characteristics of polymer foams exhibited the character of a high permeability material that can be able to dip with ceramic slurry, which was synthesized with various viscosities, during a catalyst preparation step. Next, morphology of ceramic foams was explored using scanning electron microscopy (SEM), and catalyst properties, such as; temperature profile of catalyst reduction, metal dispersion, and surface area, were also characterized by $H_2-TPR$ and $H_2-TPD$ techniques, and BET, respectively. From the results, it was found that metal-particle dispersion was relatively high about 5.89%, whereas the surface area of ceramic foam catalysts was $64.52m^2/g$. Finally, the catalytic behaviour toward hydrogen production through the dry reforming of methane using a fixed-bed reactor was evaluated under certain operating conditions. The approaches from this research provide a direction for further improvement of marketable environmental friendly catalyst production.
Background and Objectives : Sulfur dioxide gas is one of the major airborne Pollutants noxious to human in industrialized countries. The most vulnerable areas in the human respiratory system were the trachea and main bronchi and a gradient of decreasing damage was observed in the peripheral tracheobronchial tree. Induced functional alteration was increased mucosal permeability, and morphological changes were epithelial sloughing, intracellular edema, mitochondrial swelling, widened intercellular spaces, and ciliary cytoplamic extrusions. The laminins are a family of extracellular matrix glycoproteins localized in the basement membrane. Their primary role is cell-matrix attachment, but many additional biologic activities, including Promoting cell growth and migration, tumor growth and metastasis, wound repair, and graft survival, have been demonstrated. Materials and Methods : Histologic changes and expression of laminin in tracheal mucosa sacrificed at 1 day, 2 day, 3 day, 1 week, 2 weeks, and 3 weeks after continued SO2 exposure of 250 ppm for 30 minutes a day(to 7week) were studied in rats. In this study, mild immune reaction for laminin was noted at the apical cytoplasm of epithelial cells and basement membrane one day after a 7 week $SO_2$ exposure. The cilia and nucleoi of epithelial cells were normal and no immune reaction was noted in Goblet cells. The lamina propria of the tracheal tissue was infiltrated by monocytes and lymphocytes. Results : At 24 hours after exposure, all tracheal cells except Goblet cells revealed a mild immune reaction for laminin. No immune reactions were noted in the basement membrane. At 72 hours after exposure, mild or moderate immune reactions for laminin was seen in the tracheal cell cytoplasm. Irregular faint immune reaction for laminin was noted in the basement membrane. At 1 week after exposure, strong immune reaction for laminin was detected over all tracheal cells, and the basement membrane was seen clearly. At 2~3 weeks after exposure, strong immune reaction for laminin was seen in all tracheal epithelial cells except Goblet cells and a mild immune reaction was partly revealed in the basement membrane. Conclusion : Our study suggests that 502 produces histologic damage on the tracheal mucosa. Longer duration after exposure of $SO_2$ makes more progressive healing on the tracheal mucosa and increased immunoreactivity for laminin.
