Probiotics are currently considered as one of tools to modulate immune responses under specific clinical conditions. The purpose of this study was to evaluate whether oral administration of three different probiotics (Lactiplantibacillus plantarum CJLP243, CJW55-10, and CJLP475) could evoke a cell-mediated immunity in immunodeficient mice. Before conducting in vivo experiments, we examined the in vitro potency of these probiotics for macrophage activation. After co-culture with these probiotics, bone marrow derived macrophages (BMDMs) produced significant amounts of proinflammatory cytokines including interleukin-6 (IL-6), IL-12, and tumor necrosis factor-α (TNF-α). Levels of inducible nitric oxide synthase (inos) and co-stimulatory molecules (CD80 and CD86) were also upregulated in BMDMs after treatment with some of these probiotics. To establish an immunocompromised animal model, we intraperitoneally injected mice with cyclophosphamide on day 0 and again on day 2. Starting day 3, we orally administered probiotics every day for the last 15 d. After sacrificing experimental mice on day 18, splenocytes were isolated and co-cultured with these probiotics for 3 d to measure levels of several cytokines and immune cell proliferation. Results clearly indicated that the consumption of all three probiotic strains promoted secretion of interferon-γ (IFN-γ), IL-1β, IL-6, IL-12, and TNF-α. NK cell cytotoxicity and proliferation of immune cells were also increased. Taken together, our data strongly suggest that consumption of some probiotics might induce cell-mediated immune responses in immunocompromised mice.

Objective: To evaluate the radiological tumor response patterns and compare the response assessments based on immune-based therapeutics Response Evaluation Criteria in Solid Tumors (iRECIST) and RECIST 1.1 in metastatic clear-cell renal cell carcinoma (mccRCC) patients treated with programmed cell death-1 (PD-1) inhibitors. Materials and Methods: All mccRCC patients treated with PD-1 inhibitors at Henan Cancer Hospital, China, between January 2018 and April 2019, were retrospectively studied. A total of 30 mccRCC patients (20 males and 10 females; mean age, 55.6 years; age range, 37-79 years) were analyzed. The target lesions were quantified on consecutive CT scans during therapy using iRECIST and RECIST 1.1. The tumor growth rate was calculated before and after therapy initiation. The response patterns were analyzed, and the differences in tumor response assessments of the two criteria were compared. The intra- and inter-observer variabilities of iRECIST and RECIST 1.1 were also analyzed. Results: The objective response rate throughout therapy was 50% (95% confidence interval [CI]: 32.1-67.9) based on iRECIST and 30% (95% CI: 13.6-46.4) based on RECIST 1.1. The time-to-progression (TTP) based on iRECIST was longer than that based on RECIST 1.1 (median TTP: not reached vs. 170 days, p = 0.04). iRECIST and RECIST 1.1 were discordant in 8 cases, which were evaluated as immune-unconfirmed PD based on iRECIST and PD based on RECIST 1.1. Six patients (20%, 6/30) had pseudoprogression based on iRECIST, of which four demonstrated early pseudoprogression and two had delayed pseudoprogression. Significant differences in the tumor response assessments based on the two criteria were observed (p < 0.001). No patients demonstrated hyperprogression during the study period. Conclusion: Our study confirmed that the iRECIST criteria are more capable of capturing immune-related atypical responses during immunotherapy, whereas conventional RECIST 1.1 may underestimate the benefit of PD-1 inhibitors. Pseudoprogression is not rare in mccRCC patients during PD-1 inhibitor therapy, and it may last for more than the recommended maximum of 8 weeks, indicating a limitation of the current strategy for immune response monitoring.
In order to understand what leads to the appearance of harmful Chattonella algae in the Yeosu coastal waters of Korea, we measured environmental parameters every week at one station from May to November, 2006, and April to October, 2007. Four species of Chattonella appeared during the monitoring period: C. antiqua, C. globosa, C. marina and C. ovata. The range of water temperature and salinity were $15.0-27.9^{\circ}C$ and 17.6~33.0 psu, respectively, when Chattonella appeared, and their maximum cell density (4,840 cells/L) was at $27.1^{\circ}C$ and 33.0 psu. During the monitoring periods, the range of dissolved inorganic nitrogen (DIN), phosphate (DIP) and chlorophyll $\alpha$ (Chl-$\alpha$) concentrations in surface waters were $1.20-52.23\;{\mu}M$ ($8.59{\pm}8.97\;{\mu}M$), $0.03-1.56\;{\mu}M$ ($0.47{\pm}0.31\;{\mu}M$) and $0.45-31.12\;{\mu}g/L$ ($3.58{\pm}4.77\;{\mu}g/L$), respectively. Chattonella occurred at low cell density when the Chl-$\alpha$ concentration increased because of supplied nutrients, whereas their cell density increased during the periods of rapid decrease in Chl-$\alpha$. The results of growth experiments based on batch culture showed that the half saturation constant ($K_s$) of C. antiqua on ammonium (${NH_4}^-$), nitrate (${NO_3}^-$) and phosphate (${PO_4}^{2-}$) were $3.89{\mu}M$, $5.01\;{\mu}M$ and $0.63\;{\mu}M$, respectively. These Ks values are higher than those reported for diatoms and other flagellates at the DIP concentration (average $0.47{\mu}M$) of Yeosu coastal waters. Although the maximum specific growth rate (${\mu}_{max}$) of C. antiqua was lower than diatoms, it was higher than those of other flagellates. Therefore, our results indicate that the DIP level in the study area was too low to support Chattonella blooms, although Chattonella species have physiological characteristics that enable them to grow more rapidly than other flagellates when nutrient levels are higher than their $K_s$.
기존의 동물모델에서 암의 성장은 caliper를 이용하여 고형암 부피를 측정으로써 조사하였으나, 암 조직 속의괴사와 부종으로 인하여 부피측정에 신뢰성이 결여 되어 있다. 이러한 문제점을 해결하기 위해 발광 암세포를 이용하여 광학생체영상적으로 분석하는 방법이 개발 되었다. 본 연구에서는 전립선 발광 암세포를 제조하여 고형암 동물모델에서 B16 발광 암세포와 암 성장을 비교 측정하여 신규발광 암세포를 평가하였다. In vitro에서 세포 수와 발광강도는 높은 상관관계를 보였고($R^2$=0.99), 고형암 동물모델에서 암 성장 측정은 괴사에 의한 오차를 줄였다. 이러한 발광신호를 기반으로 한 측정방법은 caliper의 부피 측정에 비하여 높은 항암효과를 보임으로써 기존의 발광 암세포보다 신규 발광전립선 암세포의 유용성을 증명하였다.
현재 국내에서는 비탈면 및 구조물 안정성 확보를 목적으로 네일, 록볼트 등과 함께 그라운드 앵커 공법을 사용하고 있다. 이 중 장기목적으로 사용되는 영구앵커의 경우 사용기간 중 지지력과 내구성이 확보되어야 하나, 최근 연구 결과에 따르면 장기거동에 따른 잔존 긴장력 감소와 비탈면 변형 등의 현상이 보고되고 있다. 이와 같은 잔존 긴장력 감소 문제는 앞으로 지속적으로 증가될 것으로 전망되며, 이로 인한 유지관리 비용 증가 등의 문제가 불가피할 것으로 보인다. 이에 본 연구에서는 국내·외 문헌연구를 통해 영구앵커의 긴장력에 영향을 미치는 요인들을 파악하였으며, 과거 수행된 하중계 모니터링 자료를 분석한 선행연구들을 조사하였다. 이후, 이를 기초자료로 활용하여 실제 현장에서 수집한 하중계 계측자료를 분석하여 앵커의 긴장력 감소현황을 파악하였고, 그 장기하중감소특성을 분석하였다. 마지막으로 앞의 내용을 종합하여, 설치 직후 100일 부근의 단기 데이터를 통해 영구앵커의 장기하중감소특성을 예측하는 기법을 제안하였다.
FMS(Flexible Manufacturing System)는 로봇, 수치제어기기 등의 프로그램이 가능한 생산설비 하드웨어로 구성된 분산시스템이다. FMS의 효과적인 운용을 위해서는 생산설비 하드웨어를 통합 제어하기 위한 소프트웨어의 존재가 필수적이다. 그런데 FMS의 복잡성과 다양한 변화가능성은 통합제어 소프트웨어의 구축 및 유지, 보수를 어렵게 한다. 이를 해결하기 위하여 본 논문에서는 객체지향 FMS 통합제어 모델을 제안한다. 제안된 모델에서는 FMS에 실재하는 셀이 가상 셀 객체로 모델되어, 통합제어를 위하여 필요한 데이터와 연산들이 캡슐화되어 객체지향 데이타베이스에 관리된다. 또한 이 밖의 FMS운용을 위해 필수적인 데이타와 이들간의 관계성도 데이타베이스에 관리된다. 본 논문은 제안된 모델을 기반으로 구현한 통합제어 소프트웨어시스템인 FREE(Fms Run Time Executive Environment)를 바탕으로 통합제어 데이타베이스에 대하여 기술한다.
The purpose of this study was to determine the effects of heat application on the immune activities of the human body. To exam, furthermore, the immune effect from the healthy volunteer(male:15, female:15) by monitoring changes of immune substances such as various leukocytes[total white blood cell(WBC), eosinophil, neutrophil, basophil, monocyte, and lymphocyte], a comparative study with warm water immersion($40.8{\pm}0.3^{\circ}C$) and infrared(250W) was carried out. The plasma analysis showed that the count of white blood cell, eosinophil, and neutrophil were elevated in warm water immersion- or infrared. stimulated group compared with control group. However, the count of basophil was decreased in both warm water immersion- and infrared-stimulated group than control group. Therefore, these results suggest that the thermostimulation improved immune activity.
Fluorscence in situ hybridization with chromosome-specific probe has been shown to be a valid and rapid method for detection of chromosome rearrangements induced by chemical and physical agents. This method is useful for quantifying structural aberrations, expecially for stable ones, such as translocation and insertion, which are difficult to detect with conventional method in human lymphocyte. In order to use the FISH method as a biodosimeter for monitoring human population exposed to various chemical and physical agent, baseline level of chromosome rearragement was established. Blood from forty four healthy adults were collected and analysed with whole chromosome-specific probes by human chromosome 1,2 and 4. The frequencies of stable translocation were 2.45 per 100 cell equivalent and those of insertion, color juction, acentric and dicentric were 0.32, 3.28, 0.23 and 0.27 per 100 cell equivalent respectively. The frequencies of chromosome rearragements increased with age in both sexes except for dicenrics. From above result, stable aberrations accumulate with age and it may reflect integrated lifetime exposure of adverse environment.
The $(Sr_{0.75},\;La_{0.25})TiO_3$ (SLTO) ultra-thin films with various thicknesses have been grown on Ti-O terminated $SrTiO_3$(100) substrate using Laser-Molecular Beam Epitaxy (Laser MBE). By monitoring the in-situ specular spot intensity oscillation of reflection high energy electron diffraction (RHEED), we controlled the layer-by-layer film growth. The film structure and topography were verified by atomic force microscopy (AFM) and high resolution thin film x-ray diffraction by the synchrotron x-ray radiation. We have also investigated the electronic band structure using x-ray absorption spectroscopy (XAS). The ultra thin SLTO film exhibits thickness driven metal-insulator transition around 8 unit cell thickness when the film thickness progressively reduced to 2 unit cell. The SLTO thin films with an insulating character showed band splitting in Ti $L_3-L_2$ edge XAS spectrum which is attributed to Ti 3d band splitting. This narrow d band splitting could drive the metal-insulator transition along with Anderson Localization. In optical conductivity, we have found the spectral weight transfer from coherent part to incoherent part when the film thickness was reduced. This result indicates the possibility of enhanced electron correlation in ultra thin films.
In this study, body pressure was quantitatively detected using built-in optical sensors, inside an air cushion seat. The proposed system visualizes the effect of the body pressure distribution on the air cushion seat. The built-in sensor is based on the time-of-flight (ToF) optical method, instead of the conventional electrical sensor. A ToF optical sensors is attached to the bottom surface of the air-filled cells in the air cushion. Therefore, ToF sensors are durable, as they do not come in physical contact with the body even after repeated use. A ToF sensor indirectly expresses the body pressure by measuring the change in the height of the air-filled cell, after being subjected to the weight of the body. An array of such sensors can measure the body pressure distribution when the user sits on the air cushion seat. We implemented a prototype of the air cushion seat equipped with 7 ToF optical sensors and investigated its characteristics. In this experiment, the ToF optical pressure sensor successfully identified the pressure distribution corresponding to a sitting position. The data were accessed through a mobile device.
