파두유를 주입한 육계병아리에서 사료중 크릴밀 수준이 세포성 면역에 미치는 영향 (Effect of Dietary Krill Meal Levels on the Cell Mediated Immunity in Intra-muscularly Croton Oil Injected Broiler Chicks)

  • 근육내 파두유를 주입한 육계병아리에서 사료 중 크릴 밀 수준이 세포성 면역반응에 미치는 영향을 조사하였다. 갓 부화한 육계병아리(Ross) 수컷에 크릴 밀 0.0(기초사료), 0.5, 1.0 및 2.0% 사료를 급여하고 3주간 사육하였다. 20일령에 10㎕의 파두유를 정강이 근육내에 주입하였고, 24시간 뒤(21일령)에 혈중 TNF-α 활성과 오보트렌스훼린 수준 및 PBMC와 비장세포의 증식도를 올리브유를 주입한 대조구와 비교하였다. 크릴 밀 사료는 육계병아리의 생산성과 혈장 오보트렌스훼린 수준에 유의한 영향을 미치지 않았으나, 크릴 밀 0.0% 사료에 비해서 유의하게(p<0.0001) 혈장 TNF-α 활성을 감소시키고 PBMC 증식도를 높였다. 비장 세포증식도는 크릴 밀 1.0% 사료에서 유의하게(p=0.01) 높았다. 파두유 주입은 혈중 TNF-α 활성(p<0.0001), 오보트랜스훼린 수준 및 PBMC증식도(p<0.0001)를 높였다. 파두유 주입 시에 PBMC 증식도는 사료중 크릴 밀 수준에 따라 점차 감소(p<0.05) 하였고 비장세포 증식도는 크릴 밀 1.0 및 2.0% 사료에서 유의하게(p< 0.05) 감소하였다. 본 연구는 사료중 크릴밀이 파두유 주입으로 활성화한 선천 면역과 세포성 면역을 변화시킨다는 것을 나타내었다.   • Invasive pulmonary aspergillosis is a disease occuring predominantly in patients with defects in immunity such as neutropenia, hematologic malignancies or with defects in cell-mediated immunity. The isolation of Aspergillus from respiratory tract of normal host usually signifies tracheobronchial colonization, making this diagnosis difficult. There are isolated case reports occuring in normal hosts but most of them were diagnosed postmortem at autopsies indicating that early diagnosis of invasive aspergillosis in normal host is difficult. We describe here a case of invasive aspergillosis in a immunocompetent host diagnosed by lung biopsy which was successfully treated with Amphotericin-B. Invasive pulmonary aspergillosis should be included as one of the differential diagnosis if a patient with pneumonic consolidation does not respond to usual therapy, even if the patient does not have any defect in immunity.

  • 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has been reported to exert detrimental toxicities on various organ systems including reproductive, cardiovascular, nervous, or dermal system. Immunomodulatory effects of TCDD is thymic atrophy, downregulation of cytotoxic T or B lymphocyte differentiation and activation, which were demonstrated using experimental animals, whereas immunotoxicity in human has not been investigated well. This study was proceeded to evaluate general immunologic spectrum of the Korean Vietnam War veterans exposed to TCDD during their operation, and compare with that of the non-exposed control subjects with similar age. Regarding composition and quantity, immune cells in peripheral blood collected from the TCDD-exposed was not much different from those of the control except decreased red blood cell, hemoglobin and hematocrit level. Furthermore, plasma IgG2, G3, and G4 isotype distribution was similar between two groups, but IgG1 level was significantly lowered in the TCDD-exposed, indicating a TCDD-mediated functional alteration of B cells. Significantly enhanced level of IgE in plasma, a hallmark of dermal or respiratory allergic response, was also observed in the TCDD-exposed compared with that of the control. Elevated generation of IL-4 and IL-10 was resulted from in vitro stimulation of T cells with PMA plus ionomycin or PHA, respectively, from the TCDD-exposed in comparison to those of the control, suggesting a skewed type-2 response. In addition, the level of IFN${\gamma}$, a multifunctional cytokine for T cell-mediated immunity, was lowered in the TCDD-exposed with upregulation of tumor necrosis factor $\alpha$. The present study suggests that TCDD exposure disturbs immunohomeostasis in humans observed as an aberrant plasma IgE and IgG1 levels and dysregulation of T cell activities.

질트리코모나스(Trichomonas waginazis)에 대한 마우스 복강 대식세포의 세포독성 (Cytotoxicity of resident and Iymphokine-activated mouse peritoneal macrophage against yrichomonas vaginalis)

  • 질트리코모나스(Trichomenas vaginalis)에 대한 마우스의 복강 대식세포 및 림포카인으 로 활성화시킨 대식세포의 세포독성을 각각 관찰하였다. 세포독성은 질트리로모나스를 3H-TdR로 label시킨 후 대식세포와 반응시켜 사멸한 원충에서 방출되는 방사능 양을 비교하여 측정하였다. 대식세포의 원충에 대한 비율을 1 : 1, 5 :1, 10 : 1, 20 : 1 및 50 : 1로 증가시키고 반응시간을 12시간 및 24시간으로 변화시켰을 때, 대식세포와 원충의 비율 10 : 1 및 24시간 반응의 경우 가장 놓은 세포독성을 보였다. 마우스 비장세포를 phytohemagglutinin으로 자극시켜 얻은 림포카인으로 활성화시킨 대식세포에서는, 아무 처리를 하지 않은 대조 대식세포와 비교하였을 때 세포독성이 유의하게 증가되었으나 세포독성이 림포카인의 희석정도와는 비례하지 않았다. 또한 질트리코모나스에 세포독성을 나타내는 세포는 주로 플라스틱에 부착하는 대식세포임을 알 수 있었다.

면역억압계군에서 Salmonella gallinarum 감염에 대한 면역반응 (Study on Immunological Response on Salmonella gallinarum in Immunosuppressed Chickens)

  • The immune response against Salmonella gallinarum infection was investigated in immunosuppresed chickens. Newly hatched chickens were treated with cyclophosphamide at the first and second day after birth and were challenged intraperitoneally with S gallinarum ($1{\times}10^7CFU/m{\ell}$) on day 6. Group 1, Immunosuppresed and Challenged group, was treated with cyclophaiphamide and challenged with S gallinarum; group 2, Immunosuppressed group, was only treated with cyclophsphamide; group 3, Challenged group, was only challenged with S gallinarum; group 4, Control group. In each group, the localization of lymphocytes of the lymphoid organs and intestine was immunohistochemically compared using a variety of monoclonal antiboies ($CD4^+$, $CD8^+$, and B lymphocyte). Also, S gallinarum were assessed by Maccallum-Goodpasture stain and immunohistochemical analysis in the paraffin-embedded intestinal tissues. In S gallinarum challenged chickens, $CD4^+$ and $CD8^+$ lymphocytes of the intestinal organs such as duodenum, jejunum, ileum and colon were increased. However, in cyclophophamide treated chickens, $CD4^+$ and $CD8^+$ lymphocytes and especially B lymphocytes of the lymphoid organs such as thymus, spleen, and bursa of Fabricius were dramatically decreased. These results suggest that cyclophsophamide is an immunosuppressive agent that especially causes depletion of B lymphocytes, suppress humoral immunity and eventually suppresses avian immune responses. Its protection against S gallinarum infection is mainly dependent on both cell-mediated mechanism and the humoral immune response.

Protective Immunity of 56-kDa Type-Specific Antigen of Orientia tsutsugamushi Causing Scrub Typhus

  • Scrub typhus, caused by infection with Orientia tsutsugamushi, is a mite-borne zoonotic disease endemic to the Asian-Pacific region. In Korea, the incidence of this disease has increased with climate changes, and over 10,000 cases of infection were reported in 2013. Although this infection is treatable with antibiotics such as doxycycline and azithromycin, an effective prophylactic vaccine against O. tsutsugamushi would be more desirable for preventing scrub typhus in endemic areas. In this study, we investigated the 56-kDa type-specific antigen (TSA56), which is a major outer membrane protein of O. tsutsugamushi, as a vaccine candidate. Intranasal immunization of recombinant TSA56 (rec56) induced a higher level of TSA56-specific IgG than that induced by intramuscular immunization of tsa56-expressing DNA (p56). Both types of immunization induced a cell-mediated immune response to TSA56, as demonstrated by the splenic cell proliferation assay. Mice immunized with p56, followed by rec56 plus heat-labile enterotoxin B subunit from E. coli, had a stronger protection from a homologous challenge with the O. tsutsugamushi Boryong strain than with other combinations. Our preliminary results suggest that an effective human vaccine for scrub typhus can include either recombinant TSA56 protein or tsa56-expressing DNA, and provide the basis for further studies to optimize vaccine performance using additional antigens or different adjuvants.

실험동물을 이용한 화분섭취의 면역안전성 평가 (Immunotoxicological Evaluation of Pollen Intake Using Mice Model)

  • Pollen has been used for Prevention or treatment of certain diseases such as diabetes, arthritis, or cancer in traditional medicine. In addition, pollen is under investigation as a host cell for a gene expression. This study was undertaken to evaluate the immunologic safety of pollen intake. BALB/c mice were administered with 500, 50,5, or 0.5 mg/kg bw of lily pollen for five times a week for four weeks through gastric intubation. Comparing the control mice administered with distilled water, no significant changes were observed in body weight gain, weight of liver, spleen, lung, and his-topathological findings of liver and kidney of the mice groups administered with the pollen. Plasma level of IgG1, IgG2a, and IgE was not different among the groups. When splenic B lymphocytes were stimulated in vitro with lipopolysaccharides for 7 days, level of IgGl and IgGwa produced in the culture supernatants was not significantly different among the groups. Furthermore, no significant alteration was observed in IL-4 and $IFN{\gamma}$ producing ability with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hours between the pollen-administered and the control mice. Overall, this study suggests that the lily pollen intake is Inducing no significant modulation of humoral and cell-mediated immunity in mice.

Benzo(a)pyrene이 마우스 자연살해세포 활설에 미치는 영향 (Effects if Benzo(a)pyrene on Natural Killer Cell Activity of Mice)

  • 발암물질인 benzo(a)pyrene(B(a)P)이 암세포에 대한 세포성 방어능을 나타내는 자연살해(natural killer(NK))세포 활성에 미치는 영향을 관찰하기 위하여 사료를 C57BL/6마우스에 노출시킨 후 NK세포활성을 MTT시험법을 이용하여 측정하였다. 정상 및 시료노출군 마우스의 NK세포활성은 표적세포인 Yac-1세포에 대한 사멸세포백분율(percent dead cell)로서 측정하였다. 정상마우스의 비장세포에 B(a)P를 노출시켰을 때 노출군들은 시료농도에 따른 활성도에 큰 변화는 없었으니 낮은 농도($2.5^{\mu}g/ml$)의 노출에서도 정상에 비하여 현격히 낮은 활성치를 보였다. 정상마우스에 시료를 투여한 후에 분리한 비장세포의 NK세포활성치는 E/T ratio 200/1에서, 1회 투여보다는 2-3회 투여군에서 유의한 감소를 나타냄으로서 반복노출의 효과를 보였다. 본 시험을 통하여 B(a)P는 마우스 면역계에 미치는 효과중 암형성에 지대한 영향을 주는 NK세포활성을 크게 저하시킴을 알 수 있었다.

화살나무 및 느릅나무 추출물이 면역계세포의 활성에 미치는 영향 (Effect of Euonymus alatus and Ulmus clavidiana var japonica on the immune system)

  • We have previously shown that crude water extract of Euonymus alatus (EA) had strong prophylactic effect against chemically induced-and tumor cell implanted-cancer, and that the mechanisms responsible for its antitumor effects were due to nonspecific enhancement of the NK cell activities and the cell mediated immunity. However, it was unknown that any components of crude extract did work so, since it consisted of several components. In this paper, we fractionated the crude watar EA-extract into several fraction such as hexane-, ethylether-, ethyl acetate-, n-butanol- and water soluble-fraction, and screened the immune regulating activities of each fraction by the evaluation of lymphokine production and activated lymphocyte proliferation. As a result of the component fraction of EA-extract, it was found that n-butanol fraction was a potent immunostimulator, and the remained water soluble fraction also contained some stimulator, But, other fraction did not showed any remarkable effect. It is therefore suggested that EA-glycosides in n-butanol fraction may be new one of the potent biological response modifiers. The present study was also undertaken in an efforts to investigate the effects of elm-bark(EB, Ulmus clavidiana var japonica), which has been used for curing ulcer and inflammation as a folk medicine without any kind of experimental evidence to support this, on the cellular- and humoral-immune responses, lymphocyte function and NK cell activities in mice. Regardless of time and duration of EB-treatment, Arthus reaction and antibody response to SRBC were not modified by EB, but delayed hypersensitivity to SRBC was significantly enhanced only when EB was treated prior to SRBC-sensitization. EB slightly inhibited the proliferation responses of splenocytes to PHA-stimulation, but it significantly augmented the responses of these cells to S aureus Cowan 1 and Con A-activation, and these effects were manifested only when EB was added at culture initiation. EB did not influence Ig secretion of spleen cells but it significantly augmented the Con A-induced 1L 2 and MIF production of splenocytes. NK cell activities of splenocytes were markedly riled when effector cells were pretreated with EB and this augmentation was dine to the increase of binding affinity of effector cells to target cells and the target cell lytic activities of effector cells. These results led to the conclusion that EB triggers increase of cellular immune responses, such as delayed hypersensitivitiy, lymphokine production and NK cell activities. Also these results suggested that EB contains potent immune stimulants, which may provide the rational basis for their therapeutic use as one of the new biological response modifiers.

  • Chronic lymphocytic leukemia (CLL) is the most common type of leukemia occurring in Western nations. In CLL it is well known that the risk of a secondary malignancy is higher than in the normal population. But in Korea, CLL is a rare type of leukemia, so there have been only a few reported cases with a secondary malignancy. CLL is characterized by progressive defects in both cell-mediated and humoral immunity. It is known that defects in the immune system of patients with CLL contribute to the development of a secondary malignancy. We experienced a case of a 71-year-old man who suffered from a chronic cough and was diagnosed with small cell lung cancer coexisting with CLL. Until this case, there was no reported case in Korea of small cell lung cancer coexisting with CLL. We now report a case of small cell lung cancer coexisting with CLL and present a literature review.
