A crude extract of Smphytum officinale L. (comfrey) was for its ability to inhibit he growth of hepatocellular carcinoma cells and expression of the insulin-like growth factor I (IGF-II) gene. The DNA synthesis of hepatocellular carcinoma cell lines, Hep G2, Hep 3B, and PLC/PRF/5 was inhibited by a crude extract of Smphytum officinale in both a time- and a dose-dependent manners. This plant extract also inhibited expression of the IGF-II gene. Since IGF-II exerts a mitogenic effect on Hep G2 cells, these results suggest that the growth inhibition by Symphytum officinale extract is, in part, mediated through the inhibition of IGF-II gene expression.
The cell growth inhibitor effect of cervical cancer cells was investigated by liposome mediated transfection (pRcCMVp53/lipofectin) and by transfection using adenovirus (AdCMVp57). The papilloma virus cancer cell lines we used in this study were HPV16 positive, having inhibiter gene, wild p53 gene, CaSki, SiHa, HPV18 positive HeLa, HeLaS3 and HPV negative C33A, HT3. LacZ gene of E.coli was used as the marker gene for the transfection efficiency. The effect on the inhibition of tumor cell growth was measured by cell count and cell viability though ELISA analysis and MTT assay. The inhibition of tumor cell growth was confirmed by measuring each assay for six days, comparing with the normal control cell growth. The cell growth of cervical cancer calls by transfection was significantly reduced and showed tittle differences among the cell lines. To eliminate the potential problem of Ad(adenovirus) contamination during rAAV production, rAAV can be produced by a triple transfection of vector plasmic, packaging plasmid, and adenovirus helper plasmid. To examine the helper functions of Ad plasmids on the production of rAAV vector, we carried out cotransfection of three plasmids, AAV vector, packaging construct, and Ad helper plasmids. The optimized transfection condition for calcium phosphate method is 25ug of total DNA per 10-cm-diameter plate of 293 cell. We found that rAAV yields peaked at 48hr after Ad infection. The titer of rAAV was measured by the dot blot analysis to measure the number of particles/ml based on the quantification of viral DNA. Recent1y, Kombucha(fungi) was identified as a very potent antileukefic agent. In the present study, effect of natural toxin(plankton) and Kombucha is PSP(GTXI-3, neoSTX), on various MTT assay cervical cancer cell line. Toxin(GTX 1-3, neoSTX) also inhibited the proliferation in primary cervical cancer calls in a dose-dependent toxin concentration. These results showed that toxin was very potent in inhibiting the proliferation of cervical cancer calls in vitro. Toxins and Kombuoha exhibited a dose dependent inhibition of cellular proliferation in cancer cell line.
An Actinomycetes strain JB isolated from Mt. Hanla had a strong antimicrobial activity against gram positive bacteria and tumor cell growth inhibition. Especially, it couldn't degrade starch and casein as organic compounds. It was resist on lincomycin and rifampicin. The spore mass of strain JB which was arethrospore was white. DAP of the cell wall was L, L-DAP. Antimicrobial material was heat stable, dissolved in ethyl acetate, and not dissolved in butanol. In the pressnce of 0.1% phenol and 4% sodium chloride, strain JB could grow, but it didn't growth at below $10^{\circ}C$. Strain JB didn't use dextran, sodium acetate and sodium citrate as sole carbon source and L-cystein and L-thereonine as nitrogen source. The filtered broth of strain JB had the antimicrobial activity against gram positive bacteria, especially Staphylococcus aureus (ATCC 65389) and the growth inhibition of tumor cell line.
Effect of inhibitors on Vibrio parahaemolyticus cell growth and its urease activity was studied. The growth of the bacterium and the enzyme activity were inhibited by the addition of 0.02% p-hydroxymercuric benzoate, $HgCl_2$and $AgNO_3$. However, same concentration of boric acid, thallium acetate and $Pb(NO_3)_2$ did not affect the cell growth but inhibited urease activity by 25%, 29%, and 38%, respectively. Acetohydroxamic acid was the most potent inhibitor on cell growth by inhibiting 40% but did not affect urease activity. To investigate the effect of inhibitors on urease activity, urease was purified and confirmed on SDS-PAGE. The purified urease was inhibited 100% by the addition of 1 mM acetohydroxamic acid and $AgNO_3$but no inhibition was occurred by the addition of the same concentration of thallium acetate. and the addition of 0.01 mM of $HgCl_2$ and acetohydroxamic acid inhibited the purified urease activity by 39% and 24%, respectively. On 0.1 millimolar basic, acetohydroxamic acid and $HgCl_2$inhibited 4 times more active in urease inhibition than p-hydroxymercuric benzoate whereas no inhibition was occurred either thallium acetate or $Pb(NO_3)_2$.
This study was initially explored to procure biomaterials capable of inhibiting cancer cell growth from nine Persicaria species (Polygonaceae). The extract of P. nepalensis that was selected from the initial screenings was further fractionated to identify bioactive compounds. The ethyl acetate (EtOAc) fraction was shown to be the most active in the inhibition of cell growth against six cancer cell lines (IC50 value of 3.77-12.87 ㎍/ml). Phytochemical study led to the isolation of two galactolipids of 1,2-di-O-linolenoyl-3-O-β-D-galactospyranosyl-sn-glycerol (1) and 1-O-linolenoyl-3-O-β-D-galactospyranosyl-sn-glycerol (2) from the hexane fraction and three phenylpropanoyl sucroses of lapathoside A (3), vanicoside B (4) and lapathoside C (5) from the EtOAc fraction. These isolated compounds have not been reported from this plant. Compounds 3 and 4 exhibited the effective growth inhibition against a panel of cancer cell lines (IC50 value of 6.90-18.09 μM). In addition, the anti-inflammatory activity was evaluated to determine lipopolysaccharide (LPS)-induced nitric oxide (NO) formation in RAW264.7 mouse macrophage cells. The EtOAc fraction (IC50; 34.14 ㎍/ml) and its constituents, 3 (8.55 μM) and 4 (7.83 μM) were shown to be effective in the inhibition of LPS-induced NO production. Therefore, compounds 3 and 4 were considered to be active constituents for anti-inflammatory and antitumor activity from P. nepalensis.
Retinoids, better known as vitamin A, have been reported to inhibit the growth of several breast cancer cell lines in culture and to reduce breast tumor growth in animal models. Furthermore, retinoids can augment the action of other breast cancer cell growth inhibitors both in vitro and in vivo. Clinically, interest has increased in the potential use of retinoids for the prevention and treatment of human breast cancer. We have examine the effect of all-trans retinoic acid(tRA) and 9-cis retinoic acid(9-cis RA) on human breast cancer cell(MCF-10A, T47-D, MCF-7) proliferation using MTT assay and cell cycle analysis(FACS). Overexpression of cyclin D1 protein is observed in the majority of breast cancers, suggesting that dysregulated expression of cyclin D1 might be a critical event in breast cancer carcinogenesis. We investigated whether tRA and 9-cis RA might affect expression of cyclin D1 on human breast cancer cells(MCF-10A, T47-D, MCF-7) using RT-PCR and west-ern bolt. In MCF-10A cells, either tRA or 9-cis RA treatment did not affect the cell proliferation. In T47-D cells and MCF-7 cells, either tRA or 9-cis RA treatment showed the inhibition of the cell proliferation over control cells and also inhibit the estrogen stimulated cell proliferation when it was given together with estrogen. The effect of retinoids was dose- and time- dependent. T47-D cells treated with 1.0 $\muM$ tRA undergo G0/G1-phase arrest by Day 5. MCF-7 cells treated with 1.0 $\muM$ tRA undergo S-phase arrest by Day 5. All-trans retinoic acid(tRA) and 9-cis retinoic acid(9-cis RA) inhibited the cyelin D1 mRNA and protein expression levels of human MCF-7 and T47-D breast carcinoma cells in vitro. The data indicate that retinoids can reduce cyclin D1 expression levels in a variety of breast cell lines in vitro and result in inhibition of cell proliferation. tRA-mediated growth inhibition and cyclin D1 expression inhibition is more potent than 9-cis RA mediated that. tRA-mediated inhibition effect is more potent on T47-D cells than on MCF-7 cells. Our data suggest that retinoids activity is different according to property of cell lines. Future chemoprevention of breast cancer studies using retinoids will be necessary to determine the mechanism of the retinoids-mediated growth inhibition.
Quercetin is a dietary anticancer chemical that is capable of inducing apoptosis in tumor cells. However, little is known about its biological effect in nonmalignant hepatic cells. Using embryonic normal hepatic cell line (BNL CL.2) and its SV40-transformed tumorigenic cell line (BNL SV A.8), we evaluated the effects of quercetin on cell proliferation and apoptosis. As the results, our present study demonstrated that quercetin had a selective growth inhibition in normal versus tumorigenic hepatic cells such that BNL SV A.8 cells were very sensitive to the quercetin-mediated cytotoxicity. In particular, as evidenced by the increased number of positively stained cells in the TUNEL assay, the induction of characteristic nuclear DNA ladders, and the migration of many cells to sub-G1 phase in the BNL SV A.8 cells, quercetin treatment more sensitively induced apoptosis in BNL SV A8 cells than in BNL CL.2 cells. Collectively, our findings suggest that quercetin can be approached as a potential agent that is capable of inducing selective growth inhibition and apoptosis of hepatic cancer cells.
Various lines of evidence suggest that dietary components protect the initiation step of carcinogenesis. In this study, the ethylacetate (PGMEA), ethylether (PGMEE), butanol (PGMB) and aqueous (PGMA) soluble fractions of Punica granatum L. (PG) were screened for their growth inhibition using the MTT assay on HepG2, HeLa, C6, MCF-7 and HT-29 cells and for their activity to induce quinone reductase (QR) in HepG2 cells. Among various fractions of Punica granatum L., the PGMEE showed the strongest growth inhibition at 500 $\mu\textrm{g}$/mL which resulted 92.5% on Hela cell lines and 97.8% on C6 cell lines. The PGMEA and PGMB also showed significant growth inhibition. The assay of QR induction on HepG2 cells, grown in the presence of PGMEE at the concentration of 50$\mu\textrm{g}$/mL, was 1.4 times more effective compared with the control value of1.0. These results suggested that useful cancer chemoprevention materials could be isolated from PGMEE fraction of Punica granatum L.
This work examines the effect of treatment with Gyukhachukeotang on the growth of uterine myomal cells. Comparisons of cell growth, MAP kinase activity and expression of bcl-2 (apoptosis-related gene) were made between the control and experimental samples. The results as fallows; 1. Any concentration of Gyukhachukeotang above 0.01% yielded growth inhibition. Concentrations of 5% and 10% stopped all cell growth, demonstrating the effectiveness of Gyukhachukeotang as a growth inhibitor on uterine myomal cells. 2. The MAP kinase activity in uterine myomal cells treated with Gyukhachukeotang was decreased to a high degree at the concentration of 10%, and some inhibition of activity was detected at a concentration of 5%. 3. The expression of bcl-2, a Cell Apoptosis-related gene, in uterine myoma cells treated with Gyukhachukeotang was gradually increased with increasing concentration of Gyukhachukeotang. These results indicate the ability of Gyukhachukeotang to control uterine myomal cell growth, with concurrent reduction of MAP kinase activity. Treatment with Gyukhachukeotang appears to trigger a normal apoptosis response, as indicated by increased bcl-2 expression. This observed increase in apoptosis indicates that Gyukhachukeotang is an appropriate prescription to treat uterine myomal cells.
