The aim of this study was to investigate protein profiles related to the induction of adipogenesis within the bovine longissimus dorsi muscle (BLDM) by proteomic analysis. We analyzed BLDM proteins at different growth stages to clarify the physiological mechanisms of marbled muscle development in 20 head of Korean native cattle (11 month: 10 head, 17 month: 10 head). BLDM proteins were analyzed by two-dimensional electrophoresis and image analysis. Villin 2 was specifically identified by mass spectrometry and a protein search engine. Villin 2 protein expression in BLDM decreased during the fat development stage in test steers. In a Western blot cell culture study of spontaneously immortal bovine muscle fibroblasts, the abundance of Villin 2 was shown to be down-regulated during differentiation into muscle. In 3T3-L1 mouse embryonic fibroblasts, Villin 2 was decreased during differentiation into adipocytes. The results suggest that Villin 2 may be related to the induction of transdifferentiation and adipogenesis in bovine longissimus dorsi muscle.
Objective: The aim of this study was to investigate the expression of heat shock protein (HSP) 90, 70, and 60 in chicken muscles and their possible relationship with quality traits of meat. Methods: The breast muscles from one hundred broiler chickens were analyzed for drip loss and other quality parameters, and the levels of heat shock protein (HSP) 90, 70, and 60 were determined by immunoblots. Results: Based on the data, chicken breast muscles were segregated into low (drip loss${\leq}5%$), intermediate (5%${\geq}9.5$) drip loss groups. The expression of HSP90 and HSP60 were significantly lower in the high drip loss group compared to that in the low and intermediate drip loss group (p<0.05), while HSP70 was equivalent in abundance in all groups (p>0.05). Conclusion: Results of this study suggests that higher levels of HSP90 and HSP60 may be advantageous for maintenance of cell function and reduction of water loss, and they could act as potential indicator for better water holding capacity of meat.
Phytoplankton populations were examined at three sites in Lake Cheongpyeong, South Korea from March 2008 to December 2016, including measurement of phytoplankton communities and their dominant species, abundance and environmental factors. The annual average ranges of water temperature, dissolved oxygen and conductivity were $15.2-18.8^{\circ}C$, 10.3-12.2 mg/L, $86-140{\mu}S/cm$, respectively, with similar values at all studied sites. The highest phytoplankton cell density was observed in spring and fall, and it subsequently decreased rapidly during heavy rainfall. Diatoms were dominant in spring (mainly Stephanodiscus hantzschii, Asterionella formosa) and fall (mainly Aulacoseira granulata), while greenalgae and cyanobacteria had high appearance in early-summer and summer, respectively, indicating that water temperature is the most important factor influencing their growth. Stephanodiscus hantzschii and Asterionella formosa frequently occurred at low water temperature ($4.5-15.0^{\circ}C$ and $5.4-21.6^{\circ}C$, respectively) while Aulacoseira granulata and Anabaena spp. were favored by high water temperature (8.6-28.4 and $14.9-26.2^{\circ}C$, respectively) and phosphorus. Additionally, Fragilaria crotonensis occurred at low nutrient conditions. Rhodomonas spp. frequently appeared year-round.
Piwi proteins and Piwi-interacting RNAs (piRNAs) have been implicated in transposon control in germline from Drosophila to mammals. To examine the profile of small RNA expression in human cancer cells and explore difference in small RNA transcriptome, small RNA libraries prepared from wildtype, HILI overexpressed and HILI knockdowned Hela cells were sequenced using Solexa technology. piRNAs and other repeat-associated small RNAs were observed in Hela cells. By using in situ hybridization, piR-49322 was localized in the nucleolus and around the periphery of nuclear membrane in Hela cells. Following the overexpression of HILI, the retrotransposon elements LINE1 was significantly repressed, while LINE1-associated small RNAs decreased in abundance. The present study demonstrated that HILI along with piRNAs plays a role in LINE1 suppression in Hela cancer cell line.
This study investigated the effects of sediment removal on water quality and phytoplankton development by setting up mesocosms at Uiam Lake, South Korea, and analyzing the environmental parameters and phytoplankton communities between June and October 2015. The comparison between testbed without sediment removal (TB-1) and testbed after sediment removal (TB-2) gave similar values for water temperature, pH, dissolved oxygen (DO), and electrical conductivity. Nevertheless, the average electrical conductivities of the two testbeds were $139{\mu}S/cm$ and $135{\mu}S/cm$, which were lower than the value obtained from the external control point (TB-con; $154{\mu}S/cm$). The small difference in total phosphorus (TP) and total nitrogen (TN) concentrations between the two testbeds implied that sediment removal did not greatly reduce nutrients; however, the phytoplankton cell count had decreased by approximately 37 % in TB-2 (average 1,663 cells/mL) compared to TB-1 (average 2,625 cells/mL). Compared to TB-con, the phosphorus and nitrogen concentrations of the two testbeds had decreased by 39 % and 30 %, respectively, whereas the phytoplankton abundance had decreased by up to 73 %, perhaps because of the blocked inflow of nutrients and the stabilized body of water caused by the installation of the mesocosm. The concentration of geosmin was lower in testbeds than in the external point, because installation of the structures had reduced the cyanobacteria biomass.
Due to the shortage of the fingerling/juvenile mud loach, Misgurnus mizolepis in Korea, these fish have been imported from China. However, the mortality rate during and after their transportation is very high. In this study, we examined various physiological and histological parameters to evaluate the effect of salt treatment on the survival and recovery of mud loaches in holding farms during the quarantine process. Glucose, osmolality, $Na^+$, $Cl^-$, and histological changes were assessed for three different salinities. Non-treated fish (control 0.0%) exhibited lower levels of osmolality, and $Na^+$ and $Cl^-$ concentrations compared with those kept in solar salt solution (0.5% and 1.0%). Glucose levels in control fish were higher than those in fish exposed to 0.5% and 1.0% solar salt solution. Histologically, control fish showed thinner epidermis of skin, branchial hyperplasia and lamellar fusion with an abundance of eosinophilic granule cell-like cells. After solar salt solution treatment, damaged gill structures in the fish almost recovered within 5 days. The present study demonstrates that mud loaches transported from China suffer from skin and gill damage and physiological dysfunction which may increase the mortality and morbidity. Moreover, saline treatment might alleviate the stress responses and ionic/osmotic imbalances, and help heal gill damage.
Glucose is a major source of metabolic fuel and lipid and protein syntheses. Transport of glucose into the cell is regulated by an action of glucose transport.associated transporters, especially solute carriers 2A (Slc2a, protein symbol GLUT). The present study was focused on examination of mRNA expression of various Slc2a isoforms in the epididymis during postnatal development. Total RNAs isolated from different epididymal segments (caput, corpus, and caudal epididymis) were utilized for real-time polymerase chain reaction analyses. Results showed that Slc2a 1, 3, 4, 5, and 8 were expressed in the entire epididymal regions. In addition, the abundance of these Slc2a isoforms' transcripts was different within each epididymal regions. Moreover, the present study showed differential expression of these Slc2a isoforms among different epididymal segments according to postnatal ages. The current study suggests that glucose transport in the epididymis via various Slc2a isoforms would be necessary for maintenance of the epididymal functions.
Phlorotannins and marine algal polyphenols, including dieckol, 6,6-bieckol, phloroglucinol, phlorofucofuroeckol-A, and eckol, were isolated from brown seaweeds. These compounds have beneficial bioactivities, and Ecklonia cava has become widely used for the extraction and isolation of phlorotannins. Eckol, in particular, has been to shown to have antioxidant, anti-inflammatory, anticoagulatory, and photoprotective properties. However, due to its low abundance in weaweed, the isolation and purification of eckol are difficult. Its limited availability renders the isolation and purification of eckol labor-intensive processes. Centrifugal partition chromatography (CPC) is an efficient technique for the isolation and purification of eckol. In this study, eckol was isolated from the ethyl acetate fraction of the 70% ethanol extract of E. cava using CPC with a two-phase solvent system of a n-hexane:EtOAc:methanol:water (2:8:3:7, v/v) solution. The purity and anti-inflammatory activity of the isolated eckol were verified by high-performance liquid chromatography and by assaying lipopolysaccharide-induced inflammatory responses in an immortalized murine BV2 microglial cell line, respectively. In conclusion, CPC is a useful technique for simple and efficient isolation of eckol from E. cava.
Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Rhizoma Bletillae on the basal melanogenic activities of B16/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Rhizoma Bletillae alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Rhizoma Bletillae also suppressed the increase of ${\alpha}$-MSH (100 nM) or forskolin (20 ${\mu}M$)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Rhizoma Bletillae also inhibited the increase of forskolin(20${\mu}M$) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Rhizoma Bletillae showed less intensity than B16 melanoma cells stimulated with ${\alpha}$-MSH or forskolin. These results suggest that Rhizoma Bletillae inhibits melanogenesis and abrogates ${\alpha}$-MSH and cAMP-induced melanogenesis in B16 melanoma cells.
The following is a list of morphologic and genetic characteristics of Pinellia tuber. 1. The original plant of Pinellia tuber is Pinellia ternata$(T_{HUNB})$$B_{REIT}$. With regards to its external morphology, it is smaller than other Araceae species and its spadix is longer than its leaves, which trifurcate. 2. As regards its internal morphology, its mucous cell is elliptical and the vessel is helical or annular-shaped. Granules exist in abundance and in various shapes. 3. Distribution and size of laticifers are the key criteria on which to differentiate between domestic and imported Pinellia tuber. Laticifers are mainly distributed in the epidermis in domestic Pinellia tuber and in the cortical parenchyma in imported Pinellia tuber. The size of laticifers is somewhere between 1,3 and $8{\mu}m$ in diameter in imported Pinellia tuber bigger than its domestic counterpart. 4. RAPD markers display a great similarity in bands between domestic and Chinese Pinellia tuber. However, RAPD primers 352, 358, 365, 368 and 374 are distinctive markers for domestic Pinellia tuber. In the meantime, North Korean Pinellia tuber, morphologically similar to domestic Pinellia tuber, is genertically distinctive from its domestic counterpart in primers 354, 358, 365, 368, 374 and 379, a finding that supports the postulation that North Korean Pinellia tuber is tuber of another Araceae species.
