Induction of Cell Cycle Arrest, Apoptosis, and Reducing the Expression of MCM Proteins in Human Lung Carcinoma A549 Cells by Cedrol, Isolated from Juniperus chinensis

  • Proteins related to DNA replication have been proposed as cancer biomarkers and targets for anticancer agents. Among them, minichromosome maintenance (MCM) proteins, often overexpressed in various cancer cells, are recognized both as notable biomarkers for cancer diagnosis and as targets for cancer treatment. Here, we investigated the activity of cedrol, a single compound isolated from Juniperus chinensis, in reducing the expression of MCM proteins in human lung carcinoma A549 cells. Remarkably, cedrol also strongly inhibited the expression of all other MCM protein family members in A549 cells. Moreover, cedrol treatment reduced cell viability in A549 cells, accompanied by cell cycle arrest at the G1 phase, and enhanced apoptosis. Taken together, this study broadens our understanding of how cedrol executes its anticancer activity while demonstrating that cedrol has potential application in the treatment of human lung cancer as an inhibitor of MCM proteins.

  • Since the lips are a representative facial part that can express human attraction, aesthetic interest in them has always existed in human history. However, as lips age, they tend to form wrinkles, become thinner, and lose their volume. To counteract this phenomenon, medical procedures such as fillers or fat transplantation have been suggested. In this study, we verified that the one of main effective material complex of L G H&H L IPCERIN®, combination of cedrol, a sesquiterpene found in cedarwood, and three peptides (acetyl hexapeptide-8, acetyl tetrapeptide-9, and desamidocollagen) could ameliorate the photo-aging and reduce the wrinkles through in vitro experiments. The possibility of improving collagen and elastin expression in skin fibroblasts and reducing MMP expression under photoaging conditions was verified. In addition, it was confirmed that the amount of fat tissue in the lips can be increased by promoting adipose stem cell differentiation and increasing the amount of fat produced in the in vitro adipose stem cell differentiation experiment. Two weeks of human application tests confirmed that a combination of cedrol and peptides can improve the wrinkles, texture, elasticity, and volume of the lips. This study verified that the combination of cedrol and three peptides can be used as effective cosmetic materials to decrease the various signs of aging in the lips.

  • Skin aging is the phenomena finally expressed on the skin surface and related to the changes in the microstructure of the skin texture. Which is resulted in wrinkle formation and uneven tone of skin and so on. In this study, the synergy effect of Cedrol and a collagen-derived peptide in type III collagen synthesis was evaluated by in vitro test. The physiological skin state of 22 female volunteers was measured after using the cosmetics for 4 weeks. Results showed that Cedrol and a collagen-derived peptide had the excellent synergy effect in type III collagen synthesis. The cosmetics improved skin microrelief, star configurations, skin gloss, skin tone, hydration and elasticity except skin lightening. In conclusion, this study proved that Cedrol and collagen-derived peptide had the synergy effect of type III collagen synthesis in the cell level and cosmetics with those was improved skin aging in human volunteer test.

Isolation and Characterization of Antifungal Metabolites from Pterocarpus santalinus against Fusarium graminearum Causing Fusarium Head Blight on Wheat (자단향으로부터 밀 붉은곰팡이병균 Fusarium graminearum에 대한 항진균활성 물질의 분리 및 특성 규명)

  • Fusarium head bight (FHB) is a devastating disease on major cereal crops worldwide which causes primarily by Fusarium graminearum. Synthetic fungicides are generally used in conventional agriculture to control FHB. Their prolonged usage has led to environmental issues and human health problems. This has prompted interest in developing environmentally friendly biofungicides, including botanical fungicides. In this study, a total 100 plant extracts were tested for antifungal activity against F. graminearum. The crude extract of Pterocarpus santalinus heartwood showed the strongest antifungal activity and contained two antifungal metabolites which were identified as ${\alpha}$-cedrol and widdrol by GC-MS analysis. ${\alpha}$-Cedrol and widdrol isolated from P. santalinus heartwood extract had 31.25 mg/l and 125 mg/l of minimal inhibitory concentration against the spore germination of F. graminearum, and also showed broad spectrum antifungal activities against various plant pathogens. In addition, the wettable powder type formulation of heartwood extract of P. santalinus decreased FHB incidence in dose-dependent manner and suppressed the development of FHB with control values of 87.2% at 250-fold dilution, similar to that of chemical fungicide (92.6% at 2,000-fold dilution). This study suggests that the heartwood extract of P. santalinus could be used as an effective biofungicide for the control of FHB.

Growth-inhibiting Effects of Juniperus virginiana Leaf-Extracted Components toward Human Intestinal Bacteria

  • The growth responses of materials extracted from Juniperus virginiana leaves against Bifidobacterium bifidum, B. longum, Clostridium perfringens, Escherichia coli, Lactobacillus acidophilus, L. casei, and Streptococcus mutans were examined using impregnated paper disk agar diffusion. The biologically active constituent isolated from the J. virginiana extracts was characterized as ${\alpha}$-cedrene using various spectroscopic analyses including IR, EI-MS, and NMR. The responses varied according to the dose, chemicals, and bacterial strain tested. Methanol extracts of J. virginiana leaves exhibited a strong and moderate inhibitory activity against C. perfringens and E. coli at 5 mg/disk, respectively. However, in tests conducted with B. bifidum, B. longum, L. acidophilus, L. casei, and S. mutans, the methanol extracts showed no or weak inhibitory response. At 2 mg/disk, a-cedrene strongly inhibited the growth of C. perfringens and moderately inhibited the growth of E. coli and S. mutans, without any adverse effects on the growth of four lactic acid-bacteria. Of the commercially available compounds originating from J. virginiana leaves, cedrol and ${\alpha}$-pinene exhibited strong and moderate growth inhibition against C. perfringens, and ${\alpha}$-copaene revealed moderate growth inhibition against E. coli at 1 mg/disk. Furthermore, cedrol exhibited moderate and weak growth inhibition against S. mutans at 2 and 1 mg/disk, respectively. However, little or no activity was observed for camphene, (+)-2-carene, p-cymene, limonene, linalool, and a-phellandrene against B. bifidum, B. longum, C. perfringens, L. acidophilus, L. casei, and S. mutans at 2 mg/disk. The observed inhibitory activity of the J. virginiana leaf-extracted materials against C. perfringens, E. coli, and S. mutans may be an indication of at least one of the pharmacological actions of the J. virginiana leaf.

Comparison of Antifungal Activites of Monoterpenes and Sesquiterpenes in Essential Oil from Chamaecyparis obtusa against Dermatophytes (피부사상균에 대한 편백정유의 Mono- 및 Sesquiterpene 항진균 활성 비교)

  • This study was to compare the antifungal activitiy of monoterpenes with that of sesquiterpenes, which were major constituents of antifungal active essential oil of Chamaecyparis obtusa, to investigate which constituents had more effective against dermatophytes, and to evaluate the synergistic effect of combination of the antifungal active constituents. The antifungal activities of seven fractions (A, B, C, D, E, F, and G) from C obtusa essential oil by column chromatography were tested against Trichophyton mentagrophytes. Fraction D, E, F, and G were more active than fraction A, B, and C, and their major compounds were mono- and sesquiterpenes analyzed by GC/MS. Borneol, linalool, and ${\alpha}$-terpineol were selected as monoterpenes, and ${\alpha}$-cedrol, nerolidol, and ${\beta}$-eudesmol as sesquiterpenes, respectively. The antifungal activities of the constituents were respectively examined against Trichophyton mentagrophytes (KCTC6077), Microsporum canis (KCTC6591), and Microsporum gypseum. The constituents of sesquiterpenes were more active than those of monoterpenes. By comparing single and combined sesquiterpenes with fraction F containing the higher ratio of sesquiterpenes, combined sesquiterpenes were the most active. This result indicated that there was the synergistic antifungal effect against dermatophytes when sesquiterpenes were combined together.

  • The present study was conducted to evaluate the application of Chamaecyparis obtusa and to investigate potential utilization of essential oil from C. obtusa as plant-based medicine. The antifungal activity of essential oil from leaves and twigs of C. obtusa (Sieb. Et Zucc) was determined and the major components of active fractions against Microsporum canis (KCTC6591) and Trichophyton mentagrophytes (KCTC6077) were identified by GC/MS analysis. In treatment of essential oil from C. obtusa, the strain M. canis was more resistant than the other, T. mentagrophytes. In the agar diffusion assay, essential oil from C. obtusa inhibited hyphal growth of M. canis and T. mentagrophytes at the concentration of more than 5,000 ppm. The zones named B and C in the TLC assay of essential oil from C. obtusa showed antifungal activities. Among four sub-fractions of n-hexane extract from B and C zones, named as B-1, B-2, C-1 and C-2, the C-2 showed the highest antifungal activity. Instrumental GC/MS analysis for sub-fractions showed that a major component of C-1 was ${\alpha}$-terpineol as terpene alcohol, while C-2 contained sesquiterpenes such as elemol, cedrol and eudesmol.

  • This study was investigated to sensible characteristic and physical characteristic to estimate value of the craft for Pinus densiflora and Pinus densiflora for. erecta Uyeki wood. The sensual characteristics of wood are evaluated by wood figure, wood grain, odor, and the physical characteristics by weathering, abrasion, surface roughness and hardness. The wood figure of these species were observed at flat grain in the radial section and edge grain in the tangential section. Also, burl's figure was observed in Burl of Pinus densiflora wood. Pinus densiflora for. erecta Uyeki wood had volatile aroma compounds such as Camphene, Thymyl ether, ${\alpha}$-Longipinene, etc. Pinus densiflora wood had volatile aroma compounds such as ${\alpha}$-Longipinene, (+)-Cedrol, etc. The moisture soption, abrasion, hardness and roughness were very different between the two species.

Essential Oil Constituents of Swertia chirata Buch.-Ham.

  • The essential oil of Swertia chirata Buch.-Ham. was extracted by solvent extraction (n-pentane:diethylether, 1:1) method using simultaneous distillation-extraction (SDE) apparatus and analyzed by gas chromatography-mass spectrometry (GC/MS). The yield of essential oil obtained from S. chirata was 236.47 mg/kg. Seventy seven compounds of the essential oil belonging to chemical classes of acid (4), alcohol (21), aldehyde (15), ester (3), furan (3), hydrocarbon (7), ketone (17) and miscellaneous (7) were tentatively identified. The major volatile compounds ranged in content order were as follows: undecanoic acid (28.63%), 2-buten-2-one (20.42%), camphor (18.40%), 2-heptadecanone (14.72%), and cedrol (13.07%).

  • 지구상에는 약 40만 종의 식물이 존재하는 것으로 알려져 있으며 이 중에 는 질병의 치료를 위한 약재로 이용되거나 나무는 대부분 목재 등으로 이용된다. 향나무(Juniperus chinenesis L.)는 항균성 및 방충성이 뛰어난 상록수로서 목재는 조각재나 가구재로 사용되고 민간 및 한의학에서 다양한 증상의 약재로 쓰이고 있으며 부패한 냄새를 제거할 목적이나 향재 및 향료로도 쓰이고 있다. 열매의 정유성분은 화장품, 술, 캔디 등에 사용되며 cedrol은 향료보류제, 유분은 훈향료, 목부는 고혈압, 곽란, 심복통, 통기파혈에 쓰인다. 향나무는 폴레페놀 성분이 함유되어 있고 피톤치드가 많이 나오는 것으로 알려져 있으며 심재의 폴리페놀 화합물은 항암, 항균, 항알러지, 노화방지 및 심장질환 등을 예방하거나 지연시키는 등 광범위한 약리학적 활성을 나타내어 식품, 의약품, 화장품, 향료등 다양한 분야에 활용되고 있다. 향나무 추출물에 관한 연구로는 성분분석과 생리활성 등에 관한 의약학 분야의 다양한 연구와 항산화, 항균활성 등 약리작용에 연구가 최근 다양하게 이루어지고 있는 것을 볼 수 있다. 측백나무속에 속하는 나무들은 대부분 정유 성분을 함유하고 있는 특성으로 항균성 등 바이오 기능성 소재 개발을 위한 연구가 활발히 진행되고 있으나, 향나무 추출물의 염색성에 관한 연구는 찾아보기 어려운 실정이다. 이에 본 연구에서는 다양한 용매를 이용하여 향나무의 잎, 열매, 수피, 심재 각 부위 추출물을 이용하여 견직물에 대한 염색성을 살펴봄으로써 향나무 추출물을 활용한 천연염색과 염재로서의 가능성을 확인하고자 하였다. 실험 결과 증류수를 염액 용매로 염색한 결과 모든 추출물의 잎, 열매 염색포의 색상은 대부분 옅은 Y계열로 나타났으며 향나무의 수피와 심재 염색포는 대부분 적색기미가 강한 YR계열로 나타났다. 에탄올 혼합액을 염액 용매로 하여 염색한 결과 향나무의 잎과 열매의 색상은 대부분 Y계열의 색상을 나타냈고 유기용매 추출염색포에서 정유성분으로 추정되는 물질 때문에 균염이 어려웠으며 수피와 심재 염색포에서 대부분 R계열의 색상이 뚜렷이 나타났다.
