본 연구는 후추과 식물의 주성분인 piperine이 위암세포 AGS의 항암 효과에 미치는 영향을 확인하기 위해 수행되었다. Piperine에 의한 위암세포 AGS의 생존율을 확인하기 위해 MTT assay를 수행한 결과 농도 의존적으로 암세포의 생존율이 감소하는 것을 확인하였다. 이러한 암세포의 생존율 억제가 apoptosis에 의한 효과인지를 확인하기 위해 DAPI staining을 실시한 결과 piperine을 처치한 군에서 apoptotic body와 염색질 응축이 증가하는 것을 확인하였다. MTT assay와 DAPI staining의 결과를 바탕으로 piperine이 위암세포 AGS에서 apoptosis와 관련한 단백질 발현 양상에 미치는 영향을 확인하기 위해 western blotting을 실시하였다. 그 결과 piperine에 의해 AGS 세포에서 apoptosis를 유도하는 p53, Bax의 발현은 농도 의존적으로 증가하였고, apoptosis를 억제하는 Bcl-2, XIAP의 발현은 농도 의존적으로 감소하였다. Apoptosis의 궁극적 단계인 caspase-9와 손상된 DNA를 복구하는 PARP의 분절은 증가하였으며, apoptosis를 방해하는 인자인 Akt의 인산화는 농도 의존적으로 감소하였다. Akt 억제제인 LY294002와 piperine을 병행 또는 단독으로 처치하여 western blotting을 진행한 결과 Bcl-2의 발현은 piperine을 단독으로 처치했을 때와 유사한 감소 경향을 보였지만, p-Akt의 발현은 대조군과 비교해 LY294002와 piperine을 단독으로 처치했을 때보다 병행했을 때 더욱 감소하였다. 이와 반대로 Bax와 cleaved-PARP의 발현은 강하게 증가하였다. 본 연구의 결과를 종합해볼 때 piperine이 위암세포 AGS에서 Akt 경로를 통해 apoptosis를 유도하는 것으로 여겨지며, 향후 위암 예방제나 치료제로의 개발 가능성이 있을 것으로 생각한다.
Aloin [1,8-Dihydroxy-10-(${\beta}$-D-glucopyranosyl)-3-(hydroxymethyl)-9(10H)-anthracenone]은 알로에에서 추출한 천연 안트라퀴논이다. 다양한 유형의 인간 암세포에서 항산화, 항암 효과가 있는 것으로 밝혀졌지만 인간 대장암 세포 HT-29에서 aloin의 항암 효과는 밝혀지지 않았다. 본 연구에서는 aloin이 인간 대장암 HT-29 세포에서 세포 사멸 작용을 발휘할 수 있는 메커니즘을 조사하였다. Aloin이 세포 생존율에 영향을 미치는지 알아보기 위해 대장암 세포 HT-29, 흑색종 세포 A375SM, 위암 세포 AGS를 aloin(0, 100, 200, 300 및 $400{\mu}M$)으로 처리하였을 때, HT29에서는 농도 의존적으로 세포 생존율을 감소시켰고, A375SM과 AGS 세포에서는 암세포 생존율의 감소가 보이지 않았다. 이러한 HT-29에서의 세포 생존율 감소가 세포자멸사로 인한 감소인지 확인하기 위해 DAPI stain과 flow cytometry를 실시한 결과 apoptotic body가 유의적으로 증가하고 세포 자멸사가 증가한 것을 확인할 수 있었다. 이와 같은 결과를 바탕으로 aloin이 대장암 세포 HT-29에서 세포 사멸 관련 단백질 발현 양상에 미치는 영향을 확인하기 위해 western blotting을 실시하였다. Aloin은 Bax, PARP의 분절을 농도 의존적으로 증가시켰고, caspase3, -8을 활성화시켰지만, Bcl-2는 대조군에 비해 변화가 없었다. Aloin에 의해 유도된 세포자멸사 기전을 확인하기 위해 MAPK pathway 중 p-p38과 p-ERK의 발현을 확인한 결과, p-p38을 up-regulation시키고 p-ERK의 down-regulation을 유도했다. 따라서, aloin은 인간 대장암에서 암세포 성장 억제 효과 및 암세포 사멸 유도로 암예방 약제로서의 개발 가능성이 있다고 사료된다.
폐암은 전 세계적으로 사망률이 높은 암으로서 소세포성 폐암(small cell lung cancer, SCLC)과 비소세포성 폐암(non-small cell lung cancer, NSCLC)으로 분류된다. 오늘날까지 폐암 치료를 위해 많은 화학요법이 사용되어 왔으나 장기간 화학요법에 의한 부작용 및 항암제 내성 유발로 인해 항암 치료에 한계가 있으므로, 이와 같은 문제점을 해결하기 위해 현재는 천연물로부터 유래한 항암물질을 탐색하는 추세이다. Curcumin은 강황의 뿌리에서 추출된 polyphenol 화합물로서 항균, 항암, 항산화 및 항염증 작용이 보고되어 현재까지 꾸준히 항암 연구가 진행되어 왔으나 인간 폐암 세포주의 종류에 따른 항암 효과는 거의 연구되지 않았다. 따라서 본 연구에서는 여러 종류의 인간 폐암 세포주에 대한 curcumin의 항암 효과를 조사하고자 하였다. 그 결과, 10, 30, 50 μM 농도의 curcumin은 폐암 세포주에 따라 경미한 차이가 나타내면서 농도 의존적으로 세포 생존을 저해하였다. 또한 A549 (NSCLC) 및 DMS53 (SCLC) 세포주를 대상으로 apoptosis, autophagy, reactive oxygen species (ROS)를 flow cytometry로 측정한 결과, 농도 의존적으로 이러한 현상들이 증가되었으며 각각의 저해제를 처리했을 때 이러한 현상들이 회복되는 것이 관찰되었다. Apoptosis와 관련된 단백질인 Bax, PARP, pro-caspase-3 및 Bcl-2의 발현이 curcumin 농도 의존적으로 조절되었으며, 특히 DMS53에서 Bax/Bcl-2 비율이 더 높았다. 또한, autophagy 단백질인 p-AKT, p62 및 LC3B도 curcumin 농도 의존적으로 조절되었다. 한편, ROS 저해제인 diphenyleneiodonium 처리 시, curcumin에 의해 유도된 apoptosis와 autophagy 수준이 저해되었으며 관련 단백질의 발현도 조절되었다. 이를 통해 curcumin의 항암 작용은 curcumin에 의해 생산된 ROS를 매개로 하여 폐암 세포주의 apoptosis 및 autophagy의 유도를 통해 나타나는 것으로 확인되었다.
본 연구에서는 한약재로 널리 사용되는 건칠, 유근피 및 신석 추출물의 항암 활성을 조사하였다. 생쥐 유래 정상세포(RAW 264.7 대식세포 및 C2C12 근아세포)에서는 건칠, 유근피 및 신석 단독 및 복합 처리에 의하여 유의적인 세포생존율의 억제 현상은 관찰 할 수 없었다. 그리고 건칠, 유근피 및 신석의 복합 처리는 단독 처리군에 비하여 AGS 위암세포의 생존력을 유의적으로 억제하였으나, 폐암(A549), 대장암(HCT116), 간암(Hep3B) 및 방광암(T24) 세포에서는 그 효과가 미비하였다. 아울러 이러한 AGS 위암세포 선택적 생존 억제력은 apoptosis 유도와 밀접한 연관성이 있음을 염색질의 응축 현상, DNA 단편화 및 annexin-V 염색에 의한 flow cytometry 분석을 통하여 확인하였다. 건칠, 유근피 및 신석의 복합 처리는 Fas 및 Fas legand의 발현을 증가시켰으며, XIAP, cIAP-1 및 survivin과 같은 IAP family 단백질과 anti-apoptotic Bcl-xL의 발현은 저하시켰다. 복합 처리는 또한 mitochondrial membrane potential의 손실과 caspases (-3, -8 및 -9)의 활성에 PARP 단백질의 분절화를 유도하였다. 그러나 이러한 복합 처리에 의한 AGS 세포에서 관찰된 세포독성 및 apoptosis 유도 효과는 pan-caspases inhibitor인 z-VAD-fmk의 선처리에 의하여 차단되었다. 이상의 결과는 건칠, 유근피 및 신석의 복합 처리에 의한 AGS 위암세포 선택적 apoptosis 유도가 caspase 의존적으로 일어나고 있음을 보여주는 결과이며, in vivo 모델을 이용한 후속 연구가 진행되어야 할 것이다.
Objectives : The purpose of this study was to investigate the effect of Dangguibohyultang (DB) and its combination (DB-I; Astragali membraneus BUNGE : Angelica gigas NAKAI=5:1, DB-II; Astragali membraneus BUNGE:Angelica gigas NAKAI=1:1, DB-III; Astragali membraneus BUNGE:Angelica gigas NAKAI=1:5,) on apoptosis in human colorectal adenocarcinoma HCT116 cells. Methods : To study the cytotoxic effect of methanol extract of DB-I, DB-II and DB-III on HCT116 cells, the cell viability was determined by XTT reduction method and ttypan blue exclusion assay. To confirm the induction of apoptosis, the cleavage of poly ADP-ribose polymerase (PARP), a substrate for caspase-3 and a typical sign of apoptosis, and the activation of procaspase-3, -8 and -9 were examined by western blot analysis. Furthermore, DB-induced apoptosis was confirmed by DNA fragmentation. The release of cytochrome C from mitochondria to cytosol, the level of Bcl-2 and Bax, and the expressions of Raf/MEK/ERK were examined by western blot analysis. Results : DB-I and DB-II reduced proliferation of HCT116 cells in a dose-dependent manner. DB-I and DB-II decreased procaspase-3, -8, -9 levels in a dose-dependent manner and induced the clevage of PARP. DB-I and DB-II also triggered the mitochondrial apoptotic signaling by increasing the release of cytochrome C from mitochondria to cytosol, decreasing of anti-apoptotic Bcl-2, and increasing of pro-apoptotic Bax. DB-I and DB-II decreased the activation of Ras/Raf/MEK/ERK cascade in a dose-dependent manner. Conclusion : These results suggest that DB-I and DB-II induce apoptosis via mitochondrial pathway in HCT116 cells. Furthermore, Raf/MEK/ERK cascade is involved in DB-induced apoptosis. These results suggest that DB is potentially useful as a chemotherapeutic agent in human liver cancer.

Background: In cancer cells, autophagy is generally induced as a pro-survival mechanism in response to treatment-associated genotoxic and metabolic stress. Thus, concurrent autophagy inhibition can be expected to have a synergistic effect with chemotherapy on cancer cell death. Monensin, a polyether antibiotic, is known as an autophagy inhibitor, which interferes with the fusion of autophagosome and lysosome. There have been a few reports of its effect in combination with anticancer drugs. We performed this study to investigate whether erlotinib, an epidermal growth factor receptor inhibitor, or rapamycin, an mammalian target of rapamycin (mTOR) inhibitor, is effective in combination therapy with monensin in non-small cell lung cancer cells. Methods: NCI-H1299 cells were treated with rapamycin or erlotinib, with or without monensin pretreatment, and then subjected to growth inhibition assay, apoptosis analysis by flow cytometry, and cell cycle analysis on the basis of the DNA contents histogram. Finally, a Western blot analysis was done to examine the changes of proteins related to apoptosis and cell cycle control. Results: Monensin synergistically increases growth inhibition and apoptosis induced by rapamycin or erlotinib. The number of cells in the sub-$G_1$ phase increases noticeably after the combination treatment. Increase of proapoptotic proteins, including bax, cleaved caspase 3, and cleaved poly(ADP-ribose) polymerase, and decrease of anti-apoptotic proteins, bcl-2 and bcl-xL, are augmented by the combination treatment with monensin. The promoters of cell cycle progression, notch3 and skp2, decrease and p21, a cyclin-dependent kinase inhibitor, accumulates within the cell during this process. Conclusion: Our findings suggest that concurrent autophagy inhibition could have a role in lung cancer treatment.
천궁(C. officinale)은 예로부터 민간처방 약재로 사용되었으며, 항염증, 항산화, 항암 및 신생혈관억제 등의 효능을 가지는 것으로 알려져 있다. 하지만 혈구암세포에서 apoptosis 유발과 관련된 분자생물학적 기전에 대해서는 명확히 밝혀져 있지 않다. 본 연구에서는 인체 혈구암세포인 U937 세포에서 천궁의 열수, 에탄올 및 메탄올 추출물(WECO, EECO 및 MECO)이 유발하는 항암효과 및 항암기전을 조사하였다. 먼저 WECO, EECO 및 MECO가 유발하는 증식억제 정도를 조사한 결과 EECO가 가장 뛰어난 효능을 가진다는 것을 알 수 있었으며, 이러한 현상이 apoptosis 유발에 의한 것임을 annexin-V 염색, apoptotic body 형성, DNA 단편화 및 MMP 소실 등을 통하여 확인하였다. EECO 처리에 의한 apoptosis 유발에는 DR4의 발현 증가와 함께 cIAP-1, Bcl-2 및 total Bid의 발현감소가 관여하였으며, caspases-3, -8 및 -9의 활성화와 함께 caspases-3의 기질 단백질인 PARP, β-catenin 및 PLC γ1의 단편화도 관찰되었다. 또한 EECO는 AMPK signaling pathway를 활성화시키는 것으로 나타났으며, AMPK 억제제인 compound C를 이용하여 AMPK의 활성을 억제하였을 경우 EECO에 의하여 유발되었던 apoptosis가 현저하게 감소되는 것으로 나타났다. 이상의 결과를 살펴볼 때 인체 혈구암세포인 U937 세포에서 EECO에 의하여 유발되는 apoptosis는 AMPK가 중요한 조절자로서 작용하는 것으로 생각된다.
Objective: This study investigated whether spermine supplementation could regulate cell cycle, apoptosis, and amino acid transporter-related genes expression in the thymus and spleen of early weaned piglets. Methods: Eighty female piglets were randomly distributed to receive adequate nutrients supplemented with spermine (0.4 mmol/kg body weight/24 h) or to be provided with restricted nourishment supplemented with normal saline for 7 h or 3, 6, or 9 d in pairs. Results: Regardless of administration time, spermine supplementation significantly up-regulated cyclin A2 gene expression but down-regulated p21 and cyclin D3 mRNA levels in the thymus and spleen and reduced cyclin E2 gene expression in the thymus of piglets (p<0.05). Irrespective of the treatment period, the reduced Bax and caspase-3 gene expressions and improved Bcl-2 mRNA level were observed in the thymus and spleen of spermine-administrated piglets (p<0.05). Regardless of supplementation time, spermine intake significantly enhanced the expressions of amino acid transporter-related genes (SLC1A1, SLC1A5, SLC7A1, SLC7A7, and SLC15A1) in both thymus and spleen, as well as SLC7A9 in the spleen of piglets (p<0.05). In addition, extended spermine administration also markedly promoted cell proliferation, depressed apoptosis and modulated amino acid transport (p<0.05), and such effects were the greatest during prolonged spermine supplementation (6 d) compared to the other time periods (p<0.05). Conclusion: Spermine supplementation may regulate cell cycle during the G1/S phase, suppress apoptosis and modulate amino acid transport. A period of 6 d of spermine supplementation is required to produce the optimal effects on nutritional implications.
Thyroid cancer is the most common endocrine malignancy. Patients with well-differentiated thyroid cancers, such as papillary and follicular cancers, have a favorable prognosis. However, poorly differentiated thyroid cancers, such as medullary, squamous and anaplastic advanced thyroid cancers, are very aggressive and insensitive to radioiodine treatment. Thus, novel therapies that attenuate metastasis are urgently needed. We found that both PDGFC and PDGFRA are predominantly expressed in thyroid cancers and that the survival rate is significantly lower in patients with high PDGFRA expression. This finding indicates the important role of PDGF/PDGFR signaling in thyroid cancer development. Next, we established a SW579 squamous thyroid cancer cell line with 95.6% PDGFRA gene insertion and deletions (indels) through CRISPR/Cas9. Protein and invasion analysis showed a dramatic loss in EMT marker expression and metastatic ability. Furthermore, xenograft tumors derived from PDGFRA geneedited SW579 cells exhibited a minor decrease in tumor growth. However, distant lung metastasis was completely abolished upon PDGFRA gene editing, implying that PDGFRA could be an effective target to inhibit distant metastasis in advanced thyroid cancers. To translate this finding to the clinic, we used the most relevant multikinase inhibitor, imatinib, to inhibit PDGFRA signaling. The results showed that imatinib significantly suppressed cell growth, induced cell cycle arrest and cell death in SW579 cells. Our developed noninvasive apoptosis detection sensor (NIADS) indicated that imatinib induced cell apoptosis through caspase-3 activation. In conclusion, we believe that developing a specific and selective targeted therapy for PDGFRA would effectively suppress PDGFRA-mediated cancer aggressiveness in advanced thyroid cancers.

Sphingomyelinase (SMase) catalyzes ceramide production from sphingomyelin. Ceramides are critical in cellular responses such as apoptosis. They enhance mitochondrial outer membrane permeabilization (MOMP) through self-assembly in the mitochondrial outer membrane to form channels that release cytochrome c from intermembrane space (IMS) into the cytosol, triggering caspase-9 activation. However, the SMase involved in MOMP is yet to be identified. Here, we identified a mitochondrial Mg2+-independent SMase (mt-iSMase) from rat brain, which was purified 6,130-fold using a Percoll gradient, pulled down with biotinylated sphingomyelin, and subjected to Mono Q anion exchange. A single peak of mt-iSMase activity was eluted at a molecular mass of approximately 65 kDa using Superose 6 gel filtration. The purified enzyme showed optimal activity at pH of 6.5 and was inhibited by dithiothreitol and Mg2+, Mn2+, Ni2+, Cu2+, Zn2+, Fe2+, and Fe3+ ions. It was also inhibited by GW4869, which is a non-competitive inhibitor of Mg2+-dependent neutral SMase 2 (encoded by SMPD3), that protects against cytochrome c release-mediated cell death. Subfractionation experiments showed that mt-iSMase localizes in the IMS of the mitochondria, implying that mt-iSMase may play a critical role in generating ceramides for MOMP, cytochrome c release, and apoptosis. These data suggest that the purified enzyme in this study is a novel SMase.

서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
