The Synthesis of 1,2-Benzothiazine-3-carboxamidylhydantoin Derivatives and their Antiinflammatory and Analgesic Activities

  • A number of 4-hydroxy-2H (or alkyl)-N-(3-aralkyl-2-thio-1-hydantoinyl)-1, 2-benzothiazine3-carboxamide 1, 1-dioxides were synthesized through the reaction of 4-hydroxy-2H (or alkyl)1, 2-benzothiazine-3-carboxylic methyl ester 1, 1-dioxide and 1-amino-2-thio-3-aralkyl-4-imidazolones in xylene. The compounds synthesized were screened for antinflammatory effect on carrageenin-induced edema in rat and for analgesic effect on acetic acid-induced Writhing syndrome in mice. Most compounds were inhibots of carrageenin-induced rat foot edema and some showed significant antinflammatory activity comparable to that of indomethacin and significant analgesic activity comparable to that of indomethacin and aspirin.

  • The relationships of inhibitory activities of inflammation and wound healing of flavonoids were studied in vitro and in vivo. Generally flavonoids have not only significantly anti-inflammatory activity in carrageenin-induced paw edema and Freund's complete adjuvant-induced arthritis, but also inhibitory activity of wound healing. The more inhibitory activities of wound healing flavonoids have, the more they have the anti-inflammatory activities; apigenin > guercetin > flovone > rutin > hesperidin > naringin. Their inhibitory mechanism seems to be inhibition of the inflammatory cell infiltration and fibroblast proliferation, and so they decreased the granulomatous activity and tensile strength.

  • The anti-inflammatory activity of SEO-KYONG-TANG extract(SKTWE) was examined by using carrageenin- and acetic acid-induced edema, croton oil-induced granuloma pouch, and adjuvant arthritis in rats. In addition, the acute toxicity, analgesic and antipyretic effects of SKTWE were investigated by using general experimental methods in mice. SKTWE did not show acute toxicity at 2400 mg/kg(p.o.) nor 1200 mg/kg(i.p.). After oral administration of the SKTWE to rats, significant anti-inflammatory activity was observed on 1% carrageenin- and 5% acetic acid-induced edema. Also, it significantly inhibited granuloma and exudation in these. In the adjuvant arthritis experiment, the SKTWE decreased the hind paw edema after 3 days of oral administration. In addition, it inhibited the writhing syndromes induced by 0.7% acetic acid in mice. The antipyretic activity of SKTWE was also observed through the typhoid vaccine experiment. These results suggest that SKTWE has analgesic, anti-inflammatory and antipyretic action.

  • In folklore medicine Drymaria cordata Willd (Family-Caryophyllaceae) is reported to have laxative and anti-febrile properties along with anti-inflammatory activities. Sikkimis used this plant to treat all these ailments. The anti-inflammatory effect of the methanol extract of D. cordata was investigated against carrageenin, histamine, serotonin, dextran and $PGE_1$ induced rat hind paw oedema. It exhibited significant anti-inflammatory activity against all these phlogestic agents except $PGE_1$ in the order of carrageenin > serotonin > histamine. All these effects were compared with standard drug phenylbutazone in both the acute and chronic experimental models in albino rats.

  • The extract of Ssangkum-tang was investigated on antiinflammatory activities. The extract showed antiedematous action at a dose of 500mg/kg po in carrageenin edema test in rats. The extract also inhibited the leakage of pontamine sky blue into peritoneal cavity in mice at the doses of 300 and 500mg/kg po.

  • The whole plants of Prunella vulgaris (Labiatae) and Thesium chinense (Santalaceae) are used as Hakocho(夏枯草) in the market in Korea. In oriental medicine, these herb drugs are prescribed as a diuretic or antiinflammatory drugs. In order to investigate the efficacy of the plants, the extracts were bioassayed for antiinflammatory action. The water extracts of Prunella Herba and Thesii Herba showed remarkable anti-carrageenin effect and significant inhibition of the swellings in adjuvant arthritis in rats. However, the extracts did not show any inhibition of leucocyte emigration in CMC pouch in rats.

  • A mixture of several detoxified bacterial strains ($Sterodin^{(R)}$) has been studied for anti-inflammatory effect in Wistar rats on carrageenin, dextran and prostaglandin $E_1$ ($PGE_1$) induced edema in acute model and cotton pellet and carrageenin induced sub-acute model, while, Freund's adjuvant induced chronic model. The bacterial strains showed strong inhibitory activity in acute, sub-acute and chronic models of inflammation. Further, it reduced ${\alpha}1$ acid glycoprotein and ${\alpha}2$ macroglobulin levels in serum and prostaglandin $E_2$ in inflamed paw. These results indicated that the bacterial strains probably act through prostaglandin mediatory pathways and may be useful in treatment of inflammation.

Drug Interactions of Ibuprofen and Prednisolone in Antiinflammatory and Antipyretic Effects (항염 및 해열효과에 미치는 Ibuprofen과 Prednisolone의 약 상호 작용)

  • The authors investigated drug interaction of ibuprofen and prednisolone in antiinflammatory and antipyretic activities. We have found significant differences of the antiinflammatory and antipyretic activities between single and concurrent administration of ibuprofen and prednisolone, using Sprague-Dawley Strain rats, carrageenin as a phlogistic agent and brewer's yeast as a fever inducing agent. 1) Ibuprofen(20mg/kg) was administered to the rats orally and resulted in significant reduction of (31.70 %) the swelling of rat paw induced by carrageenin, 2) prednisolone (9mg/kg) showed significant reduction of (45.76%) the swelling, 3) concurrent administration of ibuprofen (20mg/kg) and prednisolone (9mg/kg) also reduced (57.40%) the swelling. In ibuprofen (125mg/kg) administration, the inhibition rate of edema was 39.32% and in prednisolone (1mg/kg) administration, the rate was 39.04%. In concurrent administration of ibuprofen (125mg/kg) and prednisolone (1mg/kg), the inhibition rate of edema was 63.09%. Concurrent administration of ibuprofen and prednisolone showed more anti-inflammatory effects than single administration of ibuprofen and prednisolone respectively. Prednisolone itself did not show antipyretic effect, but concurrent administration of ibuprofen and prednisolone showed more antipyretic effects than ibuprofen single administratron.

Anti-inflammatory Activity of Aqueous Extract from Gleditsiae Spina (조각자 물추출물의 항염증 작용)

  • The anti-inflammatory activity of the aqueous extract from Gleditsiae spina was investigated utilizing carrageenin-induced edema, granuloma pouch and adjuvant arthritis in rats. The effects of this agent on vascular permeability and acetic acid-induced writhing in mice were also examined. Its anti-inflammatory activity on carrageenin edema was observed with oral administration. The aqueous extract from Gleditsiae spina(400 mg/kg, 7 days) showed significant inhibitory effects on granuloma and exudate formation in rats. In the method of adjuvant arthritis, the aqueous extract(400 mg/kg), orally administered for 21 days, inhibited the development of hind paw edema in rats. The aqueous extract was also inhibited the increase in vascular permeability and the number of writhings induced by acetic acid in mice. In the present study, the aqueous extract of Gleditsiae spina was indicated to have the anti-inflammatory action.

Inhibitory Effect of Shimotsu-to, a Traditional Chinese Herbal Prescription, on Acute Inflammation in Rats and Guinea Pigs

  • We examined the effect of topical application of Shimotsu-to, a traditional Chinese herbal prescription, on carrageenin-induced edema in rats and ultraviolet radiation-induced erythema in guinea pigs. Shimotsu-to (5% in water) markedly suppressed an acute edema of rat hindpaw induced by 1% carrageenin, and was more effective than any other single crude drug componcent of Shimotsu-to, Topical treatment with this prescription also inhibited ultraviolet erythema on the back skin of guinea pigs (a human sunbrun model). These results suggest the therapeutic effect on acute inflammation by topical application of Shimotsu-to.
