Purification, Characterization and Chemical Modification of the Xylanase from Alkali-tolerant Bacillus sp. YA-14

  • The xylanase from alkali-tolerant Bacillus sp. YA-14 was purified to homogeneity by CM-cellulose, Sephadex G-50, and hydroxyapatite column chromatographies. The molecular weight of the purified enzyme was estimated to be 20, 000 Da by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The purified enzyme slightly hydrolyzed carboxymethyl cellulose and Avicel, but did not hydrolyze soluble starch, dextran, pullulan, and ${\rho}-nitrophenyl-{\beta}$-D-xylopyranoside. The maximum degree of hydrolysis by enzyme for birchwood xylan and oat spelts xylan were 47 and 40%, respectively. The Michaelis constants for birchwood xylan and oat spelts xylan were calculated to be 3.03 mg/ml and 5.0 mg/ml, respectively. The activity of the xylanase was inhibited reversibly by $HgCl_2$, and showed competitive inhibition by N-bromosuccinimide, which probably indicates the involvement of tryptophan residue in the active center of the enzyme. The Xylanase was identified to be xylose-producing endo-type xylanase and did not show the enzymatic activities which cleave the branch point of the xylan structure.

A study on the efficacy of low viscous nanosized biopolymer on the mechanical and hydraulic properties of organic silt

  • Biopolymer stabilization is a sustainable alternative to traditional techniques that cause a lesser negative impact on the environment during production and application. The study aims to minimize the biopolymer dosages by sizing the bio-additives to the nanoscale. This study combines the advantages of bio and nanomaterials in geotechnical engineering applications and attempts to investigate the behaviour of a low viscous biopolymer, nano sodium carboxymethyl cellulose (nCMC), to treat organic soil. Soil is treated with 0.25%, 0.50%, 0.75% and 1.00% of nano-bio additive, and its effect on the plastic behaviour, compaction characteristics, strength, hydraulic conductivity (HC) and compressible nature are investigated. The strength increased by 1.68 times after 90 days of curing at a dosage of 0.5% nCMC through the formation of gel threads connecting the soil particles that stiffened the matrix. The viscosity of 1% nCMC increased exponentially, deterring fluid flow through the voids and reduced the HC by 0.85 times after curing for 90 days. Also, beyond the optimum dosage of 0.50%, the nCMC forms a film around the soil particles that inhibits the inter-particle cohesion causing a reduction in strength. Experimental results show that nCMC can effectively substitute conventional additives to stabilize the soil.

A Study on the Helical Flow of Newtonian and Non-Newtonian Fluid

  • This study concerns the characteristics of helical flow in a concentric and eccentric annulus with a diameter ratio of 0.52 and 0.9, whose outer cylinders are stationary and inner ones are rotating. Pressure losses and skin friction coefficients have been measured for fully developed flows of water and 0.2% aqueous of sodium carboxymethyl cellulose (CMC), respectively, when the inner cylinder rotates at the speed of 0-500 rpm. The effect of rotation on the skin friction is significantly dependent on the flow regime. In all flow regimes, the skin friction coefficient is increased by the inner cylinder rotation. The change of skin friction coefficient corresponding to the variation of rotating speed is large for the laminar flow regime, whereas it becomes smaller as Re increases for the transitional flow regime and, then, it gradually approach to zero for the turbulent flow regime.

  • Agrochemicals for the plant-disease control are criticized severely for causing environmental pollution and residual problems, and consequently microbial disease control agents are expected to be safer and more economical for sustainable agriculture. Treatment of biological control agents to seed requires the use of effective delivery systems that allow full expression of the benefical qualities of the bioprotectant. For the activation and establishment of bioprotectant around the plant seed which are able protect the seeds and seedlings from pathogen attack, the optimal liquid coating formulation was obtained using 2% sodium carboxymethyl cellulose (binder), 20% sesame dregs (solid particulate material), and dried spore of Bacillus subtilis YBL-7 (bioprotectants, 10 mg/g of seed). Suppressive of root rot was demonstrated in pot trials with coated kidney bean (Phaseolus vulgaris L.) seeds. Coated seeds with B. subtilis YBL-7 spore in F. solani-infested soil reduced disease incidence by 85% to 90% after 30 days.

헥토라이트 점토의 수열합성 및 특성 (Synthesis and Characteristics of Hectorite Clay by Hydrothermal Process)

  • Si-Mg 첨가물, LiOH, NaOH로부터 제조한 슬러리의 수열반응으로 헥토라이트 점토를 합성하였다. 수열합성의 온도와 반응시간에 따른 합성헥토라이트 점토의 화학적 조성과 결정형태를 조사하였고, 합성헥토라이트 0.2~1.0% 수분산액의 투과도와 입경을 UV-VIS 분광광도계와 투과전자현미경으로 각각 측정하였다. 합성헥토라이트 수분산액의 투과도와 유동성은 25$0^{\circ}C$, 6시간의 합성조건과 Si/Mg/Li/Na=8.0/5.4/0.6/0.6의 조성에서 최대치를 나타내었고, Mg/Si=5.4/8.0의 슬러리 조성으로부터 합성된 헥토라이트(Hec-6)가 최적의 투명성과 용액점도를 나타내었다. 또한 Hec-6의 수분산액은 높은 전단력에서 낮은 점도의 졸상으로 전화됨으로서 요변성을 나타내었고, 1.0% Hec-6/sodium carboxymethyl cellulose(50/50) 혼합용액은 전단력이 1.0 s$^{-1}$ 이상에서 1.0% Hec-6 수분산액보다 더욱 높은 점도를 나타냄으로서 점도의 상승효과가 나타났다.

Purification and Characterization of Xylanase from Bacillus sp. Strain DSNC 101

  • A xylanase from the Bacillus sp. strain DSNC 101, isolated from soil, was purified to homogeneity by anion-exchange and hydrophobic interaction chromatography followed by gel filtration chromatography. The enzyme cleaved xylan, but not carboxymethyl cellulose, Avicel, soluble starch, and pNPX. The main product of oat spelts xylan hydrolysates was xylobiose. The xylanase had a molecular weight of 25 kDa determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Optimum temperature and pH for the xylanase activity were $50^{\circ}C$ and 6.0, respectively. $K_{m}\;and\;V_{max}$ of the enzyme for oat spelts xylan were 12.5 mg of xylan/ml and 869.5 unit/mg of protein, respectively. Xylanase was completely inhibited by Hg, Cu, and N-bromosuccinimide, but was stimulated by Ca, Co, and Mg.

  • The paper concerns an experimental study of fully developed laminar flow of a Newtonian and non-Newtonian liquid in concentric annuli with combined bulk axial flow and inner cylinder rotation. Pressure losses and skin friction coefficients have been measured for Newtonian fluid, water and non-Newtonian fluids, 0.2% aqueous of sodium carboxymethyl cellulose (CMC) and 5% bentonite solutions, when the inner cylinder rotates at the speed of $0{\sim}500$ rpm. The influences of rotation, radius ratio and working fluid on the annular flow field are investigated. And the new correlations among the skin friction coefficient, the Reynolds number and the Rossby number are presented with reasonable limits of accuracy in laminar flow regime.

  • This study was performed to evaluate the efficacy of 0.8% sodium hyaluronic acid solution and crosslinked hyaluronic acid mixture for the prevention of postoperative intraperitoneal adhesion in rats. Forty-five animals were divided into three groups ; 0.9% saline treated control group, 1% sodium carboxymethyl cellulose treated group (SCMC), and 0.8% sodium hyaluronic acid solution and crosslinked hyaluronic acid mixture treated group (SHCH). Adhesions were induced by suturing both the ileal serosa and peritoneum abrased until petechial bleeding occurred. Fourteen days later, adhesions were evaluated clinically and histopathologically. The mean tensile strength was significantly decreased in the SCMC and SHCH groups compared to the control group (p < 0.05), and the SHCH group had the lowest tensile strength. The distance of adhesion site was highest in the control group and significantly decreased in the SHCH group comparing control group (p < 0.05). The inflammatory cell infiltration, collagen hyperplasia and neovascularization of the SCMC and SHCH groups were significantly lower than the control group (p < 0.05). Therefore, it was concluded that the SHCH may be useful to prevent postoperative intraperitoneal adhesion in rats.

Study of the Rheological Properties of a Fermentation Broth of the Fungus Beauveria bassiana in a Bioreactor Under Different Hydrodynamic Conditions

  • Fermentation with filamentous fungi in a bioreactor is a complex dynamic process that is affected by flow conditions and the evolution of the rheological properties of the medium. These properties are mainly affected by the biomass concentration and the morphology of the fungus. In this work, the rheological properties of a fermentation with the fungus Beauveria bassiana under different hydrodynamic conditions were studied and the rheological behavior of this broth was simulated through a mixture of carboxymethyl cellulose sodium and cellulose fibers (CMCNa-SF). The bioreactor was a 10 L CSTR tank operated at different stir velocities. Rheological results were similar at 100 and 300 rpm for both systems. However, there was a significant increase in the viscosity accompanied by a change in the consistence index, calculated according to the power law model, for both systems at 800 rpm. The systems exhibited shear-thinning behavior at all stir velocities, which was determined with the power law model. The mixing time was observed to increase as the cellulose content in the system increased and, consequently, the efficiency of mixing diminished. These results are thought to be due to the rheological and morphological similarities of the two fungal systems. These results will help in the optimization of scale-up production of these fungi.
