  • Equinechorionic gonadotropin(eCG) is a member of the glycoprotein hormone family which includes FSH, hCG, TSH. These hormone family is characterized by a heterodimeric structure composed a common α-subunit noncovalently linked to a hormone specific β-subunit. To determine a and β-subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-molecule by fusing the carboxyl terminus of the eCG β-subunit to the amino terminus of the α-subunit was constructed and transfected into chinese hamster ovary (CHO-K1) cells. An analysis of the arm-type site binding domain of bacteriophage .lambda. integrase

  • The 356 amino acid long lambda integrase protein of bacteriophage .lambda. constains two autonomous DNA binding domains with distinct sequence specificities. The amino terminal domain of integrase is implicated to bind to the arm-type sequences and the carboxyl domain interacts with the coretype sequencess. As a first step to understand the molecular mechanism of the integrase-DNA interaction at the arm-type site, the int(am)94 gene carrying an amber mutation at the 94th codon of the int was cloned under the control of the P$\_$tac/ promoter and the lacI$\_$q/ gene. The Int(am)94 mutant protein of amino terminal 93 amino acid residues can be produced at high level from a suppressor free strain harboring the plasmid pInt(am)94. The arm-type binding activity of Int(am)94 were measured in vivo and in vitro. A comparison of the arm-type binding properties of the wild-type integrase and the truncated Int(am)94 mutant indicated that the truncated fragment containing 93 amino acid residues carry all the determinants for DNA binding at the arm-type sites.

  • MatR in Rhizobium trifolii is a malonate-responsive transcription factor that regulates the expression of genes, matABC, enabling decarboxylation of malonyl-CoA into acetyl-CoA, synthesis of malonyl-CoA from malonate and CoA, and malonate transport. According to an analysis of the amino acid sequence homology, MatR belongs to the GntR family The proteins of this family have two-domain folds, the N-terminal helix-turn-helix DNA-binding domain and the C-terminal ligand-binding domain. In order to End the malonate binding site and amino acid residues that interact with RNA polymerase, a site-directed mutagenesis was performed. Analysis of the mutant MatR suggests that Arg-160 might be involved in malonate binding, whereas Arg-102 and Arg-174 are critical for the repression activity by interacting with RNA polymerase.

  • Hyperbranched polysiloxysilanes (HBPSs), with a variety of terminal functional groups (vinyl, epoxy, carboxyl and hydroxyl), were synthesized by the self-polymerization of an $AB_3$ type monomer of tris(dimethylvinylsiloxy) silane, with subsequent end-functionalizations, such as epoxidation and radical addition reaction, respectively. The ratio of the $\alpha-and$ $\beta-addition$ linkages in the HBPS polymerized by hydrosilylation of the $AB_3$ monomer was found to be approximately 1 to 3. The thermal stability and solubility were affected by the terminal functional groups.

Cloning of cDNA Encoding PAS-4 Glycoprotein, an Integral Glycoprotein of Bovine Mammary Epithelial Cell Membrane

  • Bovine PAS-4 is an integral membrane glycoprotein expressed in mammary epithelial cells. Complementary DNA (cDNA) cloning of PAS-4 was performed by reverse-transcriptase polymerase chain reaction (RT-PCR) with oligonucleotide probes based on it's amino terminal and internal tryptic-peptides. The cloned PAS-4 cDNA was 1,852 nucleotides (nt) long and its open reading frame (ORF) was encoded 1,413 base long. The deduced amino acid sequence indicated that PAS-4 consisted of 471 amino acid residues with molecular weight of 52,796, bearing 8 potential N-glycosylation sites and 9 cysteine residues. Partial bovine CD36 cDNA from liver also was sequenced and the homology of both nucleotide sequence was 94%. Most of the identical amino acid residues were in the luminal/extracellular domains. Contrary to PAS-4, bovine liver CD36 displays 6 potential N-glycosylation sites, which were located, except for those at positions 101 and 171, at same positions as PAS-4 cDNA. Cysteine residues of PAS-4 and CD36 were same at position and in numbers. Northern blot analysis showed that PAS-4 was widely expressed, although its mRNA steady-state levels vary considerably among the analyzed cell types. PAS-4 possessed hydrophobic amino acid segments near the amino- and carboxyl-termini. Two short cytoplasmic tails of the amino- and carboxyl-terminal ends constituted of a 5-7 and 8-11 amino acid residues, respectively.

Altered APP Carboxyl-Terminal Processing Under Ferrous Iron Treatment in PC12 Cells

  • Amyloid-${\beta}$ peptide ($A{\beta}$), generated by proteolytic cleavage of the amyloid precursor protein (APP), plays a pivotal role in the pathogenesis of Alzheimer's disease (AD). The key step in the generation of $A{\beta}$ is cleavage of APP by beta-site APP-cleaving enzyme 1 (BACE1). Levels of BACE1 are increased in vulnerable regions of the AD brain, but the underlying mechanism is unknown. In the present study, we reported the effects of ferrous ions at subtoxic concentrations on the mRNA levels of BACE1 and a-disintegrin-and-metalloproteinase 10 (ADAM10) in PC12 cells and the cell responses to ferrous ions. The cell survival in PC12 cells significantly decreased with 0 to 0.3 mM $FeCl_2$, with 0.6 mM $FeCl_2$ treatment resulting in significant reductions by about 75%. 4,6-diamidino-2-phenylindole (DAPI) staining showed that the nuclei appeared fragmented in 0.2 and 0.3 mM $FeCl_2$. APP-${\alpha}$-carboxyl terminal fragment (APP-${\alpha}$-CTF) associations with ADAM10 and APP-${\beta}$-CTF with BACE1 were increased. Levels of ADAM10 and BACE1 mRNA increased in response to the concentrations of 0.25 mM, respectively. In addition, p-ERK and p-Bad (S112, S155) expressions were increased, suggesting that APP-CTF formation is related to ADAM10/ BACE1 expression. Levels of Bcl-2 protein were increased, but significant changes were not observed in the expression of Bax. These data suggest that ion-induced enhanced expression of AMDA10/BACE1 could be one of the causes for APP-${\alpha}/{\beta}$-CTF activation.

In vivo putative O-GlcNAcylation of human SCP1 and evidence for possible role of its N-terminal disordered structure

  • RNA polymerase II carboxyl-terminal domain (RNAPII CTD) phosphatases are responsible for the dephosphorylation of the C-terminal domain of the small subunit of RNAPII in eukaryotes. Recently, we demonstrated the identification of several interacting partners with human small CTD phosphatase1 (hSCP1) and the substrate specificity to delineate an appearance of the dephosphorylation catalyzed by SCP1. In this study, using the established cells for inducibly expressing hSCP1 proteins, we monitored the modification of ${\beta}$-O-linked N-acetylglucosamine (O-GlcNAc). O-GlcNAcylation is one of the most common post-translational modifications (PTMs). To gain insight into the PTM of hSCP1, we used the Western blot, immunoprecipitation, succinylayed wheat germ agglutinin-precipitation, liquid chromatography-mass spectrometry analyses, and site-directed mutagenesis and identified the $Ser^{41}$ residue of hSCP1 as the O-GlcNAc modification site. These results suggest that hSCP1 may be an O-GlcNAcylated protein in vivo, and its N-terminus may function a possible role in the PTM, providing a scaffold for binding the protein(s).

Solution Structure and Backbone Dynamics of the Biotinylation Domain of Helicobacter pylori Biotin-carboxyl Carrier Protein

  • Acetyl-CoA carboxylase (ACC) is an excellent candidate for antibiotics drug target, which mediates malonyl-CoA synthesis from acetyl-CoA through acetylation process. It is also involved in the committed step of fatty acid synthesis which is essential for living organisms. We have determined the three dimensional structure of C terminal domain of HP0371, biotin-carboxyl carrier protein of H. pyroli, in solution state using heteronuclear multi-dimensional NMR spectroscopy. The structure of HP0371 shows a flatten b-sheet fold which is similar with that of E. coli. However, the sequence and structure of protruding thumb are different with that of E. coli and the thumb shows different basis of structural rigidity based on backbone dynamics data.

  • 유연격자와 메타크릴로일 기 사이에 도입된 벤젠 고리와 메소겐 구조의 말단에 연결된 키랄 중심을 갖는 새로운 두 계열의 메타크릴레이트 단량체를 합성하였다. 이 두 계열에서 유연격자와 메소겐 기의 길이 변화에 따른 액정형성에 대한 효과를 조사하였다. 첫 번째 계열의 화합물들은 합성과정에서 얻어진 중간물질이나 최종물질에서 액정성을 보이지 않았다. 그러나 두 번째 계열에서는 중간물질 뿐만아니라 최종물질에서도 액정성을 보였다.

PU.1 유전자(cDNA)의 인위적 변이체 클로닝 (Molecular Cloning of Mutant cDNA of PU.1 Gene)

  • PU.1은 6개의 특이적인 purine-rich 염기서열 (5' -GAGGAA-3 )로 구성된 PU box에 결합하는 transcription activator이다. 이 PUol은 macro phage와 B-cell에서만 발현되어 이들 세포를 활성 화시키므로, 포유통물의 연역계를 연구하는 데 중요 한 위치를 차지하고 있다. Full length PUol cDNA 는 open reading frame 816개의 DNA 염기로 구성 되어 있으므로, 아미노산 2727~의 합성을 지령한다. PUol의 활성화는 이를 구성하고 있는 polypeptide 중 세린 잔기가 인산화되어 전사인자로서 작용한다 고 추측된다. PU.1은 22개의 세린을 함유하고 있으 며, 정확한 인산화 위치 빛 수량은 알 수 없으나, casein kinase II 에 의하여 인산화된다고 추측되는 제41,45,132'133,148번째 아미노산 세린들이 제1 차 target sites이다. 본 연구에서는 이상의 제41, 45, 132,133, 148번 아미노산 세린 codon(AGC, AGC, AGC.TCA, TCT)이 알라닌 codon(GCC, GCC, GCC.GCA, G GCT)으로 치환된 4가지의 점돌연변이체 클론 (pKKS41A, pRKS45A, pMKS132$.$133A, pMKS­1 148A)을 다음과 같이 제조하였다. Wild type PUol cDNA(template)를 해당되는 mutant DNA primers로 증폭(PCRjSOE)하여 mutant cDNA 단편을 얻었다. 이를 Hind III와 Xba I 으로 절단된 pBlu­e escript KS +에 접합시킨 후, 대장균(E. coli XLI ~ Blue)에 형질전환시켰다. 이 점돌연변이체들은 인산화 부위 및 수량은 물론 PU.1의 구조 및 기능 (Structure and Function) 연구에 기여할 것이다.
