A bacterium which grows on cyclohexanol as sole carbon and energy source was isolated from sludge of industrial areas in Taegu and identified as Acinetobacter calcoaceticus C-15. The growth medium for the optimal culture condition was composed of 0.2% cyclohexanol, 0.11% $NH_4Cl$, 0.05% $KH_2PO_4$, 0.2% $K_2HPO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, and 0.05% yeast extracts. The optimal pH value and temperature for the growth were 7.2 and $33^{\circ}C$, respectively. Specific growth rate of A. calcoaceticus C-15 at $33^{\circ}C$ on the cyclohexanol and cyclohexanone was $0.27hr^{-1}$ and $0.15hr^{-1}$, respectively. Growth yield for cyclohexanol was 1.0. The bacteria utilized ethanol, 1-butanol, 1-pentanol, and cyclohexanol as a carbon source but not methanol, 1-hexanol, m-cresol, glycerol, and cyclohexane. The bacteria grew on benzoate, adipate, acetate, and citrate, but did not on salicylate, phthalate, p-hydroxybenzoate, and gluconate. A calcoaceticus C-15 did not utilize all kind of sugars other than xylose. Cell-free extracts contained $NAD^+$-linked cyclohexanol dehydrogenase which catalized the oxidation of cyclohexanol to cyclohexanone.
Anammox (Anaerobic Ammonium Oxidation) bacteria is recently discovered microorganism which can oxidize ammonium to nitrogen gas in the presence of nitrite under anaerobic conditions. The anammox process can save an energy for nitrification and need not require a carbon source for denitrification, however, the start-up periods takes a long time more than several months due to the long doubling time (approximately 11 days). In order to find the effects of seeding microorganisms, hydrazine, and nitrite concentration on the enhancement of the anammox activity, five kinds of microorganisms were selected. Among the several kinds of seeding microorganisms, the granule from acclimated microorganisms treating high concentration of ammonia nitrogen (A-1) and sludge from piggery wastewater treatment plant (A-2) were found to have a high anammox activity. In the case of A-1, the maximum nitrogen conversion rate represented 0.4 mg N/L-hr, and the amount of nitrite utilization was high compared to those of other seeding microorganisms. The A-4 represented a higher nitrogen conversion rate to 0.7 mg N/L-hr although the ammonium concentration in the serum bottle was high as 200 mg/L. Meanwhile, the anaerobic granule from UASB reactor treating distillery wastewater showed a low anammox activity due to the denitrification by the remained carbon sources in the granule. Hydrazine, intermediate product in anammox reaction, enhanced the anammox activity by representing 1.4 times of nitrogen gas was produced in the test bottle than that of control, when 0.4 mM of $N_2H_4$ was added to serum bottle which contains 5 mM of nitrite. The high concentration of nitrite (10 mM) resulted in the decrease of the anammox activity by showing lower production of nitrogen gas compared to that of 5 mM addition of nitrite concentration. As a result of FISH (Florescence In-Situ Hybridization) experiment, the Amx820 probe showed a more than 13% of anammox bacteria in a granule (A-1).
This study investigated the physiological characteristics of biofilm microorganisms formed onto the different drinking water distribution pipe surfaces. The simulated drinking water distribution pipe system which had several PVC, STS 304, and GS coupons was operated at flow velocity of 0.08 m/sec (Re 1,950) and 0.28 m/sec (Re 7,300), respectively. At velocity of 0.08 m/sec, the number of viable heterotrophic bacteria in the biofilm over the 3 months of operation averaged $3.3{\times}10^4$, $8.7{\times}10^4$, and $7.2{\times}10^3CFU/cm^2$ for PVC, STS, and GS surfaces, respectively. The number of attached heterotrophic bacteria averaged $1.4{\times}10^3$, $5.6{\times}10^2$, and $6.5{\times}10^2CFU/cm^2$ on PVC, STS, and GS surfaces at the system with relatively high flow velocity of 0.28m/sec. The changes of physiological profile of biofilm-forming microorganisms were characterized by community-level assay that utilized the Biolog GN microplates. Biofilms that formed on different pipe surfaces displayed distinctive patterns of community-level physiological profile (CLPP), which reflected the metabolic preference for different carbon sources and/or the utilization of these carbon sources to varying degrees. The CLPP patterns have shown that the metabolic potential of a biofilm community was different depending on the pipe material. The effect of the pipe material was also characterized differently by operation condition such as flow rate. At flow velocity of 0.08 m/sec, the metabolic potential of biofilm microorganisms on GS surface showed lower levels than PVC and STS biofilms. For biofilms on pipe material surfaces exposed to water flowing at 0.28 m/sec, the metabolic potential was in order of PVC>GS>STS. Generally, the levels of the bacterial biofilm's metabolic potentials were shown to be notably higher on pipe surfaces exposed to water at 0.08 m/sec when compared to those on pipe surfaces exposed to water at 0.28 m/sec.

This study was performed to understand combustion properties four major Indonesian wood species such as Albizia, Gmelina, Mangium and Mindi were investigated by cone-calorimeter for better utilization of theses wood species. Heat release rate (HRR), total heat release (TSR), specific mass loss rate (SMLR), effective heat of combustion (EHC), time to ignition (TTI), flame time (FT), specific extinction area (SEA), smoke production rate (SPR) and CO compound production rate were measured. HRR, THR and FT were proportional to the density of woods. Albizia showed the highest HRR, while Mindi had the lowest HRR. For SPR, Albizia showed the highest value due to its higher SEA. On the other hand, Mindi had the lowest SPR due to a lower SEA value. The highest smoke emission was for Albizia at the beginning of combustion. After 300 seconds, smoke emission of Gmleina and Mangium was increased greatly. Mangium and Mindi showed the highest total carbon dioxide emission. Expecially, Gmelina released the highest carbon monoxide during the combustion period and presented three times higher $CO/CO_2$ ratio than those of other species due to incomplete combustion.
The abatement of methane emission from ruminants is an important global issue due to its contribution to greenhouse gas with carbon dioxide. Methane is generated in the rumen by methanogens (archaea) that utilize metabolic hydrogen ($H_2$) to reduce carbon dioxide, and is a significant electron sink in the rumen ecosystem. Therefore, the competition for hydrogen used for methanogenesis with alternative reductions of rumen microbes should be an effective option to reduce rumen methanogenesis. Some methanogens parasitically survive on the surface of ciliate protozoa, so that defaunation or decrease in protozoa number might contribute to abate methanogenesis. The most important issue for mitigation of rumen methanogenesis with manipulators is to secure safety for animals and their products and the environment. In this respect, prophylactic effects of probiotics, prebiotics and miscellaneous compounds to mitigate rumen methanogenesis have been developed instead of antibiotics, ionophores such as monensin, and lasalocid in Japan. Nitrate suppresses rumen methanogenesis by its reducing reaction in the rumen. However, excess intake of nitrate causes intoxication due to nitrite accumulation, which induces methemoglobinemia. The nitrite accumulation is attributed to a relatively higher rate of nitrate reduction to nitrite than nitrite to ammonia via nitroxyl and hydroxylamine. The in vitro and in vivo trials have been conducted to clarify the prophylactic effects of L-cysteine, some strains of lactic acid bacteria and yeast and/or ${\beta}$1-4 galactooligosaccharide on nitrate-nitrite intoxication and methanogenesis. The administration of nitrate with ${\beta}$1-4 galacto-oligosaccharide, Candida kefyr, and Lactococcus lactis subsp. lactis were suggested to possibly control rumen methanogenesis and prevent nitrite formation in the rumen. For prebiotics, nisin which is a bacteriocin produced by Lactococcus lactis subsp. lactis has been demonstrated to abate rumen methanogenesis in the same manner as monensin. A protein resistant anti-microbe (PRA) has been isolated from Lactobacillus plantarum as a manipulator to mitigate rumen methanogenesis. Recently, hydrogen peroxide was identified as a part of the manipulating effect of PRA on rumen methanogenesis. The suppressing effects of secondary metabolites from plants such as saponin and tannin on rumen methanogenesis have been examined. Especially, yucca schidigera extract, sarsaponin (steroidal glycosides), can suppress rumen methanogenesis thereby improving protein utilization efficiency. The cashew nutshell liquid (CNSL), or cashew shell oil, which is a natural resin found in the honeycomb structure of the cashew nutshell has been found to mitigate rumen methanogenesis. In an attempt to seek manipulators in the section on methane belching from ruminants, the arrangement of an inventory of mitigation technologies available for the Clean Development Mechanism (CDM) and Joint Implementation (JI) in the Kyoto mechanism has been advancing to target ruminant livestock in Asian and Pacific regions.

It is likely that maltose could provide a good substrate for the bacteria in the intestine, when the pathogenic bacteria invade and colonize in human gut. For better understanding of this organism's maltose metabolism, a mutant that was not able to grow with maltose as a sole carbon source was screened from a library of mutants constructed by a random transposon mutagenesis. By a transposon-tagging method, malPQ genes encoding a maltodextrin phosphorylase and a 4-${\alpha}$-glucanotransferase, were identified and cloned from Vibrio vulnificus. The deduced amino acid sequences of malPQ from V. vulnificus were 48 to $91\%$ similar to those of MalP and MalQ reported from other Enterobacteriaceae. Functions of malPQ genes were assessed by the construction of mutants whose malPQ genes were inactivated by allelic exchanges. When maltose was used as the sole carbon source, neither malP nor malQ mutant was able to grow to a substantial level, revealing that the MalP and MalQ are the only enzymes for metabolic utilization of maltose. The malQ mutant exhibited decreased adherence toward intestinal epithelial cells in vitro, but there was no difference in the $LD_{50}s$ of the wild-type and the malQ mutant in mice. Therefore, it appears that MalQ is less important in the pathogenesis of V. vulnificus than would have been predicted by considering maltose as a most common sugar in the intestine, but not completely dispensable for virulence in mice.
Yeast strains useful for the production of wine using mandarine orange, Citrus unshiu, as a main substrate were screened, and their primary ability to decompose citric acid that affects directly wine quality was investigated. Total eleven strains were selected for brewing orange wine. Five wild strains were from soil-based collections and identified: four of them were Saccharomyces cerevisiae and one of them was S. ellipsoideus. The rest of six strains were from among eighteen laboratory strains: three of them were S. cerevisiae, and the other three were S. coreanus, S. uvarum, and S. sake. Two strains of S. cerevisiae out of these selections were chosen and their decomposition of citric acid was investigated. Citric acid was not utilized as sole carbon source for cellular growth. However, when both citric acid and glucose were added together as carbon sources, decrease of citric acid concentration was observed after incubation. Shaking incubation was more effective for the reduction of citric acid than standing incubation. Utilization of citric acid did not contribute to the increase of ethanol concentration during fermentation. On the other hand, it appeared that citric acid caused partial inhibition of cellular growth of the yeasts.

The depletion of conventional oil reserves and the increasing energy need in developing countries such as China and India result in exceeding oil demand over supply. As a solution of the problem, the efficient utilization of heavy oil has been receiving more and more interest. In order to utilize heavy oil, upgrading processes are required. Among the upgrading processes, thermal decomposition is thought to be relatively simple and economical. In this study, to understand basic characteristics of thermal decomposition of heavy oil, we conducted pyrolysis experiments of deasphalted oil (DAO) produced by a solvent deasphalting process. DAO is a mixture of many components and consists mainly of materials of carbon number 20~40. For the comparison with results of DAO pyrolysis, additional pyrolysis experiments with single materials of carbon number 30 ($C_{30}H_{62}$, $C_{30}H_{58}O_4S$, $C_{30}H_{63}O_3P$) were conducted. Pyrolysis experiments were carried out non-isothermally with variation of heating rate (10, 50, $100^{\circ}C$/min) in a thermogravimetric analyzer. Average pyrolysis activation energy determined by using Arrhenius method, Ingraham and Marrier method, and Coats and Redfern method was 72~99 kJ/mol. In the activation energy calculated by Ozawa-Flynn-Wall method, DAO had wider variation than other single materials.
This experiment was conducted to investigate the effect of the reuse of sawdust fermented with swine excretion as bed material on the growth performance of pigs. The sawdust which was already fermented with swine excretion in the pig house for eight months was transported to a fermentation facility for secondary fermentation. A total of 96 pigs with average 30kg of initial body weight were randomly assigned in the $2{\times}2$ factorial design with two levels(0%, 1.5%) of probiotics added for secondary fermentation and two levels(0%, 1.5%) of probiotics in feed. The results obtained are as follows : 1. Total-nitrogen(T-N), $K_2O$, total-carbon(T-C), and carbon/nitrogen(C/N) in sawdust bed showed no significant difference within treatments, but phosphate increased by 57% in average compared to the initial. 2. There was no significant difference in temperature in the sawdust bed treatments. 3. The internal parasite eggs detected were Trichuris suis, Strong. ransomi, Ascaris suum, Coccidia and Balantidium coli. 4. The utilization period of sawdust fermented with swine excretion was 52, 26, 16, 4, 5, 3 days, respectively, with increase of body weight. 5. Average daily gain and feed conversion were significantly improved by adding probiotics in the feed(P<0.05), but there was no difference between fermented sawdust with or without probiotics. 6. There was no significant difference in carcass weight and backfat thickness of pig among treatments(P>0.05).
The realization of carbon neutrality requires cooperation from various stakeholders and the utilization of a governance system. The criteria for participating members are crucial for the successful operation of governance, and it is especially necessary for experts who can provide scientific advice for policy implementation to share a framework for successful consensus. In this study, governance model theory and model structure, governance common success factors by case, and the application of governance cases in the climate and environmental sector of Seoul, were investigated and analyzed to derive common success factors in order to present the activity standards of the science and technology experts participating in governance. The study of the model theory suggested that the model structure is commonly composed of a basic condition-process-result structure, and it was confirmed that common success factors can be derived at the process stage which is the activity period of members. Through the case study of common success factors, overlapping factors were found to be reliability, accountability, transparency, networks, and related factors. The validity of the common success factors was verified using the analysis results of satisfaction survey data from Seoul Governance Committee participants. The results confirmed that reliability was the most valuable factor followed by networks, transparency, and responsibility, and it was found that the related factors were appropriately derived. The findings of this study are expected to be used as an activity factor for science and technology experts to increase the acceptability and effectiveness of carbon-neutral policies in the future.
