Effects of Chrysanthemum boreale M. Water Extract on Serum Liver Enzyme Activities and Kupffer Cells of Carbon Tetrachloride-Induced Rats

  • Effects of water extract obtained from Chrysanthemum boreale M. (CE) on serum enzyme activities and Kupffer cells of carbon tetrachloride ($CCl_4$)-induced rats were investigated. Thirty-two healthy male Sprague-Dawley rats were divided into normal (N), $CCl_4$-induced (T), CE-supplemented (C), and $CCl_4$-induced and CE-supplemented (TC) groups. $CCl_4$ injection significantly increased aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, and alkaline phosphatase activities in serum. Significant increases in total cholesterol and triglyceride concentrations were also observed in $CCl_4$-induced rats. Oral administration of CE at 300 mg/kg body weight significantly decreased serum enzyme levels and suppressed $CCl_4$ hepatotoxicity-induced lipid profile changes. Histological findings showed fatty change, fibrosis and increased number of Kupffer cells in T group. Electron microscopic examination showed increased lysosome content and dilation of rough endoplasmic reticulum within Kupffer cells in T group, whereas CE supplement attenuated liver injury in $CCl_4$-induced liver. These results indicated CE could significantly alleviate CC4-induced hepatotoxicity injury.

어성초 분획물이 사염화탄소로 유발된 흰쥐의 간손상에 대한 보호효과 (Protective Effects of Houttuynia cordata Thunb on Carbon Tetrachloride-induced Hepatotoxicity in Rats)

  • This study was performed to investigate the protective effect of Houttuynia cordata Thunb on hepatotoxicity in carbon tetrachloride$(CCI_4)$ intoxicated rats. The examined effects hexane, chloroform, butanol and water fractions prepared from the Houttuynia cordata Thunb methanol exlract and rats were administrated with those orally once a day for successive 6 days, fellowed by treaoent with $CCl_4$ on the sixth day. After 6 days, the activities of aminotransferase, alkalinephosphatase, ${\gamma}-glutamyl$ transpeptidase, lactate dehydrogenase and contents of triglyceride, hepatic lipid peroxide in butanol fraction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, also depletion glutathione content induced by treatment with $CCl_4$ was prevented by butanol fraction pretreated rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in butanol fiaction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, but the activity of hepatic glutathione-S-transferase was not significantly effect. These results suggest that butanol fiuction of Houttuynia cordata Thunb methanol extract have potent hepatoprotective effect against carbon tetrachloride intoxicated rats.

  • Calvert et al. formulated the hypothesis that carbon tetrachloride ($CCl_4$) acted on the central nervous system to produce and intensify sympathetic discharge which resulted in anoxic necrosis of the liver. Recknagel suggested that the essential feature of $CCl_4$ hepatotoxicity depended on the cleavage of it to $CCl_3$(free radical) and the peroxidative decomposition of cytoplasmic membrane structural lipids. And there are many reports which show the increase of adrenergic activity in hyperthyroidism. In this paper, the influence of thyroxine on the hepatotexicity of carbon tetrachloride was investigated in mice. The results obtained were summarized as follows; 1) Hepatic total lipid and lipid peroxide contents were slightly decreased by L-sodium thyroxine injection(4mg/kg/day for 4days or 6days), but hepatic glycogen content was significantly decreased. 2) Hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity were significantly increased by $CCl_4$ (4 ml/kg single dose or triple dose: 4ml/kg/day for 3days), but hepatic glycogen content was significantly decreased. 3) The increase of hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity induced by $CCl_4$ were significantly inhitited by the pretreatment of thyroxine. 4) The decrease of hepatic glycogen induced by $CCl_4$ was not affected by the pretreatment of thyroxine.

Bergenin 및 Acetylbergenin의 사염화탄소 유발 간독성에 대한 치료효과I (Therapeutic Effects of Bergenin and Acetylbergenin on Carbon Tetrachloride-Induced Hepatotoxicity in Rats)

  • Bergenin is a C-glucoside of 4-O-methyl gallic acid that has been isolated from the cortex of Mallotus japonicus (Euphorbiaceae). Acetylbergenin was synthesized by acetylation from bergenin to increase lipophilic and physiological activities. The therapeutic effects of bergenin and acetylbergenin were evaluated against carbon tetrachloride ($CCl_4$)-induced hepatotoxicity in rats. Bergenin and acetylbergenin were administered orally once daily for successive 5 days, after the intraperitoneal injection of a mixture 0.5 m1/kg of $CCl_4$ in olive oil (1:1). The substantially elevated serum enzymatic activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and ${\gamma}$-glutamyltransferase induced by $CCl_4$ were restored towards normalization by posttreatment with bergenin and acetylbergenin. Bergenin and acetylbergenin also significantly prevented the elevation of hepatic malondialdehyde formation and depletion of glutathione content induced by $CCl_4$ in a dose dependent fashion. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored towards normalization. These results suggest that therapeutic effects of bergenin and acetylbergenin may be related complex mechanisms that involve prevention of lipid peroxidation and preservation of hepatic GSH. The results of this study clearly indicate that bergenin and acetylbergenin have potent hepatothrapeutic action against $CCl_4$-induced hepatotoxicity in rats. In addition, acetylbergenin 50 mgHg showed almost the same levels of hepatoprotective activity as those of bergenin 100 mgAg, indicating the fact that lipophilic acetylbergenin is more effective in the hepatoprotective action against $CCl_4$ than bergenin.

예덕나무피엑스의 사염화탄소 및 갈락토사민 유발 간독성에 대한 보호 및 치료효과 (Protective and Therapeutic Effects of Malloti Cortex Extract on Carbon Tetrachloride- and Galactosamine-induced Hepatotoxicity in Rats)

  • Hepatoprotective effects of Malloti cortex extract (MCE) from Mallotus japonicus against the carbon tetrachloride (CCl$_{4}$) and galactosamine (GalN) were investigated. Whereas serum aspartate aminotransferase and alanine aminotransferase levels were markedly elevated after CCl$_{4}$ and GalN administration, pretreatment and posttreatment with MCE before and after the injection of CCl$_{4}$ and GalN resulted in decreases in elevated serum aminotransferase activities. Whereas CCl$_{4}$ and GalN treatment caused 3~7 fold increases in sorbitol dehydrogenase and ${\gamma}$-glutamyltransferase activities, pretreatment and posttreatment with MCE resulted in the blocking of CCl$_{4}$ and GalN-induced liver toxicity. The hepatoprotective effect of MCE was in part due to MCE-induced elevation of hepatic glutathione levels. Pretreatment and posttreatment with MCE also reduced increased lipid peroxidation induced by CCl$_{4}$ and GalN. These results suggest that MCE may be useful for the prevention and therapy of hepatotoxic pathogenesis. It is presumed that protective and therapeutic effects of MCE due to be inducible glutathione S-transferase and glutathione reductase activities, involving in glutathione-medicated detoxication and maintainment of glutathione content, respectively.

보원탕(保元湯)이 CCl4로 유도된 간중독 Mouse에 미치는 영향 (Effect of Bowontang on Mouse Hepatoxicity Induced by Carbon tetrachloride)

  • 본 연구는 보원탕(保元湯)이 $CCl_4$로 유도된 간중독 mouse에 미치는 효과를 조사하였다. 실험동물은 3개의 군(정상군, 대조군, 약물투여군)으로 나누었다. 정상군은 마우스용 고형사료와 물을 제한 없이 공급하였으며, 대조군은 7일간 마우스용 고형 사료와 물을 제한 없이 공급한 후 carbon tetrachloride($CCl_4$)를 실험 동물의 체중 kg당 0.6ml을 1일간 복강 주사하여 마우스용 고형사료와 물을 2주간 제한없이 공급하였으며 약물투여군은 7일간 마우스용 고형 사료와 물을 제한 없이 공급한 후 carbon tetrachloride($CCl_4$)를 실험 동물의 체중 kg당 0.6ml을 1일간 복강 주사하여 마우스용 고형사료와 보원탕(保元湯) 추출물을 kg당 900mg씩 2주간 음용시켰다. 보원탕(保元湯) 추출물은 지질과산화 반응에 주요원인이 되는 자유기에 대한 직접적인 소거능을 나타내었고 혈청중 GPT, GOT, ALP 및 LPO는 대조군에 비해 약물투여군이 유의성 있는 감소를 나타내었으며 GSH 함량 및 catalase 활성은 약물투여군이 대조군에 비해 유의성 있게 증가하였다. 이 결과들로 보아 $CCl_4$로 유도된 mouse의 간손상에 보원탕(保元湯)이 유효한 것으로 확인되었으며 앞으로 이에 대한 심도 있는 연구가 계속되어야 할 것으로 사료된다.

  • These studies were attempted to investigate the therapeutic effect earthworm (Lamnodrilus gotai Hatai) extract (LS) on the liver damage induced by carbon tetrachloride ($CCl_4$) in rats. Male rats (Sprague-Dawley) were treated with sub-chronic concentration of $CCl_4$, which was 0.2 ml/kg of $CCl_4$ via peroral administration twice a week. LS administration to rats (6 ml/kg,po) prevented the development of fatty liver, necrosis and fibrosis induced by $CCl_4$. Furthermore, as compared with rats suspended after treated with $CCl_4$ only, LS treatment significantly decreased the increment in serum enzyme activities, liver enlargement and improved growth rate. This protective effect was observed when LS was given monthly for 4 months after $CCl_4$ treated for 2 months. These results suggest that earthworm extract has appreciable therapeutic effect on $CCl_4$-induced hepatotoxicity.

  • The brown alga Saccharina japonica is consumed as a foodstuff in many countries. Carbon tetrachloride ($CCl_4$) is a potent hepatotoxin that is used to assess hepatotoxicity in animal models. This study assessed the protective effect of S. japonica extract (SJE) on $CCl_4$-induced acute liver injury in rats. Experimental rats were divided into the following three groups: control, $CCl_4$, and $CCl_4$+SJE; the latter two groups were given 150 or 300 mg SJE/kg orally for 10 days. Three hours after the final treatment, all rats-except for those in the control group-were administered intraperitoneal injections of $CCl_4$. One day later, blood and liver samples were collected for evaluation biomarker of levels. Aspartate transaminase (APT; GPT) and alanine aminotransferase (ALT; GOT) levels were markedly lower in the $CCl_4$+SJE group than the $CCl_4$ group. The hepatic superoxide dismutase (SOD) activity of the $CCl_4$+SJE group was significantly lower than that of the $CCl_4$ group. In comparison, glutathione S-transferase (GSH) and catalase (CAT) levels were significantly higher in the $CCl_4$+SJE group. Western blotting revealed that SJE attenuated the $CCl_4$-induced EGFR and MAPK activity in the liver. Thus, we conclude that SJE protects against $CCl_4$-induced hepatotoxicity.

Tungtungmadic Acid Isolated from Salicornia herbacea Suppresses the Progress of Carbon Tetrachloride-induced Hepatic Fibrosis in Mice

  • Tungtungmadic acid(3-caffeoyl, 4-dihydrocaffeoyl quinic acid: CDCQ) is a new chlorogenic acid derivative isolated from the Salicornia herbacea. The suppressive effects of CDCQ on the progress of acute carbon tetrachloride($CCl_4$)-induced hepatic fibrosis were investigated in mice. CDCQ significantly suppressed $CCl_4$-induced hepatic necrosis and inflammation, as determined by serum enzymatic activities of alanine and aspartate aminotransferase and serum TNF-$\alpha$ levels in a dose-dependent manner. In addition, increased hepatic lipid peroxidation and fibrosis after acute $CCl_4$ treatment were suppressed by the administration of CDCQ. CDCQ also significantly prevented the elevation of hepatic hydroxyproline and collagen content and ${\alpha}$-smooth muscle actin(${\alpha}$-SMA) expression in the liver of $CCl_4$-intoxicated mice. These results suggest that the suppressive effects of CDCQ against the acute $CCl_4$-induced hepatic fibrosis possibly related to its ability to block both hepatic inflammation and the activation of hepatic stellate cells.

Evaluation of the Efficacy of Kochiae fructus Extract in the Alleviation of Carbon Tetrachloride-induced Hepatotoxicity in Rats

  • Hepatoprotective effects of the extract of Kochiae fructus (KF), a traditional oriental medicinal plant, were evaluated against carbon tetrachloride($CCl_4$)-induced liver damage in rats. Male Sprague-Dawley rats were divided into control, $CCl_4,\;CCl_4$ plus methanol extract of KF (KFM-$CCl_4$), and $CCl_4$ plus butanol extract of KF (KFB-$CCl_4$) groups. KFM and KFB were orally administered once a day (200 mg/kg body weight) for 14 days. A mixture of 0.2 mL/100 g body weight of $CCl_4$ in olive oil was injected at 30 minutes after the final administration of KFM and KFB. The KFB pretreatment resulted in a significant decrease in the serum transaminase and lactic dehydrogenase levels in the $CCl_4$-treated rats. The $CCl_4$ treatment significantly lowered the activities of glutathione, glutathione reductase (GR), glutathione-S-transferase (GST), superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-Px). However, pretreatment with KFM and KFB resulted in a significant increase in the glutathione, GR and GST levels. KFB increased the activities of SOD, catalase and GSH-Px, but KFM did not alter them. Pretreatment with KFM and KFB resulted in a significant decrease in the production of aminopyrine N-demethylase in the $CCl_4$-treated rats. KF extract would appear to contribute to alleviate the adveISe effect of $CCl_4$ treatment by enhancing the hepatic antioxidant defense system.
