Roles of the meta- and the ortho-Cleavage Pathways for the Efficient Utilization of Aromatic Hydrocarbons by Sphingomonas yanoikuyae Bl

  • Catabolic pathways for the degradation of various aromatics by Sphingomonas yanoikuyae Bl are intertwined, joining at the level of substituted benzoates, which are further degraded vita ring cleavage reactions. The mutant strain EK497, which was constructed by deleting a large DNA region containing most of the genes for biphenyl, naphthalene, m-xylene, and m-toluate degradation, was unable to grow on all of the aromatics tested except for benzoate as the sole source of carbon and energy.S. yanoikuyae EK497 was found to possess only catechol ortho-ring cleavage activity due to deletion of the genes for the meta-cleavage pathway. Wild-type S. yanoikuyae Bl grown on benzoate has both catechol orthoand meta-cleavage activity. However, m-xylene and m-toluate, which are metabolized through methylbenzoate, and biphenyl, which is metabolized through benzoate, induce only the meta-cleavage pathway, suggesting the presence of a substrate-dependent induction mechanism.

  • To optimize the culture conditions for Beauveria bassiana 726, the effects of culture medium, pH, and temperature on mycelium and spore production were investigated. The optimum temperature and pH for the cultivation of B. bassiana 726 were 28 $^{\circ}C$ and 5.0, respectively. The optimized medium was composed of 1.0~2.0% total sugar from molasses, 0.5% corn steep liquor and 0.05% KH$_2$PO$_4$. In the cultivation of B. bassiana 726 with the optimum medium, the specific growth rate and substrate utilization were well-fitted with the proposed kinetic model in the shake flask and stirred tank reactor. When the fed-batch cultivation using carbon suorce, nitrogen source, and mineral salt as a feeding medium was compared with batch cultivation in stirred tank reactor, mycelium (12.7 g/L) and spore production (5.4$\times$$10^8/mL$) were enhanced up to 110% and 85%, respectively.

Correlation between Phylogeny and Zn-Resistance in Methylobacterium Species Isolated from Non-Polluted Soil Environments

  • Zn-resistant Methylosobacterium strains were isolated from several non-polluted soil samples collected in all over Japan. Zn-resistant Methylosobacterium strains were predominantly detected in all soil samples and they were also characterized as a UV-tolerant bacteria. The MIC test revealed that the isolates have high zinc resistance in comparison with that of reference Methylosobacterium strains obtained from culture collections. The 16S rDNA-based phylogenetic analysis showed that all strains were divided into two clusters designated as cluster A and cluster B in the present study. All isolates were distributed only in the cluster A. The phylogenetic clustering also well coincided with the differences in the pattern of carbon source utilization.

  • Xinlei studies on the production of pullulan by Aureobasidium pullulans using batch culture in a 15L bioreactor were carried out. The mathematical models were obtained in this study, which provided a reasonable description for the biomass, the product, and the substrate variation with time. The values frets the mathematical models were satisfactorily coincided with the experimental data for the biomass of A. pullulans, the production of pullulan and the utilization of sucrose as the sole carbon source.

Succinic Acid Production by Anaerobiospirillum succiniciproducens ATCC 29305 Growing on Galactose, Galactose/Glucose, and Galactose/Lactose

  • Succinic acid-producing Anaerobinspirillum succiniciproducens was anaerobically grown on galactose, galactose/glucose, or galactose/lactose in order to study its galactose fermentation. Unlike a previous report, A. succiniciproducens was found to efficiently metabolize galactose as the sole carbon source at a rate of 2.4 g/g-DCW/h and produced succinic acid with as high a yield of 87% as with using glucose. When glucose and galactose were present, A. succiniciproducens metabolized both sugars simultaneously. Furthermore, when lactose and galactose coexisted, lactose did not inhibit the galactose fermentation of A. succiniciproducens. Therefore, co-utilization of galactose and other sugars can improve the productivity and economy of bio-based succinic acid processes.

  • To find microorganisms producing esterase inhibitors, microbes were isolated from soil samples that were collected at different locations in Korea and screened for inhibitory activities. One of the inhibitor-producing strains was named strain DMC-498. This strain was found to be a new species of the genus Streptomyces by comparison with the characteristics of morphology and metabolisms of the other species of the genus.

Enhanced pectinase and β-glucosidase enzyme production by a Bacillus subtilis strain under blue light-emitting diodes

  • Bacillus subtilis B22, a chemotrophic and aerobic bacterial strain was isolated from homemade kimchi, identified by 16S rRNA gene sequencing. B22 was primarily screened by biochemical, carbon source utilization tests. B22 was used to produce pectinase and ${\beta}$-glucosidase by submerged fermentation under different light sources. B22 was incubated in pectin media and basal media (pH 7.0) under blue, green, red and white light-emitting diodes (LEDs), fluorescent white light, and in darkness at $37^{\circ}C$, orbital shaker 150 rpm for 24 hours. Fermentation under blue LEDs maximized pectinase production ($71.59{\pm}1.6U/mL$ at 24 h) and ${\beta}$-glucosidase production ($56.31{\pm}1.6U/mL$ at 24 h). Further, the production of enzyme increased to pectinase ($156{\pm}1.28U/mL$) and ${\beta}$-glucosidase ($172{\pm}1.28U/mL$) with 3% glucose as a carbon source. Activity and stability of the partially purified enzymes were higher at pH 6.0 to 8.0 and $25-55^{\circ}C$. The effect on the metal ions $Na^+$ and $K^+$ and (moderateactivity) $Mn^{2+}$ and $Ni^{2+}$ increased activity, while $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, and $Fe^{2+}$ inhibited activity. EDTA, phenylmethylsulfonyl fluoride and 5,5-dithiobis (2-nitrobenzoicacid) reduced activity, while tetrafluoroethylene and 1,10-phenanthroline inhibited activity. The amylase was highly tolerant of the surfactants TritonX-100, Tween-20, Tween-80 and compatible with organic solvents methanol, ethanol, isoamylalcohol, isopropanol, t-butylalcohol and the oxidizing agents hydrogen peroxide, sodium perborate and sodium hypochlorite, although potassium iodide and ammonium persulfate reduced activity. These properties suggest utility of pectinase and ${\beta}$-glucosidase produced by B. subtilis B22 under blue LED-mediated fermentation for industrial applications.

Synergistic Utilization of Dichloroethylene as Sole Carbon Source by Bacterial Consortia Isolated from Contaminated Sites in Africa

  • The widespread use and distribution of chloroethylene organic compounds is of serious concern owing to their carcinogenicity and toxicity to humans and wildlife. In an effort to develop active bacterial consortia that could be useful for bioremediation of chloroethylene-contaminated sites in Africa, 16 combinations of 5 dichloroethylene (DCE)-utilizing bacteria, isolated from South Africa and Nigeria, were assessed for their ability to degrade cis- and trans- DCEs as the sole carbon source. Three combinations of these isolates were able to remove up to 72% of the compounds within 7 days. Specific growth rate constants of the bacterial consortia ranged between 0.465 and $0.716\;d^{-1}$ while the degradation rate constants ranged between 0.184 and $0.205\;d^{-1}$ with $86.36{\sim}93.53\;and\;87.47{\sim}97.12%$ of the stoichiometric-expected chloride released during growth of the bacterial consortia in cis- and trans-DCE, respectively. Succession studies of the individual isolates present in the consortium revealed that the biodegradation process was initially dominated by Achromobacter xylosoxidans and subsequently by Acinetobacter sp. and Bacillus sp., respectively. The results of this study suggest that consortia of bacteria are more efficient than monocultures in the aerobic biodegradation of DCEs, degrading the compounds to levels that are up to 60% below the maximum allowable limits in drinking water.

Physiological Effects of GLT1 Modulation in Saccharomyces cerevisiae Strains Growing on Different Nitrogen Sources

  • Saccharomyces cerevisiae is one of the most employed cell factories for the production of bioproducts. Although monomeric hexose sugars constitute the preferential carbon source, this yeast can grow on a wide variety of nitrogen sources that are catabolized through central nitrogen metabolism (CNM). To evaluate the effects of internal perturbations on nitrogen utilization, we characterized strains deleted or overexpressed in GLT1, encoding for one of the key enzymes of the CNM node, the glutamate synthase. These strains, together with the parental strain as control, have been cultivated in minimal medium formulated with ammonium sulfate, glutamate, or glutamine as nitrogen source. Growth kinetics, together with the determination of protein content, viability, and reactive oxygen species (ROS) accumulation at the single cell level, revealed that GLT1 modulations do not significantly influence the cellular physiology, whereas the nitrogen source does. As important exceptions, GLT1 deletion negatively affected the scavenging activity of glutamate against ROS accumulation, when cells were treated with H2O2, whereas Glt1p overproduction led to lower viability in glutamine medium. Overall, this confirms the robustness of the CNM node against internal perturbations, but, at the same time, highlights its plasticity in respect to the environment. Considering that side-stream protein-rich waste materials are emerging as substrates to be used in an integrated biorefinery, these results underline the importance of preliminarily evaluating the best nitrogen source not only for media formulation, but also for the overall economics of the process.
