구제역 매몰지역 침출수에서 발생하는 난분해성 유기물질 거동 (Behavior of Refractory Organic Matter in Leachate from Landfill Contaminated by Foot-and-mouth Disease)

  • 구제역 매몰지에서 발생하는 침출수는 지하수, 호소수 및 하천수 등의 자연수에서 나타나는 유기물질의 성상(친수성 유기물질: 22~27%)과 달리 친수성유기물질을 44~50%로 포함하고 있었다. 자연수에서는 유기탄소가 미생물의 대사 등에 의해 감소되는 경향을 나타낸다. 하지만, 매몰 초기에 발생하는 구제역 침출수(leachate-1, 2)에서는 RTOC와 RDOC의 농도가 초기 TOC, DOC 농도보다 높게 나타나는 특성이 있다. 매몰시간이 경과하면서 RTOC와 RDOC는 초기 TOC와 DOC보다 낮게 검출되었다. 구제역 침출수 중 386일이 경과한 leachate-6의 RDOC는 RTOC 중에 91%를 차지하였다. 이것은 안정화된 매몰지 환경에서도 침출수 중에는 여전히 미생물 등의 활동에 의해 제거되기 어려운 형태의 난분해성 유기물질이 유기물질의 대부분을 차지하고 있음을 알 수 있다. 따라서 구제역 침출수의 난분해성 유기물질의 검출수준과 성상은 지표수 및 지하수에 대한 영향을 파악하는 정보로서 유효하며, 침출수 내 환경을 이해하는 데에도 기여할 것으로 기대된다.

Dietary Modification for Reducing Electrical Conductivity of Piggery Wastewater

  • A total of 108 pigs (including 36 starters, 36 growers, and 36 finishers) were randomly allocated to six treatments, which involved a 2 (Crude Protein (CP): 100 and 80% of control diet)${\times}$3 (Ca, P, Salt (CPS): 100, 80 and 60% of control diet) factorial design to evaluate the effectiveness of reducing CP and CPS in reducing wastewater EC in different stages. Another 72 starters were adopted to examine the effect of the six treatment diets (as mentioned above) on the growth performance of pigs. Activated carbon and Reverse Osmosis System (RO) were adopted to examine the reducing efficiency of wastewater EC, and ion analysis was also applied to compare with the wastewater EC in different stages of the metabolism trial. The results of wastewater EC of the six treatment diets in different stages of metabolism trial demonstrated that diminishing dietary CP or CPS decreased wastewater EC. The largest decrease of EC was approximately 30%, and was achieved with 20 and 40% reduced dietary CP and CPS, respectively. Pig growth performance deteriorated somewhat when dietary CP or CPS was diminished. Wastewater ion concentration was not always consistent with dietary CP or CPS content, except for $NO_2^{-}$, $NH_4^{+}$ and $K^{+}$, which were positively correlated with dietary CP or CPS in different stages. Activated carbon is not effective for reducing wastewater EC, while, RO system is effective (90% elimination rate) in reducing wastewater EC, but the EC of concentrated (excreted) water is around 10% higher than that of intact wastewater, representing an additional problem besides the high cost of RO system treatment.

Carbon Storage Regulator A (csrA) Gene Regulates Motility and Growth of Bacillus licheniformis in the Presence of Hydrocarbons

  • The global carbon storage regulator (Csr) system is conserved in bacteria and functions as a regulator in the exponential and stationary phases of growth in batch culture. The Csr system plays a role in the central carbon metabolism, virulence, motility, resistance to oxidative stress, and biofilm formation. Although the Csr was extensively studied in Gram negative bacteria, it has been reported only in the control of motility in Bacillus subtilis among Gram positive bacteria. The goal of this study was to explore the role of the csrA gene of Bacillus licheniformis M2-7 on motility and the bacterial ability to use hydrocarbons as carbon source. We deleted the csrA gene of B. licheniformis M2-7 using the plasmid pCsr-L, harboring the spectinomycin cassette obtained from the plasmid pHP45-omega2. Mutants were grown on culture medium supplemented with 2% glucose or 0.1% gasoline and motility was assessed by electron microscopy. We observed that CsrA negatively regulates motility by controlling the expression of the hag gene and the synthesis of flagellin. Notably, we showed the ability of B. licheniformis to use gasoline as a unique carbon source. Our results demonstrated that CsrA is an indispensable regulator for the growth of B. licheniformis M2-7 on gasoline.

Effect of Carbon Dioxide Concentration on Malate and Titratable Acidity in Pereskia aculeata and Kalanchoe rosea

  • The induction of crassulacean acid metabolism(CAM) characterized by day/night acid fluctuation was investigated in leaves from 10 days exposure to elevated $CO_2$ concentration(1,000 ${\mu}L\;L^{-1}CO_2).$ For Pereskia aculeata, have $C_3-like$ gas exchange pattern in well watered condition and shift into CAM-like in water stress, showed a more typical CAM-like diurnal acid fluctuation. Whereas the massive diurnal fluctuation of acidity in typical CAM of Kalanchoe rosea was declined. The effect of short-term exposure to various concentrations of $CO_2$ on titratable acidity in P. aculeata and K. rosea was also investigated. To investigate the response of various $CO_2$ concentrations, four different $CO_2$ levels(350, 700, 1,000 and 1,500 ${\mu}L\;L^{-1})$ were imposed for 24hr and measured the titratable acidity at 06:00, when the acidity was maximum, and 14:00, when the acidity was minimum. The accumulation of acid in P. aculeata was enhanced markedly by higher concentration of $CO_2,$ while the level of acidity in f rosea did not highly respond to $CO_2.$ A notable difference between P. aculeata and K. rosea was the response of de-acidification to a higher concentration of $CO_2$ Increasing with $CO_2,$ the degree of do-acidification of P. auleata was increased while that of K. rosea was depressed.

Proteome analysis of chloroplast proteins in stage albinism line of winter wheat (triticum aestivum) FA85

  • The "stage albinism line of winter wheat" FA85 was a specific natural mutant strain on leaf color. This physiological mutation was controlled by cytogene. In order to reveal the genetic and biochemical mechanism of albinism, 2-DE was used to investigate the difference of chloroplast protein expression pattern between FA85 and its parent wheat Aibian 1. From the results of 2-DE gels analysis, approximately 683 spots were detected on each gel, and 57 spots were expressed differently at least two-fold. Using MALDI-TOF/TOF MS, 14 of 57 spots were identified, which could be categorized into four classes: carbon metabolism, energy metabolism, defense/stress response and signal transduction. Compared with the parent wheat, the expression of ATPase-$\gamma$ and GP1-$\alpha$ was up-regulated in FA85, and of other proteins was down-regulated. Together, we concluded that the expression of chloroplast proteins had changed obviously in FA85, which might be related to the leaf color mutant.

Urea Kinetics in Wethers Exposed to Different Ambient Temperatures at Three Dietary Levels of Crude Protein

  • Eighteen Suffolk wether lambs (BW = 24 kg) were chronically exposed to temperatures of cold (2$^{\circ}C$) or warm (22$^{\circ}C$). The experimental design consisted of a 2${\times}$3 factorial with a single crossover of environment treatment. The sheep were closely shorn and were housed in individual metabolic crates in controlled environment rooms. Sheep consumed pelleted diets ad libitum, which consisted of mainly barley grain and brome grass, and diets contained 7, 11 or 14% crude protein (CP). Animals were catheterized via one jugular vein with a PVC catheter and received a single injection of 60-65 Ci of $^{14}$C]urea. Plasma urea-N (PUN), urinary urea (UU), and carbon specific radioactivity were measured. Urea metabolism was not affected by environment. Percent urea recycling and urea space clearance were highest (p<0.05) on the low nitrogen diet. Urea pool was increased (p<0.10) for the 14% CP diet. Both UU and PUN concentration were positively related (p<0.01) with diet CP content. Therefore, dietary CP content significantly influenced urea metabolism, however, cold exposure did not alter those parameters.

Fasting Heat Production of Growing Buffalo Calves

  • Fasting heat production (FHP) of growing buffalo calves (Bubalus bubalis) in the body weight range of 76 to 236 kg was determined using open circuit respiration chamber. The details of the chambers, calibration of gas analysers and operation of the systems are described. Animals were fasted for 96 hrs during which only water was provided. FHP was determined during next 24 hrs. The mean oxygen consumed, carbon dioxide and methane produced and urinary N excretion per 24 h was $17.03{\ell}$, $11.70{\ell}$, and $0.12{\ell}$ and 0.35 g respectively. The mean respiratory quotient ranged from 0.68 to 0.71, which indicated that post absorptive stage is reached after 96 hrs in growing buffalo calves previously fed ammoniated straw-based ration. Mean FHP of calves was $331.4kJ/kg\;W^{0.75}$. FHP of calves with range of mean body weights of 167 to 235 kg, although nonsignificant but, was almost 12% higher than of calves having mean body weight of 101 kg. Suitable exponent to body weight to describe FHP of buffalo calves was 0.87.

호열성 사상균 Thermoascus aurantiacus의 알코올분해대사 관련 효소학적 특성 (Enzyme Production Related to Alcohol Metabolism from Thermophilic Fungus Thermoascus aurantiacus)

  • 본 균의 생육 및 효소생산에 유용한 탄소원으로서 자연계의 식물에 풍부한 펙틴을 탄소원으로 할 경우, 그 생육도는 전분보다 뛰어났으며, alcohol oxidase와 catalase의 생산량도 높아지는 것으로 나타났다. 특히 alcohol oxidase의 경우는 전분의 15배 이상의 생산량을 보여 본 균과 펙틴 이용성과의 관계를 시사하였고, 세포외 pectin esterase, pectinase등의 높은 활성이 검출되어 이를 증명하였다. 또한 alcohol oxidase 반응에서 생성되는 물질인 formaldehyde를 산화하는 formaldehyde dehydrogenase와, formate를 산화하여 $CO_2$를 생성하는 formate dehydrogenase의 반응을 발견하여, 본 균의 pectin 이용성과 관련한 일련의 에너지 대사계의 존재를 추정할 수 있었다.

Isolation and Characterization of 4-(2,4-Dichlorophenoxy)Butyric Acid-Degrading Bacteria from Agricultural Soils

  • Eight numerically dominant 4-(2,4-dichlorophenoxy) butyric acid (2,4-DB)-degrading bacteria and three pairs of bacteria showing syntrophic metabolism of 2,4-DB were isolated from soils, and their phylogenetic and phenotypic characteristics were investigated. The isolates were able to utilize 2,4-DB as a sole source of carbon and energy, and their 2.4-DB degradative enzymes were induced by the presence of 2.4-DB. Analysis of 16S rDNA sequences indicated that the isolates were related to members of the genera, Variovorax, Sphingomonas, Bradyrhizobium, and Pseudomonas. The chromosomal DNA patterns of the isolates obtained by polymerase-chain-reaction (PCR) amplification of repetitive extragenic palindromic (REP) sequences were distinct from each other. Four of the isolates had plasmids, but only one strain, DB 1, rad a transmissible 2,4-D degradative plasmid. When analyzed with PCR using primers targeted to the tfdA, B, and C genes, only strains DB2 and DB9a produced DNA bands of the expected sizes with the tfdA and C primers, respectively. All of the isolates were able to degrade 2,4-D as well as 2,4-DB, suggesting that the degradation pathways of these compounds were closely related to each other, but respiratory activities of many isolates adapted to 2,4-DB metabolism were quite low with 2,4-D.

Gene-Diet Interaction on Cancer Risk in Epidemiological Studies

  • Genetic factors clearly play a role in carcinogenesis, but migrant studies provide unequivocal evidence that environmental factors are critical in defining cancer risk. Therefore, one may expect that the lower availability of substrate for biochemical reactions leads to more genetic changes in enzyme function; for example, most studies have indicated the variant MTHFR genotype 677TT is related to biomarkers, such as homocysteine concentrations or global DNA methylation particularly in a low folate diet. The modification of a phenotype related to a genotype, particularly by dietary habits, could support the notion that some of inconsistencies in findings from molecular epidemiologic studies could be due to differences in the populations studied and unaccounted underlying characteristics mediating the relationship between genetic polymorphisms and the actual phenotypes. Given the evidence that diet can modify cancer risk, gene-diet interactions in cancer etiology would be anticipated. However, much of the evidence in this area comes from observational epidemiology, which limits the causal inference. Thus, the investigation of these interactions is essential to gain a full understanding of the impact of genetic variation on health outcomes. This report reviews current approaches to gene-diet interactions in epidemiological studies. Characteristics of gene and dietary factors are divided into four categories: one carbon metabolism-related gene polymorphisms and dietary factors including folate, vitamin B group and methionines; oxidative stress-related gene polymorphisms and antioxidant nutrients including vegetable and fruit intake; carcinogen-metabolizing gene polymorphisms and meat intake including heterocyclic amins and polycyclic aromatic hydrocarbon; and other gene-diet interactive effect on cancer.
