여대생의 식품 및 영양소 섭취량 조사 (A Survey of Food and Nutrient Intakes in Female College Students)

  • Purpose: The study was done to investigate Food and Nutrient Intakes of female college students and define differences between teen-age groups and women in their twenties. Method: The subjects of this study were 821 female college students. They were eighteen to twenty nine years old. Nutrient intakes were measured by the Moon, S. J. Scale (1980). Data was collected from June 13 to June, 27. Data was analyzed with the SPSS program by Chi-test, t-test and Pearson's correlation coefficients. Result: Female college students' mean intake of calories, protein, fat, and carbohydrates were $1560.7{\pm}367.71kcal,\;62.0{\pm}19.03g,\;33.1{\pm}9.85g,\;and\;253.7{\pm}57.52g$ respectively. Their mean intake of calcium, phosphorus and iron were $592.9{\pm}221.82mg,\;918.2{\pm}284.24mg\;and\;13.7{\pm}4.50mg$ respectively. Intake of calories, calcium, iron and vitamine B1 were below their Percent of Recommended Intake. Nutrient intakes showed a significant difference between age groups. Teen-age female college students had more nutrient intakes than the women in their twenties. In addition, there was a difference in food consumption behaviors between the teen-age group and the twenty-something group. Conclusion: The nutrient intake of female college students changed by age. Many teen-age female college students ate breakfast well, thus their intake of calories, protein, fat, carbohydrates, minerals such as calcium, phosporus and vitamins was significantly higher than the over twenty years old students. Teen-age female college students like fruits and sugars but over twenty years college students preferred coffee. This study can be useful in directing a dietary consumption status of women to improve nutritional health.

Effects of microbial enzymes on starch and hemicellulose degradation in total mixed ration silages

  • Objective: This study investigated the association of enzyme-producing microbes and their enzymes with starch and hemicellulose degradation during fermentation of total mixed ration (TMR) silage. Methods: The TMRs were prepared with soybean curd residue, alfalfa hay (ATMR) or Leymus chinensis hay (LTMR), corn meal, soybean meal, vitamin-mineral supplements, and salt at a ratio of 25:40:30:4:0.5:0.5 on a dry matter basis. Laboratory-scale bag silos were randomly opened after 1, 3, 7, 14, 28, and 56 days of ensiling and subjected to analyses of fermentation quality, carbohydrates loss, microbial amylase and hemicellulase activities, succession of dominant amylolytic or hemicellulolytic microbes, and their microbial and enzymatic properties. Results: Both ATMR and LTMR silages were well preserved, with low pH and high lactic acid concentrations. In addition to the substantial loss of water soluble carbohydrates, loss of starch and hemicellulose was also observed in both TMR silages with prolonged ensiling. The microbial amylase activity remained detectable throughout the ensiling in both TMR silages, whereas the microbial hemicellulase activity progressively decreased until it was inactive at day 14 post-ensiling in both TMR silages. During the early stage of fermentation, the main amylase-producing microbes were Bacillus amyloliquefaciens (B. amyloliquefaciens), B. cereus, B. licheniformis, and B. subtilis in ATMR silage and B. flexus, B. licheniformis, and Paenibacillus xylanexedens (P. xylanexedens) in LTMR silage, whereas Enterococcus faecium was closely associated with starch hydrolysis at the later stage of fermentation in both TMR silages. B. amyloliquefaciens, B. licheniformis, and B. subtilis and B. licheniformis, B. pumilus, and P. xylanexedens were the main source of microbial hemicellulase during the early stage of fermentation in ATMR and LTMR silages, respectively. Conclusion: The microbial amylase contributes to starch hydrolysis during the ensiling process in both TMR silages, whereas the microbial hemicellulase participates in the hemicellulose degradation only at the early stage of ensiling.

Direct Evidence for the Radioprotective Effect of Various Carbohydrates on Plasmid DNA and Escherichia coli Cells

  • Damage to cells exposed to radiation is primarily attributed to direct effects on the structure of cellular DNA. Radiation-induced damage of pBluescript SK plasmid DNA and Escherichia coli $DH5\alpha$ were examined in the presence of various branched oligosaccharides, polysaccharides, and/or 8-MOP (8-methoxypsoralen). Branched oligosaccharides efficiently protected DNA and cells exposed to ultrasoft X-ray and UV irradiation. In the presence of 0.2% (w/v) branched oligosaccharides and polysaccharides, DNA can be protected from damage due to W and ultrasoft X-ray by a factor of 1.3-2.1 fo1d and 3.2-8.3 fold, respectively. The protective effect of cells exposed to UV or ultrasoft X-ray was also observed by branched oligosaccharides. The combination of MOP, a photoreagent, with carbohydrates increased the protective effects for DNA and cells, compared with that of a single use of MOP or carbohydrate alone.

  • Fast and dynamic biochemical, enzymatic and morphological changes occur during the so-called generative development and during the vegetative processes in seeds. The most characteristic biochemical and compositional changes of this period are the formation and decline of storage components or their precursors, the change of their degree in polymerization and an extensive change in water content. The aim of the present study was to detect the maturation processes in seed nondestructively and to verify the applicability of near infrared spectroscopic methods in the measurement of physiological, chemical and biochemical changes in wheat seed. The amount and variation of different water “species” has been changed intensively during maturation. Characteristic changes of three water absorption bands (1920, 1420 and 1150 nm) during maturation were analysed. It was concluded that the free/bound transition of water molecules could be followed sensitively in different region of NIR spectra. Kinetic changes of carbohydrate reserves were characteristic during maturation. An intensive formation and decline of carbohydrate reserves were observed during early stage of maturation (0 -13 days, high energy demand). An accelerated formation of storage carbohydrates (starch) was detected in the second phase of maturation. Five characteristic absorption bands were analysed which were sensitive indicators the changes of carbohydrates occurred during maturation. Precursors of protein synthesis and the synthesis of reserve proteins and their kinetic changes during maturation were followed from NIR spectra qualitative and qualitatively. Dynamic formation of amino acids and the changes of N forms were detected by spectroscopic, chromatographic and by capillary electrophoresis methods. Calibration equations were developed and validated in order to measure the optimal maturation time protein and moisture content of developing wheat seeds. The spectroscopic methods are offering chance and measurement potential in order to detect fine details of physiological processes. The spectra have many hidden details, which can help to understand the biochemical background of processes.

The Effects of a High-fat or High-sucrose Diet on Serum Lipid Profiles, Hepatic Acyl-CoA Synthetase, Carnitine Palmitoyltransferase-I, and the Acetyl-CoA Carboxylase mRNA Levels in Rats

  • The purpose of this study was to investigate the effects of altering relative intakes of fat and carbohydrates on serum lipid profiles, hepatic acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-I (CPT-I), and the acetyl-CoA carboxlyase (ACC) mRNA level in Sprague-Dawley rats. For four weeks the rats were fed either an AIN-76 diet or one of its modified diets that were supplemented with 20% beef tallow (high-fat diet, HF) and 66.3% sucrose (highsucrose diet, HS). The HS group had significantly higher serum triglyceride and total cholesterol concentrations when compared with the other groups. Serum LDL-cholesterol concentrations in the HS and HF groups were significantly higher when compared to the normal diet (ND) group. Serum HDL-cholesterol levels of the ND and HS groups were significantly higher than those of the HF group. The hepatic total lipid level of the HF group was significantly higher than those of other groups; triglyceride levels of the HS and HF groups were significantly higher than those of the ND group. Hepatic ACS mRNA levels of the HF group were significantly higher than those of the ND group. Hepatic CPT-I mRNA levels were higher in the HF group than other groups. Also, ACC mRNA levels in the liver increased in the HF group. In conclusion, changes in the composition of dietary fat and carbohydrates could affect the hepatic ACS, CPT-I, and ACC mRNA levels. These results facilitate our understanding of the coordinated regulation of the ACS, CPT-I, and ACC mRNA levels and will serve to enhance our understanding of the molecular mechanisms that underlie the regulation of fatty acid metabolism.

미숙아를 출산한 산모의 임신시 영양소 섭취실태에 관한 연구 (I) (A Study on Nutrient Intake During Pregnancy of Women of Premature Delivery (I))

  • Inadequate nutritional status of pregnancy can cause underweight and premature birth, undergrowth and deliverance of physically and mentally defected babyies. The purpose of this study is to provide guidelines for preventing preterm delivery in the aspect of nutritional factors. The nutrient intakes were compared between a preterm delivery group and a normal term delivery group to recognize risk factors of preterm delivery. The results obtained are summarized as follows. The pregnancy period was statistically longer in the normal term group (p<0.0001). Weight increase was statistically higher in the normal term group (p<0.0001). Calories (p<0.05), carbohydrates (p<0.0005), dietary fibers (p<0.0001), potassium (p<0.0005), vitamin $B_1$ (p<0.0005), vitamin $B_6$ (p<0.05), vitamin C (p<0.0001), and folic acid (p<0.05) intakes were statistically higher in the normal term group. Nutrient density of vitamin $B_1$ (p<0.05) and vitamin C (p<0.0001) in the normal term group was statistically higher. Nutrient adequacy ratio of zinc (p<0.05), vitamin $B_1$ (p<0.05) and folic acid (p<0.05) were statistically higher in the normal term group. Index of Nutritional Quality of vitamin $B_1$ (p<0.05) and vitamin C (p<0.0001) were statistically higher in the normal term group. In this study, the normal term delivery showed higher intakes of calories, carbohydrates, dietary fiber, crude fiber, potassium, vitamin $B_1$, vitamin $B_6$ vitamin C and folic acid than the preterm delivery group. Deficiencies in various nutrients may lead to preterm delivery, therefore, balanced nutrient intake is recommended to prevent preterm delivery.

수종의 스포츠 음료를 토끼에 경구 투여한 후 수분 흡수에 대한 비교연구 (Comparative Study on the Absorption of Water after Oral Administration of Various Sports Drinking Beverages to Rabbits)

  • After oral administration of various drinking solutions, the initial absorption rate of water through gastrointestinal tract of the rabbits was evaluated using tritinated water $(^3H_2O)$ as a marker to develop the sports drinking beverage for Korean people. The polynomial curve fitting over 20 min was performed using computer program to obtain the initial absorption rate of water from the tangent line of the fitted equation because initial absorption rate of water was more critical compared to elimination rate during exercise. The amount of water absorbed was increased but a large variation was observed among testing preparations in a small study group $(2{\leq}n{\leq}6)$. The initial absorption rate of water from isotonic sports drinking beverages was statistically significant when compared to hypertonic cola but was not significant when compared to hypotonic solutions (potable water and barley water). In case of hypertonic sports dringking beverages (i.e. Takeda), initial absorption rate of water was not improved and efficient when compared to other isotonic sports dringking beverages. The initial absorption rate of water from prescribed isotonic sample solution containing electrolytes, carbohydrates, and vitamins was not statistically significant when compared to other isotonic drinking beverages but showed similar absorption profile. It was obvious that isotonic solutions simultaneously containing electrolytes, vitamins and carbohydrates (sugar and glucose) had a tendency to increase the initial absorption of water compared to hypotonic (potable water and barley water) and hypertonic preparations (orange juice and cola). Although statistical significance of initial absorption rate of water between isotonic sports drinking beverages and hypotonic potable and barley water was not observed, unlike the hypertonic solutions, isotonic sports drinking beverages may aid not only to replenish loss of water, electrolytes and other nutrients during the exercise but also to prevent dehydration and muscle fatigue, resulting in improved physical performance in an exhausted condition.

Residual frying oil in the diets of sheep: intake, digestibility, nitrogen balance and ruminal parameters

  • Objective: The objective of this study was to evaluate the intake and nutrient digestibility, nitrogen balance and ruminal ammonia nitrogen in lambs of diets containing different levels of residual frying oil. Methods: Levels of 0, 20, 40, 60, and 80 g/kg dry matter (DM) base of residual frying oil in the diets of lambs were evaluated. Five castrated lambs with initial body weights of $36.8{\pm}3.3kg$, distributed in a Latin square ($5{\times}5$) design, were used. Results: There was a decreasing linear effect on the intake of DM, organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), total carbohydrates (TCH), and nonfibrous carbohydrates (NFC). There was an increased linear intake of ether extract (EE). The apparent digestibility of DM, OM, CP, NDF, TCH, and NFC, as well as urine nitrogen excretion, nitrogen balance and ruminal parameters, were not influenced by different levels of residual frying oil in the diet. EE digestibility presented a crescent linear effect. Conclusion: It can be concluded that the addition of residual frying oil to the diets of sheep can affect nutrient intake without affecting the digestibility of most nutrients (with the exception of EE), nitrogen balance and ruminal ammonia nitrogen concentration.

  • 경기도 광릉에서 채집한 자실체로부터 분리한 Lentinus edodes IY105균을 배양하여 얻은 균사체를 알칼리로 추출하여 얻은 물질이 면역계와 sarcoma 180이 이식된 mouse에 미치는 영향을 검토하였다. Lentinus edodes IY105 배양균사체의 알칼리 추출물은 항보체활성과 alternative complement hemolytic 활성을 가지고 있었고, carbon clearance 활성을 가지고 있었고, carbon clearance 활성을 나타내는 것으로 보아 mouse의 망내계에 존재하는 macrophage를 활성화시켜 이 물질에 대한 탐식기능을 증가시킴을 알수 있었다. 또한, 이물질은 mouse의 용혈반 형성 세포수를 증가시키는 것으로 보아 항체생산 자극효과가 있는 것으로 추측되었으며, 종양세포인 sarcoma 180이 이식된 mouse의 암을 50 정도 억제하는 효과를 나타내었다. 알칼리 추출물의 성분을 분석한 결과, 당 17, 단백질 42로 구성된 단백질다당체가 주성분으로 단백다당체는 16종의 아미노산과 52종류의 당으로 구성되어 있었다.

Xylanase를 생산하는 Streptomyces sp. YB914의 특성과 효소 생산성 (Characterization and Xylanase Productivity of Streptomyces sp. YB914)

  • 토양으로부터 세포외로 xylanase를 분비 생산하는 방선균 YB-914가 분리되었으며, 형태, 배양, 생화학적 특성을 조사한 결과 Streptomyces 속 균주로 확인되었다. 분리균의 배양상등액에 존재하는 xylanase는 pH 5.5과 $55^{\circ}C$의 반응조건에서 반응성이 가장 높았으며, pH 4.5~7.0 범위에서 최대활성의 80% 이상을 나타냈다. Xylanase의 생산을 위한 배지를 최적화하기 위해서 G.S.S 배지성분을 여러 종류의 탄수화물로 대체하였다. Oat spelt xylan, corn cob xylan, 밀기울 및 유당과 같은 탄수화물은 Streptomyces sp. YB914의 xylanase 생산성을 증가시키는 것으로 확인되었으며, galactose와 arabinose는 효소 생산을 크게 억제하였다. Oat spelt xylan(1%)와 유당(1.5%)을 함유한 변형배지에서 xylanase의 최대생산성이 48 U/mL로 확인되었다.
