화학적 처리방법에 의한 변성전분의 이화학적 특성 (The Physico-Chemical Characteristics of Modified Starch Made by Chemical Treatment)

  • Developing carbohydrate fat replacer as materials for low-calorie and low-fat food made of Korean potato, it is expected that the new demand of fat replacer will be created. Potato starch was modified by chemical modification. Observing modified starch(treated in different method) by SEM, EZ(treated by enzyme) showed shape of deformed round oval, AC(treated by acetylation,), HPR(treated by hydropropylation) showed shape similar to that of NL(N-Lite), the commercial fat replacer. In the modified starch such as AC, peak in B and C type similar to those of general starch was found, but EZ showed non-crystalline shape. Compared to other modified starch, HPR, the chemically produced denatured starch showed very peculiar peak and structure in V-form. While the order of contents of amylopectin was in the order HPR > EZ > AC showed extremely high contents. Measuring the degree of gelatinization per the modified method, the degree of gelatinization of HPR as much higher than others. The water binding activity of modified starch was 240% in HPR. Measuring viscosity by producing general starch and modified starch as gel of 10% concentration, the CPS showed very high viscosity of 30.30 ${\times}$ 10$^3$ cp. Showing viscosity of 38.60, 31.60 10$^3$ ${\times}$ cp, the modified starch was in the order of HPR. While the calorie of starch of GPS was measured to be 3.0 Kcal/g, very low calorie those of chemically modified starch, HPR showed 2.5 Kcal/g respectively, suggesting that calorie is decreased by modified treatment. The appropriateness of processing food was experimented by substituting the existing oil and fat containing food with saturated gel of starch and modified starch in constant rate through utilization of modified starch. Therefore, research and development for materials and related products which maintain the existing quality and reduce fat contents will be constantly performed in the future.

  • 제한통성 메탄올 자호세균인 Methylovorus sp. strain SS1은 최적 성장 조건하에서는 소량의 세포외 다당류(EPS)를 생산하였지만, 질소원이 결핍된 성장 조건하에서는 성장속도는 느렸지만 다량의 EPS를 생산하였다. EPS는 배지내의 탄소대 질소 비율이 5.2일 때 가장 많이 생산되었다. EPS 생산을 위한 최적 온도는 30^{\circ}C.$이고 최적 pH는 6.5였다. EPS는 탄수화물과 단백질 및 약간의 피루브산으로 구성되어 있었고, 환원당으로는 다량의 포도당과 소량의 mannose가 존재하였다. 에탄올을 처리한 EPS(EPSae)에는 에탄올을 처리하지 않은 EPS(EPSbe)에 존재하던 피루브산이 존재하지 않았고, EPS보다 단백질의 양도 적고 점성도 낮았다. EPSbe의 점성은 NaCl에 의해 큰 영향을 받았는데, 0.5%(w/v) 농도의 NaCl 용액에서도 점성이 크게 떨어졌으며, 높은 온도에서는 점성이 비가역적으로 크게 증가하였다. Gel filtration 방법으로 조사한 EPSae의 분자량은 $2.5{\times}$10^6$ - $3.5{\times}$10^6$이었다. 냉동건조한 다당류를 전자현미경으로 관찰하였을 때, EPSbe는 섬유모양을 하였고, EPSae는 벌집모양의 망상구조를 하고 있었다.

주목열매 추출물 구조분석 (Structure Determination of the Extractives from the Taxus Cuspidata Fruits)

  • 주목열매에서 분리한 과육과 씨를 95% EtOH 용액을 사용하여 각각 1.15 kg과 0.94 kg을 유리 용기에 침지하여 추출하고, 농축된 추출용액을 용매의 극성에 따라 n-hexane, dichloromethane, ethyl acetate 및 수용성으로 순차 분획한 후, 동결건조하였다. 혼합물로부터 순수한 화합물을 분리하기 위하여 ethyl acetate 및 수용성 분획에 대하여 Sephadex-LH 20 칼럼크로마토그래피를 실시하였다. 단리된 화합물들의 구조는 flavan 화합물의 경우 $^1H$-NMR, $^{13}C$-NMR 및 EI-MS 스펙트럼을 측정하였으며, 탄수화물은 보다 정확한 구조를 결정하기 위해 COSY, HSQC와 같은 2-D NMR과 LC/MS 스펙트럼을 이용하여 구조를 결정하였다. 또한 구성당 분석을 위해 acid hydrolysis, permethylation을 실시하여 구성당의 성분과 결합위치를 확인하였다. 위의 실험을 통하여 (+)-catechin (1), (-)-epicatechin (2), (+)-gallocatechin (3), (-)-epigallocatechin(4) 및 ${\beta}$-D-fructofuranose-($2{\rightarrow}4$)-O-${\beta}$-D-glucopyranose($1{\rightarrow}4$)-O-${\alpha}$-D-glucopyranose-($1{\rightarrow}2$)-O-${\beta}$-D-fructofuranose (5)의 구조를 결정하였다.

Protein molecular structure, degradation and availability of canola, rapeseed and soybean meals in dairy cattle diets

  • Objective: The aims of this study were to reveal the magnitude of the differences in protein structures at a cellular level as well as protein utilization and availability among soybean meal (SBM), canola meal (CM), and rapeseed meal (RSM) as feedstocks in China. Methods: Experiments were designed to compare the three different types of feedstocks in terms of: i) protein chemical profiles; ii) protein fractions partitioned according to Cornell Net Carbohydrate and Protein System; iii) protein molecular structures and protein second structures; iv) special protein compounds-amino acid (AA); v) total digestible protein and energy values; vi) in situ rumen protein degradability and intestinal digestibility. The protein second structures were measured using FT/IR molecular spectroscopy technique. A summary chemical approach in National Research Council (NRC) model was applied to analyze truly digestible protein. Results: The results showed significant differences in both protein nutritional profiles and protein structure parameters in terms of ${\alpha}-helix$, ${\beta}-sheet$ spectral intensity and their ratio, and amide I, amide II spectral intensity and their ratio among SBM, CM, and RSM. SBM had higher crude protein (CP) and AA content than CM and RSM. For dry matter (DM), SBM, and CM had a higher DM content compared with RSM (p<0.05), whereas no statistical significance was found between SBM and CM (p = 0.28). Effective degradability of CP and DM did not demonstrate significant differences among the three groups (p>0.05). Intestinal digestibility of rumen undegradable protein measured by three-step in vitro method showed that there was significant difference (p = 0.05) among SBM, CM, and RSM, which SBM was the highest and RSM was the lowest with CM in between. NRC modeling results showed that digestible CP content in SBM was significantly higher than that of CM and RSM (p<0.05). Conclusion: This study suggested that SBM and CM contained similar protein value and availability for dairy cattle, while RSM had the lowest protein quality and utilization.

도토리전분(澱粉)의 이화학적(理化學的) 성질(性質)에 관(關)한 연구(硏究) (Studies on Some Physico-Chemical Properties of the Acorn Starch)

  • 3종(種)의 도토리 Quercus crispula Blume, Quercus serrata Thunberg 및 Quercus mongolica Fischer를 공시재료(供試材料)로 하여 그의 이화학적성질(理化學的性質), 호화도(糊化度), 점도(粘度) 및 X-ray회절등(回折等)을 실시(實施)한 결과(結果)는 다음과 같다. 1. 도토리는 지방(脂肪) $3.5{\sim}5.0%$, 단백질(蛋白質) $7.2{\sim}7.7%$, 당질(糖質) $71.6{\sim}76.3%$, 회분(灰分) $1.9{\sim}2.2%$였다. 2. Ether처리(處理)보다 85% methanol열처리(熱處理)가 보다더 순수(純粹)한 정제전분(精製澱粉)을 얻을 수 있었다. 3. 도토리 조제전분(粗製澱粉)은 일반(一般)으로 갈색(褐色)이나 정제전분(精製澱粉)은 회백색(灰白色)을 띄었다. 4. 호화(糊化)는 비교적 낮은 온도(溫度) 곧 $61{\sim}68^{\circ}C$이고, 호화개시(糊化開始)는 Q.C.B.>Q.S.T>Q.M.F.의 순(順)으로 빨랐고 점도(粘度)는 Q.M.F>Q.S.T.>Q.C.B.의 차례로 높았다. 5. 도토리전분(澱粉)은 그 결정구조(結晶構造)가 감자전분(澱粉)과 옥수수전분(澱粉)의 중간성질(中間性質)을 띈 것으로 추정(推定)된다. 또 고온습열처리(高溫濕熱處理)로서 면간격(面間隔)이 긴쪽 부터 먼저 결정구조(結晶構造)가 파괴되었다.

수도품종의 위고현상과 생리 및 형태해부학적 구조와의 관련성에 관한 연구 (Relationships of Physiologieal Activity and Anatomical Structure to the Wilting Phenomena in Rice Plant 1. Reappearance of the Wilting Phenomena concerning to Physiological Aspects and Environment)

  • 본 시험은 신품종인 유신을 공시하여 실소와 가리의 시비수준을 달리하고 출수후 차광처리를 하여 급성위고현상의 발생을 재현시고, 위고현상의 발생과 식물체내 무기성분의 함양 및 근의 생리적 활역과의 관련성을 명백히 함과 동시에 1976년에 전국적으로 .유신품종에 대발생한 위고현상과 1977년에 전혀 발생이 없었던 기상적인 요인을 비교 검토하여 위고현상의 발생요인을 구명코자 실시하였다. 그 결과를 요약하면 다음과 같다. 1. 유신품종은 실소증비에 의해 엽신중당 영화수 (Sink/source)가 현저히 증가하고, 상위절간에 비해 하위절간의 신장률이 뚜렷한 품종적인 특징이 있다. 2. 유신품종은 실업증비에 따라 엽신의 호흡양이 타품종에 비해 현저히 증대하는 특성이 있음을 인정 할 수 있었다. 3. 위고극다비( $N_{30}$, $K_{10)}$ 하에서 근발육과 근의 생리적 활력이 저하했으며, 실소극다비+차광처리에서 더욱 현저한 저하를 보였다. 4. 또한 실고극다비+차광처리는 도체내 엽초와 간내의 K_2 O함량을 저하시키고, 특히 간내 K_2O/N 비 및 탄수화물의 함량을 명백히 저하시켰다. 5. 유신의 위고현상은 실소극다비구에서 경미하게 발생하였고, 실고극다비+차광에서 50 % 처리구에서 격심한 발생을 유발 재현시킬 수 있었다. 6. 본 시험결과로 미루어 1976년에 대발생한 유신의 급성위고현상은 도작기간 일사양이 현저히 부족했던 기상적인 요인이 주요인이며, 농민의 실소과잉시용이 위고현상을 조장하는 부요인으로 해석되었다.

간호학 교육에서 기초의과학 교과목별 목표와 내용에 대한 연구 (Objectives and Contents of Basic Medical Sciences in Nursing Education)

  • The purpose of this study was to investigate the objectives and contents of basic medical sciences at department of nursing in college of nursing, and junior college of nursing, thus ultimately providing the basic data to standardize the curriculum of the basic medical sciences in nursing education. Seventy eight professors who were in charge of teaching basic medical sciences to at 22 colleges of nursing/ department of nursing, and 20 junior colleges of nursing responded to the questionnaires that consisted of the questions regarding objectives and contents, of basic medical sciences. Based on the description of objectives, the description related to nursing, nurse, nursing science was cathegorized as on objective applicable to nursing science, the description related to medicine or clinical medicine as medical model, the description without description related to medicine was cathegorized as knowledge acquisition. The number of schools corresponding to each category were summerized in descending order. The objectives of basic medical sciences were categorized by concepts and number of schools corresponding to the categorized concept. The findings of the study are as follows ; 1. The subjects of basic medical science identified were physiology, anatomy, biochemistry, pathology, microbiology, and pharmacology in most colleges of nursing and junior colleges. Two colleges of nursing/department of nursing (9.1%) and 19 junior colleges of nursing(95%) did not offer biochemistry, 1 college of nursing /department of nursing(5%) did not offer pathology & pharmacology. 2 junior colleges of nursing (10%) did not offer pharmacology, 1 junior college of nursing(5%) did not offer pathology. The other 1 junior college of nursing did not offer microbiology. 2. Objectives of physiology were to acquire knowledge and understanding on human function in both 6 (50%) colleges and 5 junior colleges. Objectives of anatomy were to acquire knowledge on human structure in both 4 (57%) colleges and 2 (50%) junior colleges; knowledge applicable to nursing sciences in both 3 (42.8%) colleges and 2 (50%) junior colleges. Objectives of biochemistry was to obtain knowledge and understanding on biochemistry, and understanding of basic concepts about biochemistry. Objectives of pathology were to obtain knowledge and understanding on pathology in both 4 (57.1%) colleges and 5(62.5%) junior colleges. Objectives of microbiology were to acquire knowledge and understanding on microbiology in both 5(83.8%) colleges and 6(85.7%) junior colleges. Objectives of pharmacology were to acquire knowledge on pharmacology in both 7(100%) colleges and 8(100%) junior colleges. 3. Contents of physiology in 19 (100%) schools were membrane transport, digestion, circulation, nervous system and respiration. In 16(84.2%) were kidney and muscle, that in 13(68.4%) were endocrine physiology. In 11(57.9%) were introduction and that in 9(47.4%) were structure and function of cells. Contents of anatomy in 11(100%) schools were skeletal system, muscle system, digestive system, circulatory system, concepts regarding human structure. In 10(90.9%) schools were endocrine system and nervous system, and in 5(45.5%) schools were blood, urinary system and cell. Contents of biochemistry in 6(100%) schools were history of biochemistry, body regulating factor, bioenergy, health and nutrition, nutrition of cell, energy production system. In 5(83.3%) schools were metabolism of protein and carbohydrate and enzyme, and in 3(50%) schools were metabolism of energy and fat. Contents of microbiology in 13(100%) schools were environment and influenc of bacteria, virus, G(-) rods, purulent cocci, G(+) rods. In 10 (76.9%) were immunity, diphtheria, enterobacteria, and in 9(69.2%) were spirochete, rickettsia and clamydia, and that in 6(46.2%) were sterilization and disinfection. Contents of pathology in 14(100%) schools were cell injury and adaptation, inflammation, respiratory diseases, circulatory diseases. In 10(71.4%) were neurological disorders, in 8(57.1%) were immunity and disease, and in 7 (50%) were tumor and progressive changes. Contents of pharmacology in 15(100%) were cardivascular drugs, introduction to pharmacology, hypnotics, analgesics, local anesthetics, an ticonvulsants. In 12(80%) were drugs activity on sympathetic and parasympathetic nervous system, and in 11(73%) were sulfa drugs, antibiotics, drug abuse and addiction.

기능성 막 제조를 위한 DPPC 지질막과 탄수화물 혼합막에 관한 연구 (A Study on DPPC Lipid Membrane and its Carbohydrate Mixture Membrane for Preparation of a Functional Membrane)

  • 막의 기능설계를 하기 위하여 DPPC(dipalmitoylphosphatidylcholine)인 지질로 제조한 liposome과 DPPC와 DTAB(dodecyltrimethylammonium bromide)를 혼합하여 liposome을 제조하여 그 구조변화를 조사하였다. 막 구조의 변화는 CF(carboxylfluorescein)를 이용하여 DPPC의 상전이 온도($t_c=41^{\circ}C$) 이상과 이하에서 측정하여 평가하였다. DPPC liposome에 함유된 CF의 유출에 의한 형광강도는 $45^{\circ}C$에서 증가되었지만 $20^{\circ}C$에서는 그 변화는 관측되지 않았다. 동등한 조건에서 DPPC/DTAB liposome에서 CF 유출에 의한 형광강도의 변화는 DPPC liposome에 비하여 큰 것이 관측되었다. 이 결과는 DPPC/DTAB liposome이 불규칙한 배열을 하고 있다는 것을 시사한다. 한편 $Ca^{2+}$ 존재하에서 DPPC liposome과 DTAB/DPPC liposome에서 Quin 2의 형광강도는 $45^{\circ}C$에서 현저히 증가되었지만 $20^{\circ}C$에서는 거의 변화하지 않았다. 그리고 형광강도의 변화는 DPPC/DTAB liposome보다 DPPC liposome 쪽이 더 크게 나타났다. 이것은 DPPC/DTAB liposome의 구조가 DPPC liposome보다 더 안정하다는 것을 의미한다. 또한 형광물질인 ANS를 이용하여 막 표면의 양상을 조사한 결과 DPPC liposome과 DPPC/DTAB liposome에서 ANS의 형광강도는 $45^{\circ}C$$20^{\circ}C$에서 다른 양상을 보였다. 이 양상은 상전이 온도의 이상과 이하에서 DPPC liposome과 DPPC/DTAB liposome의 막유동성을 의미하고 그 변화량은 DPPC liposome이 DPPC/DTAB liposome보다 더 컸다. 또한 DSC에 의해 측정된 상전이 온도는 DPPC liposome의 경우 $41^{\circ}C$이었으며 DPPC/DTAB liposome의 경우에는 $32^{\circ}C$이었다. 이상의 결과로 DPPC/DTAB liposome의 분자배열 상태가 불규칙한 것을 의미한다. 그러므로 DPPC/DTAB liposome은 불규칙하게 배열되어 있지만 안정된 구조를 갖고 있다고 생각된다.

어육(정어러) 발포건조제품가공에 관한 연구 1. 원료$\cdot$첨가물의 배합 및 가공조건 (Dehydration of Foamed Fish (Sardine)-Starch Paste by Microwave Heating 1. Formulation and Processing Conditions)

  • Sardine and mackerel so called dark muscled fish have been underutilized due to the disadvantages in bloody meat color, high content of fat, and postmortem instability of protein. Recent efforts were made to overcome these defects and develope new types of product such as texturized protein concentrates and dark muscle eliminated minced fish. Approach of this study is based on the rapid dehydration of foamed fish-starch paste by dielectric heating. In process comminuted sardine meat was washed more than three times by soaking and decanting in chilled water and finally centrifuged. The meat was ground in a stone mortar added Ivith adequate amounts of salt, foaming agent, and other ingredients for aid to elasticity and foam stability. The ground meat paste was extruded in finger shape and heated in a microwave oven to give foamed, expanded, and porous solid structure by dehydration. Dielectric onstant $(\varepsilon')$ and dielect.ic loss $(\varepsilon")$ values of sardine meat paste were influenced by wavelength and moisture level. Those values at 100 KHz and 15 MHz were ranged 2.25-9.86; 2.22-4,18 for E' and 0.24-19.24; 0.16-1.20 for E", respectively, at the moisture levels of $4.2-13.8\%$. For a formula for fish-starch paste preparation, addition of $20-30\%$ starch (potato starch) to the weight of fish meat, $2-4\%$ salt, and $5-10\%$ soybean protein was adequate to yield 4-5 folds of expansion in volume when heated. Addition of e99 yolk was of benefit to micronize foam size and better crispness. In order to provide better foaming and dehydration, addition of $0.2-0.5\%$sodium bicarbonate, foaming agent, was proper to result in foam size of 0.5-0.7 mm and foam density of $200-400\;/cm^2$ which gave a good crispness. Heating time was depended upon the moisture level of fish-starch paste. For a finger shaped paste (1.0cm. $D\times10cm.L$) heating for 150-200 sec. in a microwave oven (700W. 2.45GHz) was sufficient to generate foams, expand, and solidify the porous structure of fish-starch paste. When the moisture content was above $55\%$ browning and scorching was deepened due to over-expansion and over-heating whereas the crispness was hardened by insufficient expansion at lower moisture content. In quality evaluation of the product, chemical composition of $30\%$ starch and $3\%$ salt added product was moisture $8.8\%$, lipid $2.4\%$, carbohydrate $46.7\%$, protein $36.1\%$, and ash $6.0\%$. Eleven membered panel test evaluated that fish-starch paste was acceptable in color, crisp-ness, taste, except a trace of fishy odour which could be masked by the addition of spice extracts.

  • 연구배경 : Lectin은 탄수화물의 특정한 당잔기에 높은 친화성과 특이성을 가지므로 이러한 lectin-sugar 상호작용을 이용하여 사람 폐결핵결절에서 이를 구성하는 주된 성분인 건락성 괴사 상피양세포, 림프구 및 섬유층에 존재하는 탄수화물의 종류와 분포에 대해 알아보고자, 본 연구를 실시하였다. 방법 : 사람 폐결핵 절제 조직에서, 13종의 lectin을 사용하여 조직화학염색을 시행하였다. 결과: 1) 건락성 괴사부에서 BS $I-B_4$는 모든 구성성분에서, BS I는 변연부에 있는 일부의 세포성분을 제외하고 나머지 구성성분 모두에서 음성반응을 나타내었다. 그러나 나머지 lectin들은 건락성 괴사부에서 다양한 형태의 렉틴결합반응을 나타내었다. 2) 상피양세포들은 세포질에서의 반응 양상에 따라 크게 세 군으로 나눌 수 있었다. 첫째, DBA, UEA I 및 LCA는 대부분 중등도 이상의 강한 양성반응을 나타내었다. 둘째, WGA, s-WGA, PNA, SBA, VVL, ECL, PHA-L 및 PHA-E는 세포에 따라 다양한 형태의 반응양상을 나타내었다. 셋째, BS I 및 BS $I-B_4$는 음성반응을 나타내었다. 상피양세포의 세포막에 대한 결합반응을 살펴보았을 때, 첫째, ECL, PHA-L 및 PHA-E는 대부분 강한 양성반응을 보여주었다. 둘째, WGA, s-WGA 및 PNA는 세포에 따라 양성 및 음성반응이 혼재하는 양상을 나타내었다. 셋째, BS I, BS $I-B_4$, UEA I, DBA 및 VVL은 음성반응을 나타내었다. 3) 상피양세포사이물질들은 건락성 괴사부의 세포사이물질에 대한 lectin 결합 양상과 유사한 반응 양상을 나타내었다. 4) WGA, ECL, PHA-L, PHA-E 및 LCA들은 림프구에 강한 양성반응을 나타내었다. 5) SBA는 건락성 괴사부를 둘러싸는 섬유층의 외측에 위치하는 혈관 내피층에서 대체로 양성반응을 보였으나, 섬유층내의 혈관 내피층에서는 대체로 음성반응을 나타내었다. 6) PNA는 섬유층의 외측지역에서 양성반응을 나타내었지만 그와는 대조적으로 내측지역의 세포사이물질에서 음성반응을 나타내었다. 결론 : 이상의 결과와 같이, 렉틴결합특성을 이용한 조직화학적 연구는 사람 폐결핵 결절의 여러 성분들에 존재하는 복합당질의 종류, 특성 및 분포, 그리고 연결핵결절 형성과정 동안에 일어나는 복합당질 조성의 변화에 관한 유용한 정보를 제공해 주었으며, 이러한 결과들은 결핵결절의 병리학적 연구에 도움이 될 것으로 사료된다.
