수종 살충제가 줄지렁이 치사에 미치는 영향평가 (Assessment of the Effects of Some Insecticides on Mortality of Earthworm (Eisenia fetida))

  • The toxic effects of 12 commercially available insecticides on the earthworm, Eisenia fetida (Savigny), were evaluated using artificial soil, surface sprayed soil, immersion, and contact filter tests. The risk to earthworm was assessed by the TER (toxic exposure ratio) value, which was calculated by the formula, TER=NOEC/PEC (predicted environmental concentration). TER was 3 for methomyl SL, 20 for carbaryl WP, 20 for phosphamidon SL, 30 for imidacloprid WP, and 60 for dichlorvos EC the in artificial soil test. At recommended, the earthworm mortality to methomyl SL reached 50% in the surface sprayed soil test, 72% in the immersion test, 30% in the contact filter paper test, whereas that to imidacloprid WP reached 56, 32, and 100 respectively. As a result of the four methods, methomyl SL and imidacloprid WP would be toxic insecticides to earthworm.

중공사 나노여과막을 이용한 방향족 농약의 배제 특성 (Rejection Properties of Aromatic Pesticides by a Hollow Fiber NF Membrane)

  • The rejection properties of 6 aromatic pesticides were evaluated by a continuous flow system equipped with a hollow fiber NF membrane. Different from the separation experiment of batch cell, the rejection and the removal could be calculated exactly because the concentration of feed, permeate and retentate was separately obtained. The lowest and the highest rejection were found in carbaryl(54.8%) and methoxychlor(99.2%), respectively, and the removals were always shown higher than rejections. This may be caused by some reasons such as the solute adsorption on the membrane, the variation of feed concentration. Although molecular weight, molecular width regarded as solute characteristics and log P(n-octanol/water partition coefficient) as hydrophobicity could be applied to explain the rejection property, these factors should be considered together for better analysis. According to the higher relationship between log B(solute permeability) and molecular weight, it was revealed that the solute separation with this membrane was influenced more by molecular weight.

율무의 조명나방 방제체계 (Control System of Asian corn borer, Ostrinia furnacalis (Guenee) in Coix lacryma-jobi L. var. ma-yuen Stapf)

  • 본 연구는 율무 주산단지인 경기도 북부의 연천 지역에서 율무에 크게 피해를 주는 조명나방에 대한 발생 생태 조사 및 방제적기에 관한 연구를 수행한 결과는 다음과 같다. 율무 조명나방은 연천지역에서 성충 발생은 년간 $2{\sim}3$ 회였고 1, 2, 3 화기 발생최성기는 각각 5월 하순${\sim}$6월 중순, 7월 하순${\sim}$8월 상순, 8월 하순이었으며, 율무 조명나방에 의한 줄기 피해는 $7{\sim}8$월부터 시작하여 10월에 가장 심하였다. 조병나방에 대한 방제약제는 할로스린유제가 93.9%로 방제효과가 가장 높았으며, 효과적 방제적기는 할로스린유제를 7 월하순 fb 8 월 상순 , 8월상순 fb 8월 중순 2회 처리시 방제가가 92.4, 93.5% 로 높았으며 10a당 수량은 무처리구 216kg에 비하여 각각 81%, 82% 증수하였다.

콩 재배포장의 노린재류 발생 및 콩 피해실태 (Field Occurrence of Stink Bug and its Damage in Soybean)

  • 콩에 발생하는 노린재류의 피해실태와 약제방제효과를 구명하기 위하여 1997부터 1998년까지 경북농업기술원 전작포장에서 실시한 시험의 결과를 요약하면 다음과 같다. 1. 경북 남부지역 콩포장에 발생하는 노린재류는 톱다리개미허리노린재, 풀색노린재, 썩덩나무노린재, 가로줄노린재, 알락수염노린재 등 9증이었다. 2. 콩 품종별 노린재류의 피해립율은 두유콩, 신팔달콩2호, 삼남콩, 만리콩, 단경콩 등 단경품종군에서 15.2-27.0%, 한남콩, 장수콩, 푸른콩, 무한콩 등 장경품종 군이 36.1-50.0%였고, 피해협율도 같은 경향이었으며, 또한 100개체당 생충밀도는 단경품종군에서 5.0-12.5마리, 장경품종군에서는 10.3-30.0마리로 단경품종군이 노린재류의 선호도가 낮았지만 노린재류에 대한 저항성을 보인 품종은 없었다. 3. 톱다리개미허리노린재, 풀색노린재는 대체로 기온이 높을때 발생밀도가 높아 협에 피해가 많았고, 썩덩나무노린재, 가로줄노린재, 알락수염노린재는 기온이 서늘할 때 발생이 많았으며 대체로 종실에 피해가 많았다. 4. 8월 24일부터 9월 17일 까지 10일 간격으로 3회 약제살포후의 노린재류 생충밀도는 100개체당 무처리 40.3마리에 비해 아조포유제 3.7, 메프수화제 5.3, 나크수 화제 3.0마리로 약제방제효과가 높았다. 5. 약제방제후 피해립율은 무처리 33.8%에 비해 약제처리구는 4.4-10.3%였고, 아조포유제 83.4%, 메프수화제 69.5, 나크수화제 87.0%의 방제가를 보였다. 수량은 ha당 무처리구 1.92MT에 비해 약제처리구에서 38-45%증수되었다.

Genetic and Phenotypic Diversity of Carbofuran-Degrading Bacteria Isolated from Agricultural Soils

  • Thirty-seven carbofuran-degrading bacteria were isolated from agricultural soils, and their genetic and phenotypic characteristics were investigated. The isolates were able to utilize carbofuran as a sole source of carbon and energy. Analysis of the 16S rRNA gene sequence indicated that the isolates were related to members of the genera Rhodococcus, Sphingomonas, and Sphingobium, including new types of carbofuran-degrading bacteria, Bosea and Microbacterium. Among the 37 isolates, 15 different chromosomal DNA patterns were obtained by polymerase chain reaction (PCR) amplification of repetitive extragenic palindromic (REP) sequences. Five of the 15 representative isolates were able to degrade carbofuran phenol, fenoxycarb, and carbaryl, in addition to carbofuran. Ten of the 15 representative isolates had 1 to 8 plasmids. Among the 10 plasmid-containing isolates, plasmid-cured strains were obtained from 5 strains. The cured strains could not degrade carbofuran and other pesticides anymore, suggesting that the carbofuran degradative genes were on the plasmid DNAs in these strains. When analyzed with PCR amplification and dot-blot hybridization using the primers targeting for the previously reported carbofuran hydrolase gene (mcd), all of the isolates did not show any positive signals, suggesting that their carbofuran hydrolase genes had no significant sequence homology with the mcd gene.

Toxicological Aspects of Carboxylesterases -A Sensitive Biomarker of Organophosphate Toxicity-

  • Egasyn is accessory protein of ${\beta}$-glucuronidase(${\beta}$-G) in the liver microsomes. Liver microsomal ${\beta}$-G is stabilized within the luminal site of the microsomal vesicles by complexation with egasyn which is one of carboxylesterase isozymes. We investigated the effects of organophosphorus compounds(OPs) such as insecticides on the dissociation of egasyn-${\beta}$-glucuronidase(EG) complex. The EG complex was easily dissociated by administration of OPs, i.e., Fenitrothion, EPN, Phenthionate, and bis-p-nitrophenyl phosphate(BNPP), and resulting ${\beta}$-G dissociated was released into blood, leading to the rapid and transient increase of plasma ${\beta}$-G level with a concomitant decrease of liver microsomal ${\beta}$-G level. In a case of phenthionate treatment, less increase in plasma ${\beta}$-G level was observed, as compared with those of other OPs. This may be explained by a fact that phenthionate was easily hydrolyzed by carboxylesterase. Similarly, carbamate insecticides such as Carbaryl caused rapid increase of plasma ${\beta}$-G level. In contrast, no significant increase of plasma ${\beta}$-G level was observed when pyrethroid insecticides were administered to rats. This is due to a fact that pyrethroids such as Phenthrin and Allethrin were easily hydrolyzed by A-esterase as well as carboxylesterase. On the other hand, addition of OPs to the incubation mixture containing liver microsomes caused the release of ${\beta}$-G from microsomes to the medium. From these in vivo and in vitro data, it is concluded that increase of the plasma ${\beta}$-G level after OPs administration is much more sensitive biomarker than cholinesterase inhibition to acute intoxication of OPs and carbamates.

  • 가. 해충과 거미에 대한 반치사약량및 상대독성 조사 (1) BPMC와 MIPC는 벼멸구와 황산적거미에 대한 상대독성치가 각각 $18.4\~29.6,\;8.0\~10.3$으로 비교적 높은 선택독성을 보였다. (2) 이화명충과 황산적거미에서는 thiocyclam, chlorpyrifos-methyl이 각각 727, 14의 상대독성치를 보였으며 벼밤나방에서는 thiocyclam이 189.3으로 선택독성이 높았다. (3) 끝동매미충과 황산적거미에 대해서도 BPMC의 상대독성치가 8.2로 양호하였다. 나. 약제형성 및 처리회구가 해충 및 천적밀도에 미치는 영향(폿트시험) (1) 이화명충 방제약제중 cartap이 이화명충 방제효과도 높고, 거미밀도도 높았으며, 제형간에서는 유제가 분제보다 거미에 미치는 영향이 적었다. (2) MIPC 처리구에서 끝동매미충 방제효과가 높고 거미 밀도에 미치는 영향도 적었다. (3) pyridaphenthion 1회 처리구가 2회 처리구에 비하여 거미밀도에 미치는 영향이 적었으나 혹명나방 방제효과는 비슷하였다.

Carassius auratus(goldfish)를 이용한 Carbofuran의 단기간 생물농축계수의 측정 (Determination of Short-term Bioconcentration Factor on Carbofuran in Carassius auratus (goldfish))

  • The Bioconcentration factor(BCF) is used as an important criterion in the risk assessment of environmental contaminants. Also it can be used as indicator of biomagnification of environmentally hazardous chemicals through food-chain as well as a tool for ranking the bioconcentration potential of the chemicals in the environment. This paper reports the measured BCF value on carbofuran in Carassius auratus(goldfish), under steady state, and examined corelation between the BCF value and the depuration rate constant. Carassius auratus(goldfish) was chosen as test organism and test periods were 1-day, 3-day and 5-day. Experimental concentrations were 0.05, 0.10 and 0.50 ppm. Carbofuran in fish tissue and in test water was extracted with n-hexane and acetonitril. GC-ECD was used to detect and quantitate carbofuran. The depuration rate of carbofuran from the whole body of goldfish is determined over the 24-h period after treatment. The obtained results were as follows: 1. It was possible to determine short term BCFs of carbofuran through relatively simple procedure in environmental concentrations. 2. $BCF_1$ of carbofuran in concentration of 0.05, 0.10 and 0.50 ppm were 1.66, 1.64 0.61, $BCF_3$ were 2.08, 2.14, 0.66 and $BCF_5$ were 2.21, 2.57, 0.86, respectively. 3. Carbofuran concentration in fish extract was increased as increasing test concentration and prolonging test period, but $BCF_s$ in concentration of 0.50 ppm was greately decreased. 4. Determined deputation rate constants of carbofuran in concentration of 0.05, 0.10, 0.50 ppm were 0.076, 0.082 and 0.089, respectively. 5. It is considered that great decrease of $BCF_s$ in concentration of 0.50 ppm is due to high water solubility and stability of carbofuran in testwater. 6. It is suggested that low BCF of carbofuran is due to its relatively high water solubility and depuration rate, compared to BPMC, carbaryl and chlorothalonil.

고도산화와 정밀여과막 혼성공정을 이용한 먹는 물에 존재하는 발암원인으로 의심되는 유기화학성분의 분해 및 제거분석에 관한 연구 (A Study on Dissolve and Remove Analysis of Pollutants in Drinking Water by Suspected Cancer Causing Organic Chemicals using AOPs (Advanced Oxidation Processes) & M/F Hybird Process)

  • The AOPs research defined by creating a sufficient amount of OH radicals from the dissolution of organic materials through photoxidation and research for a complete elimination of residual organic materials by membrane are actively ongoing. This research focuses on the hybrid processing of AOPs and M/F membrane to dissolve and eliminate organic chemicals in drinking water which are suspected of carcinogens. For this purpose, underground water was used as a source of drinking water for the hybrid processing of AOPs oxidation and M/F membrane, and a pilot plant test device was installed indoor. Carcinogenic chemicals of VOCs and pesticide were artificially mixed with the drinking water, which was then diluted close to natural water in order to examine treatment efficiency and draw optimal operation conditions. The samples used for this experiment include four chemicals phenol, chloroform, in VOCs and parathion, carbaryl in pesticide. As a result of the experiments conducted with simple, and compound solutions, the conditions to sufficiently dissolve and eliminate carcinogenic chemicals from the hybrid processing of where carcinogens were artificially added are : (hydrogen peroxide) prescribed solution 100 mg/L under pH 5.5~6.0, and the temperature $12{\sim}16^{\circ}C$, at the normal temperature and pressure. $d-O_3$ volume of 5.0 ppm and above and 30-40 minutes of reaction time are most appropriate and using MF/UF for membrane was ideal.

영산강 및 섬진강 수계 중 농약 분포 조사 (Monitoring of Pesticides in the Yeongsan and Seomjin River Basin)

  • 영산강과 섬진강의 지천을 중심으로 총 22개 지점에서 4회에 걸쳐 농약(aldicarb, carbaryl, carbofuran, chlorpyrifos, 2,4-D, MCPA, methomyl, metolachlor, molinate)의 실태조사를 수행하였다. 하천수 시료에서 농약을 추출하기 위해 Oasis HLB 카트리지를 사용하였고 LC/ MS/MS를 이용하여 분석하였다. 시험법은 matrix-matched 내부표준검량곡선과 방법검출한계, 정량한계, 정확도, 정밀도, 회수율 등으로 정도관리를 수행하였다. 각 화합물의 방법검출한계는 모든 화합물이 국립환경과학원에서 제안하는 GV/10을 만족하였다. 모든 화합물의 상관계수(r2)는 0.9965- 0.9999로써 직선성이 매우 높았고 정확도와 정밀도는 각각 89.4-113.6%와 3.1-14.0%로써 매우 양호함을 알 수 있었으며 평균 회수율은 90.8-106.2%로 매우 우수하였다. Aldicarb를 제외한 모든 성분이 검출되었고, 특히 carbofuran과 metolachlor의 검출농도와 빈도가 높았는데 이들의 광범위한 적용 범위에 따른 많은 사용량에 기인한 것으로 판단되었다. 2,4-D와 MCPA는 1(5월), 2차(7월) 시료에서만 검출되었는데 이는 두 농약의 사용 적기와 일치하였다. Methomyl과 molinate는 사용 금지된 농약이었으나 일부 지역에서 검출되었고 이는 두 농약이 불법적으로 사용되었거나 과거에 사용되어 수계로 유입, 저질에 흡착된 일부분이 지금까지 용출되어 검출되었을 것으로 판단되었다. 영산강 및 섬진강 수계의 많은 하천수 시료에서 농약이 검출되었고 그 원인이 영농활동 중 농약의 사용이라고 단정할 수 없으나, 영산강과 섬진강 수계 인근의 주된 산업활동이 농업활동이고 농약의 주된 사용이 영농활동에 이루어지며 검출된 농약이 농작물에 대한 약제의 사용 시기와 부합하는 것을 미루어 보아 영산강과 섬진강 유역 농경지에서 사용된 농약이 수계로 유입되어 비점오염원으로 작용하는 것으로 판단되었다. 따라서 수생태계 보존을 위하여 농약에 대한 수질기준 설정이 절실히 요구되었고, 이를 위해 보다 많은 실태조사 결과의 축적과 꾸준한 연구 수행이 필요한 것으로 판단되었다.
