Protein Structural Characterization by Hydrogen/Deuterium Exchange Mass Spectrometry with Top-down Electron Capture Dissociation

  • This study tested the feasibility of observing H/D exchange of intact protein by top-down electron capture dissociation (ECD) mass spectrometry for the investigation of protein structure. Ubiquitin is selected as a model system. Local structural information was obtained from the deuteration levels of c and $z^{\cdot}$ ions generated from ECD. Our results showed that ${\alpha}$-helix region has the lowest deuteration level and the C-terminal fraction containing a highly mobile tail has the highest deuteration level, which correlates well with previous X-Ray and HDX/NMR analyses. We studied site-specific H/D exchange kinetics by monitoring H/D exchange rate of several structural motives of ubiquitin. Two hydrogen bonded ${\beta}$-strands showed similar HDX rates. However, the outer ${\beta}$-strand always has higher deuteration level than the inner ${\beta}$-strand. The HDX rate of the turn structure (residues 8-11) is lower than that of ${\beta}$-strands (residues 1-7 and residues 12-17) it connects. Although isotopic distribution gets broader after H/D exchange which results in a limited number of backbone cleavage sites detected, our results demonstrate that this method can provide valuable detailed structural information of proteins. This approach should also be suitable for the structural investigation of other unknown proteins, protein conformational changes, as well as protein-protein interactions and dynamics.

  • Genosensor technology utilizes a patterned array of DNA molecules immobilized on solid supports for biomedical analysis. The detection capability of the sensor depended mainly on the way the capture probes are attached to the support as well as the sequence. We compared two different. coupling methods currently used to covalently graft DNA molecules onto a glass surface.

  • Fine particle capture is facing a challenge since traditional filtration which relies on the combination of impaction, interception, diffusion has a limited efficiency for fine particle capture particularly in size from 0.1 to $0.5{\mu}m$. This paper reviewed the collection efficiency of above mechanisms, as well as magnetic mechanisms for ferromagnetic particles, and mainly studied the influencing factors of magnetic filtration. Filtration velocity, magnetic field intensity and fiber size were found to be the most important parameters for magnetic filtration.

  • The Avengers film produced by Marvel Comics shows visual effects that were impossible to produce in the past. Companies that produce film special effects were initially equipped with large personnel and equipment, but technology is gradually evolving to be feasible for smaller companies that do not have high-priced equipment and a large workforce. The development of hardware and software is becoming increasingly available to the general public as well as to experts. Equipment and software which were difficult for individuals to purchase before quickly popularized high-performance computers as the game industry developed. The development of the cloud has been the driving force behind software costs. As augmented reality (AR) performance of mobile devices improves, advanced technologies such as motion tracking and face recognition technology are no longer implemented by expensive equipment. Under these circumstances, after implementing mobile-based facial capture technology in animation projects, we have identified the pros and the cons and suggest better solutions to improve the problem.

  • The design of a 7 degree of freedom motion capture device(MCD) has been presented. The newly designed MCD overcomes the shortcomings of the existing CADEN-7 exoskeleton robot by implementing various ergonomic design. To improve ease of operation, light-weight high-strength materials such as carbon pipes and engineering plastics were used to reduce weight of the MCD and arm-length adjustment mechanism was also added. The MCD showed consistent measurement results in designed experiments involving change of arm posture from nominal configuration to either elbow-side or arm-front configurations. Furthermore, captured motion in more natural tennis swing appeared to agree well with visual observations made.

Capture Myopathy in a Red-Necked Wallaby (Macropus rufogriseus) (붉은목왈라비에서 발생한 포획근병증)

  • This case report describes capture myopathy in an 8-month-old female red-necked wallaby (Macropus rufogriseus) that died after a 14-day history of depression, lameness and astasia following a bathing protocol for dermatitis. In a blood test performed the day after the wallaby fell down, serum chemistry showed a normal rangeof LDH but elevated CK, ALT and AST. Upon histopathologic examination after the animal's death, we found degeneration and necrosis of myofibers in skeletal muscle and myocardium. According to history that the wallaby was pressed by other individuals, suspected vitamin E insufficiency in the diet, physical stress during bathing as well as results of a blood test and microscopic examination, we diagnosed this case as capture myopathy. Although capture myopathy is a common problem in marsupials, this is the first reported case in Korea and understanding this case will help to manage future cases of capture myopathy in captive settings in Korea.

Development of Device Driver for Image Capture and Storage by Using VGA Camera Module Based on Windows CE (WINDOWS CE 기반 VGA 카메라 모듈의 영상 획득과 저장을 위한 디바이스 드라이버 개발)

  • In this paper device driver for camera capture in hand held mobile system is implemented based on microsoft windows CE operating system. We also study the storage device driver based on the FAT fie system by using NAND flash memory as a storage device. We use the MBA2440 PDA board for implementing the hardware for image capture by using CMOS camera module producted by PixelPlus company. This camera module has VGA $640{\times}480$ pixel resolution. We also make application program which can be cooperated with the device driver for testing its performance, for example image capture speed and quality of captured image. We check that the application can be cooperated well not only with the device driver for camera capture but also with the device driver for FAT file system designed especially for the NAND flash memory.

  • Widespread deployment of infrastructure WLANs has made Wi-Fi an integral part of today's Internet access technology. Despite its crucial role in affecting end-to-end performance, past research has focused on MAC protocol enhancement, analysis, and simulation-based performance evaluation without sufficiently considering a misbehavior stemming from capture effect. It is well known that the capture effect occurs frequently in wireless environment and incurs throughput unfairness between nodes. In this paper, we propose a novel Fair MAC algorithm which achieves fairness even under physically unfair environment. While satisfying the fairness, the proposed algorithm maximizes the system throughput. Extensive simulation results show that the proposed Fair MAC algorithm substantially improves fairness without throughput reduction.

Exon Capture - Principle and Applications to Phylogenomics and Population Genomics of Fishes (엑손 포획 - 원리와 어류의 계통유전체학 및 집단유전체학으로의 응용)

  • Phylogenetic reconstruction based on one locus or a few loci can be misleading due to gene-tree/species-tree discordance. Species delimitation and intraspecific studies also often suffered from low resolution because of insufficient statistic power when few loci were used. Exon capture method is one of the most efficient way to collect genome-scale data, which can significantly augment studies that aimed to investigate patterns and histories of organisms at both intraspecific and high level. Here, I showed the advancement of shifting from single-gene method to genomic approach and the benefit of applying exon capture method comparing to alternative genomic techniques. Then, I explained the principle of exon capture method as well as providing detailed recommendations for applying this method. Finally, I demonstrated exon capture method using two applications and discussed future perspectives of this technology.
