7종의 수용성 비타민 즉, 티아민, 리보플라빈, 피리독신, 시아노코발라민, 아스코르브산, 엽산, 니코틴산을 free solution capillary electrophoresis로 정량분석하였다. 최적 분리조건 설정을 위해 완충용액의 pH, 모세관의 온도 그리고 적용한 전압에 대해 실험한 결과 완충용액의 pH는 6에서는 겹치는 peak가 있었으며 pH 8에서는 최적의 분리를 보여주었고 pH가 더 올라갈수록 분리도는 저하되었다. 모세관의 온도와 적용한 전압은 둘 다 높아질수록 분리시간은 단축되었으며 온도 $25^{\circ}C$, 전압 20 kV에서 최적분리능을 보여주었고 그 이상의 각 조건에서는 두 경우 모두 분리능이 저하되었다. 앞의 최적조건에서 수용성비타민은 8분내에 분리되었으며 각 peak의 migration time의 편차는 $0.01{\sim}0.1$분(CV : 0.9%)이었다.
We wish to develop micro-total analysis system (TAS) on a chip, and to make a trial approach to solve the important problem that is to detect ions separated by the electric field. We propose an idea, which is as for rotational motions of dipolar ions, which are affected by the ion atmosphere in outer regions. This is a new kind of the ion-sensitive field effect transistor (ISFET). We wish to develop the ISFET chips, and give more effective, fast and sensitive, capillary electrophoresis is designed in near future.
An analytical method using capillary electrophoresis (CE) was developed for quantitation of water soluble vitamin contents in various vitamin products. The method includes the optimization of separation of 11 water soluble vitamins changing the micellar concentration and pH of running buffer, applied voltage and sample preparation. Best resolution was obtained with 25 mM phosphate buffer (pH=8.0) containing 50 mM sodium dodecyl sulphate (SDS) as micellar phase. At optimum condition, water soluble vitamins were determined in orange juice and vitamin products such as vitamin C pulvis, vitamin injection, coated multivitamin tablet. The quantitative analysis of water soluble vitamins with CE was suitable for quality control of pharmaceutical products with sound reproducibility.
The quantitation of inorganic anions in various drinking waters were investigated using capillary electrophoresis(CE) and the results were compared with ion chromatography(IC). With CE, in contrast to IC, was demonstrated rapid analysis, good efficiency, a low detection limit and the low comsumption of a solvent and samples. CE analysis was used 5 mM sodium chromate(pH 8.0) containing 20 mM tetraalkylammoniumbromide at -25kV applied voltage with indirect UV detection at 254 nm. This results in exceedingly short analysis time within 3 min. with efficiencies approaching 200,000 theoretical plates. The coefficients of variants of migration time are less than 0.8% and those of peak area are less than 2.3%. Detection limits for quantitative determination were 300 ppb-50 ppm level. These optimum conditions are applicable to various samples without pretreatment.
The stability constants for the inclusion complexes between carboxymethyl-$\beta$-cyclodextrin (CM-$\beta$-CD) and five $\beta$-blockers, such as atenolol (ATE), bisoprolol (BIS), metoprolol (MET), pindolol (PIN) and propranolol (PRO) were determined by capillary electrophoresis. The magnitude of stability was decreased as following order; PRO>MET>BIS>ATE>PIN. Among them PRO showed the highest affinity towards CM-$\beta$-CD with stability constants of 383 and 371 $M^{-l}$ for (R)- and (S)-enantiomer, respectively. PIN enantiomers showed the lowest stability towards CM-$\beta$-CD, while the selectivity between (R)- and (S)-enantiomer was higher than any other tested $\beta$-blocker.r.
This paper reports low-cost PDMS/glass based DNA microbiochip for the restriction enzyme reaction and its products detection using the capillary electrophoresis. The microbiochip ($25mm{\times}75mm$) has the heater integrated reactor ($5{\mu}{\ell}$) for DNA restriction enzyme reaction at $37^{\circ}C$ and the microchannel ($80\;{\mu}m{\times}100\;{\mu}m{\times}58mm$) for the capillary electrophoresis detection. It is experimentally confirmed that the digestion of the plasmid ($pGEM^{(R)}-4Z$) by the enzyme (Hind III and Sca I) is performed for less than 10 min and its electrophoresis detection is able to sequentially on the fabricated microbiochip.
In the non-cyanide-based electroless Au plating solution using thiomalic acid as a complexing agent and aminoethanethiol as a reducing agent, analysis of each component constituting the plating solution is essential for the analysis of the reaction mechanism. And component analysis in the plating solution is important for monitoring component changes in the plating process and optimizing the management method. Capillary Electrophoresis (CE) method is rapid, sensitive and quantitative and could be readily applied to analysis of Aun+ ion, complexing agent and reducing agent in electroless Au plating solution. In this study, the capillary electrophoresis method was used to analyze each component in the electroless Au plating solution in order to elucidate the complex bonding form and the plating mechanism of the non-cyanide-based electroless Au plating bath. The purpose of this study was to establish data for optimizing the monitoring and management method of plating solution components to improve the uniformity of precipitation and stability. As a result, it was confirmed that the analysis of thiomalic acid as a complexing agent and Aun+ ions and the analysis of aminoethanethiol as a reducing agent were possible by capillary electrophoresis. In the newly developed non-cyanide-based electroless Au plating solution, it was confirmed that Aun+ ions exist in the form of Au+ having a charge of +1, and that thiomalic acid and Au+ are combined in a molar ratio of 2 : 1. In addition, it was confirmed that aminoethanethiol can form a complex by combining with Au+ ions depending on conditions as well as acting as a reducing agent.
Determination of chloride ion in concentrated NaOH solution by capillary electrophoresis has been studied. The analysis was performed by indirect UV absorption detection using chromate buffer at 254nm. The matrix effect of the sample has been observed so that the sensitivity in strong NaOH solutaion has decreased up to 10% of that in distilled water. The pH effect of the sample on the sensitivity of CE peaks has been investigated. The method for increasing the sensitivity have been investigated and the optimum pH and concentration of the buffer were 7.5 and 10mM, respectively. A cationic surfactant cetyltrimethylammonium bromide(CTAB), was added to a buffer solution in order to reverse the electroosmotic flow(EOF) in the capillary. This results in a short analysis time and better peak shapes. Using this optimum condition, the determination of chloride ion in real environmental sample has been performed, which is captured in strong NaOH absorbent prepared for absorbing gas from chimney. The standard addition method has been applied for the quantitative analysis, and it was obtained the good reproducibility.
We have studied the migration behavior of single-stranded DNA using capillary gel electrophoresis under various conditions. It was found that optimum electric fields should be less than 150 V/cm for the good tradeoff between the separation time and the resolution. It seems that the gel matrix with the combination of different polymer average molecular weights is important to extend the maximum readable DNA bases. The total gel concentration less than 3.1% in the mixed gel system showed good separation efficiency up to 600 bases. The best result was obtained with the poy(ethylene)oxide (PEO) gel concentration of 1.2% of Mr 8,000,000 and 1.8% of Mr 600,000. We observed that the capillary length between 50 cm to 100 cm (effective length) should be employed for the optimization between the total DNA migration time and the maximum readable length. A trizma base-boric acid-ethlyenediaminetetraacetic acid (EDTA) (TBE) buffer was commonly used for DNA sequencing, but we found that 3-[tris(hydroxymethyl)methyl amino]-1-propane sulfonic acid (TAPS) buffer worked as well for the single-stranded DNA separation. Especially, TAPS buffer showed a good resolution for very short DNA bases (1 to 30 bases).
Mixtures of cyano complexes of palladium(II) and platinum(II) were separated by capillary electrophoresis using a fused silica capillary as a separation column and 30 mM phosphate buffer (pH 7) containing 15 wt. % acetonitrile as a running buffer. By virtue of the high ionic mobilities of the negatively charged cyano complexes of Pd(II) and Pt(II), they were separated using a cathodic injection and anodic detection scheme. The metal complexes eluted through the capillary were detected by direct UV absorption at 214 nm. A linear relationship between peak area and concentration was obtained for both ions and the detection limit was lower than $10^{-14}$ mole. The proposed method was applied to real sample, e.g., anode slime obtained from an electrolytic copper refinary, as a method for the simultaneous determination of palladium and platinum.
