고려인삼(Panax ginseng C.A.Meyer) 조직배양에서 2.4-dichloro phenoxyacetic acid와 kinetin첨가가 Isoperoxidase 변이에 미치는 영향 (Effect of 2,4-Dichlorophenoxyacetic Acid and Kinetin on Peroxidase Isoenzymes in Ginseng(Panax ginseng C. A. Meyer) Callus Cultures)

  • This study was undertaken to investigate the influence of kinetin and 2, 4-dichlorophenoxyacetic acid on the rate of growth, the contents of RNA, DNA, and protein. And also the effect of plant growth regulator on isoperoxidases in callus derived from root (root-callus) and petiole (petiolecallus) was investigated. The rate of growth in petiole-callus was higher than the rootcallus at 0.1 mg/l kinetin and 1mgfl 2,4-D. At 1mgll kinetic, the rate of growth increased, but at high concentration the rate of growth decreased fast. The contents of RNA, DNA and protein also increased, but it did not coincide with the increase of the growth rate of callus. The isoperoxidases of callus grown at various amounts of 2,4-D and kinetic occurred in an almost fashion, but those of root-callus appeared different from those of petiole-callus.

수수로부터 allelopathy성 물질의 기내선별 (In vitro bioassay for allelopathic substances of Sorghum ( Sorghumbicolor L.))

  • These experiments were conducted to determine the effects of Sorghum allelopathic substances on the callus growh of several weeds and crops. 1. When substances extracted from allelopathic Sorghum(Sorghum bicolor L.) were treated on medium, growth of callus of several weeds and crops were in-hibited. The degree of inhibition differed depending on the genotypes, ranging from 50 to 90% com-pared with that of control. 2. The extracts of above 5% Sorghum inhibited the callus growth of Che-nopodium albun L., Commelina communis L., and .Ammaranthus retroflexus L.and showed in-hibition rate of above 70% in callus growth. These results indicate that we could investigate theallelopaihy effect by using in vitro system. 3. The suitable explant for callus induction fromallelopathic plants was immature embryos, the callus induction rate differed depending on the geno-type, growth regulators and concentrations. In general, the addition of 2, 4-D and NAA onto medium increased the rate and amount of callus.

형개의 조직배양에 의한 정유생산 (Production of Essential Oils by Tissue Culture of Schizonepeta tenuifolia)

  • The callus was induced from the seedlings of Schizonepeta tenuifolia Brig. (Labiatae) and the effects of culturing conditions on growth rate and essential oil formation of the callus were experimented. It was found in the experiments, that the proper culturing temperature is $23^{\circ}C$ and the addition of biosynthetic precursors(leucine, mevalonic acid lactone) inhibits the growth of the callus. The growth rate of the callus and the amount of essential oils of the callus in the medium containing NAA were higher than the medium containing 2,4-D. The essential oils from the callus and the leaves of the cultivated Schizonepeta tenuifolia showed different GC pattern, but pulegone was found in both oils.

  • Callus was derived from the seedlings of Agastache rugosa(Labiatae). The growth rate of callus and the production of essential oil were studied with the variation of culturing conditions. 2, 4-D 2ppm in the medium was more effective for the production of essential oil than NAA 2ppm. The growth rate of callus and the production of essential oil were inhibited by the illumination of the light. The essential oils from Agastache rugosa and the callus cultivated on the medium containing 2, 4-D 2 ppm and kinetin 0.2 ppm were analysed by TLC, gas chromatography and mass spectrometry. These two oils showed different compositions. The main component of the plant oil, methyl chavicol was not contained in the callus oil.

  • 고추과실의 placenta 조직(組織)을 합성배양(合成培養)기에서 배양(培養)코저 callus발생(發生)에 적합(適合)한 배지(培地)를 선택하고 callus의 증식 속도를 검토한 결과 다음의 결과(結果)를 얻었다. callus발생(發生)에는 Linsmaier and Skoog RM 1964 기본 배지에 2, 4-D $(10^{-5}M)$과 kinetin $(10^{-6}M)$을 함유한 고형배지(固形培地)가 가장 효과적(效果的)이었다. 이들 callus는 배양후 2주일부터 3주일 사이에 가장 증식이 빨랐고 그 이후는 완만한 속도를 보였다.

  • Callus was derived from the shoots of Cnidium officinale. The growth rate of callus and the production of essential oils were studied under different culture conditions. The essential oils in the rhizome of Cnidium officinale and the cultivated callus were analyzed and compared by gas chromatography and mass spectrometry. It appeared that NAA induced higher growth rate and production of essential oils than 2,4-D. The compositions of essential oils were influenced by the illumination. Butyl phthalide, cnidilide, senkyunolide, butylidene phthalide, ligustilide, grandisol, tricosane, 3-methylphenol and 2-pentylthiophene were identified in the cultivated callus.

Multiple Shoots Regeneration and In vitro Bulblet Formation from Garlic Callus

  • The leaf segments of garlic (Allium sativum L.) were cultured in vitro and determined optimal concentration of plant growth regulators and sugars for callus induction, multiple shoots regeneration and in vitro bulblet formation. Highest yield of callus was observed in the leaf segment culture on Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-D, 30 g/L sucrose and 8 g/L agar. Regeneration rate of multiple shoots from callus was high in the MS medium supplemented with kinetin 3.0 + NAA 3.0 mg/L or SA 1.0 + NAA 3.0 mg/L, containing 30 g/L sucrose. High rate of bulblet formation was observed as the concentration of jasmonic acid increased from 0.5 to 2.0 mg/L in medium, whereas addition of gibberellic acid significantly suppressed bulblet formation. The rate of in vitro bulbing was as high as $96\%$ in MS medium supplemented with 2.0 mg/L jasmonic acid and 120 g/l sucrose after two month culture at $25{\pm}1^{\circ}C$ under 16 hours day light.

  • The present work has been carried out mainly to determine the effect of lead on the growth and cell organelles of Carrot (Daucus carota L.) callus. 1 mM of lead nitrate is added to the culture media R-2 and callus cells are cultured for 16 days. The growth rate is measured by fresh weight and structural changes of cell organelles is observed by electron microscope at every 4 days. The results show that lead inhibits the growth of Carrot callus and disturbs the structure of mitochondria remarkably.

Methyl Jasmonate-mediated Enhancement of Phenylethanoid Glycoside in Callus from Abeliophyllum distichum (cultivar Okhwang1)

  • Abeliophyllum distichum, one of the Korean endemic plant, is a significant pharmaceutical plant resource. A. distichum with phenylethanoid glycoside can use to regulate the development of cancer, DNA damage with radicals, and the generation of inflammatory mediators. In this study, we investigated whether the biomass, content of phenylethanoid glycoside, and growth rate of callus derived from A. distichum (cultivar Okhwang1, CAD) change in the absence or presence of plant hormones (2,4-Dichlorophenoxyacetic acid; 2, 4-D and 1-Naphthaleneacetic acid; NAA). The results showed that the best biomass, the growth rate of callus, and the contents of phenylethanoid glycoside were cultivated on Murashige and Skoog (MS) growth medium fortified with 1 ppm 2,4-D + 2 ppm NAA after 4 weeks. In a further study, CAD was cultivated on MS growth medium fortified with an elicitor (Methyl Jasmonate, MeJA). The results showed that CAD turned to brown color and fragile form with the elicitor. HPLC-PDA analysis revealed that the contents of phenylethanoid glycoside in the elicitor-treated group were higher than in the elicitor-non-treated group. These results are consistent with the findings of Arano-Varela H et al.,'s study which is that acteoside production can increase after the treatment of MeJA. Therefore, this study can be used to develop an effective and sustainable production of useful substances as an alternative to plant cultivation.

Effects of Cold Pretreatment and Medium Composition on Anther Culture Initiation in Strawberry

  • Callus culture initiation of strawberry (Fragaria${\times}$ananassa Duch.) was investigated at different Murashige and Skoog (MS) medium strengths, types and concentrations of plant growth regulators, and incorporating a cold pretreatment period to determine the optimal nutritional and environmental conditions. No high quality callus was induced on MS media without auxin regardless of medium strength. When 6-benzylaminopurine (BA) was combined with indole acetic acid (IAA), naphthalene acetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D), high quality callus were highly induced compared to medium supplemented with auxin alone. When $0.5mg{\cdot}L^{-1}$ BA was combined with IAA, NAA, and 2,4-D, high quality callus induction was more effective than the medium supplemented with the other BA concentrations. The best combination of auxin and cytokinin for high quality callus induction was $1.0mg{\cdot}L^{-1}$ NAA and $0.5mg{\cdot}L^{-1}$ BA. Although the differences in callus induction were not significant, high quality callus induction at half strength MS medium was more effective than at full strength medium. When $30g{\cdot}L^{-1}$ sucrose was added to the half strength MS medium, the rate of high quality callus induction increased. The optimum cold pretreatment temperature and period for high quality callus induction were $4^{\circ}C$ and 72 h, respectively. Regeneration rate of high quality callus increased in MS medium supplemented with thidiazuron.
