Monitoring and Risk Assessment of Heavy Metals in Edible Mushrooms (국내 유통 버섯 중 중금속 함량 조사 및 위해성 평가)

  • BACKGROUND: Many edible mushrooms are known to accumulate high levels of heavy metals. This research was focused on health risk assessment to investigate the mushrooms in Korea, arsenic (As), cadmium (Cd), lead (Pb) and mercury (Hg) contaminations in edible mushrooms in cultivated areas were investigated, and health risk was assessed through dietary intake of mushrooms. METHODS AND RESULTS: The heavy metals in mushrooms were analyzed by ICP/MS after acid digestion. Probabilistic health risk were estimated by Monte-Carlo simulation techniques. The average contents of As, Cd, Pb, and Hg were $0.035{\pm}0.042$ mg/kg, $0.017{\pm}0.020$ mg/kg, $0.043{\pm}0.013$ mg/kg, and $0.004{\pm}0.004$ mg/kg, respectively. The results showed that contents of Cd and Pb did not exceed maximum residual levels established by European Uion regulation (Cd 0.20 mg/kg and Pb 0.30 mg/kg). For health risk assessment, estimated intakes in all age populations did not exceed the provisional tolerable daily intake of As and Hg, provisional tolerable monthly intake of Cd, provisional tolerable weekly intake of Pb. The Hazard Index (HI) were ranged from $0.03{\times}10^{-4}{\sim}0.01{\times}10^{-3}$ for As, $0.02{\times}10^{-3}{\sim}0.81{\times}10^{-3}$ for Cd, $0.06{\times}10^{-3}{\sim}0.38{\times}10^{-3}$ for Pb, and $0.08{\times}10^{-4}{\sim}0.14{\times}10^{-3}$ for Hg at general population. CONCLUSION: The HI from the ratio analysis between daily exposure and safety level values was less than 1.0. This results demonstrated that human exposure to heavy metals through dietary intake of mushrooms might not cause adverse effect.

  • This study was undertaken to investigate the eftects of Cd administered ad libitum for 6 weeks on the cellular immune responses of Balb/c mice. The results were summarized as follows; 1. The mice fed 25, 50 and 100ppm Cd drank as much as control, but the mice fed 200ppm Cd drank significantly less water after Cd exposure than did control. Increasing rates of body weight of Cd-fed mice for 6 weeks were as this, control group 27.0%, Cd administered groups(25, 50, 100 and 200ppm) 28.54%, 28.31%, 20.49% and 18.04%, respectively. 2. Absolute spleen to body weight(mg/g) of control, 25, 50, 100 and 200ppm Cd administered groups were $4.34{\pm}0.23$, $4.20{\pm}0.54$, $4.80{\pm}0.87$, $4.25{\pm}0.32$ and $4.40{\pm}0.32$, respectively. Splenic cellularity(${\times}10^7$) of control was $24.29{\pm}5.98$ but increased to $27.72{\pm}5.48$, $32.96{\pm}8.44$, $28.32{\pm}8.76$ and $29.64{\pm}4.08$ in 25, 50, 100 and 200ppm Cd-fed groups, respectively. 3. Total $CD_4{^+}$ cells(${\times}10^7$) of control, 25, 50, 100 and 200ppm Cd-fed groups were $9.15{\pm}2.24$, $10.40{\pm}2.04$, $12.04{\pm}3.08$, $10.20{\pm}3.16$ and $10.80{\pm}1.48$, respectively and total $CD_8{^+}$ cells(${\times}10^7$) of these groups were $2.32{\pm}0.56$, $2.54{\pm}0.27$, $3.12{\pm}0.80$, $2.25{\pm}0.70$ and $2.24{\pm}0.28$, in order. On the other hand, $CD_4{^+}/CD_8{^+}$ ratios in total cells were increased significantly except for 50ppm Cd-fed group($3.88{\pm}0.01$). And that of control was $3.97{\pm}0.02$, but those of 25, 100 and 200ppm were $4.35{\pm}0.01$, $4.54{\pm}0.03$ and $4.81{\pm}0.03$. 4. Phagocytosis rates of peritoneal macrophages were increased significantly in 25 and 50ppm Cd groups($36.34{\pm}9.45$ and $37.15{\pm}9.22$, respectively), but 100 and 200ppm groups showed similar rates($18.20{\pm}3.04$ and $19.48{\pm}3.22$ respectively) to that of control($21.43{\pm}3.62$). 5. In mitogen-induced splenocyte proliferation, various concentraions of $CdCl_2(10^{-4}-10^{-7}M)$ were added to mitogen-stimulated culture in vitro. Splenocyte proliferation induced by LPS was decreased dose dependently, but proliferation by Con-A was increased slightly in concentrations of $10^{-7}-10^{-6}M$. 6. Significant cytotoxicity of splenocytes with $CdCl_2$ were shown at $10^{-4}M$ treated group, especially at 24 hrs. From these results, it could be concluded that Cd might modulate the immune responses by modifying a distribution of T cell subpopulations.

Monitoring of Heavy Metal Content in Alcoholic Beverages (국내 유통 주류 중 중금속 실태조사)

  • This study was conducted to estimate the contents of heavy metals including lead, cadmium, arsenic and total mercury in alcoholic beverages in Korea. Concentration of Hg was analyzed by gold amalgamation method, using mercury analyzer, while concentration of Pb, Cd and As was analyzed by ICP-MS. Concentration (${\mu}g/kg$) of heavy metal in fermented liquors were; for Pb $9.9\;{\pm}\;8.4$(0~38.0), Cd $5.8\;{\pm}\;4.9$(0~5.4), As $28.6\;{\pm}\;19.4$(1~96.4), Hg $0.7\;{\pm}\;1.2$(0~10.6). Concentration (${\mu}g/kg$) of distilled liquors were ; for Pb $4.4\;{\pm}\;5.7$(0~29.3), Cd $2.0\;{\pm}\;2.5$(0~10.3), As $12.0\;{\pm}\;17.0$(0~95.6), Hg $0.2\;{\pm}\;0.3$(0~2.3). Concentration(${\mu}g/kg$ of other liquous were ; for Pb $7.5\;{\pm}\;5.1$(0~13.7), Cd $5.8\;{\pm}\;3.9$(0.6~11.2), As $25.2\;{\pm}\;39.0$(0.5~103.3), Hg $0.3\;{\pm}\;0.1$(0.1~0.5). The present study showed that difference of the amount of constituent in a same category of food are not affect to the content of heavy metals among them. The residual levels of takju, yakju, sake, beer, fruit wine, soju, whiskey, brandy, general distilled liquor, liquor, other liquors are within the maximum levels, prescribed by Korea food code. It is given that heavy metal exposure of Pb, Cd, As, Hg from consumption of alcoholic beverages (takju, yakju, sake, beer, fruit wine, soju, whiskey, brandy, general distilled liquor, liquor, other liquors) are less than 0.03%, 0.06%, 0.01%, 0.01% (mean) in provisional tolerable weekly intake (PTWI) respectively, indicated by FAO/WHO.

  • Littorina brevicula, a common small herbivorous gastropod, inhabits in almost every rocky and/or boulder shores of Korea with high density. The survival rates and oxygen consumption rates of this species exposed to tributyltinchloride (TBTCl), mercury (Hg) and cadmium (Cd) were investigated according to temperatures (15, $23^{\circ}C$) and individual sizes (12$\pm$0.5, 5$\pm$1 mm). At temperature $15^{\circ}C$, acute inhibitory concentrations for large (12$\pm$0.5 mm) and small (5 $\pm$1 mm) size individual showed that 13 day-$LC_{50}$ for TBTC1 were 0.87 and 0.65 ppm, respectively, 11 day-$LC_{50}$ of Hg were 5.55 and 2.85 ppm, respectively and 9 day-$LC_{50}$ of Cd were 13.77 and 8.46 ppm, respectively. At $23^{\circ}C$, acute inhibitory concentrations of pollutants on the large and the small size individual showed that 8 day-$LC_{50}$ of TBTCl were 0.68 and 0.15 ppm, respectively, 5 day-$LC_{50}$ of Hg were 10.41 and 5.73 ppm, respectively, and 4 day-$LC_{50}$ of Cd were 13.31 and 4.47 ppm, respectively. The order of toxicity on the species was TBTCl > Cd > Hg. Oxygen consumption rates during exposure to TBTCl, Hg and Cd toxicity were decreased more in small size than in large size individuals at 15 and $23^{\circ}C$.

An Optimization Method of Measuring Heart Position in Dynamic Myocardial Perfusion SPECT with a CZT-based camera (동적 심근관류 SPECT에서 심장의 위치 측정방법에 대한 고찰)

  • Purpose Cadmium-zinc-telluride (CZT) camera with semiconductor detector is capable of dynamic myocardial perfusion SPECT for coronary flow reserve (CFR). Image acquisition with the heart positioned within 2 cm in the center of the quality field of view (QFOV) is recommended because the CZT detector based on focused multi-pinhole collimators and is stationary gantry without rotation. The aim of this study was to investigate the optimal method for measuring position of the heart within the center of the QFOV when performing dynamic myocardial perfusion SPECT with the Discovery NM 530c camera. Materials and Methods From June to September 2018, 45 patients were subject to dynamic myocardial perfusion SPECT with D530c. For accurate heart positioning, the patient's heart was scanned with a mobile ultrasound and marked at the top of the probe where the mitral valve (MV) was visible in the parasternal long-axis view (PLAX). And, the marked point on the patient's body matched with the reference point indicated CZT detector in dynamic stress. The heart was positioned to be in the center of the QFOV in rest. The coordinates of dynamic stress and rest were compared statistically. Results The coordinates of the dynamic stress using mobile ultrasound and those taken of the rest were recorded for comparative analysis with regard to the position of the couch and analyzed. There were no statistically significant differences in the coordinates of Table in & out, Table up & down, and Detector in & out (P > 0.05). The difference in distance between the 2 groups was measured at $0.25{\pm}1.00$, $0.24{\pm}0.96$ and $0.25{\pm}0.82cm$ respectively, with no difference greater than 2 cm in all categories. Conclusion The position of the heart taken using mobile ultrasound did not differ significantly from that of the center of the QFOV. Therefore, The use of mobile ultrasound in dynamic stress will help to select the correct position of the heart, which will be effective in clinical diagnosis by minimizing the image quality improvement and the patient's exposure to radiation.

Risk Assessment of Heavy Metals Migrated from Plastic Food Utensils, Containers, and Packaging Distributed in Korea (국내 유통 식품용 플라스틱 기구 및 용기, 포장의 중금속 위해도 평가)

  • Heavy metals can be intentionally or unintentionally introduced into plastic food utensils, containers, and packaging (PFUCP) as additives or contaminants, which can be ingested with food by humans. Here, seven-heavy metals (lead, cadmium, nickel, chromium, antimony, copper, and manganese) with toxicity concerns were selected, and risk assessment was done by establishing their migration from 137 PFUCP products made of 16 materials distributed in Korea. Migration of heavy metals was examined by applying 4% acetic acid as a food simulant (70℃, 30 minutes) to the PFUCP products. Inductively coupled plasma mass spectrometry (ICP-MS) was employed for the analysis of migrated heavy metals, and the reliability of quantitative results was confirmed by checking linearity, LOD, LOQ, recovery, precision, and expanded uncertainty. As a result of monitoring, heavy metals were detected at a level of non-detection to 8.76 ± 11.87 ㎍/L and most of the heavy metals investigated were only detected at trace amounts of less than 1 ㎍/L on average. However, antimony migrated from PET products was significantly higher than other groups. Risk assessment revealed that all the heavy metals investigated were safe with a margin of exposure above 311. Collectively, we demonstrated that heavy metals migrated from PFUCP products distributed in Korea appear to be within the safe range.

Monitoring of Pesticide Residues and Heavy Metals in Fermented Liquor in Ulsan (울산지역 유통 발효주의 잔류농약 및 중금속 실태조사)

  • In this study, we investigated in pesticide residues and heavy metals in fermented liquor products (wine, beer, makgeolli). Targeted analysis of 400 pesticide residues in the sample was performed using the quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method, followed by gas chromatography-tandem mass spectrometry (MS/MS) and LC-MS/MS. The contents of heavy metals (Pb, Cd) were determined by ICP-MS using the microwave method. The mercury was measured using a mercury analyzer. From the analysis of 150 cases, 102 (68.0%) cases of fermented liquor were detected, and 35 pesticide residues (including metalaxyl, mandipropamid, azoxystrobin, and fenhexamid) were detected among the 400 pesticide residues tested. Pb, Cd, and Hg were tested in 150 samples. Lead was detected in 73 samples (48.7%), cadmium in 9 samples (6.0%), and mercury in 36 samples (24.0%). Exposure assessment was conducted to determine the safety of the detected pesticide residues and heavy metals. According to this assessment, the pesticide residues and heavy metals showed very low %ADI values (less than 1%).
