유구(維鳩)-광천(廣川) 금은광화대지역(金銀鑛化帶地域)에서의 토양(土壤) 및 농작물(農作物)의 중금속오염(重金屬汚染) (Heavy Metal Contamination of Soils and Crop Plants in the Yugu-Kwangcheon Au-Ag Metallogenic Province)

  • Yugu-Kwangcheon Au-Ag Province is connected to the southwestern part of the Cheonan Province and includes the Gubong (one of the biggest gold mine in past), Daebong and Samgwang mines. Cadmium concentrations in sphalerite of Yugu-Kwancheon Province is higher than those of other Au-Ag provinces and could be the source of the heavy metal contaminations. Heavy metals are present in rock-soil-plant-human system from natural geological materials, and dietary intake through this system makes a significant contribution to the ingestion of heavy metals by man. In order to examine the degree and extent of heavy metal contaminations in the Gubong, Daebong and Samgwang mine areas of Yugu-Kwangcheon Province, soil, stream water, sediment and crop plant samples were taken and analyzed for Cd, Cu, Pb and Zn by Atomic Absorption Spectrometry (AAS) and Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES). Cadmium and Pb concentrations in paddy/farm soils and stream sediments from the Gubong and Samgwang mine areas are higher than those in uncontaminated soils. Lead concentrations in crop plants cultivated in these soils are high up to $0.38{\mu}g/g$ in rice grain and $4.5{\mu}g/g$ in sesame. In these mine areas, regular consumption of crop plants by the local population would seem to pose a potential health problems from long-term Pb exposure. The biological absorption coefficient in rice grain for heavy metals varies in the order $C_d=Zn>Cu>Pb$ and the relationship between Cd concentrations in soils and crop plants is expressed by the equation $Cd_{plant}=0.11245\;Log\;Cd_{soil}+0.13472$.

다결정 CdTe 기반의 광계수형 X선 센서 제작 및 특성평가 (The Fabrication and Property Evaluation of Poly-crystalline CdTe based Photon Counting X-ray Sensor)

  • 종래의 방사선 의료영상 센서는 방사선에 의해 생성된 전하신호를 화소내에서 적분하는 방식이다. 본 연구에서는 저선량에서 우수한 해상력이 가능한 광계수형 영상센서를 개발하기 위해 진공 열증착법을 이용하여 다결정 CdTe(p-CdTe) 필름을 제작하였다. 또한, 광계수형 센서의 성능평가를 위하여 물리적 특성(SEM, XRD) 및 전기적 특성(leakage current, x-ray sensitivity, SNR) 평가를 하였다. 측정 결과, $1V/{\mu}m$ 이하의 인가전압에서 $5nA/cm^2$ 이하의 누설 전류밀도를 보였으며, X선 발생신호량은 $7{\mu}C/cm^2{\cdot}R$으로 광계수형 센서로의 적용에 적합한 것으로 평가되었다. 또한, 신호대잡음비는 동작영역에서 5000 이상의 값을 보였다.

  • 망그로브 생태계는 수생태계로 유입되는 중금속을 받아들이는 기능을 가지고 있다. 중금속에 오염된 퇴적물에 노출됨에도 불구하고 망그로브는 중금속에 내성을 가지고 있다. 이 연구에서 우리는 망그로브로부터 중금속 저항성 관련 유전자를 클로닝하고, 중금속 노출에 유전자 발현 변화를 분석하였다. 미크로네시아 축라군의 웨노섬에서 채취한 Rhizophora stylosa의 잎과 뿌리조직으로부터 CTAB 방법을 이용하여 RNA를 분리하였고, gene specific primers를 이용하여 phytochelatin synthase 1(PCS1) 유전자를 클로닝하였다. R. stylosa 태생종자를 100 ppb의 Cd과 10 ppb의 Cu에 노출하였을 때 각각 1.91배 및 2.72배 발현이 증가하였다. 이러한 결과는 PCS1 유전자의 발현분석이 망그로브 생태계의 건강성을 평가하기 위한 좋은 도구가 될 수 있음을 나타낸다.

서울지역에 서식하는 집비둘기 Columba livia의 깃털을 이용한 중금속오염 모니터링 (Monitoring of Heavy Metal Contaminations using Feathers of Feral Pigeons Columba livia in Seoul)

  • 집비둘기 Columba livia 깃털의 중금속 농도를 파악하기 위하여 서울시에 서식하는 집비둘기를 서식지 환경에 따라 공단, 상업, 공원 및 주거지역으로 나누어 각 지역에 서식하는 개체들을 채집, 중금속 농도를 측정하였다. 깃털 중 철(Fe)의 농도는 공원지역에서, 망간(Mn)은 공업지역과 공원지역에서, 구리(Cu)는 공원지역에서 다른 지역에 비해 월등히 높게 나타났으며, 각 원소별로 지역간에 유의차가 존재하였다(ANOVA, p<0.05). 또한 아연(Zn)은 도심상업지역에서, 납(Pb)은 공업지역에서, 카드뮴(Cd)은 주거지역이 다른지역에 비해 약간 높았지만 지역별로 유의적인 차이가 나타나지 않았다. 간에서의 중금속농도와 깃털에서의 중금속농도와의 상관관계는 구리(r²=0.983, p<0.01)와 카드뮴(r²=0.632, p<0.01)에서 고도의 유의차가 나타났지만, 다른 원소에서는 유의성이 나타나지 않았다.

환경 바이오모니터링에서의 혈중 중금속 분석 숙련도 시험을 위한 표준물질 제조 및 적용성 평가 (Development and Validation of Reference Material for Proficiency Testing of Blood Heavy Metals in Environmental Biomonitoring)

  • Objectives: This study developed and validated reference materials (RMs) to analyze metal compounds in blood. Methods: We referred to KoNEHS (Korea National Environmental Health Survey) to estimate concentrations of blood metals (cadmium, Cd; lead, Pb; mercury, Hg) and applied analytical methods (inductively coupled plasma - mass spectroscopy, ICP-MS, for Cd and Pb; graphite furnace - atomic absorption spectrometry, GF-AAS, for Cd and Pb; and direct mercury analyzer, DMA, for Hg). Homogeneity and stability tests were carried out. In addition, certified values and uncertainties of RMs were calculated through internal and external experiments. All RMs were developed and assessed in various forms according to element, analytical method, and two types of concentration levels high concentration for occupational exposure and low concentration for environmental exposure. Results: All samples showed acceptable homogeneity, except for low concentration of Cd in the GF-AAS method. Short- and long-term stabilities were satisfied by ANOVA testing. In the inter-laboratory comparison, robust medians were lower than the certified values of all RMs (robust median/reference value; $1.301/1.327{\mu}g/L$ for Cd, ICP-MS, low concentration; $3.152/3.388{\mu}g/L$ for Cd, ICP-MS, high concentration; $1.219/1.301{\mu}g/L$ for Cd, GF-AAS, low concentration; $3.074/3.321{\mu}g/L$ for Cd, GF-AAS, high concentration; $14.473/14.516{\mu}g/L$ for Pb, ICP-MS, low concentration; $50.069/50.114{\mu}g/L$ for Pb, ICP-MS, high concentration; $12.881/14.147{\mu}g/L$ for Pb, GF-AAS, low concentration; $47.015/47.591{\mu}g/L$ for Pb, GF-AAS, high concentration; $4.059/4.218{\mu}g/L$ for Hg, DMA, low concentration; $11.474/11.181{\mu}g/L$ for Hg, DMA, high concentration). Conclusion: This study demonstrates procedures for developing and validating RMs for biomonitoring in the field of the environmental health.

카드뮴과 아연 노출에 따른 야외종 장수깔따구(Chironomus plumosus)의 생태독성학적 반응 (Ecotoxicological Response of Cd and Zn Exposure to a Field Dominant Species, Chironomus plumosus)

  • 인간의 인위적 활동으로 인해 발생하는 중금속 중 카드뮴(Cd)과 아연(Zn)은 다양한 경로를 통해 하천으로 유입되어 서식생물에게 유해 영향을 준다. 본 연구에서는 도심 하천에서 우점하는 장수깔따구 (Chironomus plumosus)를 이용하여 Cd와 Zn 노출에 따른 생태독성학적 반응을 연구하였다. 생활사를 고려하여, 생존율, 성장율, 우화율, 성비와 하순기절 기형을 관찰하였다. 장수깔따구 생존율은 Cd와 Zn 노출에 따라 시간, 농도의존적인 경향을 나타냈다. 성장율은 Cd 노출 후 Day 4와 Day 7, 농도의존적인 감소를 보였으며, 고농도인 $50mg\;L^{-1}$에서 대조군에 비해 급격한 감소를 나타냈다. 또한, 우화율은 10, $50mg\;L^{-1}$ Cd에서 대조군에 비해 감소함을 관찰하였다. 성비 변화는 $100mg\;L^{-1}$ Zn에서 암컷의 비율이 증가였으나 농도가 높아짐에 따라 수컷의 비율이 높아짐을 보이며 성비불균형이 관찰되었다. 게다가, 장수깔따구의 하순기절에서는 대조군에 비해 Cd와 Zn 노출된 장수깔따구에서 기형이 관찰되었다. 이와 같은 결과는 수생태계로 유입되는 중금속이 하천의 하상저층에 서식하는 저서무척추동물에게 유해한 영향을 주며, 중금속 노출에 따른 생물학적 분석을 위한 현장 지표종으로서의 가능성을 보여주었다.

국내 유통 식품용 유리제의 중금속 노출 평가 (Exposure Assessment of Heavy Metals Migrated from Glassware on the Korean Market)

  • 본 연구는 국내 유통 되는 식품용 유리제 총 110개 제품을 대상으로 유리제에서 식품으로 이행될 우려가 있는 납, 카드뮴 및 바륨에 대하여 이행량을 조사하고 안전성 평가를 실시하였다. 납, 카드뮴 및 바륨은 ICP-OES로 측정되었고, 시험법은 직선성, 검출한계, 정량한계, 회수율, 정밀도 및 측정불확도 추정에 의해 검증되었다. 납과 바륨의 이행량은 각각 불검출 ~ 0.7949 mg/L 및 불검출 ~ 0.1823 mg/L 이었으며, 카드뮴은 모든 시료에서 검출되지 않았다. 식이를 통한 이행물질의 일일추정섭취량 및 위해도를 확인한 결과 국내 유통되는 식품용 유리제는 안전하게 관리되고 있음을 확인하였다.

Physiological Responses and Phytoextraction Potential of Pinus thunbergii on Cd-contaminated Soil

  • We investigated physiological responses and phytoextraction ability of Pinus thunbergii in cadmium contaminated soil as part of our efforts in identifying plant materials for the restoration and revegetation of forest soil contaminated by heavy metals. Thirty seedlings (ten per treatment) were assigned to three treatments (control, 0.3 and 0.6 mM $CdSO_4$ solution) at first year experiment. At second year, ten seedlings per treatment treated with Cd during the first year experiment were divided by two groups (no Cd-treated and consecutive Cd-treated group). At first experiment, photosynthetic pigment content, and superoxide dismutase (SOD) and glutathione reductase (GR) activities have significantly reduced by Cd application, and the reduction rate was increased much higher as the rate of Cd application increased. On the other hand, thiol and malondialdehyde (MDA) content were significantly increased at the application of 0.6 mM of Cd. At the second year experiment, a general increase in chlorophyll and carotenoid content was observed with Cd treatment while SOD and GR activities showed a relative reduction compared to the control. Similar to the first year measurement, thiol and MDA contents also increased considerably due to Cd treatment. At harvest, dry matter was significantly reduced by Cd treatment especially at the rate of 0.6 mM Cd, but dry yield of P. thunbergii treated with 0.3 mM Cd was less affected and it was comparable with the control seedling. Cadmium concentration in seedling tissues increased with increasing Cd application rate while Cd uptake was higher in seedlings supplied with 0.3 mM Cd, which could be ascribed to their high dry matter. Overall, our study has demonstrated the unique physiological response of P. thunbergii to Cd-prolonged exposure by showing that the changes in photosynthetic pigment content and antioxidative enzyme activities were dependent on the concentration and duration of treatment. In addition, our results have demonstrated the potential of P. thunbergii to withstand up to 0.3 mM Cd (equivalent to cumulative Cd concentration of 134.4 to 268 mg $kg^{-1}$) without showing growth reduction, hence it might be used for phytoremediation of Cd contaminated areas.

  • Certain heavy metals which may lead peoples to poisonous status are widely used in industry and their uses have been increasing along with rapid industrialization of this country. Such an increasement of metal uses aggravates the status of environmental pollution affecting foodstuffs which are the most important life supporting factor of animal and humanbeing. Concerning the safety measures to minimize food-borne transmission of such hazardous metals, surveillance is the backbone of them and probably more so with a potential problem such as intoxication. Theoretically, this surveillance should include the determination of levels of heavy metal toxicants in foods, the determination of food consumption patterns and typical total diet, and the estimation of total load of the metal contaminant from all sources of exposure including air, water, and occupational sources. In recent year, actually, such estimates on the total daily intake of some heavy metals from foods have been made in several developed countries and a wide variation of date by season, locality, and research method was recognized. Also in this country, this kind of research data is vitally needed to make up for the serious shortage or lack of references to estimate the total amount of heavy metal intake of the people. In this study, a modification model for estimation of the total daily intake of cadmium copper, nickel, zinc, and lead through foods was applied and concentrations of the above metals in crops cultivated in this country were measured with atomic absorption spectro photometer to get the following results. 1. Level of heavy metal concentration in crops Generally, the levels of such metals in essential crops such as rice, cucumber, radish. chinese cabbage, apple, pear, grape, and orange are similar or lower than those in Japan and other developed countries. By the way, a striking result on cadmium concentration was increasement of its concentration in rice from $0{\sim}0.035ppm$ in 1970 to 0.11ppm in this study. However, the value is still far below the. Japanese Permissible Lebel of 1.0ppm. 2. Estimation of total daily intake per capita from foods A new model for estimation was devised utilizing levels of metal concentration in foods, amount of food consumed, and other food factors. Based on the above method, the daily intake of cadmium was estimated to be $70.53{\mu}g/man/day$ in average which was as high as the Limit Value of ILO/WHO(up to $71.4{\mu}g/man/day$). Also, 3.89mg of Zinc, 1.65mg of cuppor, 0.32mg of lead were given as the total daily intake per capita by this research. 3. Efficacy of washing or skinning to decrease the amount of metals in crops After washing the crops sufficiently with commercial linear alkylate sulfonate, the concentration of heavy metals could be reduced to $50{\sim}80%$ showing decreasement rate of $20{\sim}50%$. Also, after skinning the fruits, decreasement rate of the heavy metal concentration shelved $0{\sim}50%$.

Comparative physiological and proteomic analysis of leaf in response to cadmium stress in sorghum

  • Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.
