The effect of acute cadmium-neuropathy on phospholipid metabolism in rat sciatic nerve was investigated. An animal model of cadmium neuropathy was induced by feeding diet containing cadmium to Sprague-Dawley rat for two weeks. Four weeks aged Sprague-Dawley rats were divided into four groups : normal control group, 10ppm-cadmium treated group, 100ppm-cadmium treated group, 1000ppm-cadmium treated group, reference drug, myo-inositol-treated group. All rats were sacrificed at the end of two weeks. The rate of incorporation of 2-[3H]myo-inositol into polyphosphinositide was significantly decreased while the rates of incorporation into phospholipid of titratedserine, ethanolamine and choline were unchanged in sciatic nerve obtained from cadmium-treated rat. Continuously the activities of three enzymes concerned with inositol phospholiped metabolism were measured in homogenates of rat sciatic nerves. Cystidine diglyceride transferase and phophatidylinositol kinase showed significantly decreased activities while phosphatidylinositol-4-phosphate kinase did not show any significant change in activity by cadmium treatment. However these deficits of inositol phospholipid metabolism were ameliorated by myo-inositol administration and these effectiveness were more potent in lower dose cadmiumtreated rats than higher dose cadmium-treated rats. These results suggest that cadmium intoxicated peripheral nerve with perturbation of the ployphosphoinositide metabolism and alteration of the enzyme activity which concerned with myo-inositol metabolism.
This study aimed to find out the effect of freeze-drying leek against cadmium poisoning on the cholesterol and enzyme activities in serum and superoxide radical, SOD and catalase in liver and kidney of the male rats during the administrered period. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 3 groups which were fed differently either for 5 weeks or for 10 weeks:basal diet, basal diet and cadmium in water and 3 % leek added diet and cadmium in water. Cadmium was administered ad libiturn 100 ppm CdCl$_{2}$ in water. The followings are the results of this experiment. 1. Leek reduced the cholesterol and the activities of GPT increase resulted from cadmium treatment. 2. Leek reduced the rate of cadmium in liver and kidney. 3. Leek reduced the activities of SOR and catalase in liver and kidney, while it enhanced the activities of SOD. 4. Leek reduced the necrosis and swelling in liver and kidney casused by cadmium treatment. This experiment showed that leek-addition group had protective effect against cadmium poisoning and increased ALPase activities in serum. Leek alleviated GPT activities in serum and cadmium concentration, necrosis, and swelling in liver and kidney. Therefore, this experiment concluded that leek has defensive power against cadmium poisoning.

Metallothionein (MT), a small protein molecule which can bind or release metal ions, is involved in the regulation of cellular metal homeostasis. This study was investigated the accumulation of cadmium in blood, tissue (liver, kidney and brain), and the effect of cadmium on several key genes (MT-I, MT-II, ZnT-1) in zinc metabolism in the mouse. Mouses weighing 20∼25 g were randomly assigned to control and cadmium treated group (Cd group). Cd group was intraperitoneally injected with cadmium 2, 4, 8 mg/kg and control group was administerd with saline. Mouses of each group were sacrificed by decapitation 4 hours after the administration of cadmium. Cadmium contents in blood, liver, kidney and brain were increased by a dose-dependent manner. Accumulation of cadmium was mainly occurred in liver and kidney. Induction of MT-I and MT-II protein was increased, but ZnT-1 expression was decreased in a dose-dependent manner by the treatment of 2∼8 mg/kg cadmium. These results suggested that cadmium can be transported to brain and alter the expression of several key genes in zinc homeostasis.
The influence of selenium to several toxic effects of cadmum, including lethality has been shown following pretreatment with cadmium, zinc and seleniurm Five groups of rats, each consisting of 16 rats, were studied and each group was divided into four subgroups, 4 rats for each subgroup. After subcutaneous pretreatment during 5 days with saline, CdCl$_{2}$ (0.5mg/kg, ZnCl$_{2}$ (13.0mg/kg) and $Na_{2}SeO_{3}$(1.0mg/kg), rats were given intraperitioneal administration of various dosage of or cadium of cadmium and selenium. After giving the challenge dose, cadmium and metallothionein(MT) concentrations were determined in liver and kidney The concentration of cadmium in liver and kideny increased proportionally to the increase of challenge dosage. The simultaneous administration of cadmium and selenium significantly more decrease cadmium concentrations in liver and kidney than those of the administration of cadmium only. However, MT concentrations in liver and kideny were increased by the pretreatment of cacmium, zinc and selenium. Our results suggest that increasing cadmium concentrations, gradully accumulating in the tissues of liver and kidney as a result of the pretreatment, served to induced the synthesis of MT, thus making them resistant to the challenge from cadmium.
A cadmium-binding protein was purified the cell-free extract of extreme cadmium tolerant Hansenula anomala B-7. The molecular weight was determined to be approximately 33, 000 and was composed two kinds of subunits having a molecular weight of 18, 000 and 14, 000, respectively. The extinction coefficient of the cadmium-binding protein was calculated to be 19.58. The amount of cadmium in the cadmium-binding protein was $9.26{\mu}{\textrm{g}}$ per $100{\mu}{\textrm{g}}$ of protein. A total of 14 amino acids were detected in the cadmium-binding protein, including aspartic acid, glycine and alanine that were present in a high quantity, but proline, valine and methionine were not found. The purified cadmium-binding protein contained a high quantity of cysteine and cadmium, and therefore this protein showed clearly the characteristics of metallothionein.
This study was performed to investigate the effect of dietary calcium level on cadmium and lead toxicity in rats. Fifty-four male rats of Sprague-Dawely strain weighing 152$\pm$12g were blocked into 9 groups according to body weight, and were raised for 30 days. Nine experimental diets different with cadmium(0%, 0.04%), lead (0%, 0.071%) and calcijm(0.5%, 1.0%, 1.5%) levels were prepared. The results are summarized as follow. Weight gain, F.E.R.(food efficiency ratio), and weights of liver, kidney and femur were lower in cadmium exposed groups than those of heavy metal free groups. Weight gain F.E.R. and ash weight of lead groups were lower than those of heavy metal free groups. But, these were increased with increasing dietary calcium level. Cadmium and lead concentrations in blood, liver, kidney and femur were lower in rats fed 1.5% calcium than 0.5% calcium diet. Fecal cadmium and lead excretions were remarkably increased in 1.5% calcium groups, and cadmium and lead retention rates were decreased in 1.5% calcium groups. Metallothionein concentrations in liver, kidney and small intestine were higher in rats exposed to cadmium and lead. Calcium content in blood, femur and daily urinary and fecal calcium excretion were decreased by cadmium and lead additions, and increased in 1.5% calcium groups. Creatinine clearance were decreased with cadmium administratino and calcium addition. In conclusion, weight gain and organ weights were decreased with cadmium or lead administration. But, cadmium administration was more toxic than lead adminstration. Cadmium or lead toxicity was alleviated by increasing dietary calcium level. Especially, lead toxicity was alleviated in proportion to dietary calcium level.
This experiment was conducted to investigate the effect of cadmium levels on weight gain, nutrient digestibility and the retention of iron, copper and zinc in tissues of growing pigs. A total of one hundred and ninety-two crossbred pigs (barrows, Duroc$\times$Landrace$\times$Yorkshine, 27.67$\pm$1.33 kg of average initial body weight) were randomly allotted to four treatments. Each treatment had three replicates with 16 pigs per pen. The corn-soybean basal diets were supplemented with 0, 0.5, 5.0, 10.0 mg/kg cadmium respectively, and the feeding experiment lasted for eight-three days. Cadmium chloride was used as cadmium source. The results showed that pigs fed the diet containing 10.0 mg/kg cadmium had lower ADG and FCR than any other treatments (p<0.05). Apparent digestibility of protein in 10.0 mg/kg cadmium-treated group was lower than that of other groups (p<0.05). There was lower iron retention in some tissues of 5.0 mg/kg and 10.0 mg/kg cadmium treatments (p<0.05). However, pigs fed the diet 10.0 mg/kg cadmium had higher copper content in most tissues than that of any other groups (p<0.05). There was a significantly increase of zinc retention in kidney of 10.0 mg/kg cadmium additional group (p<0.05) and zinc concentrations in lymphaden, pancreas and heart of 10.0 mg/kg cadmium treatment were lower than those of the control (p<0.05). This study indicated that relatively high cadmium level (10.0 mg/kg) could decrease pig growth performance and change the retention of iron, copper and zinc in most tissues during extended cadmium exposure period.
A pot experiment was conducted to find out the effects of phosphorous on the retardation of cadmium uptake by rice plant using the soil with 8.8 ppm of available phosphorous. Soils were treated with cadmium concentrations of 3, 6, 9 ppm and phosphorous amounts of 10, 20, 40 kg/10a respectively. The results were summarized as followings: 1. There was no correlation between cadmium and phosphorous content in the rice plant. 2. Even though the greater amounts of cadmium were absorbed and translocated to the brown rice than leaves, the cadmium contents between brown rice and leaves were positively correlated. 3. In soil which contained 3, 6 and 9 ppm cadmium ranges on soil, phosphorous was applied 13, 17.6 and 27.6 kg/10a respectively, and so the cadmium content fell below 0.4 ppm in the brown rice. 4. The growth and yield of rice plant were normal in soils with cadmium content below 9 ppm.
This study was designed to investigate the antibody production to sheep red blood cells(SRBC) and proliferation of mitogen-stimulated spleen cells in Balb/c mice which received cadmium chloride. The mice were divided into three independent groups which were one control and two experimental groups by the cadmium treatment or not. No specific treatment was done for the control group. One of two experimental groups, which is called 'pre-treatment group' in this paper, was subcutaneously injected with low dose of cadmium chloride(0.5 mg/kg/day) for 5 consecutive days before the primary SRBC immunization. The other called 'non-pretreatment group' was only pretreated with normal saline. Both experimental groups were intraperitoneally injected with high dose of cadmium chloride(5 mg/kg) 8 hours before the primary immunization. Mice were intraperitoneally immunized twice with 2% SRBC suspension containing $10^8$ cells. The results obtained were as follows, 1. The PFG responses to SRBC were significantly increased in two experimental groups, cadmium pretreatment and non-pretreatment compared with that of control group(p<0.05). 2. The total antibody titers to SRBC in cadmium treated groups were similar to that of control group, but titers of IgG antibody were significantly elevated(p<0.01). 3. The proliferation response of spleen lymphocytes to various mitogens was suppressed in proportion to the concentration of cadmium and the degree of cadmium accumulation in liver was increased in the cadmium treated groups. These results suggest that cadmium chloride could affect on mouse immune response, especially its cell mediated immune response could be decreased while its humoral immune response could be increased, which may not be influenced by the administration methods or pretreatment of cadmium to mouse.
In this study the authors investigated the distribution of cadmium accumulated in cadmium-iun tolerant Hansenula anomala B-7 cells and also the effect of cadmium on protein synthesis. 84.9% of the cadmium accumulated was distributed in the soluble fraction (cytosol, etc.). The intracellular protein content was decreased by cadmium (1,000 $\mu g$/ml), but the content of soluble protein preeipitated by ammonium sulfate (30-75% saturation) was increased compared with the content of it obtained from the cells grown without cadmium. Furthermore, in the cells grown with 1,000 $\mu g$/ml of cadmium t h higher molecular weight soluble protein was increased compared with the cells grown without caa, mium, but the lower molecular weight soluble protein was decreased. These results suggested that the protein synthesis was inhibited by cadmium, but synthesis of higher molecular weight soluble protein was remarkably stimulated by cadmium.
